Methicillin resistance has emerged in clinical isolates of Staphylococcus pseudintermedius from cats in Switzerland. Three cats suffering from urinary tract infections were infected with methicillin-resistant S. pseudintermedius (MRSP). Phenotypic and genotypic characterization of the resistance profile showed that the isolates displayed resistance to all beta-lactams and cephalosporins (blaZ, mecA), fluoroquinolones, tetracyclines [tet(K)], macrolides, lincosamides and streprogramins B [erm(B)], chloramphenicol (catpC221), trimethoprim [dfr(G)] and the aminoglycosides gentamicin [aac(6')-Ie-aph(2')-Ia], kanamycin and neomycin [aph(3')-III] and streptomycin [ant(6)-Ia]. They also harbor the leukocidin gene lukS-I. MRSP represents a new challenge for antibiotic therapy and this zoonotic bacteria may rapidly spread to animals and humans.

We have produced and analyzed transgenic birdsfoot trefoil (Lotus corniculatus L.) plants harboring antisense dihydroflavonol reductase (AS-DFR) sequences. In initial experiments the effect of introducing three different antisense Antirrhinum majus L. DFR constructs into a single recipient genotype (S50) was assessed. There were no obvious effects on plant biomass, but levels of condensed tannins showed a statistical reduction in leaf, stem, and root tissues of some of the antisense lines. Transformation events were also found, which resulted in increased levels of condensed tannins. In subsequent experiments a detailed study of AS-DFR phenotypes was carried out in genotype S33 using pMAJ2 (an antisense construct comprising the 5′ half of the A. majus cDNA). In this case, reduced tannin levels were found in leaf and stem tissues and in juvenile shoot tissues. Analysis of soluble flavonoids and isoflavonoids in tannin down-regulated shoot tissues indicated few obvious default products. When two S33 AS-DFR lines were outcrossed, there was an underrepresentation of transgene sequences in progeny plants and no examples of inheritance of an antisense phenotype were observed. To our knowledge, this is the first report of the genetic manipulation of condensed tannin biosynthesis in higher plants.

Activation of anion channels by blue light begins within seconds of irradiation in seedlings and is related to the ensuing growth inhibition. 5-Nitro-2-(3-phenylpropylamino)-benzoic acid (NPPB) is a potent, selective, and reversible blocker of these anion channels in Arabidopsis thaliana. Here we show that 20 μm NPPB blocked 72% of the blue-light-induced accumulation of anthocyanin pigments in seedlings. Feeding biosynthetic intermediates to wild-type and tt5 seedlings provided evidence that NPPB prevented blue light from up-regulating one or more steps between and including phenylalanine ammonia lyase and chalcone isomerase. NPPB was found to have no significant effect on the blue-light-induced increase in transcript levels of PAL1, CHS, CHI, or DFR, which are genes that encode anthocyanin-biosynthetic enzymes. Immunoblots revealed that NPPB also did not inhibit the accumulation of the chalcone synthase, chalcone isomerase, or flavanone-3-hydroxylase proteins. This is in contrast to the reduced anthocyanin accumulation displayed by a mutant lacking the HY4 blue-light receptor, as hy4 displayed reduced expression of the above enzymes. Taken together, the data indicate that blue light acting through HY4 leads to an increase in the amount of biosynthetic enzymes, but blue light must also act through a separate, anion-channel-dependent system to create a fully functional biosynthetic pathway.

Management of fisheries resources requires information on the fish stocks; the magnitude, distribution and trends of fishing effort and the trends of fish catches. Catch Assessment Surveys (CAS) in Lake Victoria are one of the ways through which the partner states sharing the lake are generating the key information required. In the Uganda part of the lake, CAS activities are carried out at 54 fish landing sites in the 11 riparian districts. The National Fisheries Resources Research Institute (NaFIRRI), the Department of Fisheries Resources (DFR), and the Districts of Busia, Bugiri, Mayuge, Jinja, Mukono, Wakiso, Kampala, Mpigi, Masaka, Kalangala and Rakai jointly conduct the surveys, The CAS enumerators are recruited from the fishing communities and work under direct supervision of sub-county fisheries officers. The National Fisheries Resources Research Institute (NAFIRRI), the Department of Fisheries Resources (DFR) and the districts in and around Lake Victoria jointly conduct Catch Assessment Surveys (CAS) with a major objective of providing information on the magnitude of fish catches in the lake. Fisheries contribute as high as 12% to Uganda's GDP. Catch assessment surveys are some of the avenues used to collect information for monitoring fish population dynamics as well as the magnitude, distribution and trends of fishing effort and fish catches. They are necessary for making sound decisions for the management of Lake Victoria fisheries. This information provides the Catch Per Unit of Effort (CPUE). The CPUE is used together with frame survey data to estimate total fish catches.

The electric power systems are getting more complex and covering larger areas day by day. This fact has been contribuiting to the development of monitoring techniques that aim to help the analysis, control and planning of power systems. Supervisory Control and Data Acquisition (SCADA) systems, Wide Area Measurement Systems and disturbance record systems. Unlike SCADA and WAMS, disturbance record systems are mainly used for offilne analysis in occurrences where a fault resulted in tripping of and apparatus such as a transimission line, transformer, generator and so on. The device responsible for record the disturbances is called Digital Fault Recorder (DFR) and records, basically, electrical quantities as voltage and currents and also, records digital information from protection system devices. Generally, in power plants, all the DFRs data are centralized in the utility data centre and it results in an excess of data that difficults the task of analysis by the specialist engineers. This dissertation shows a new methodology for automated analysis of disturbances in power plants. A fuzzy reasoning system is proposed to deal with the data from the DFRs. The objective of the system is to help the engineer resposnible for the analysis of the DFRs’s information by means of a pre-classification of data. For that, the fuzzy system is responsible for generating unit operational state diagnosis and fault classification.
