No presente trabalho, foi investigado o comportamento do extrato aquoso e alcoólico da beterraba (Beta vulgaris) como um indicador ácido-base, tendo como objetivo principal despertar o interesse pelo uso dos indicadores naturais, como alternativa didática para transmissão dos conceitos de titulação, equilíbrio químico e a lei de Lambert-Beer. Para a realização dos experimentos foram utilizadas soluções padrão de ácido clorídrico (HCl), ácido acético e hidróxido de sódio (NaOH). Inicialmente foram feitos testes para avaliar a reversibilidade do sistema, e observou-se que a coloração passa de vermelho para amarela com adição de base e retorna para vermelho com adição do ácido, sendo possível sua utilização para explicação do equilíbrio químico. As análises de espectroscopia UV-vis mostraram que os espectros de absorção molecular na região do visível, apresentam diferentes característica, dando indícios que ocorreu modificação na estrutura do indicador em meio ácido e básico. Após as análises volumétricas constatou-se que os pontos de equivalência determinados com o indicador natural tiveram concordância com os obtidos pelo método potenciométrico. Além disso, os extratos aquosos e alcoólicos da beterraba apresentam potencial didático para a explicação da lei de Lambert-Beer, sendo que no meio ácido houve uma melhor correlação comparado com o meio básico.

OBJETIVO: Avaliar a factibilidade e os resultados de uma técnica pouco invasiva para a pesquisa do linfonodo sentinela (LFNsn) com o uso do corante vital azul patente. MÉTODO: A pesquisa do LFNsn foi realizada em 12 pacientes portadores de adenocarcinoma gástrico que não apresentavam linfonodos suspeitos de metastases durante o estadiamento clínico pré-operatório. Injetou-se nos quatro quadrantes do tumor, 0,5 ml de corante vital azul patente. Os linfonodos que se coraram de azul foram classificados como LFNsn e foram obtidos após a gastrectomia com linfadenectomia D2. Utilizou-se a coloração HE para avaliação anatomopatológica dos LFNsn, e nos casos com LFNsn negativo para HE, foi realizada imuno-histoquímica com pan-citoqueratinas AE1/AE3. RESULTADOS: Dos 12 pacientes, oito eram mulheres, com média de idade de 64,5 (48-87) anos. Identificaram-se em média 3,25 (2-6) LFNsn por paciente. A factibilidade da técnica foi de 100%. A acurácia do método foi de 91,6% . Em 11 de 12 casos foi possível predizer as características linfonodais regionais pela concordância da análise anatomopatológica entre os LFNsn e não sentinelas(LFNñsn). CONCLUSÃO: A pesquisa do linfonodo sentinela em câncer gástrico é factível com o uso da técnica do corante azul patente. O método mostrou ser promissor como técnica minimamente invasiva para estadiar tumores gástricos, nesta casuística inicial.

A biópsia do linfonodo sentinela no tratamento do câncer de mama vem substituindo a linfadenectomia em casos iniciais. Este tratamento pode ser realizado com o corante azul vital e/ou radiofármaco. Há relatos de reações alérgicas ao corante com diferentes graus de severidade. Relata-se um caso de reação anafilática severa após injeção intradérmica do corante azul patente, em paciente com diagnóstico de carcinoma ductal in situ. O corante azul patente, apesar de facilitar a detecção do linfonodo, apresenta risco de desencadear reações anafilactóides. É necessário que a equipe envolvida tenha preparo para diagnosticar e tratar esta complicação.

O presente trabalho compara processos de purificação de enterotoxina estafilocócica A, utilizando cromatografia de afinidade com corante Red A em relação a troca iônica (SP - Sephadex C-25) - permeabilidade em gel (Sephadex G-75). Aplicou-se nas colunas o sobrenadante da cultura de Staphylococcus aureus 722 em caldo contendo 3% de triptona e suplementado com 1% de extrato de levedura, previamente concentradas com Amberlite CG-50. O processo capturou rapidamente a EEA, porém a proporção de 15 mg de resina para 150 mg de toxina causou saturação, recuperando apenas 10 a 30% de toxina do sobrenadante. A cromatografia de afinidade com Red A permitiu a recuperação de 60,87% de toxina aplicada em 76 horas, em relação a 114 horas requeridas para purificação utilizando coluna de troca iônica e permeabilidade em gel, com rendimento de 6,5%. O perfil eletroforético das amostras purificadas indicaram que, a toxina obtida da coluna Red A apresentou teor de pureza superior, na ordem de 90%, em relação a 60% atingida pelo método clássico.

