Neyraiella distinctus n. sp. was found parasitizing nymphs of the cricket Gryllodes laplatae Sauss in City Bell, Argentina. This species was characterized by having the excretory pore in the posterior end of the basal bulb, vulva protruding with one lip well developed in the 1/3 end of the body, anus of the female with wings, male with a single spicule without any sculpture, gubernaculum and bursa are absent, six pairs of genital papillae arranged in two preanal pairs, one adanal pair and three postanal pairs, and the tail appendage in both sexes was short and conic.

A new genus and a new species of Heligmonellidae nematodes are described parasiting the stomach of three agoutis (two Dasyprocta fuliginosa and one D. leporina) captured in the middle and high Negro river microregion, state of Amazonas, Brazil. The new genus, as well as its type-species, are closely related to the trichostrongylids included in Fuellebornema, particularly on what concerns the pattern of the caudal bursa, but differing from them by the characteristics of the synlophe, that presents a poorly developed carene, when compared to the referred number of body ridges in Freitastrongylus n. gen. and consequently in F. angelae n. sp.,in which the ridges are well developed and the carene at mid-body has a similar size when compared to the ridge situated in front of the right field (ridge no. 5). Caudal bursa is of the type 1-4, with rays 9 shorter than rays 10, with a very long genital cone.

Angiostrongylus cantonensis is the etiologic agent of eosinophilic meningoencephalitis in humans. Cases have been recorded in many parts of the world, including Brazil. The aim of this study was to compare the differences in the biology and morphology of two different Brazilian haplotypes of A. : ac8 and ac9. A significantly larger number of L1 larvae eliminated in the faeces of rodents at the beginning of the patent period was observed for ac9 haplotype and compared to the total of L1 larvae eliminated, there was a significant difference between the two haplotypes. The ac9 haplotype showed a significant difference in the proportion of female and male specimens (0.6:1), but the same was not observed for ac8 (1.2:1). The morphometric analysis showed that male and female specimens isolated from ac8 haplotype were significantly larger with respect to body length, oesophagus length, spicule length (male) and distance from the anus to the rear end (female) compared to specimens from ac9. The morphological analysis by light microscopy showed little variation in the level of bifurcations at the lateral rays in the right lobe of the copulatory bursa between the two haplotypes. The biological, morphological and morphometric variations observed between the two haplotypes agree with the observed variation at the molecular level using the cytochrome oxidase subunit I marker and reinforce the possible influence of geographical isolation on the development of these haplotypes.

Members of the tumor necrosis factor (TNF) family play key roles in the regulation of inflammation, immune responses and tissue homeostasis. Here we describe the identification of the chicken homologue of mammalian B cell activating factor of the TNF family (BAFF/BLyS). By searching a chicken EST database we identified two overlapping cDNA clones that code for the entire open reading frame of chicken BAFF (chBAFF), which contains a predicted transmembrane domain and a putative furin protease cleavage site like its mammalian counterparts. The amino acid identity between soluble chicken and human BAFF is 76%, considerably higher than for most other known cytokines. The chBAFF gene is most strongly expressed in the bursa of Fabricius. Soluble recombinant chBAFF produced by human 293T cells interacted with the mammalian cell-surface receptors TACI, BCMA and BAFF-R. It bound to chicken B cells, but not to other lymphocytes, and it promoted the survival of splenic chicken B cells in culture. Furthermore, bacterially expressed chBAFF induced the selective expansion of B cells in the spleen and cecal tonsils when administered to young chicks. Our results suggest that like its mammalian counterpart, chBAFF plays an important role in survival and/or proliferation of chicken B cells.

