Triple staining with fluorochromes (DA/DAPI/CMA) and C-banding were used to characterize the composition of Pseudonannolene strinatii heterochromatin. C-banding showed C+ bands of different labeling intensity on chromosomes 1 and 2 in some cells. Fluorochrome staining revealed DAPI+ regions corresponding to the C-banding pattern, indicating that the heterochromatin of this species is abundant in AT-rich sequences.

Abstract The final disposal of residues generated at sewage treatment plants (STPs) has become a major problem for cities, due to the increase in the amount of treated sewage. One of the alternatives for the residue, labeled sewage sludge, is its reuse in agriculture and in degraded soil. However, not all pathogens and metals present in it are eliminated during treatment. Diplopods have been used as bioindicators in ecotoxicological tests as they are constantly in close contact with the soil. Owing to this fact, the purpose of this study was to expose specimens of the diplopod Rhinocricus padbergi to substrate containing sewage sludge collected at STPs to analyze morphological alterations in their parietal and perivisceral fat body, where substances are stored. The exposures were held for 7, 15, or 90 days at different concentrations of sewage sludge (control, 1%, 10%, and 50%). The parietal fat body showed no alterations in any of the three exposure periods or concentrations. Alterations in the perivisceral fat body were observed for all exposure periods. According to the results, we suggest that the sludge used has toxic agents responsible for changing the animal's perivisceral fat body. © 2012 Microscopy Society of America.

This study presents the ovarian morphology and the dynamics of the vitellogenesis process in the oocytes of the diplopod Leptodesmus dentellus. The oocytes are arranged in clusters called ovisacs which are distributed in pairs along the midline of the body forming the ovary. Regions similar to the germarium appear paired in the anterior region of the ovary; however, the development of the oocytes of this species does not follow a regionalisation in the reproductive organ. Cells in three developmental stages are found throughout the length of the ovary. Calcium, proteins, lipids and neutral polysaccharides were detected in the oocytes of L. dentellus. The polysaccharides and the proteins found in the oocytes have a double origin: endogenous, with the participation of the germinative vesicle, and exogenous, from follicular epithelium. The origin of the lipids is exogenous, i.e., they are incorporated into the oocytes, probably derived from the perivisceral fat body, which are highly developed in this region. The deposition of calcium is pre-vitellogenic and probably functions as a reserve during the juvenile stages. © 2012 Koninklijke Brill NV, Leiden.

Millipedes are macro-arthropods of soil which perform a important role in maintaining the balance of the ecosystem, since they feed on decaying organic matter and because they promotes the soil enrichment, aeration and humidification. These animals are usually found in humid with low light places, under rocks and trunks. The diplopods exoskeleton is much resistant and impregnated with calcium salts. Due the secretion of defense, with strong and unpleasant smell, the animals of this class have few or no predators. Large diplopods populations are not observed, but explosions population can occur for unbalances enviromnmental, by climate change and by use of pesticides that may eliminate potential competitors. In this sense, the millipede Urostreptus atrobrunneus Pierozzi & Fontanetti, 2006, has presented points of infestation in urban centers of Sao Paulo state, causing many disorders of the human population. Because it is a newly described species, little is known of its aspects biological and morphological. Thus, the purpose of this study was to describe the structure and function of the digestive tract of the species U. atrobrunneus by means of histological, histochemistry and ultrastructural techniques. The results demonstrated that the digestive tract of U. atrobrunneus is similar to the descriptions given for other species. The foregut consists of a simple epithelium, composed of cells of different heights, covered by a cuticular intima, leaning on a thin basal membrane, followed by a well-developed muscular layer and an external membrane, around the foregut have a pair of salivary glands. The midgut epithelium has a pseudostratified, supported by a thick basal membrane, followed by a muscular layer and a layer of hepatic cells. Human activities generate a great amount of sewage daily, which is dumped into the sewer system. After sewage-treatment processes, sewage sludge is generated. Such byproduct can be treated by different methods; the result of treatment is a stabilized compost of reduced pathogenicity that has a similar inorganic chemical composition to the raw sewage sludge. After such pretreatment, sewage sludge is called a biosolids, and it can be used in agriculture. In this contest, the present study evaluated the effects of a sample of biosolids on the perivisceral fat body of a diplopod. These invertebrates are soil organisms that play an important role in the dynamics of terrestrial ecosystems, and as a consequence, they are in contact with xenobiotics present in this environmental compartment. Special emphasis is given on the interpretation of the effects of complex mixtures in target organs of diplopods. A semiquantitative analysis for the evaluation of histopathological changes in the perivisceral fat body was proposed. The sample-induced histopathological and ultrastructural changes in individuals exposed to it, and the severity of the effects was positively related to the exposure time, resulting in the deaths of exposed individuals after 90 days. Thus, the results indicate the need for caution in the use of biosolids as well as the need for improving waste management techniques, so they will produce environmentally innocuous final products.