A tendência do consumidor atual é utilizar alimentos de fácil preparo que, adicionalmente à qualidade nutritiva, tragam benefícios à saúde. Neste trabalho, uma massa alimentícia fresca funcional foi desenvolvida pela incorporação de Isolado Protéico de Soja (IPS) e polidextrose, utilizando páprica como corante. O consumo de IPS pode contribuir para a diminuição dos níveis de colesterol, enquanto a polidextrose, funcionando como fibra, pode promover a redução de risco de câncer de cólon. A otimização da formulação foi realizada utilizando-se um delineamento fatorial completo 23, considerando como variáveis independentes os teores de: i) IPS; ii) polidextrose; e iii) páprica. Os efeitos destas variáveis na qualidade da massa alimentícia foram avaliados em função das características físico-químicas (cor e teste de cozimento), textura instrumental e propriedades sensoriais. Na avaliação dos parâmetros físicos, observou-se que a cor instrumental da massa crua foi influenciada apenas pelo teor de páprica; já o tempo de cozimento (2,5 minutos), o aumento de massa (1,40 a 1,64%) e a perda de sólidos (3,80 a 5,56%) não foram influenciados pelos ingredientes utilizados; e a elasticidade (parâmetro da textura instrumental) foi reduzida com o aumento do teor de polidextrose. Na avaliação sensorial da massa crua, o aumento na porcentagem de IPS e a redução na porcentagem de polidextrose afetaram positivamente a aceitação global e a intenção de compra; e os aumentos nas porcentagens de IPS e de páprica aumentaram a aceitação da cor. Na avaliação sensorial da massa cozida, observou-se que o IPS teve influência positiva sobre a textura, enquanto que a polidextrose e a páprica afetaram negativamente este parâmetro. Entretanto, na avaliação da intenção de compra da massa cozida, não houve influência dos ingredientes adicionados. Os percentuais ideais de adição de polidextrose, isolado protéico de soja e páprica foram de 3,5, 8,0 e 1,5%, respectivamente.

The objective of this work was to evaluate biological aspects of Diatraea saccharalis fed on artificial diet containing different concentrations of the Sudan B Red dye and the possibility to mark the parasitoid Cotesia flavipes, when submitted to the parasitism of dyed caterpillars. For that, were added to the artificial diet four concentrations of Sudan Red B dye (100, 200, 300 and 400 ppm) and control (no dye addition). It was evaluated larval and pupal period, larval and pupal viability, longevity, sex rate, pupal weigh, eggs per female, eggs per day, number of eggs per egg mass, egg viability and embrionary period; besides same were accomplished measurements in the caterpillars (bioassay I). Caterpillars of 17 days old (30) of each treatment were removed from the tubes and exposed to the parasitism of C. flavipes (bioassay 2). The egg-pupae period, sex rate, pupal period and viability, number of females, males, total of emerged adults and longevity were evaluated. The data were submitted to the multivariate analisys methods: cluster analysis, two-way and principal component analysis. Based on analysis, it was observed that the treatment of 100 ppm was the least harmful to the biology of the sugar cane borer larvae by groping to the control and did not influence negatively its biological aspects. The concentration of 400 ppm affected negatively the biology of C. flavipes. The Sudan Red B it is ended doses marked the caterpillars and the adults, however the concentration of 100 ppm is the most suitable to dye D. saccharalis. None of the tested concentration marked adults of C. flavipes, despite to affect negatively its biology. It is unviable to increase the concentration seeking futures tests, for that dye to be harmful to the biological aspects of D. saccharalis and C. flavipes.

The food industry is interested in natural products. Anthocyanins are phenolic antioxidants of great importance with health-relevant applications. Several studies have linked the intake of fruits and vegetables with reduced risk of chronic degenerative diseases because of its antioxidant properties. This study aimed to compare different strategies for obtaining natural pigments from red jambo (Syzygium malaccence) and analyze its functional potential. Two different strategies were studied: (1) solid-liquid extraction (SLE) in reactor with controlled parameters, (2) powder obtention. The investigation of the functional potential was conducted taking into account the total phenolic content (TPC), the antioxidant activity (AA), the total anthocyanins concentration (TA) and α-amylase and α-glucosidase inhibition. The best extracts obtained by SLE showed TPC of 174.15 mg GAE/100g, AA of 3.56 μmol Trolox eq/g and TA of 133.59 mg cyd-3-glu/100 g. The best results for the second strategy were TPC of 1024.22 mg GAE/100 g, AA of 29.03 μmol Trolox eq/g and TA of 1193.41 mg cyd-3- glu/100 g. It was observed moderate amylase inhibition (26.30%) and high glucosidase inhibitory activity (97.47%). Skin extracts showed, in general, superior results when compared to whole red jambo, with superior values for dehydrated products. Based on our result, red jambo can be considered as a rich source of phenolic antioxidants, as well on amylase and glucosidase inhibitors