The chicken represents the best-characterized animal model for B cell development in the so-called gut-associated lymphoid tissue (GALT) and the molecular processes leading to B cell receptor diversification in this species are well investigated. However, the mechanisms regulating B cell development and homeostasis in GALT species are largely unknown. Here we investigate the role played by the avian homologue of B cell-activating factor of the tumor necrosis factor family (BAFF). Flow cytometric analysis showed that the receptor for chicken B cell-activating factor of the tumor necrosis factor family (chBAFF) is expressed by mature and immature B cells. Unlike murine and human BAFF, chBAFF is primarily produced by B cells both in peripheral lymphoid organs and in the bursa of Fabricius, the chicken's unique primary lymphoid organ. In vitro and in vivo studies revealed that chBAFF is required for mature B cell survival. In addition, in vivo neutralization with a decoy receptor led to a reduction of the size and number of B cell follicles in the bursa, demonstrating that, in contrast to humans and mice, in chickens BAFF is also required for the development of immature B cells. Collectively, we show that chBAFF has phylogenetically conserved functions in mature B cell homeostasis but displays unique and thus far unknown properties in the regulation of B cell development in birds.

The brown lacewings (Neuroptera, Hemerobiidae) of northwestern Turkey with new records, their spatio-temporal distribution and harbouring plants. The occurrence and spatio-temporal distribution of brown lacewing species (Neuroptera, Hemerobiidae) in Bursa province, northwestern Turkey, was investigated during 1999-2011. A total of 852 brown lacewing specimens of 20 species, including the genera of Hemerobius, Megalomus, Micromus, Sympherobius, and Wesmaelius were collected. Of these, 12 species were new records for northwestern Turkey while Sympherobius klapaleki is a new record for the Neuroptera fauna of Turkey. The most widespread species were Hemerobius handschini and Sympherobius pygmaeus with percent dominance values of 42.00 and 15.96%, respectively. Wesmaelius subnebulosus was the earliest emerging hemerobiid species and had the longest flight activity lasting from March to October. The species of southern origin characterized by the Mediterranean elements constituted 55% of the hemerobiid fauna and prevailed over the species of northern origin that belong to the Siberian centres. The total number of hemerobiid species reached a peak in July with captures of 15 species per month. There were 11, 13, 18 and 5 hemerobiid species occurring at altitudes between 1-500, 501-1000, 1001-1500 and 1500-2000 m, respectively. In addition, plant species harbouring hemerobiids are given for each species, and their association with the hemerobiid fauna is discussed.

Se estudió la bioacumulación de cobre total en organismos bentónicos como, moluscos y crustáceos, en la bahía de Huarmey y se relacionó con las acciones naturales y antrópicas que ocurren en ella. El objetivo fue conocer qué organismo bentónico bioacumula cobre total en mayor cantidad para justificar su propuesta como organismo centinela de éste metal. Tomando como base las capturas de organismos bentónicos realizadas por buceo en el periodo 2006 al 2012, complementadas con muestras de sedimento marino, sedimento de río y agua de río se determinó cobre total mediante espectrofotometría de absorción atómica en horno de grafito. Las mayores concentraciones media de cobre total se encontraron en crustáceos y moluscos, como el “cangrejo peludo,” Cancer setosus (MOLINA, 1782) (110,70 μg/g), el “caracol negro,” Stramonita chocolata (DUCLOS, 1832) (108,79 μg/g), la “lapa,” Fisurella crassa (LAMARCK, 1822) (99,52 μg/g), el “caracol turbante,” Tegula atra (LESSON, 1830) (88,27 μg/g), el “caracol rosado,” Bursa ventricosa (BRODERIP, 1833) (78,54 μg/g) y el“cangrejo violáceo,” Platyxanthus orbignyi (H. MILNE EDWARS Y LUCAS, 1843) (46,30 μg/g). La concentración de cobre total en el caracol negro se relaciona con las concentraciones de cobre total en el sedimento marino, en el sedimento de río y en el agua de río. La ecuación de regresión lineal múltiple evidenció dicha asociación (R=0,998).

