Whipping cream, skim milk powder and soft cheese were produced throughout the year. Whipping cream manufactured in spring and winter produced significantly higher overrun and better serum stability, and whipping time was related to buffering capacity of raw milk. Heat stability of reconstituted skim milk powder (RSMP) at 9% TS was greater in summer and autumn, and greater than 25% TS throughout the year. It was positively related to the protein content of raw milk, but negatively with fat. In contrast to other dairy products, no significant effect of season on the properties of soft cheese was found.

The isoforms of the Na+/H+ exchanger present in T84 human colon cells were identified by functional and molecular methods. Cell pH was measured by fluorescence microscopy using the probe BCECF. Based on the pH recovery after an ammonium pulse and determination of buffering capacity of these cells, the rate of H+ extrusion (J(H)) was 3.68 mM/min. After the use of the amiloride derivative HOE-694 at 25 mu M, which inhibits the isoforms NHE1 and NHE2, there remained 43% of the above transport rate, the nature of which was investigated. Evidence of the presence of NHE1, NHE2, and NHE4 was obtained by reverse transcriptase polymerase chain reaction (RT-PCR) (mRNA) and Western blot. There was no decrease of J(H) by the NHE3 inhibitor S3226 (1 mu M) and no evidence of this isoform by RT-PCR was found. The following functional evidence for the presence of NHE4 was obtained: 25 mu M EIPA abolished J(H) entirely, but NHE4 was not inhibited at 10 mu M; substitution of Na by K increased the remainder, a property of NHE4; hypertonicity also increased this fraction of J(H). Cl--dependent NHE was not detected: in 0 Cl- solutions J(H) was increased and not reduced. In 0 Cl- cell volume decreased significantly, which was abolished by the Cl- channel blocker NPPB, indicating that the 0 Cl- effect was because of reduction of cell volume. In conclusion, T84 human colon cells contain three isoforms of the Na+/H+ exchanger, NHE1, NHE2, and NHE4, but not the Cl-dependent NHE.

A capillary electrophoresis method for organic acids in wine was developed and validated. The optimal electrolyte consisted of 10 mmol/L 3,5-dinitrobenzoic acid (DNB) at pH 3.6 containing 0.2 mmol/L cetyltrimethylammonium bromide as flow reverser. DNB was chosen because it has an effective mobility similar to the organic acids under investigation, good buffering capacity at pH 3.6, and good chromophoric characteristics for indirect UV-absorbance detection at 254 nm. Sample preparation involved dilution and filtration. The method showed good performance characteristics: Linearity at 6 to 285 mg/L (r > 0.99); detection and quantification limits of 0.64 to 1.55 and 2.12 to 5.15 mg/L, respectively; separation time of less than 5.5 min. Coefficients of variation for ten injections were less than 5% and recoveries varied from 95% to 102%. Application to 23 samples of Brazilian wine confirmed good repeatability and demonstrated wide variation in the organic acid concentrations. (C) 2008 Elsevier Ltd. All rights reserved.

Neste trabalho, avaliou-se a fermentação da cana-de-açúcar queimada, ensilada com ou sem uso de aditivo seco. Os tratamentos (seis no total) consistiram da silagem de cana crua ou queimada, adicionada de 0, 50 ou 100 g/kg de milho desintegrado com palha e sabugo (MDPS), com base no peso verde da forragem. Foram determinados os teores de MS, PB, nitrogênio insolúvel em detergente ácido (NIDA), FDN, FDA, celulose, hemicelulose e lignina. Na avaliação das características fermentativas, foram determinados os valores de carboidratos solúveis, o poder tamponante, o pH e as concentrações de nitrogênio amoniacal e etanol. Como características microbiológicas, avaliou-se o desenvolvimento de leveduras. A inclusão de MDPS elevou os teores de MS e reduziu discretamente os teores de N-NH3 e etanol das silagens, não ocasionando efeito nos valores de pH e na população de leveduras. A presença do fogo reduziu a concentração de MS das silagens, elevou os teores de etanol e leveduras e diminuiu os teores de N-NH3. A fermentação etanólica durante a ensilagem não foi controlada com a inclusão de aditivo seco ou com o uso do fogo.