En investigación se determinó la densidad, número de familia y composición de familias de macroinvertebrados encontradas en tres microcuencas de la Subcuenca del Río Estelí y dos microcuenca de la Subcuenca del Río Viejo. En el marco del proyecto UNA/CARE/MARENA/PIMCHAS, 2010. Al mismo tiempo se realizó comparación de la densidad con respecto al uso de suelo. De igual manera, se determinó el cambio de macroinvertebrados en tres profundidades de muestreo (hojarasca, 1 -10cmy 10-25cm) y se agruparon por sus hábitos alimenticios. Para la toma de datos se exploraron los macroinvertebrados en 171 monolitos de 20x20x25 cm repartidos en los sitios de acción del proyecto UNA-CARE-MARENA- PIMCHAS de las microcuencas El Coyote, Las Chichiguas, La Pita, La Laguneta y Tomabú. Los 2,920 individuos identificados se determinaron en 52 familias, 22 órdenes, 7 clases y 4 filums. El número de familias y su composición no varió significativamente entre microcuencas ni entre sitios. Las familias que obtuvieron los mayores valores de abundancia en general fueron: Carabidae, Staphylinidae, Curculionidae, miembros de la clase Diplopoda- d, Formicidae y Scolopendridae. La densidad y el número de familias no variaron significativamente en las tres profundidades estudiadas , pero sí varió la composición de familias. Los individuos con hábitos alimenticios fitófagos y depredadores, además de estar entre los grupos con más individuos por m2 , junto a los fungívoros y xilófagos, tuvieron la mayor variedad de familias. Los usos de tierra en donde se reportaron los mayores números de individuos por m 2 fueron de manera descendente: café con sombra, bosque, granos básicos y pasto. El número de familias no varió significativamente entre los usos, sin embargo la composición de familia si varió.

We investigated the soil arthropod communities of urban and suburban holm oak (Quercus ilex L.) stands in a small (Siena) and a large Italian city (Naples) and tested whether the abundance and diversity of higher arthropod taxa are affected by the biotic and abiotic conditions of urban forest soils, including pollution. Acarina and Collembola were the dominant taxa in both cities. In Siena the total number of arthropod individuals collected in the samples was over 1/3 greater than in Naples, but all diversity indices scored higher in Naples than in Siena, probably in response to the higher heterogeneity of microclimatic and pedological conditions found in Naples study area. Oribatids resulted twice more abundant in Siena and so were the total mites with respect to Collembola. While “taxonomic richness” per site increased with distance from road traffic, entropy and evenness indices scored higher at the two ends of the impact gradient in both cities. The overall variation in basic pedological and microbiological soil parameters positively correlated with the total abundance of arthropods, and negatively correlated with their taxonomic richness. At the resolution employed, no significant relation emerged between anthropogenic factors, such as traffic load and soil pollution, and the arthropod fauna density and variety. These results are consistent with conclusions drawn from a previous study on the enchytraeid fauna examined at species level, which is remarkable considering the different taxonomic resolutions of the two studies. CCA results suggest that the higher abundance of Oribatid mites, Protura and Thysanura and the lower abundance of Diplopoda and Symphyla in Siena could depend on a higher fungi/bacteria ratio. This observation can be interpreted in terms of differences in fungi and bacteria between the two cities: Siena is shifted towards the fungal decomposition channel, which supports taxa such as oribatid mites, while Naples is shifted towards the bacterial channel, which supports chiefly detritivorous groups, such as diplopods.

