Resumo:
Este documento faz a análise do associativismo em Cabo Verde nos territórios rurais da ilha de Santiago. Apresenta o estudo de caso de três associações comunitárias da ilha, focaliza os principais parceiros, as actividades desenvolvidas, o seu funcionamento e o contributo dos mesmos no desenvolvimento local/comunitário. A Analise comparativa demonstra uma proximidade dessas associações quanto a origem, (impulso da ajuda externa), objectivos e as acções desenvolvidas apesar de localizarem em áreas geográficas distintas. As acções desenvolvidas variam de acordo com a necessidade de cada comunidade e são desenvolvidas com base nos contratosprograma. A participação defendida como o ideal para a implementação dos projectos e o funcionamento da organização ficou explícita no trabalho de campo, que na realidade muitos dos membros não o exercem. Espera-se que este documento venha a contribuir para o conhecimento da actividade associativa em Cabo Verde em geral e na ilha de Santiago em particular.

O estudo foi realizado no Triângulo Mineiro e teve como objetivo avaliar as modificações morfológicas e físicas de um Latossolo Vermelho-Amarelo argiloso, quando comparadas uma pastagem antiga de baixa produtividade (BCO) e duas pastagens renovadas (BRP e BRC), utilizando uma área de Cerrado (CER) como referência. As análises físicas e morfológicas permitem concluir que: (a) existem impedimentos estruturais para o desenvolvimento radicular na pastagem antiga (BCO); (b) a porosidade formada por fissuras nas pastagens renovadas compensa a menor porosidade encontrada nos torrões; (c) a macrofauna de invertebrados teve importante papel na formação e na estabilidade de agregados e na regeneração dos volumes estruturais compactos; (d) nas pastagens renovadas, formadas somente por gramíneas ou consorciadas com Stylosanthes guianensis cv. Mineirão, os impedimentos estruturais deixam de existir; (e) o método do perfil cultural é recomendável em avaliações morfológicas de solos cultivados com pastagens.

O objetivo do presente trabalho foi avaliar o efeito da variabilidade da precipitação sobre o escoamento direto e sobre o aporte de sedimento na bacia do rio Pipiripau (A = 235 km²), no Distrito Federal. Cinco anos típicos da série de precipitação foram selecionados: o ano mais úmido, o ano mais seco, o ano médio e dois anos que representaram os desvios-padrão da série (médio-seco e médio-úmido). Depois de o modelo ser calibrado com dados diários de chuva e vazão da bacia, simulações foram realizadas para cada evento chuvoso de cada um dos cinco anos selecionados da série, e o escoamento direto e o aporte de sedimento da bacia foram calculados com a MUSLE em três diferentes cenários de uso e manejo do solo: o cenário atual, onde predomina o preparo convencional do solo (PC) nas áreas de agricultura da bacia; o passado recente, em que predominava o Cerrado nativo (CER); e o cenário de plantio direto (PD), nas áreas de agricultura da bacia. Apesar de as vazões médias anuais observadas terem apresentado relação linear com os volumes de precipitação pluvial anual, em todos os cenários, o escoamento direto e o aporte de sedimento apresentaram aumento exponencial com o aumento da precipitação pluvial anual, sendo esse mais pronunciado na condição de preparo convencional (PC) do que nos outros cenários estudados (CER e PD). O aumento observado pode ser explicado pela não-linearidade do modelo MUSLE ao volume de precipitação e ao volume de escoamento.

Nutrients are basically transported to the roots by mass flow and diffusion. The aim of this study was to quantify the contribution of these two mechanisms to the acquisition of macronutrients (N, P, K, Ca, Mg, and S) and cationic micronutrients (Fe, Mn, Zn, and Cu) by maize plants as well as xylem exudate volume and composition in response to soil aggregate size and water availability. The experiment was conducted in a greenhouse with samples of an Oxisol, from under two management systems: a region of natural savanna-like vegetation (Cerradão, CER) and continuous maize under conventional management for over 30 years (CCM). The treatments were arranged in a factorial [2 x (1 + 2) x 2] design, with two management systems (CER and CCM), (1 + 2) soil sifted through a 4 mm sieve and two aggregate classes (< 0.5 mm and 0.5 - 4.0 mm) and two soil matric potentials (-40 and -10 kPa). These were evaluated in a randomized block design with four replications. The experiment was conducted for 70 days after sowing. The influence of soil aggregate size and water potential on the nutrient transport mechanisms was highest in soil samples with higher nutrient concentrations in solution, in the CER system; diffusion became more relevant when water availability was higher and in aggregates < 0.5 mm. The volume of xylem exudate collected from maize plants increased with the decrease in aggregate size and the increased availability of soil water in the CER system. The highest Ca and Mg concentrations in the xylem exudate of plants grown on samples from the CER system were related to the high concentrations of these nutrients in the soil solution of this management system.
Planossolos e Gleissolos Utilizados na Fabricação de Cerâmica Artesanal no Semiárido de Minas Gerais

