O gênero Steno pertence à Ordem Cetartiodactyla, Família Delphinidae, e compreende apenas uma espécie: o golfinho-de-dentes-rugosos, Steno bredanensis. O golfinho-de-dentes-rugosos é encontrado nos Oceanos Atlântico, Pacífico e Índico, em águas profundas tropicais, subtropicais e temperadas quentes. Entretanto, em algumas localidades como as regiões Sudeste e Sul do Brasil, esta espécie é conhecida por apresentar hábitos costeiros, o que a torna suscetível a ameaças antropogênicas como a degradação do hábitat, as capturas acidentais e diversos tipos de poluição. Conhecer a magnitude destes impactos e o grau de diferenciação genética das populações usando marcadores moleculares são aspectos importantes para a conservação da espécie. Os marcadores moleculares são segmentos específicos de DNA que podem ou não fazer parte de um gene e que apresentam grau de polimorfismo adequado para responder questões sobre as relações genéticas de indivíduos, populações ou diferentes espécies. O DNA mitocondrial é um dos marcadores moleculares mais utilizados em estudos sobre estrutura populacional, sistemática e filogenia de cetáceos. Estudos genéticos têm mostrado que várias espécies de delfinídeos apresentam estrutura populacional genética, entre e dentro das bacias oceânicas. No presente estudo foi investigada a diferenciação genética do golfinho-de-dentes-rugosos usando sequências da região controle mitocondrial de várias localidades em todo o mundo (Oceano Pacífico Centro-Sul: N=59; Pacífico Tropical Leste: N= 4; Pacífico Noroeste: N=1; Oceano Índico: N=1; Atlântico - Caribe: N=3; Atlântico Sudoeste: N=44; N total = 112). Análises preliminares indicaram grande diferenciação genética entre os Oceanos Atlântico e Pacífico/Índico (distância p = 0,031), que foram posteriormente investigadas utilizando sequências do citocromo b e mitogenomas completos. As análises filogenéticas de Neighbor-Joining e Bayesianas não foram conclusivas sobre a existência de especiação críptica em Steno. No entanto, a grande diferenciação entre as bacias oceânicas merece uma análise mais aprofundada, utilizando outros marcadores genéticos (por ex., sequências nucleares) bem como dados morfológicos. Não obstante, as análises AMOVA e FST par-a-par revelaram forte diferenciação populacional, não só entre os oceanos Atlântico e Pacífico, mas também no Atlântico, onde foram detectadas três populações: Caribe, região Sudeste e região Sul do Brasil. As populações detectadas no Atlântico Sudoeste devem ser aceitas como Unidades de Manejo (Management Units, MU) e dados demográficos básicos precisam ser levantados para essas MU, a fim de possibilitar uma melhor avaliação dos impactos antrópicos sobre elas. Este estudo fornece a primeira perspectiva sobre a diferenciação genética mundial de S. bredanensis.

O CPAA esta testando um metodo de conservacao fundamentado em aspectos genetico-ecologicos. Em uma area de 0,25ha, procurou-se representar as estrategias utilizadas pelas especies autoctones para manterem-se em equilibrio no ecossistema tropical: diversidade, densidade e variabilidade genetica. A pesquisa, iniciada em janeiro de 1994, estuda a conservacao de recursos geneticos de sete especies frutiferas (Theobroma grandiflorum, T. cacao, Bactris gasipaes, Euterpe spp, Myrciaria dubia, Rollinia mucosa e Couma utilis) e cinco florestais (Hevea spp, Ceiba pentandra, Jacaranda copaia, Buchenavia huber, Trattinickia burserifolia), utilizando-se tres espacamentos. As caracteristicas biometricas das plantas sao anualmente medidas e, apos cinco anos, a viabilidade tecnica sera avaliada quanto a sobrevivencia e capacidade das especies em deixar descendentes. Preliminarmente, as especies que apresentaram valores superiores na avaliacao de estabelecimento foram C. pentandra entre as florestais (medias de 4,76m de altura e 11,46cm de diametro, a 50cm do solo) e R. mucosa entre as fruteiras (media de 3,71m de altura e 6,28cm de diametro, a 50cm do solo).