1. The establishment of grassy strips at the margins of arable fields is an agri-environment scheme that aims to provide resources for native flora and fauna and thus increase farmland biodiversity. These margins can be managed to target certain groups, such as farmland birds and pollinators, but the impact of such management on the soil fauna has been poorly studied. This study assessed the effect of seed mix and management on the biodiversity, conservation and functional value of field margins for soil macrofauna. 2. Experimental margin plots were established in 2001 in a winter wheat field in Cambridgeshire, UK, using a factorial design of three seed mixes and three management practices [spring cut, herbicide application and soil disturbance (scarification)]. In spring and autumn 2005, soil cores taken from the margin plots and the crop were hand-sorted for soil macrofauna. The Lumbricidae, Isopoda, Chilopoda, Diplopoda, Carabidae and Staphylinidae were identified to species and classified according to feeding type. 3. Diversity in the field margins was generally higher than in the crop, with the Lumbricidae, Isopoda and Coleoptera having significantly more species and/or higher abundances in the margins. Within the margins, management had a significant effect on the soil macrofauna, with scarified plots containing lower abundances and fewer species of Isopods. The species composition of the scarified plots was similar to that of the crop. 4. Scarification also reduced soil- and litter-feeder abundances and predator species densities, although populations appeared to recover by the autumn, probably as a result of dispersal from neighbouring plots and boundary features. The implications of the responses of these feeding groups for ecosystem services are discussed. 5. Synthesis and applications. This study shows that the management of agri-environment schemes can significantly influence their value for soil macrofauna. In order to encourage the litter-dwelling invertebrates that tend to be missing from arable systems, agri-environment schemes should aim to minimize soil cultivation and develop a substantial surface litter layer. However, this may conflict with other aims of these schemes, such as enhancing floristic and pollinator diversity.

The management of pheasants (Phasianus colchicus) in pens and their subsequent release within woodland for game shoots is widely practised in Britain. With the exception of ground flora and songbirds, the impacts on other taxa have not been well documented. We investigated the effects of pheasants on invertebrate abundance and community composition, using pitfall trapping. We compared release pens with control plots located in the same woods and in woods where no pheasant releasing had occurred for at least 25 years. Conditions for invertebrates within release pens were altered, with more annual plants and disturbance-tolerant perennials and a reduced leaf litter layer. No major differences in invertebrate abundance, or Carabidae or Staphylinidae richness, were found in spring at either the pen scale or the wood scale. However, pheasant release pens resulted in significant changes in the species composition of Carabidae, with shifts towards species typical of arable fields and grassland. Carabid species active in spring and those that are very large (N17.0 mm) declined at pheasant release densities higher than 1000 birds/ha. Both effects are likely to be due to predation by pheasants at the peak of release in July–August, operating separately on larvae and adults respectively. There was an overall increase in the abundance of detritivores, including Diplopoda, Oniscoidea, Gastropoda (snails), at higher release densities. Mean release density in our study was 1489 ± 126 birds/ha (range 174–3409, n = 37 pens) and we suggest that detrimental effects on specialist woodland invertebrates would be minimized if releasing was conducted at the recommended density of 700 birds/ha.

In diplopods, the presence of calcium-containing structures seems to be a common finding in some species, with its formation being similar to that observed for other intracellular mineralization systems. In the present study, using histochemistry and transmission electron microscopy, a large amount of calcium was observed in the oocytes of Rhinocricus padbergi. Calcium was detected in both less and well developed oocytes, i.e., the occurrence of calcium coincided with the beginning of vitellogenesis. Calcium was observed as fine granulation distributed within the cytoplasm or deposited in spherical structures apparently formed by overlapping calcium layers. Some authors have suggested that these structures represent a type of reserve used for the calcification of the embryo exoskeleton, whereas others believe that calcium inclusions are a mechanism of organism detoxification as a result of excess calcium ingested by animals during soil turnover. We suggest in this paper that the first hypothesis could be occurring in R. padbergi since at the juvenile stages of the individuals the uptake of calcium is low and because the oocyte is a specialized cell not associated with detoxification.
