Familial Systemic Mastocytosis With Germline Kit K509i Mutation Is Sensitive To Treatment With Imatinib, Dasatinib And Pkc412.

Mastocytosis are myeloproliferative neoplasms commonly related to gain-of-function mutations involving the tyrosine kinase domain of KIT. We herein report a case of familial systemic mastocytosis with the rare KIT K509I germ line mutation affecting two family members: mother and daughter. In vitro treatment with imatinib, dasatinib and PKC412 reduced cell viability of primary mast cells harboring KIT K509I mutation. However, imatinib was more effective in inducing apoptosis of neoplastic mast cells. Both patients with familial systemic mastocytosis had remarkable hematological and skin improvement after three months of imatinib treatment, suggesting that it may be an effective front line therapy for patients harboring KIT K509I mutation.

Expression of constitutively activated human c-Kit in myb transformed early myeloid cells leads to factor independence, histiocytic differentiation, and tumorigenicity

The cDNAs encoding wild type (WT) human receptor tyrosine kinase c-Kit and a constitutively activated mutant, V816Kit, were introduced into granulocyte-macrophage colony-stimulating factor (GM-CSF)-dependent early murine hemopoietic cells, which had been transformed with activated Myb, WTKit cells were able to grow in the presence of the human ligand for Kit, stem cell factor (SCF), but displayed reduced growth and clonogenic potential in either SCF or GM-CSF compared with the parental cells in GM-CSF. In contrast, V816Kit cells grew without factor at a higher rate than the parental cells in GM-CSF and displayed increased clonogenicity. Dissection of the growth characteristics in liquid culture showed that in the presence of appropriate factors, the different populations had similar proliferation rates, but that V816Kit profoundly increased cell survival compared with WTKit or parental cells, This suggests that the signals transduced by WTKit activated with SCF, and by V816Kit, were not identical. Also, WTKit and V816Kit-expressing cells both varied from the early myeloid progenitor phenotype of the parental cells and gave rise to a small number of large to giant adherent cells that expressed macrophage (alpha-naphthyl acetate) esterase and neutrophil (naphtol-AS-D-chloroacetate) esterase, were highly phagocytic and phenotypically resembled histiocytes. Thus, WTKit activated by SCF and V816Kit were able to induce differentiation in a proportion of Myb-transformed myeloid cells. The factor independent V816Kit cells, unlike the parental and WTKit expressing cells, were shown to produce tumors of highly mitotic, invasive cells at various stages of differentiation in syngeneic mice. These results imply that constitutively activated Kit can promote the development of differentiated myeloid tumors and that its oncogenic effects are not restricted to lineages (mast cell and B-cell acute lymphoblastic leukemia), which have been reported previously. Furthermore, the mixed populations of cells in culture and in the tumors phenotypically resembled the leukemic cells from patients with monocytic leukemia with histiocytic differentiation (acute myeloid leukemia-M5c), a newly proposed subtype of myeloid leukemia. Inactivation of a c-Myb/estrogen receptor fusion protein in transformed primary cells leads to granulocyte/macrophage differentiation and down regulation of c-kit but not c-myc or cdc2

Primary murine fetal hemopoietic cells were transformed with a fusion protein consisting of the ligand-binding domain of the estrogen receptor and a carboxyl-terminally truncated c-Myb protein (ERMYB), The ERMYB-transformed hemopoietic cells exhibit an immature myeloid phenotype when grown in the presence of beta-estradiol. Upon removal of beta-estradiol, the ERMYB cells display increased adherence, decreased clonogenicity and differentiate to cells exhibiting granulocyte or macrophage morphology, The expression of the c-myc, c-kit, cdc2 and bcl-2 genes, which are putatively regulated by Myb, was investigated in ERMYB cells grown in the presence or absence of beta-estradiol. Neither c-myc nor cdc2 expression was down-regulated after removal of beta-estradiol demonstrating that differentiation is not a consequence of decreased transactivation of these genes by ERMYB. While bcl-2 expression was reduced by 50% in ERMYB cells grown in the absence of beta-estradiol, there was no increase in DNA laddering, suggesting that Myb was not protecting ERMYB cells from apoptosis, In contrast, a substantial (200-fold) decrease in c-kit mRNA level was observed following differentiation of ERMYB cells, and c-kit mRNA could be partially re-induced by the re-addition of beta-estradiol. Furthermore, a reporter construct containing the c-kit promoter was activated when cotransfected with a Myb expression vector, providing further evidence of a role for Myb in the regulation of c-kit.