Knowledge of the role of origin-related, environmental, sex, and age factors on host defence mechanisms is important to understand variation in parasite intensity. Because alternative components of parasite defence may be differently sensitive to various factors, they may not necessarily covary. Many components should therefore be considered to tackle the evolution of host-parasite interactions. In a population of barn owls (Tyto alba), we investigated the role of origin-related, environmental (i.e. year, season, nest of rearing, and body condition), sex, and age factors on 12 traits linked to immune responses [humoral immune responses towards sheep red blood cells (SRBC), human serum albumin (HSA) and toxoid toxin TT, T-cell mediated immune response towards the mitogen phytohemagglutinin (PHA)], susceptibility to ectoparasites (number and fecundity of Carnus haemapterus, number of Ixodes ricinus), and disease symptoms (size of the bursa of Fabricius and spleen, proportion of proteins that are immunoglobulins, haematocrit and blood concentration in leucocytes). Cross-fostering experiments allowed us to detect a heritable component of variation in only four out of nine immune and parasitic parameters (i.e. SRBC- and HSA-responses, haematocrit, and number of C. haemapterus). However, because nestlings were not always cross-fostered just after hatching, the finding that 44% of the immune and parasitic parameters were heritable is probably an overestimation. These experiments also showed that five out of these nine parameters were sensitive to the nest environment (i.e. SRBC- and PHA-responses, number of C. haemapterus, haematocrit and blood concentration in leucocytes). Female nestlings were more infested by the blood-sucking fly C. haemapterus than their male nestmates, and their blood was less concentrated in leucocytes. The effect of year, season, age (i.e. reflecting the degree of maturation of the immune system), brood size, position in the within-brood age hierarchy, and body mass strongly differed between the 12 parameters. Different components of host defence mechanisms are therefore not equally heritable and sensitive to environmental, sex, and age factors, potentially explaining why most of these components did not covary.

Ha sido evaluado el efecto de benzoilfenylureia sobre la capacidad copuladora de machos de Spodoptera littoralis (Boisd.) (Lepdoptera: Noctuidae). Dos grupos de larvas de 3º estadio fueron tratados con el flufenoxuron, uno por ingestión y el otro por contacto, con una concentración del producto igual a la DL60. Cuando hubo la emergencia de los adultos, 10 machos por tratamiento fueron individualizados en cilindros de papel de filtro y puestos a copular con hembras vírgenes, que eron sustituídas diariamente, hasta la muerte de las mismas. Las hembras que fueron ofrecidas a los machos, al morir, eron dissecadas para verificar se estaban o no copuladas, a través de la observación de la presencia de espermatóforos en la bursa copulatrix. Fue observado que los machos de S. littoralis provenientes de larvas tratadas con el flufenoxuron, no tuvieron su capacidad copuladora alterada. La metodología fue considerada adecuada para evaluar la atividad copuladora y el número de copulaciones realizadas por el macho de esta especie.

Many eukaryote organisms are polyploid. However, despite their importance, evolutionary inference of polyploid origins and modes of inheritance has been limited by a need for analyses of allele segregation at multiple loci using crosses. The increasing availability of sequence data for nonmodel species now allows the application of established approaches for the analysis of genomic data in polyploids. Here, we ask whether approximate Bayesian computation (ABC), applied to realistic traditional and next-generation sequence data, allows correct inference of the evolutionary and demographic history of polyploids. Using simulations, we evaluate the robustness of evolutionary inference by ABC for tetraploid species as a function of the number of individuals and loci sampled, and the presence or absence of an outgroup. We find that ABC adequately retrieves the recent evolutionary history of polyploid species on the basis of both old and new sequencing technologies. The application of ABC to sequence data from diploid and polyploid species of the plant genus Capsella confirms its utility. Our analysis strongly supports an allopolyploid origin of C. bursa-pastoris about 80 000 years ago. This conclusion runs contrary to previous findings based on the same data set but using an alternative approach and is in agreement with recent findings based on whole-genome sequencing. Our results indicate that ABC is a promising and powerful method for revealing the evolution of polyploid species, without the need to attribute alleles to a homeologous chromosome pair. The approach can readily be extended to more complex scenarios involving higher ploidy levels.

Synovial chondromatosis is a benign condition characterized by synovial proliferation and metaplasia, with development of cartilaginous or osteocartilaginous nodules within a joint, bursa or tendon sheath. In the shoulder, synovial osteochondromatosis may occur within the glenohumeral joint and its recesses (including the tendon sheath of the biceps long head), and in the subacromial-deltoid bursa. Such condition can be identified either by radiography, ultrasonography or magnetic resonance imaging, showing typical features according to each method. Radiography commonly shows ring-shaped calcified cartilages and periarticular soft tissues swelling with erosion of joint margins. Ultrasonography demonstrates hypoechogenic cartilaginous nodules with progressive increase in echogenicity as they become calcified, with development of posterior acoustic shadow in case of ossification. Besides identifying cartilaginous nodules, magnetic resonance imaging can also demonstrate the degree of synovial proliferation. The present study is aimed at describing the imaging findings of this entity in the shoulder.