O MHC (Major Histocompatibility Complex) é um sistema genético importante para a manutenção de espécies ameaçadas, uma vez que baixa variabilidade para locos MHC tem sido associada a uma menor capacidade de resposta a doenças e diminuição do sucesso reprodutivo. Deste modo, pesquisas sobre a variabilidade genética do MHC têm demonstrado ser bastante informativas em estudos populacionais voltados para aspectos referentes à conservação. No presente trabalho foi investigada a variabilidade genética do MHC para três espécies de mamíferos marinhos (toninha, baleia franca austral e lobo marinho sul-americano) do sul do Brasil, com intensa mortalidade provocada por atividades humanas atuais ou passadas. As amostras foram coletadas de animais mortos encalhados na costa, de animais capturados acidentalmente por barcos pesqueiros, e também através de um sistema de biópsia. A região variável do exon 2 do gene DQB do MHC foi amplificada por PCR (Polymerase Chain Reaction) em 109 amostras de toninhas (Rio de Janeiro n=32, Rio Grande do Sul n=52, Argentina n=25), 35 amostras de lobo marinho sul-americano e 30 amostras de baleia franca austral, utilizando-se um par de primers heterólogos. O fragmento resultante de 172 pares de bases foi analisado quanto ao polimorfismo de seqüência através da técnica de SSCP (Polimorfismo de Conformação de Fita Simples) em todas as amostras de toninha e de lobo marinho sul-americano e 14 amostras de baleia franca austral. Dificuldades associadas à amplificação resultaram em padrões de SSCP pouco informativos para as amostras de lobo marinho sul-americano e baleia franca austral Todas as amostras de toninha apresentaram um padrão de pelo menos 4 bandas por indivíduo. As 4 bandas de um único indivíduo do Rio Grande do Sul foram seqüenciadas, tendo sido possível verificar que 2 seqüências relacionadas ao genes DQB estão sendo amplificadas com estes primers. Pelas análises de SSCP foi possível detectar ausência de variabilidade para as amostras de toninha provenientes do Rio de Janeiro e diferenciá-las da população da Argentina, que é polimórfica. A população do Rio Grande do Sul parece apresentar níveis intermediários de variação em relação aos extremos da distribuição da espécie. Analisando as três populações amostradas, conclui-se que a espécie apresenta baixos níveis de variabilidade para o loco DQB, a exemplo do que é reportado para os genes de MHC de outros mamíferos marinhos.

O gênero Paspalum L. compreende aproximadamente 400 espécies no mundo e cerca de 220 no Brasil. Paspalum é ecologicamente e economicamente importante e tem sido utilizado como pastagem. Paspalum notatum Flügge (grama-forquilha) é uma valorosa gramínea forrageira nos subtrópicos. Esta espécie consiste de vários biótipos sexuais (diplóides) e apomíticos (tetraplóides, ocasionalmente tri e pentaplóides). Neste trabalho, os Inter Simple Sequence repeat (ISSR) foram utilizados para acessar a diversidade genética da grama-forquilha (Paspalum notatum). Os tecidos vegetativos de 95 acessos de grama-forquilha foram obtidos de vários locais da América do Sul (Brasil, Argentina e Uruguai). Um total de 91 de fragmentos reproduzível ISSR foi observado. Oitenta e nove fragmentos (97,5% do total observado) foram polimórficos. A análise de agrupamento (UPGMA) foi realizada para o conjunto de dados ISSR. Os resultados ilustram as relações genéticas entre 95 acessos de Paspalum notatum. A comparação entre dados moleculares, morfológicos e nível de ploidia foi realizada. Em resumo, os marcadores moleculares ISSR mostraram-se eficientes para distinção dos genótipos analisados e observou-se uma variabilidade ampla para a espécie. Estes resultados adicionam novas informações sobre a diversidade genética em Paspalum notatum, conseqüentemente contribuindo para o conhecimento biológico desta espécie e fornecendo subsídios para futuros programas de melhoramento genético e para programas de conservação.