Signals from Eph and ephrin proteins: a developmental tool kit

Interactions between Eph receptors and their ligands the ephrin proteins are critically important in many key developmental processes. Emerging evidence also supports a role for these molecules in postembryonic tissues, particularly in pathological processes, including tissue injury and tumor metastasis. We review the signaling mechanisms that allow the 14 Eph and nine ephrin proteins to deliver intracellular signals that regulate cell shape and movement. What emerges is that the initiation of these signals is critically dependent on which Eph and ephrin proteins are expressed, the level of their expression, and, in some cases, which splice variants are expressed. Diversity at the level of initial interaction and in the downstream signaling processes regulated by Eph-ephrin signaling provides a subtle, versatile system of regulation of intercellular adhesion, cell shape, and cell motility.

An overview of drugs and ancillary equipment for the dentist's emergency kit

The concept of a basic (i.e., essential) medical emergency kit suitable for a general dental practitioner varies somewhat between different authorities. A practitioner's choice is also dependant on the proximity of medical aid and the nature of the dental practice. Over recent years the trend has been to restrict the items to a minimum, in the interest of both common sense and safety, for example, just oxygen, adrenaline 1:1000, an oral carbohydrate source, glyceryl trinitrate and aspirin as first options. Ancillary equipment should include an oxygen therapy facemask, a pocket mask and a set of oral (Guedel) airways. Two further medication options for consideration are an aerosol bronchodilator and, in certain circumstances, an injectable antihypoglycaemic agent. This paper provides a selective overview of the subject. An absolute necessity is for dentists to be competent in Basic Life Support skills, and to maintain a complete and current medical history for all patients.

Assentar a primeira pedra

A presente dissertação tem como objectivo o estudo da primeira fase da institucionalização da investigação geológica em Portugal. São analisados os primeiros organismos responsáveis pelo levantamento geológico do território de Portugal continental, ou seja, a Comissão Geológica e Mineralógica, criada em 1848 no contexto da Academia Real das Ciências de Lisboa e chefiada pelo Engenheiro francês Charles Bonnet (1816-1867), e a Comissão Geológica do Reino, fundada em 1857 como secção dos Serviços Geodésicos, no quadro do Ministério das Obras Públicas Comércio e Indústria, liderada por Carlos Ribeiro (1813-1882) e Pereira da Costa (1809-1888). Sem ser efectuada uma análise exaustiva, são ainda abordadas as reformas implementadas depois da cessação de funções da Comissão Geológica do Reino, por se considerar a sua menção indispensável para a compreensão das consequências dos acontecimentos que conduziram à suspensão desta instituição. Neste estudo deu-se particular relevo aos objectivos, estrutura, organização e enquadramento institucional das Comissões Geológicas, analisando-se as relações estabelecidas com os organismos que as tutelavam e os factores que condicionaram o seu funcionamento, em especial aqueles que levaram à suspensão da Comissão Geológica e Mineralógica, em 1858, e à extinção da Comissão Geológica do Reino, dez anos depois. No que se refere à investigação da estrutura institucional das Comissões Geológicas, foi utilizada uma abordagem proposta por Bruno Latour no quadro dos “science studies”, por parecer a mais adequada à compreensão da estrutura e organização das duas Comissões, e do seu enquadramento na administração do Estado. Estas linhas orientadoras foram ainda complementadas com uma análise das intenções e motivações individuais dos protagonistas envolvidos, por terem sido consideradas determinantes no esclarecimento das razões que conduziram à dissolução da Comissão Geológica do Reino. No que se refere às actividades desenvolvidas por ambas as Comissões, serão analisados os respectivos planos e métodos de trabalho, os resultados da sua acção, as relações internacionais dos seus membros, bem como as suas contribuições científicas tendo em conta os padrões internacionais de prática geológica da época. Neste âmbito, demonstra-se que a Comissão Geológica do Reino produziu mais e melhores resultados, que se constituíram num “ciclo de acumulação”. Para este processo, foi determinante a criação de uma rede de apoio, sobretudo no estrangeiro, que garantiu não só a aquisição de colecções, mapas, periódicos e livros, mas também a circulação de conhecimento e o intercâmbio com instituições congéneres estrangeiras, o que permitiu aos geólogos da CGR ver o seu trabalho reconhecido na comunidade científica internacional.