The aim of this study was to compare microbiological and salivary variables possibly related to caries risk in treated and untreated mouthbreathing syndrome (MBS) children and control children. Thirty control children, 30 mouthbreathers and 25 treated mouthbreathers were studied for the numbers of lactobacilli, mutans streptococci and yeasts in their saliva. Snyder's test, salivary flow and buffering capacity were also evaluated. Levels of immunoglobulins to Candida albicans and Streptococcus mutans in the saliva were quantified using ELISA. Considering the results obtained for the microbiological and salivary caries risk tests, no significant differences were observed among the proportions of patients with small/negative and high/moderate caries risk in the studied groups. The level of IgG to S. mutans was significantly higher in the treated MBS group in relation to MBS patients. on the other hand, the median anti-S. mutans IgM level was lower in the treated MBS patients than in the other groups. For the studied anti-Candida immunoglobulins, IgM level was significantly lower in the treated MBS group than in the other groups. No differences were observed for anti-S. mutans and anti-Candida IgA levels among the groups. The findings suggest that mouthbreathing cannot be considered a risk factor for dental caries. Statement of problem. The oral mucosa has been reported to show a variety of changes in subjects with diabetes mellitus.Purpose. The purpose of this study was to compare diabetic and nondiabetic subjects wearing complete dentures with regard to salivary flow, salivary buffering capacity, denture retention, and oral mucosal lesions.Material and methods. Sixty subjects, 30 with and 30 without a diagnosis of diabetes, were matched for gender, race, and age. Salivary flow, salivary buffering capacity, glycemia, blood pressure, presence of mucosal lesions, denture retention, use of medications, and behavioral factors (controlled or uncontrolled diet, alcohol consumption, and smoking) reported by the subjects, were evaluated. For the salivary buffering capacity test, 1 mL of saliva was pipetted into a test tube containing 3 mL 0.005 N of hydrochloric acid, and the pH was measured with indicator strips. Group differences were statistically analyzed using the Student t test and the Mann-Whitney test for quantitative variables and the chi-square test for qualitative variables (alpha = .05).Results. Mean (SD) salivary flow was 1.14 (0.87) mL/min in the nondiabetic subjects and 0.95 (0.61) mL/min in the diabetic subjects. Evaluation of self-reported denture retention revealed no significant difference between groups. Denture retention was observed in 66.7% (20/30) of the control group and in 50% (15/30) of the diabetic group. The prevalence of mucosal lesions was 90% (27/30) in the control group and 83.3% (25/30) in the diabetic group. Salivary buffering capacity was 5.80 (0.85) in the control group and 5.26 (0.83) in the diabetic group (P = .017).Conclusions. Within the limitations of this study, no significant differences were observed in salivary flow, denture retention, or oral lesions in diabetic and nondiabetic subjects.

Avaliaram-se as alterações da fração fibrosa e as características químicas dos fenos de braquiária decumbens (Brachiaria decumbnes Stapf) e jaraguá (Hyparrhenia rufa Ness Stapf) não-tratados, tratados com uréia (U-5,4% da MS), uréia (UL-5,4% da MS) mais labe-labe (Lablab purpureus L. Sweet, cv. Highworth-3,0% da MS) ou amônia anidra (NH3 -3,0% da MS). O tratamento químico com uréia ou NH3 aumentou o pH e a digestibilidade in vitro verdadeira dos fenos. A amonização não alterou os teores de fibra em detergente ácido e celulose, mas diminuiu os de fibra em detergente neutro, hemicelulose e lignina. O uso do labe-labe como fonte adicional de urease não aumentou a eficiência da uréia no tratamento dos volumosos. As avaliações do conteúdo de umidade, do poder tampão e da atividade ureática são técnicas que podem auxiliar na previsão das respostas dos volumosos à amonização com o uso de uréia.