Brazil is one of the major centers of diversity for polyploid cotton plants; these plants belong to the genus Gossypium, which has three known species: G. hirsutum, G. barbadense and G. mustelinum. The Northeast is the only region where the three species occur, the last group being endemic. Northeast s cotton plants can be important sources of variability for genetic breeding. It is believed that great part of local diversity is being lost, due to economic, political, cultural and agricultural problems. In an attempt to mitigate this loss and delineate conservation strategies it is necessary to know how the species are found where they occur. The objective was to characterize and determine how plants are maintained in situ in the states of Maranhão, Piauí, Ceará, Rio Grande do Norte and Paraíba at the beginning of the XXI century. The in situ characterization of G. hirsutum and G. barbadense was conducted through structured interviews with the cotton plants owners and through the analysis of the environment. The data were collected during expeditions undertaken between the years 2004 to 2005. Twenty-two plants were collected in the state of Paraíba, forty-four in the state of Rio Grande do Norte, one hundred and forty-six in the state of Ceará, forty in the state of Maranhão and ninety-one plants in the state of Piauí. All plants collected in the states of Paraíba and Rio Grande do Norte belonged to moco type. Moco cotton plants also predominated in the other states, representing 92%, 62% and 78% of plants collected in Ceará, Piauí and Maranhão, respectively. The other cotton plants collected belong to the species G. barbadense. The cotton plants were found in situ as dooryard plants, roads side, feral populations, cultivation or local varieties. Great part were dooryard plants (45.2%), being major in Piauí and Maranhão. Cultivation predominated in Ceará; in Rio Grande do Norte feral populations were the most frequent and, in Paraíba, local varieties. The maintenance of moco plants is related, mainly, to the phytotherapic domestic use (20.9%) and to confection of lamp wicks (29.7%). Few inhabitants in Paraíba, Rio Grande do Norte, Piauí and none in Maranhão used harvest the plants, storage the seeds or gin; however, in Ceará, 40.5% of owners affirmed that they harvested and commercialized the fiber. It was found that the maintenance of species is dependent of the fragile cultural habits of local inhabitants, therefore the maintenance in situ is not a suitable way to conservation of genetic resources. Gossypium mustelinum Miers ex Watt is the only cotton species native from Brazil. It is endemic of the semi-arid region from North-east of the country, where it occur near from resilient water sources. The threats to the in situ conservation of the populations are caused by human interference in its habitat, mainly by excessive cattle graze and deforestation. Establish efficient strategies of in situ conservation depend on the accomplishment of a diagnosis of how the specie is found in its natural environment, and the knowledge about the genetic structure of the populations. The objectives of this work were i) to determine the in situ conditions of two populations present in rivers from basin of Rio Paraguaçu at the Bahia State, ii) to evaluate the structure and genetic variability presented in both populations, iii) to establish in situ and ex situ conservation strategies. It were realized collection in november 2007, when was realized in situ characterization of G. mustelinum. SSR markers were used for analyze 218 genotypes deriving from two populations of the G. mustelinum, localized at Tocó river and the Capivara river. The allelic frequencies, the heterozigosity and the F statics were estimated. All the plants were classified as wild and natives, and there was no evidence of the use the plants or its parts. The populations showed different conservation conditions in situ. Few plantlets were found in sites with excessive cattle feed, an indication that the damages in young plants should be high enough to compromise the renovation of the populations. On the other hand, populations were well preserved when the anthropic damages was low or inexistent. The 14 SSR primer pairs amplified 17 loci with a medium number of 5 alleles per locus (a total of 85 alleles). The high level of endogamy estimated (FIS=0,808) and the low observed heterozygosity (H0=0,093) were indicatives that the populations reproduce mainly by selfing, geitonogamy and crosses between related individuals. The genetic diversity was high (HE=0,482) and the differentiation between the populations was very high (FST=0,328). At least two sites from both populations of G. mustelinum must be preserved to achieve suitable in situ conservation. Actions that preserve the gallery forest and keep the cattle away should implemented, and could be as simple as erecting a fence. It is not possible anticipated if the in situ preservation will be possible. Therefore collections and ex situ preservation of representative specimens are essential to conserve the genetic diversity of native G. mustelinum