O conhecimento etnopedológico tem fornecido informações importantes sobre o modo de vida das populações rurais a respeito de suas tradições ancestrais, como a arte de elaborar peças artesanais a partir do barro advindo de solos com características próprias a esse uso. O objetivo deste trabalho foi avaliar física, química e mineralogicamente Planossolos e Gleissolos explorados para a produção de artefatos de cerâmica artesanal em Minas Gerais. Nos barreiros, foram coletados dois perfis de Planossolos (P1 e P2) e um Gleissolo (P3) usados como matéria-prima na produção artesanal de cerâmica. Foram realizadas análises físicas e químicas, limites de liquidez (LL) e plasticidade (LP), índice de plasticidade (IP) e de atividade coloidal (IA), além da mineralogia da fração argila. Os horizontes selecionados pelos ceramistas para a fabricação de cerâmica artesanal (BA, Btg e BCg, do P1; Btg1 e Btg2, do P2; e C2g e C3g, do P3) apresentaram os maiores teores de argila e silte, IP e IA, importantes para a qualidade final da cerâmica. O horizonte Cg do perfil P1 possui potencial de ser utilizado para a produção artesanal, em virtude do seu IP, superior aos dos horizontes normalmente usados, além dos teores de argila, silte e areia fina e suas características mineralógicas. A proporção ideal das frações areia, silte e argila e a porcentagem de matéria orgânica na definição de um bom material para cerâmica são difíceis de estabelecer e variam principalmente em razão de aspectos quantitativos e qualitativos da argila nos solos.

O papel fundamental da matéria orgânica (MO) justifica o crescente interesse pela identificação de sistemas de uso e manejo que melhorem o estoque orgânico em solos tropicais. O objetivo deste trabalho foi analisar variações quantitativas e qualitativas da MO e caracterizar compartimentos orgânicos em um Latossolo Vermelho-Escuro argiloso sob vegetação natural antropizada (CER), pastagem de longa duração (PAL), pastagem degradada (PAD), e pousio (PAC), comparados com culturas sob preparo convencional (CCL) e plantio direto (PD). Foi encontrada pouca variação dos estoques orgânicos na camada superficial, explicada pela antropização da vegetação em CER, pela não-exportação dos resíduos em PD e CCL e pela prática de pousio em PAC. Fracionamento granulométrico, considerando os compartimentos: resíduos vegetais (20-2.000 µm), organo-siltoso (2-20 µm) e organo-argiloso (0-2 µm), mostrou diferenças na qualidade da MO quando comparadas situações edafoambientais semelhantes. Mesmo com pequenas variações, o compartimento resíduos vegetais foi um indicador da evolução dos estoques orgânicos, permitindo a caracterização da degradação nas pastagens e do efeito do plantio direto, quando comparado ao sistema convencional. PD favoreceu a estocagem de C no compartimento organo-argiloso. Solos estudados diferem de outros solos argilosos tropicais pela mais elevada relação C/N encontrada nas frações 0-20 µm.

