La technique de clonage par transfert nucléaire de cellules somatiques (SCNT) présente une page importante dans les annales scientifiques, mais son application pratique demeure incertaine dû à son faible taux de succès. Les anomalies placentaires et de développement fœtal se traduisent par des pertes importantes de gestation et des mortalités néonatales. Dans un premier temps, la présente étude a caractérisé les changements morphologiques des membranes fœtales durant la gestation clonée en les comparant à des gestations contrôles obtenues à partir de l’insémination artificielle. Les différentes anomalies morphologiques des placentomes telles que l’œdème chorioallantoique, la présence de zones hyperéchoiques et irrégulières dans la membrane amniotique et la présence de cellules inflammatoires dégénérées compromettent le développement fœtal normal de la gestation clonée. L’examen ultrasonographique représente une technique diagnostique importante pour faire le suivi d’une gestation et de caractériser les changements placentaires dans le cadre d’évaluation globale du bien-être fœtal. Le profil hormonal de trois stéroïdes (progestérone (P4), estrone sulfate (E1S), et œstradiol (E2)) et de la protéine B spécifique de gestation (PSPB) dans le sérum des vaches porteuses de clones SCNT a été déterminé et associé aux anomalies de gestations clonées. Une diminution de la P4 sérique au jour 80, une élévation du niveau de la concentration de la PSPB au jour 150, et une augmentation de la concentration d’E2 sérique durant le deuxième et troisième tiers de la gestation clonée coïncident avec les anomalies de gestation déjà reportées. Ces changements du profil hormonal associés aux anomalies phénotypiques du placenta compromettent le déroulement normal de la gestation clonée et gênent le développement et le bien-être fœtal. Sur la base des observations faites sur le placenta de gestation clonée, le mécanisme moléculaire pouvant expliquer la disparition de l’épithélium du placenta (l’interface entre le tissue maternel et le placenta) a été étudié. L’étude a identifié des changements dans l’expression de deux protéines d’adhérence (E-cadhérin et β-catenin) de cellules épithéliales pouvant être associées aux anomalies du placenta chez les gestations clonées. Le tissu de cotylédons provenant de gestations clonées et contrôles a été analysé par Western blot, RT-PCR quantitatif, et par immunohistochimie. Les résultats présentaient une diminution significative (p<0.05) de l’expression des dites protéines dans les cellules trophoblastiques chez les gestations clonées. Le RT-PCR quantitatif démontrait que les gènes CCND1, CLDN1 et MSX1 ciblés par la voie de signalisation de la Wnt/β-catenin étaient significativement sous exprimés. La diminution de l’expression des protéines E-cadherin et β-catenin avec une réduction de l’activation de la protéine β-catenin durant le période d’attachement de l’embryon peut potentiellement expliquer l’absence totale ou partielle de l’attachement des membranes fœtales au tissu maternel et éventuellement, l’insuffisance placentaire caractéristique des gestations clonées chez la vache. La caractérisation morphologique et fonctionnelle du placenta durant les gestations clonées à haut risque est essentielle pour évaluer le statut de la gestation. Les résultats de la présente étude permettront de prédire le développement et le bien-être fœtal de façon critique à travers un protocole standardisé et permettre des interventions médicales pour améliorer le taux de succès des gestations clonées chez les bovins.

Different studies has aimed the understanding of the causes that lead some cities, regions or territories to develop themselves, whereas others remain stagnant or get back. One starts from the presupposed that the development results from the standard of social territorial organization, this one capable to provoke collective territorial innovations, as a result of the institucional density, that is, from the local capacity to constitute relations in chain. The present análisis is centered in the municipality of Sarandi/RS/ Brasil, that from the nineties has enterprised a trajectory of uncomum development. From a serious situation of social economical crisis in the previous decades, its social economical and institutional actors, in a way of chain, were capable to reason a collection of initiatives that resulted in the structure of a microcluster in the department of clothes industry, counting today around 50 companies and the institutions of support minimally necessaries.