Brazil has been considered one of the diversity centers of Gossypium barbadense species. It is believed that a relatively big erosion genetic process occurs with the species, due to economic, cultural and agricultural problems. A local diagnostic about species situation is the first step for reducing the diversity loss and establishing conservation strategies in situ. This research aimed the identification of the presence of Gossypium populations, characterization, determination of the main risks and collection of the accesses to store in germoplam banks, in Para and Amapa States. Expeditions were conducted in November 2004. An interview was carried out with the plant proprietor for characterizing in situ of G. barbadense species and of the environment where the plants were inserted. On hundred seventy nine plants in 22 municipal districts were collected in Para State and 117 plants in nine municipal districts in Amapa State. The majority of plants belong to G. barbadense species (98% in Amapa and 94% in Para). Plants occur in back yards, beside roads and spontaneously. That ones from back yards were more abundant (97% in Amapa and 95% in Para) and maintained as medicinal plants as the principal reason. Plants in natural environments in both states evaluated were not found, therefore, the creation of reserves and the application of others conventional methods of maintenance in situ are not applicable. The plant proprietors do not use to store or process seeds. Seed storage was reported as a practice by only 1% of the plant proprietors from Para and 11% from Amapa. The most plants collected were from two to three years of age (58% in Amapa and 93% in Para). As conclusions G. barbadense is the species most spread in the two studied states and are found in back yards. In Amapa State the botanical variety barbadense or Quebradinho is predominant, whereas in Para State the predominant variety is brasiliense or Rim-de-boi. The objective of this study was to identify DNA polymorphisms at the genes leptin, β-lactoglobulin and pituitary-specific transcription factor in three genetic groups of Holstein x Guzerat dairy cows and investigate the relationship between their genotypes and the composition and quality of milk of dairy cows. Samples were collected in August 2009, being 113 blood samples from lactating crossbred cows and 58 milk samples. For analysis of DNA polymorphisms blood samples were collected, analyzed later in the Genetic Laboratory affiliated to the Zootechny Institute of São Paulo and individual milk samples were collected according to standards established by the laboratory of Management Program of Northeast Dairy Herds (PROGEN), at Federal Rural University of Pernambuco (UFRPE) for analysis of milk composition and quality. The characterization of genotypes was performed by PCR-RFLP, for which were designed specific primers for each studied gene and restriction enzymes Kpn2I, HaeIII and HinfI that cut the DNA of the following genes: leptin, β-lactoglobulin and a PIT, respectively. The leptin estimate genotypic frequence were CC 0.112, TT 0.225 and CT 0.661, for β-lactoglobulin were AA 0.136, AB 0.323 and BB 0.539, and for PIT were ++ 0.655, -- 0.311 and +- 0.032. The results show that the population is in Hardy-Weinberg disequilibrium for leptin, β-lactoglobulin and a PIT due to excess of heterozygotes in the population, however, as these genes are associated with the milk production it is considered that the animals have genetic potential for milk production in the Brazilian semi-arid conditions. Through the characterization of the studied herd there were not found implications of the polymorphism of leptin, β-lactoglobulin and PIT in the composition and quality of milk from cows in the different genetic groups 1/2, 3/4 and 7/8 Holstein x Guzerat. Among the traits of economic importance to dairy cattle livestock those related to sexual precocity and longevity of the herd are essential to the success of the activity, because the stayability time of a cow in a herd is determined by their productive and reproductive lives. In Brazil, there are few studies about the reproductive efficiency of Swiss-Brown cows and no study was found using the methodology of survival analysis applied to this breed. Thus, in the first chapter of this study, the age at first calving from Swiss-Brown heifers was analyzed as the time until the event by the nonparametric method of Kaplan-Meier and the gamma