Systems biology is a new, emerging and rapidly developing, multidisciplinary research field that aims to study biochemical and biological systems from a holistic perspective, with the goal of providing a comprehensive, system- level understanding of cellular behaviour. In this way, it addresses one of the greatest challenges faced by contemporary biology, which is to compre- hend the function of complex biological systems. Systems biology combines various methods that originate from scientific disciplines such as molecu- lar biology, chemistry, engineering sciences, mathematics, computer science and systems theory. Systems biology, unlike “traditional” biology, focuses on high-level concepts such as: network, component, robustness, efficiency, control, regulation, hierarchical design, synchronization, concurrency, and many others. The very terminology of systems biology is “foreign” to “tra- ditional” biology, marks its drastic shift in the research paradigm and it indicates close linkage of systems biology to computer science. One of the basic tools utilized in systems biology is the mathematical modelling of life processes tightly linked to experimental practice. The stud- ies contained in this thesis revolve around a number of challenges commonly encountered in the computational modelling in systems biology. The re- search comprises of the development and application of a broad range of methods originating in the fields of computer science and mathematics for construction and analysis of computational models in systems biology. In particular, the performed research is setup in the context of two biolog- ical phenomena chosen as modelling case studies: 1) the eukaryotic heat shock response and 2) the in vitro self-assembly of intermediate filaments, one of the main constituents of the cytoskeleton. The range of presented approaches spans from heuristic, through numerical and statistical to ana- lytical methods applied in the effort to formally describe and analyse the two biological processes. We notice however, that although applied to cer- tain case studies, the presented methods are not limited to them and can be utilized in the analysis of other biological mechanisms as well as com- plex systems in general. The full range of developed and applied modelling techniques as well as model analysis methodologies constitutes a rich mod- elling framework. Moreover, the presentation of the developed methods, their application to the two case studies and the discussions concerning their potentials and limitations point to the difficulties and challenges one encounters in computational modelling of biological systems. The problems of model identifiability, model comparison, model refinement, model inte- gration and extension, choice of the proper modelling framework and level of abstraction, or the choice of the proper scope of the model run through this thesis.

My research deals with agent nouns in the language of the works of Mikael Agricola (ca. 1510–1557). The main tasks addressed in my thesis have been to describe individual agent noun types, to provide a comprehensive picture of the category of agent nouns and to clarify the relations between different types of agent nouns. My research material consists of all the agent nouns referring to persons in the language of Agricola’s works, together with their context. The language studied is for the most part translated language. Agent nouns play an important role both in the vocabulary of natural language and in broader sentence structures, since in a text it is constantly necessary to refer to actors re-ferring to persons in the text. As a concept and a phenomenon, the agent noun is widely known in languages. It is a word formed with a certain derivational affixes, which typical-ly refers to a person. In my research the agent noun category includes both deverbal and denominal derivatives referring to persons, e.g. kirjoittaa > kirjoittaja (to write > writer), asua > asuva (to inhabit > inhabitant), imeä > imeväinen (to suck > suckling), juopua > juopunut (to drink > drunkard), pelätä > pelkuri (to fear > one who fears ‘a coward’), apu > apulainen (help/to help > helper); lammas > lampuri (sheep > shepherd). Besides original Finnish expressions, agent noun derivatives taken as such from foreign languages form a word group of central importance for the research (e.g. nikkari, porvari, ryöväri, based on the German/Swedish for carpenter, burgher, robber). Especially important for the formation of agent nouns in Finnish are the models offered by foreign languages. The starting point for my work is predominantly semantic, as both the criteria for collecting the material and the categorisation underlying the analysis of the material are based on semantic criteria. When examining derivatives, aspects relating to structure are also inevitably of central importance, as form and meaning are closely associated with each other in this type of vocabulary. The alliance of structure and meaning can be described in an illustrative manner with the help of structural schemata. The examination of agent nouns comprises on the one hand analysis of syntactic elements and on the other, study of cultural words in their most typical form. The latter aspect offers a research object in which language and the extralinguistic world, referents, their designations and cultural-historical reality are in concrete terms one and the same. Thus both the agent noun types that follow the word formation principles of the Finn-ish language and those of foreign origin borrowed as a whole into Finnish illustrate very well how an expression of a certain origin and formed according to a certain structural model is inseparably bound up with the background of its referent and in general with semantic factors. This becomes evident both on the level of the connection between cer-tain linguistic features and text genre and in relation to cultural words referring to per-sons. For example, the model for the designations of God based on agent nouns goes back thousands of years and is still closely linked in 16th century literature with certain text genres. This brings out the link between the linguistic feature and the genre in a very con-crete manner. A good example of the connection between language and the extralinguistic world is provided by the cultural vocabulary referring to persons. Originally Finnish agent noun derivatives are associated with an agrarian society, while the vocabulary relat-ing to mediaeval urbanisation, the Hansa trade and specialisation by trade or profession is borrowed and originates in its entirety from vocabulary that was originally German.