The generation of effluent from the finishing process in textile industry is a serious environmental problem and turned into an object of study in several scientific papers. Contamination with dyes and the presences of substances that are toxic to the environment characterize this difficult treatment effluent. Several processes have already been evaluated to remove and even degrade such pollutants are examples: coagulation-flocculation, biological treatment and advanced oxidative processes, but not yet sufficient to enable the recovery of dye or at least of the recovery agent. An alternative to this problem is the cloud point extraction that involves the application of nonionic surfactants at temperatures above the cloud point, making the water a weak solvent to the surfactant, providing the agglomeration of those molecules around the dyes molecules by affinity with the organic phase. After that, the formation of two phases occurred: the diluted one, poor in dye and surfactant, and the other one, coacervate, with higher concentrations of dye and surfactants than the other one. The later use of the coacervate as a dye and surfactant recycle shows the technical and economic viability of this process. In this paper, the cloud point extraction is used to remove the dye Reactive Blue from the water, using nonionic surfactant nonyl phenol with 9,5 etoxilations. The aim is to solubilize the dye molecules in surfactant, varying the concentration and temperature to study its effects. Evaluating the dye concentration in dilute phase after extraction, it is possible to analyze thermodynamic variables, build Langmuir isotherms, determine the behavior of the coacervate volume for a surfactant concentration and temperature, the distribution coefficient and the dye removal efficiency. The concentration of surfactant proved itself to be crucial to the success of the treatment. The results of removal efficiency reached values of 91,38%, 90,69%, 89,58%, 87,22% and 84,18% to temperatures of 65,0, 67,5, 70,0, 72,5 and 75,0°C, respectively, showing that the cloud point extraction is an efficient alternative for the treatment of wastewater containing Reactive Blue

This study aimed to evaluate the potential use of smectite clays for color removal of textile effluents. The experiments were performed by testing exploratory/planning method factorial and fractional factorial where the factors and levels are predetermined. The smectite clays were used originating from gypsum hub of the region Araripe-PE, and the dye used was Reactive Yellow BF-4G 200%. The smectite clay was collected and transported to the Laboratory of Soil Physics of UFRPE, where it held its preparation through air drying, lump breaking and classification in sieve to then submit it to the adsorption process. Upon completion of 22 complete factorial design it was concluded that the values of (96, 96,5 and 95,8%) corresponding to the percentage of of removal for "in-kind", chemically and thermally activated, respectively and adsorbed amounts of (4,80, 4,61 and 4,74 mg/g) for three clays. Showed that the activation processes used did not increase the adsorption capacity of smectite clay. The kinetic data were best fitted to the Freundlich isotherm, with an exponential distribution of active sites and that shows above the Langmuir equation for adsorption of cations and anions by clays. The kinetic model that best adapted to the results was the pseudosecond order model. In the factorial design study 24-1, at concentrations up to 500 mg/L obtains high percentage of color removal (92,37, 90,92 and 93,40%) and adsorbed amount (230,94, 227,31 and 233,50 mg/g) for three clays. The kinetic data fitted well to Langmuir and Freundlich isotherms. The kinetic model that best adapted to the results was the pseudosecond order model

The industries of food, medicine and cosmetic apply microencapsulation for many reasons, among them, stabilize the active, control the release of encapsulated and separate incompatible components of the formulation. In this context, microencapsulation techniques have been used in the food industry to provide stable liquid and solid ingredients. Anthocyanins have high antioxidant potential, but they are photodegradable. The challenges are therefore directed to the research for techniques that could make this potential remaining active and bioavailable and could be used as a vehicle for the delivery release of bioactive and micronutrients in appropriate conditions and levels. This work has as main objective to propose a method to encapsulate the anthocyanins in the extract of mountain apple using the interfacial polymerization technique. As well as to define the ideal conditions of temperature and agitation system for this procedure. The microparticles were characterized for size, morphology, active distribution, surface charge, degradation, composition and stability. The results, like particle diameter of 5.94 μm and Zeta potential of 7.03 mV, showed that the technique used to obtain these microparticles was satisfactory and has potential for application in cosmetics and food