shared frailty model, under the survival analysis methodology. Survival and hazard rate curves associated with this event were estimated and identified the influence of covariates on such time. The mean and median times at the first calving were 987.77 and 1,003 days, respectively, and significant covariates by the Log-Rank test, through Kaplan-Meier analysis, were birth season, calving year, sire (cow s father) and calving season. In the analysis by frailty model, the breeding values and the frailties of the sires (fathers) for the calving were predicted modeling the risk function of each cow as a function of the birth season as fixed covariate and sire as random covariate. The frailty followed the gamma distribution. Sires with high and positive breeding values possess high frailties, what means shorter survival time of their daughters to the event, i.e., reduction in the age at first calving of them. The second chapter aimed to evaluate the longevity of dairy cows using the nonparametric Kaplan-Meier and the Cox and Weibull proportional hazards models. It were simulated 10,000 records of the longevity trait from Brown-Swiss cows involving their respective times until the occurrence of five consecutive calvings (event), considered here as typical of a long-lived cow. The covariates considered in the database were age at first calving, herd and sire (cow s father). All covariates had influence on the longevity of cows by Log-Rank and Wilcoxon tests. The mean and median times to the occurrence of the event were 2,436.285 and 2,437 days, respectively. Sires that have higher breeding values also have a greater risk of that their daughters reach the five consecutive calvings until 84 months

About 40% of the earth is occupied by tropical and subtropical forests, including 42% of dry forests, where there is Caatinga Bioma, contemplating tree forests and shrubs, with xerophytic characteristics. Study and conservations of Caatinga biologic diversity is one of the greatest challenges of Brazilian science because those are, proportionally, the less studied among natural areas, with most of the scientific effort centered in very few points around the main cities in the area and also because it is the less protected natural Brazilian area. The environmental degradation is constantly increasing and has its rhythm accelerated by the men appropriation to meet or not their own needs. Therefore, species conservation should be based in three principles: the use of natural resources by present generation, waste prevention and use of the natural resources to benefit the majority of the citizens. Among the strategies to species conservation, we can mention the ex situ conservation , in which the conservation of genetic resources may be realized outside of the natural environment in which the species occur, and in situ conservation , or, in other words, in the places where the species occur. In ex situ conservation, the germplasm collections are maintained in the field and/or in laboratories (conservation chambers), and this mainly conserves intraspecific diversity (genetic variance), the ex situ collections are continuously enriched by collection activities, introduction and germplasm interchange; the in situ conservation preserving ecosystems and habitats, maintaining and recovering native population of species of interest. So, the objective of this paper is the search for strategies to the conservation of Mimosa caesalpiniifolia B. (sabiá) using instruments of environmental perception and plant biotechnology, as mechanisms of in situ and ex situ conservation. To environmental perception, were realized open, semi-structured and qualitative interviews. The questions included socioeconomic data and knowledge of Sabiá specie. To plant biotechnology, Sabiá seed collection were realized in different location to formation of a germplasm bank. The specie micropropagation was made from nodal segment of plants from the matrizeiro. About the knowledge of rural populations and the use of Sabiá plant, some preferences occurred from speeches that the plant possesses a firm wood, not attacked by termites, legalized for exploration by the Brazilian environmental organ (IBAMA), and is a native specie. This research found the rural population has knowledge about Sabiá specie and the natural resources are exhausting. The proposal that the rural community brought was the donation of the Sabiá specie seeding initiating on the rain season, in which the seeding would be plated between the lots, in individual plantations. To the formation of a matrix bank, plant biothecnology brought answers favorable to Sabiá specie seeding, with the formation of multiple shoots