Realizou-se a classificação morfofuncional de plântulas de 122 espécies arbóreas da Estação Ecológica do Panga. Espécies de três formações florestais contíguas - mata de galeria, mata mesófila semidecídua e cerradão - tiveram suas plântulas classificadas segundo suas características cotiledonares de posição, textura e exposição. O estudo objetivou verificar se as espécies estudadas enquadravam-se no sistema de classificação utilizado e se existiria alguma relação entre os tipos morfofuncionais de plântulas com grupos sucessionais, síndrome de dispersão, peso das sementes, micro-habitat, sistemas sexuais, épocas de frutificação e relação entre a biomassa de raízes e partes aéreas. As espécies foram incluídas em cinco categorias, sendo 63 espécies do tipo fanero-epígeo-foliáceo (PEF), 20 fanero-epígeo-armazenador (PER), 10 fanero-hipógeo-armazenador (PHR), 28 cripto-hipógeo-armazenador (CHR) e apenas uma espécie do tipo cripto-epígeo-armazenador (CER). A classificação morfofuncional mostrou-se eficaz, representando praticamente todas as espécies. Não houve relações significativas dos grupos morfofuncionais com a síndrome de dispersão, época de frutificação, biomassa raiz/parte aérea ou local de ocorrência das espécies estudadas. Por outro lado, foi possível relacionar os grupos sucessionais, peso das sementes e os sistemas sexuais aos grupos morfofuncionais. Espécies pioneiras, em sua maioria com sementes pequenas e leves apresentaram quase exclusivamente plântulas PEF, enquanto espécies climácicas tolerantes à sombra, usualmente com um menor número de sementes maiores e com maior quantidade de reservas, apresentaram predominantemente plântulas CHR. Este tipo também foi predominante entre espécies dióicas. Os resultados demonstraram que, pelo menos no estágio inicial, a morfologia desempenha funções determinantes nos processos de desenvolvimento e estabelecimento das plântulas de espécies arbóreas.

Cholesterol (Chol) is an important lipid in cellular membranes functioning both as a membrane fluidity regulator, permeability regulator and co-factor for some membrane proteins, e.g. G-protein coupled receptors. It also participates in the formation of signaling platforms and gives the membrane more mechanical strenght to prevent osmotic lysis of the cell. The sterol structure is very conserved and already minor structural modifications can completely abolish its membrane functions. The right interaction with adjacent lipids and the preference of certain lipid structures over others are also key factors in determining the membrane properties of cholesterol. Because of the many important properties of cholesterol it is of value to understand the forces and structural properties that govern the membrane behavior of this sterol. In this thesis we have used established fluorescence spectroscopy methods to study the membrane behavior of both cholesterol and some of its 3β-modified analogs. Using several fluorescent probes we have established how the acyl chain order of the two main lipid species, sphingomyelin (SM) and phosphatidylcholine (PC) affect sterol partitioning as well as characterized the membrane properties of 3β-aminocholesterol and cholesteryl phosphocholine. We concluded that cholesterol prefers SM over PC at equal acyl chain order, indicating that other structural properties besides the acyl chain order are important for sphingomyelin-sterol interactions. A positive charge at the 3β position only caused minor changes in the sterol membrane behavior compared to cholesterol. A large phosphocholine head group caused a disruption in membrane packing together with other membrane lipids with large head groups, but was also able to form stable fluid bilayers together with ceramide and cholesterol. The Ability of the large head group sterol to form bilayers together with ceramide was further explored in the last paper where cholesteryl phosphocholine/ceramide (Chol-PC/Cer) complexes were successfully used to transfer ceramide into cultured cells.