Elephant grass (Pennisetum purpureum Schum.), was cut after nine weeks of regrowth and mixed with 10, 20, 30 and 40% of sugar cane bagasse (SCB) with the objective of reducing the moisture content of the ensiled mass. Willing of the grass for eight and twelve hours was used as a comparative treatment. Initial dry matter of the grass (13%) increased in the forage mass to 17, 23, 24 and 27% by the addition of 10, 20, 30 and 40% of the SCB respectively. Wilting for eight and twelve hours increased initial dry matter to 18 and 24% respectively. Buffering capacity of elephant grass was reduced by the addition of 40% of SCB. Clostridium spores in the ensiled mass tended to be lower due to the effect of the two pre-treatments. Initial dry matter and number of spores of Clostridium were negatively correlated although without statistic significance. The addition of SCB reduced (P < 0.0.5) soluble carbohydrates and crude protein percentages in the ensiled mass. It was concluded that wilting is more effective than the addition of SCB in the practice of ensiling elephant grass.

Silages from three cultivars of triticale (X Triticosecale wittimack) were evaluated at the UNESP, Jaboticabal, Brazil. The cultivars FCA VJ-CB-01, FCA VJ-CB-02, and FCA VJ-CB-03 were harvested for silage in three growing stages of maturity; beginning of flowering, (S1), milk stage (S2), and dough stages (S3). Data were analyzed by randomized complete block with three replications. The DM (%) values increased while the CP (%DM) and buffering capacity (me HCl/100 g DM) decreased with plant development. Silages of plants harvested at S3 stage had higher pH and N-NH3 values compared to the S1 and S2 silages. The soluble carbohydrates contents (%DM) were higher at the S2 stage (16.9%) and were not different at the S1 (8.7%) and S3 (9.2%) stages. The crude energy contents (Kcal/kg MS) increased while the ADF, NDF, cellulose, and hemicellulose (DM%) decreased due to the presence of dough grains. This was not observed with the lignin contents. The IVDMD values were 66.3, 60.1 and 58.9%, for plants harvested at the S1, S2, and S3 stages, respectively. The results showed that there was no difference among for chemical composition, crude energy, and for IVDMD.

This work had the objective of evaluating the effects of different percentages of corn (Zea mays L.) cob on the quality of the silage of high-moisture corn grains. The following treatments were studied: percentages of corn cob in the silage (0, 5, 10, 15 and 20%); period of sampling after opening the silos (zero, two, four and six days). The factorial arrangement 5 × 4 was studied according to a completely randomized block design with three replications. The variables studied were not affected by the sampling period. The buffering capacity and the pH were not affected by the cob, while the percent soluble carbohydrates and amoniacal nitrogen increasing until 1.2 and 1.89 unit percent, respectively. Increasing the amount of corn cobs reduced the contents of DM (from 63.9% to 58.6%), CP (from 10.0% to 7.3%), EE (from 4.87% to 3.92%) and the values of DMIVD (from 90.5% to 79.1%) in the silages and increasing the contents of acid detergente fiber (ADF) (from 3.3% to 12.9%) and neutral detergente fiber (NDF) (from 15.16% to 26.1%). The values of brute energy (BE) were not affected (P>0.01) by the cob corn in the silage.

The aim of this in vitro study was to evaluate the erosive capacity of fermented milk beverages, as well as some of their properties that affect the demineralization of dental enamel (pH, buffering capacity, fluoride, calcium and phosphorus contents). Three different batches of 6 commercial brands of fermented milk beverages were analyzed. pH evaluation was accomplished using a potentiometer. The buffering capacity was measured by adding 1 mol L -1 NaOH. Fluoride concentration was assessed by an ion specific electrode after hexamethyldisiloxane-facilitated diffusion, and calcium and phosphorus concentrations were assessed by a colorimetric test using a spectrophotometer. Sixty specimens of bovine enamel were randomly assigned to 6 groups (n = 10). They were exposed to 4 cycles of demineralization in the fermented milk and remineralization in artificial saliva. Enamel mineral loss was determined by surface microhardness (%SMHC) and profilometric tests. The samples' pH ranged from 3.51 to 3.87; the buffering capacity ranged from 470.8 to 804.2 μl of 1 mol L -1 NaOH; the fluoride concentration ranged from 0.027 to 0.958 μgF/g; the calcium concentration ranged from 0.4788 to 0.8175 mgCa/g; and the phosphorus concentration ranged from 0.2662 to 0.5043 mgP/g. The %SMHC ranged from-41.0 to -29.4. The enamel wear ranged from 0.15 μm to 0.18 μm. In this in vitro study, the fermented milk beverages did not promote erosion of the dental enamel, but rather only a superficial mineral loss.