The Caatinga biome has a high diversity of potential and their conservation constitutes one of the greatest challenges of Brazilian science. The sustainable management of the Caatinga emerges as an alternative that through the formation of systems agrossilvipastoris, enables the use of forest resources sustainably, ensuring their conservation, regeneration and recovery. In RN this technique has been developed mainly in settlements of Agrarian Reform, such as P. A. Moaci Lucena, and their impacts go beyond the environmental aspect and reverberate socially and economically on the quality of life of family farmers. Despite the efficiency of the Sustainable Management of the Caatinga in the conservation of native species, many forests species of this biome faces serious problems of propagation and for this reason have become vulnerable to extinction, as is the case of Mimosa caesalpiniifolia Benth . Thus , it is evident the need to use sustainable alternatives to overcome the difficulties of propagation of this species and enable their replacement in areas where their existence is threatened. The Plant Biotechnology is considered as a promising alternative in this sense, considering that by micropropagation enables the large-scale production of seedlings with high health genetics status. This work has the following objectives: evaluate the perception of family farmers of P. A. Moaci Lucena in relation to social, environmental and economic impacts of the Sustainable Management of the Caatinga and check the conditions of germination and in vitro propagation of Mimosa caesalpiniifolia Benth that enabling the production of seedlings of this specie on a large scale. To achieve the first objective, semi-structured interviews showed that in the perception of farmers PA Moaci Lucena, the Sustainable Management of the Caatinga was responsible for generating many social, environmental and economic impacts that affected directly in the improvement in the quality of life of the families of the Settlement Project Moaci Lucena. Have to achieve the second objective, were investigated the influence of different substrates and concentrations of growth regulator BAP in the germination and shoot induction in vitro of Mimosa caesalpiniifolia Benth. The vermiculite was presented as the most suitable substrate for germination of this species, because it provided a more rapid germination, higher growth rates and higher dry matter accumulation. Regarding micropropagation, the concentration of 17.76 μmol/L of BAP presented a more responsive in relation to multiplication rate and the number of shoots in M. caesalpiniifolia, thus constituting the most suitable concentration for the in vitro propagation of this specie

Brazil has about 8,500 km of coastline and on this scale, fishing is a historically important source of animal protein for human consumption. The national fishing background shows a growth of marine fishery production until 1985 and within this period it was recorded a steady decline. From the year 2003 fishing statistics aim to some "recovery" of the total fisheries production, which probably is related to a change in industry practice. The target of commercial fishing became smaller species with low commercial value, but very abundants. The coney, Cephalopholis fulva (Serranidae), is one of these species that have been suffering a greater fishing pressure in recent years. In order to provide data about the current situation of the genetic diversity of these populations, several molecular markers have been being used for this purpose. The prior knowledge of genetic variability is crucial for management and biodiversity conservation. To this end, the control region sequences (dloop) of mtDNA from Cephalopholis fulva (Serranidae) from five geographical points of the coast of Brazil (Ceará, Rio Grande do Norte, Bahia and Espírito Santo) and the Archipelago of Fernando de Noronha (FN) were sequenced and their genetic diversity analyzed. The FST values were very low (0.0246 to 0.000), indicating high gene flow between the sampled spots. The indices h and indicate a secondary contact between previously allopatric lineages differentiated or large and stable populations with long evolutionary history. Tests of Tajima and Fu showed expansion for all populations. In contrast, the mismatch distribution and SSD indicated expansion just for coastal populations. Unlike other species of the Atlantic which have been deeply affected by events on later Pleistocene, the population-genetic patterns of C. fulva may be related to recent events occurred approximately 130,000 years ago. Moreover, the data presented by geographical samples of the specie C. fulva showed high genetic diversity, also indicating the absence of deleterious effects of over-exploitation on this specie, as well as evidence of complete panmixia between all sampled populations