Nous présentons dans cette thèse des théorèmes d’existence pour des systèmes d’équations différentielles non-linéaires d’ordre trois, pour des systèmes d’équa- tions et d’inclusions aux échelles de temps non-linéaires d’ordre un et pour des systèmes d’équations aux échelles de temps non-linéaires d’ordre deux sous cer- taines conditions aux limites. Dans le chapitre trois, nous introduirons une notion de tube-solution pour obtenir des théorèmes d’existence pour des systèmes d’équations différentielles du troisième ordre. Cette nouvelle notion généralise aux systèmes les notions de sous- et sur-solutions pour le problème aux limites de l’équation différentielle du troisième ordre étudiée dans [34]. Dans la dernière section de ce chapitre, nous traitons les systèmes d’ordre trois lorsque f est soumise à une condition de crois- sance de type Wintner-Nagumo. Pour admettre l’existence de solutions d’un tel système, nous aurons recours à la théorie des inclusions différentielles. Ce résultat d’existence généralise de diverses façons un théorème de Grossinho et Minhós [34]. Le chapitre suivant porte sur l’existence de solutions pour deux types de sys- tèmes d’équations aux échelles de temps du premier ordre. Les résultats d’exis- tence pour ces deux problèmes ont été obtenus grâce à des notions de tube-solution adaptées à ces systèmes. Le premier théorème généralise entre autre aux systèmes et à une échelle de temps quelconque, un résultat obtenu pour des équations aux différences finies par Mawhin et Bereanu [9]. Ce résultat permet également d’obte- nir l’existence de solutions pour de nouveaux systèmes dont on ne pouvait obtenir l’existence en utilisant le résultat de Dai et Tisdell [17]. Le deuxième théorème de ce chapitre généralise quant à lui, sous certaines conditions, des résultats de [60]. Le chapitre cinq aborde un nouveau théorème d’existence pour un système d’in- clusions aux échelles de temps du premier ordre. Selon nos recherches, aucun résultat avant celui-ci ne traitait de l’existence de solutions pour des systèmes d’inclusions de ce type. Ainsi, ce chapitre ouvre de nouvelles possibilités dans le domaine des inclusions aux échelles de temps. Notre résultat a été obtenu encore une fois à l’aide d’une hypothèse de tube-solution adaptée au problème. Au chapitre six, nous traitons l’existence de solutions pour des systèmes d’équations aux échelles de temps d’ordre deux. Le premier théorème d’existence que nous obtenons généralise les résultats de [36] étant donné que l’hypothèse que ces auteurs utilisent pour faire la majoration a priori est un cas particulier de notre hypothèse de tube-solution pour ce type de systèmes. Notons également que notre définition de tube-solution généralise aux systèmes les notions de sous- et sur-solutions introduites pour les équations d’ordre deux par [4] et [55]. Ainsi, nous généralisons également des résultats obtenus pour des équations aux échelles de temps d’ordre deux. Finalement, nous proposons un nouveau résultat d’exis- tence pour un système dont le membre droit des équations dépend de la ∆-dérivée de la fonction.

L’apoptose est une forme de mort cellulaire essentielle au développement et au maintien de l’homéostase chez les animaux multicellulaires. La machinerie apoptotiq ue requiert la participation des caspases, des protéases conservées dans l’évolution et celle des organelles cytoplasmiques. Les lysosomes subissent des ruptures partielles, labilisation de la membrane lysosomale (LML), qui entraînent l’activation des cathepsines dans le cytoplasme de cellules cancéreuses humaines en apoptose induite par la camptothecin (CPT), incluant les histiocytes humains U-937. Ces modifications lysosomales se manifestent tôt durant l’activation de l’apoptose, concomitamment avec la perméabilisation de la mitochondrie et l’activation des caspases. Une étude protéomique quantitative et comparative a permis d’identifier des changements précoces dans l’expression/localisation de protéines lysosomales de cellules U-937 en apoptose. Lors de deux expériences indépendantes, sur plus de 538 protéines lysosomales identifiées et quantifiées grâce au marquage isobarique iTRAQ et LC-ESIMS/ MS, 18 protéines augmentent et 9 diminuent dans les lysosomes purifiés de cellules en cours d’apoptose comparativement aux cellules contrôles. Les candidats validés par immuno-buvardage et microscopie confocale incluent le stérol-4-alpha-carboxylate 3- déhydrogénase, le prosaposin et la protéine kinase C delta (PKC-d). Des expériences fonctionnelles ont démontrées que la translocation de PKC-d aux lysosomes est requise pour la LML puisque la réduction de son expression par ARN interférents ou l’inhibition de son activité à l’aide du rottlerin empêche la LML lors de l’apoptose induite par la CPT. La translocation de PKC-d aux lysosomes conduit à la phosphorylation et l’activation de la sphingomyelinase acide lysosomale (ASM), et à l’accroissement subséquent du contenu en céramide (CER) à la membrane lysosomale. Cette accumulation de CER endogène aux lysosomes est un évènement critique pour la LML induite par la CPT car l’inhibition de l’activité de PKC-d ou de ASM diminue la formation de CER et la LML.Ces résultats révèlent un nouveau mécanisme par lequel la PKC-d active l’ASM qui conduit à son tour à l’accumulation de CER à la membrane lysosomale et déclenche la LML et l’activation de la voie lysosomale de l’apoptose induite par la CPT. En somme, ce mécanisme confirme l’importance du métabolisme des sphingolipides dans l’activation de la voie lysosomale de l’apoptose.