The increasing demand for natural dyes in place of synthetic ones is justified by the non-toxicity or low toxicity of the former. The synthetic dyes are associated with diseases like cancer as well as when released in the environment takes longer to degrade and the intermediates could be still more toxic. The Annatto (Bixa Orellana L.) is a carotenoid and one of the more important natural dyes used in the food industry. In the form of dye, it represents nearly 70% of the world natural dye production and 90% in Brazil. In the present work, annatto seeds were used of the species peruana paulista, which had nearly 2.1% of bixin. The process of dye extraction with ethyl alcohol showed 4% of dye in the form of powder with particle diameter of 28mm. The extraction process did not alter the chemical composition of the dye, which was confirmed by the electronic spectrum of absorption. Dyeings were carried out with different mordents to study the total colour difference as well as the wash fastness properties and friction fastness properties under wet and dry conditions. The samples treated with copper sulphate showed colour difference but at the same time showed better fastness results. The samples treated with resin (no formaldehyde) did not alter the colour significantly still better the fastness properties. From the results, it could be stated that the resin could be an alternative for heavy metallic mordents

In this work, electrochemical technology was used to treat synthetic wastewater containing Methyl Red (MR) and Blue Novacron (BN) by anodic oxidation using anodes platinum (Pt) and real samples of textile effluents using DDB anodes and platinum (Pt). The removal of color from the galvanostatic electrolysis of synthetic wastewater MR and BN, and the actual sample has been observed under different conditions (different current densities and temperature variation). The investigation of these parameters was performed in order to establish the best conditions for removal of color and chemical oxygen demand (BOD). According to the results obtained in this study, the electrochemical oxidation processes suitable for the degradation process of color and COD in wastewater containing such textile dyes, because the electrocatalytic properties of Pt and BDD anodes consumption energy during the electrochemical oxidation of synthetic solutions AN and MR and real sample, mainly depend on the operating parameters of operation, for example, the synthetic sample of MR, energy consumption rose from 42,00kWhm-3 in 40 mAcm-2 and 25 C to 17,50 kWhm-3 in 40mAcm-2 and 40 C, from the BN went 17,83 kWhm-3 in 40mAcm and 40°C to 14,04 kWhm- 3 in 40mAcm-2 and 40 C (data estimated by the volume of treated effluent). These results clearly indicate the applicability of electrochemical treatment for removing dyes from synthetic solutions and real industrial effluents

The phenomenon of adsorption is of fundamental importance for the treatment of textile effluents and removal of dyes. Chitosan is characterized as an excellent adsorbent material, not only for its adsorption capacity but also the low cost production. Equilibrium and kinetic studies were developed in this study to describe the mechanism of adsorption of the anionic azo dye Orange G in chitosan, with the isotherms obtained from the variation of the concentration of dye in the continuous phase. The kinetics of the process was analyzed based on models involving the adsorption of molecules of the dye in nonpolar and polar sites. Adsorption experiments were carried out in water and in saline media with different NaCl concentrations, both for the determination of the equilibrium time as isotherms for making kinetic curves in which the amount of dye adsorbed measured indirectly varied with time. The experiments revealed the opening of the biopolymer structure with increasing concentration of Orange G, accompanied by high pH values and change on the type of interaction between the dye and the adsorbent surface, suggesting behavior advocated by the Langmuir equation in a certain range of concentration of the adsorbate and following the Henry's Law at higher concentrations, from the increased number of sites available for adsorption. The studies conducted showed that the saline medium reduces the chitosan s adsorption capacity according to a certain concentration, the occurrence of the cooperative adsorption process steps kinetic mechanism suggested as a new alternative for the interpretation of the phenomenon

Textile activity results in effluents with a variety of dyes. Among the several processes for dye-uptaking from these wastewaters, sorption is one of the most effective methods, chitosan being a very promising alternative for this end. The sorption of Methyl Orange by chitosan crosslinked particles was approached using equilibrium and kinetic analyses at different pH s. Besides the standard pseudo-order analysis normally effectuated (i.e. pseudo-first-order and pseudo-second-order), a novel approach involving a pseudo-nth-order kinetics was used, nbeing determined via non-linear regression, using the Levenberg-Marquardt method. Zeta potential measurements indicated that electrostatic interactions were important for the sorption process. Regarding equilibrium experiments, data were well fitted to a hybrid Langmuir-Freundlich isotherm, and estimated Gibbs free energy of adsorption as a function of mass of dye per area of chitosan showed that the process of adsorption becomes more homogeneous as the pH of the continuous phase decreased. Considering the kinetics of sorption, although a pseudo-nth-order description yielded good fits, a kinetic equation involving diffusion adsorption phenomena was found to be more consistent in terms of a physicochemical description of the sorption process