A dissertação “Reintegração Social de Adolescentes Privados de Liberdade em Teresina” tem como objetivo analisar como se materializam os itinerários de reintegração social de adolescentes privados de liberdade e inseridos no Centro Educacional Masculino (CEM), em Teresina, Estado do Piauí, tendo sido considerada para a análise dos dados a articulação entre as possibilidades e oportunidades oferecidas aos adolescentes e as suas reais necessidades, desejos e sonhos para uma reintegração segura ao meio social. A pesquisa foi desenvolvida na cidade de Teresina, junto aos adolescentes envolvidos com a prática de atos infracionais, sentenciados judicialmente e submetidos à Medida Socioeducativa de Internação, que segundo a Lei Brasileira n. 8.069 de 13 de Julho de 1990 – Estatuto da Criança e do Adolescente (ECA), as pessoas que compõem esse segmento da sociedade brasileira são consideradas sujeitos de Direitos e se encontram em situação peculiar de desenvolvimento pessoal e social. Os resultados apresentados revelam que o programa oficial de reintegração social de adolescentes do Piauí apresenta uma proposta de trabalho elaborada dentro do modelo determinado pelo ECA. Porém o desenvolvimento do trabalho de modo geral não ocorre de forma a garantir o que teoricamente apresenta, isto é, ações concretas que garantam o desenvolvimento psicossocial dos jovens, pois verificamos uma visível desarticulação entre necessidades e ofertas de oportunidades, bem como, a ausência de um grupo formado concretamente de educadores preparados para propor e executar ações que ofereçam meios para o adolescente moderar-se e viabilizar-se como pessoa e cidadão produtivo e assim poder construir seu projeto de vida e garantir seu retorno seguro ao meio social, familiar e comunitário.

We analyze how the characteristics of El Niño-Southern Oscillation (ENSO) are changed in coupled ocean–atmosphere simulations of the mid-Holocene (MH) and the Last Glacial Maximum (LGM) performed as part of the Paleoclimate Modeling Intercomparison Project phase 2 (PMIP2). Comparison of the model results with present day observations show that most of the models reproduce the large scale features of the tropical Pacific like the SST gradient, the mean SST and the mean seasonal cycles. All models simulate the ENSO variability, although with different skill. Our analyses show that several relationships between El Niño amplitude and the mean state across the different control simulations are still valid for simulations of the MH and the LGM. Results for the MH show a consistent El Niño amplitude decrease. It can be related to the large scale atmospheric circulation changes. While the Northern Hemisphere receives more insolation during the summer time, the Asian summer monsoon system is strengthened which leads to the enhancement of the Walker circulation. Easterlies prevailing over the central eastern Pacific induce an equatorial upwelling that damps the El Niño development. Results are less conclusive for 21ka. Large scale dynamic competes with changes in local heat fluxes, so that model shows a wide range of responses, as it is the case in future climate projections.

Background: Early microbial colonization of the gut reduces the incidence of infectious, inflammatory and autoimmune diseases. Recent population studies reveal that childhood hygiene is a significant risk factor for development of inflammatory bowel disease, thereby reinforcing the hygiene hypothesis and the potential importance of microbial colonization during early life. The extent to which early-life environment impacts on microbial diversity of the adult gut and subsequent immune processes has not been comprehensively investigated thus far. We addressed this important question using the pig as a model to evaluate the impact of early-life environment on microbe/host gut interactions during development. Results: Genetically-related piglets were housed in either indoor or outdoor environments or in experimental isolators. Analysis of over 3,000 16S rRNA sequences revealed major differences in mucosa-adherent microbial diversity in the ileum of adult pigs attributable to differences in earlylife environment. Pigs housed in a natural outdoor environment showed a dominance of Firmicutes, in particular Lactobacillus, whereas animals housed in a hygienic indoor environment had reduced Lactobacillus and higher numbers of potentially pathogenic phylotypes. Our analysis revealed a strong negative correlation between the abundance of Firmicutes and pathogenic bacterial populations in the gut. These differences were exaggerated in animals housed in experimental isolators. Affymetrix microarray technology and Real-time Polymerase Chain Reaction revealed significant gut-specific gene responses also related to early-life environment. Significantly, indoorhoused pigs displayed increased expression of Type 1 interferon genes, Major Histocompatibility Complex class I and several chemokines. Gene Ontology and pathway analysis further confirmed these results.