Biology is turning into an information science. The science of systems biology seeks to understand the genetic networks that govern organism development and functions. In this study the chicken was used as a model organism in the study of B cell regulatory factors. These studies open new avenues for plasma cell research by connecting the down regulation of the B cell gene expression program directly to the initiation of plasma cell differentiation. The unique advantages of the DT40 avian B cell model system, specifically its high homologous recombination rate, were utilized to study gene regulation in Pax5 knock out cell lines and to gain new insights into the B cell to plasma cell transitions that underlie the secretion of antibodies as part of the adaptive immune response. The Pax5 transcription factor is central to the commitment, development and maintenance of the B cell phenotype. Mice lacking the Pax5 gene have an arrest in development at the pro-B lymphocyte stage while DT40 cells have been derived from cells at a more mature stage of development. The DT40 Pax5-/- cells exhibited gene expression similarities with primary chicken plasma cells. The expression of the plasma cell transcription factors Blimp-1 and XBP-1 were significantly upregulated while the expression of the germinal centre factor BCL6 was diminished in Pax5-/- cells, and this alteration was normalized by Pax5 re-introduction. The Pax5-deficient cells further manifested substantially elevated secretion of IgM into the supernatant, another characteristic of plasma cells. These results for the first time indicated that the downregulation of the Pax5 gene in B cells promotes plasma cell differentiation. Cross-species meta-analysis of chicken and mouse Pax5 gene knockout studies uncovers genes and pathways whose regulatory relationship to Pax5 has remained unchanged for over 300 million years. Restriction of the hematopoietic stem cell fate to produce T, B and NK cell lineages is dependent on the Ikaros and its molecular partners, the closely related Helios and Aiolos. Ikaros family members are zinc finger proteins which act as transcriptional repressors while helping to activate lymphoid genes. Helios in mice is expressed from the hematopoietic stem cell level onwards, although later in development its expression seems to predominate in the T cell lineage. This study establishes the emergence and sequence of the chicken Ikaros family members. Helios expression in the bursa of Fabricius, germinal centres and B cell lines suggested a role for Helios in the avian B-cell lineage, too. Phylogenetic studies of the Ikaros family connect the expansion of the Ikaros family, and thus possibly the emergence of the adaptive immune system, with the second round of genome duplications originally proposed by Ohno. Paralogs that have arisen as a result of genome-wide duplications are sometimes termed ohnologs – Ikaros family proteins appear to fit that definition. This study highlighted the opportunities afforded by the genome sequencing efforts and somatic cell reverse genetics approaches using the DT40 cell line. The DT40 cell line and the avian model system promise to remain a fruitful model for mechanistic insight in the post-genomic era as well.

O objetivo deste estudo foi avaliar as alterações do aparato podotroclear de 22 cavalos da raça Quarto de Milha acometidos pela síndrome do navicular utilizando o acesso transcuneal. Esses equinos foram submetidos ao exame radiográfico e ultrassonográfico. No exame radiográfico foi observado a presença de invaginação sinovial de diferentes tipos, além de calcificação da bursa podotroclear e tendão flexor digital profundo, entesiófitos, osteófitos e alteração do limite córtico-medular. No exame ultrassonográfico observou-se alterações da bursa podotroclear, aderência e irregularidades nos bordos do tendão flexor digital profundo, diminuição do coxim digital, calcificação do ligamento sesamóide distal ímpar e irregularidade da superfície flexora do osso sesamóide distal. Todos os equinos que apresentavam alterações radiográficas também apresentaram alterações no exame ultrassonográfico compatíveis com a síndrome do navicular. O exame ultrassonográfico utilizando o acesso transcuneal foi um método prático e eficiente para avaliação das lesões do aparato podotroclear dos eqüinos, sendo um método complementar ao exame radiográfico.