The research outlined in this paper highlights the importance of the early nutrition of vegetable crops, and its long-term effects on their subsequent growth and development. Results are also presented to demonstrate how the nutrient supply during the establishment stages of young seedlings and transplants can be enhanced by targeting fertiliser to a zone close to their developing roots. Three different precision fertiliser placement techniques are compared for this purpose: starter, band or side-injected fertiliser. The use of each of these methods consistently produced the same (or greater) yields at lower application rates than those from conventional broadcast applications, increasing the apparent recovery of N, P and K, and the overall efficiency of nutrient use, while reducing the levels of residual nutrients in the soil. Starter fertilisers also advanced the maturity of some crops, and enhanced produce quality by increasing the proportions of the larger and/or more desirable marketable grades. The benefits of the different placement techniques are illustrated with selected examples from research at Warwick HRI using different vegetable crops, including lettuce, onion and carrot.

In the vertebrate brain, the thalamus serves as a relay and integration station for diverse neuronal information en route from the periphery to the cortex. Deficiency of TH during development results in severe cerebral abnormalities similar to those seen in the mouse when the retinoic acid receptor (ROR)α gene is disrupted. To investigate the effect of the thyroid hormone recep-tors (TRs) on RORalpha gene expression, we used intact male mice, in which the genes encoding the α and beta TRs have been deleted. In situ hybridization for RORalpha mRNA revealed that this gene is expressed in specific areas of the brain including the thalamus, pons, cerebellum, cortex, and hippocampus. Our quantitative data showed differences in RORalpha mRNA expression in different subthalamic nuclei between wild-type and knock-out mice. For example, the centromedial nucleus of the thalamus, which plays a role in mediating nociceptive and visceral information from the brainstem to the basal ganglia and cortical regions, has less expression of RORalpha mRNA in the knockout mice (-37%) compared to the wild-type controls. Also, in the dorsal geniculate (+72%) and lateral posterior nuclei (+58%) we found more RORalpha mRNA in dKO as compared to dWT animals. Such differences in RORalpha mRNA expression may play a role in the behavioral alterations resulting from congenital hypothyroidism.

This research analyses the model of management for results from Minas Gerais State. The theoretical research begins with the origin of the management for results, then analyses its influences and tools. A case study about Minas Gerais State is presented and the model analysis tool is applied. This tool was developed by the Inter-American Development Bank (IADB). It is being applied in many countries and states. After that, interviews with the main people of Minas Gerais government were carried out to get information about the current model of management. With all this information collected, the model is analysed to verify if it is adequate or not to the standards of the management for results.

OBJETIVO: Investigar a ação hìstolítica de uma solução composta de fenol, glicerina e ácido acético na ascite neoplásica em cobaias. MÉTODOS: Foram utilizadas 32 cobaias, distribuídas por sorteio, em grupos experimentais e controles e estudados os efeitos da injeção peritonial da solução teste. Nos grupos controles empregou-se solução fisiológica. Foram estudadas alterações bioquímicas, anatomopatológicas (coração, pulmões, rins, baço e serosa peritonial), com 24 horas e 4 semanas de evolução. RESULTADOS: Verificou-se que a solução E quando instilada na cavidade peritonial não provocou nenhuma alteração clinica, histologica ou laboratorial nestes animais, quando comparados com o grupo controle. CONCLUSÃO: Frente aos resultados obtidos, consideramos interessante estudar os efeitos da solução proposta em casos de ascite neoplásica experimental em animais, com posterior estudo em seres humanos.
