Emerging parasitoids of aphids encounter secondary plant chemistry from cues left by the mother parasitoid at oviposition and from the plant-feeding of the host aphid. In practice, however, it is secondary plant cheinistry oil the Surface of the aphid mummy which influences parasitoid olfactory behaviour. Offspring of Aphidius colemani reared oil Myzus persicae on artificial diet did no distinguish between the odours of bean and cabbage, but showed a clear preference for cabbage odour if sinigrin had been painted oil the back of the mummy. Similarly Aphidius rhopalosiphi reared on Metopolophium dirhodum on wheat preferred the odour of wheat plants grown near tomato plants to odour of wheat alone if the wheat plants oil which they had been reared had been exposed to the volatiles of nearby tomato plants. Aphidius rhopalosiphi reared on M dirhodum, and removed from the mummy before emergence, showed a preference for the odour of a different wheat cultivar if they had contacted a mummy from that cultivar, and similar results were obtained with A. colemani naturally emerged from M. persicae mummies. Aphidius colemani emerged from mummies oil one crucifer were allowed to contact in sequence (for 45 min each) mummies from two different crucifers. Artificial diet studies were used to differentiate among physical and chemical mechanisms affecting the suitability to diamondback moth (Plutella xylostella L.), of 16 food substrates obtained by growing four different brassicas in the glasshouse or field and measuring the pest's performance on either leaf discs or a diet incorporating leaf powders. Leaves of Chinese cabbage and the cabbage cultivar 'Minicole' were, respectively, the most and least suitable leaves for the insect, but this ranking was reversed on artificial diet. Leaves of glasshouse-grown plants were more suitable than those of plants grown in the fields. Differences in the suitability of leaves to diamondback moth appeared to be largely determined by leaf toughness and surface wax load. Concentrations of individual glucosinolates in the brassicas probably acted as phagostimulants, so increasing their intrinsic susceptibility to diamondback moth, but the effect of the physical factors appeared more important.

The characterization and identification of proteolytic bacteria from the gut of the velvetbean caterpillar (Anticarsia gemmatalis) were the objectives of this study. Twelve aerobic and anaerobic isolates of proteolytic bacteria were obtained from the caterpillar gut in calcium caseinate agar. The number of colony forming units (CFUs) of proteolytic bacteria was higher when the bacteria were extracted from caterpillars reared on artificial diet rather than on soybean leaves (1.73 +/- 0.35 X 10(3) and 0.55 +/- 0.22 X 10(3) CFU/mg gut, respectively). The isolated bacteria were divided into five distinct groups, according to their polymerase chain reaction restriction fragment-length polymorphism profiles. After molecular analysis, biochemical tests and fatty acid profile determination, the bacteria were identified as Bacillus subtilis, Bacillus cereus, Enterococcus gallinarum, Enterococcus mundtii, and Staphylococcus xylosus. Bacterial proteolytic activity was assessed through in vitro colorimetric assays for (general) proteases, serine proteases, and cysteine proteases. The isolated bacteria were able of hydrolyzing all tested substrates, except Staphylococcus xylosus, which did not exhibit serine protease activity. This study provides support for the hypothesis that gut proteases from velvetbean caterpillar are not exclusively secreted by the insect cells but also by their symbiotic gut bacteria. The proteolytic activity from gut symbionts of the velvetbean caterpillar is suggestive of their potential role minimizing the potentially harmful consequences of protease inhibitors from some of this insect host plants, such as soybean, with implications for the management of this insect pest species.

A proteinaceous trypsin inhibitor was purified from Crotalaria pallida seeds by ammonium sulphate fractionation, affinity chromatography on immobilized Trypsin-Sepharose and TCA precipitation. The trypsin inhibitor, named ITC, had Mr of 32.5 kDa by SDS-PAGE and was composed by two subunits with 27.7 and 5.6 kDa linked by disulphide bridges, a typical characteristic of Kunitz-Inhibitor family. ITC was stable until 50°C, and at 100°C its residual activity was of about 60%. Also, ITC was stable at pHs 2 to 12. The inhibition of trypsin by ITC was non-competitive, with a Ki of 8,8 x 10-7M. ITC inhibits weakly other serine proteinases such as chymotrypsin and elastase. The inhibition of papain (44% of inhibition), a cysteine proteinase was an indicative of the bi-functionality of ITC. In vitro assays against digestive proteinases from several Lepdoptera, Diptera and Coleoptera pests were made. ITC inhibited in 100% digestive enzymes of Ceratitis capitata (fruit fly), Spodoptera frugiperda and Alabama argillacea, the last one being a cotton pest. It also inhibited in 74.4% Callosobruchus maculatus (bean weevil) digestive enzymes, a Coleoptera pest. ITC, when added in artificial diet models, affected weakly the development of C. capitata larvae and it had a WD50 of 2.65% to C. maculatus larvae

One Kunitz-type trypsin inhibitors (PmTI) was purified from Piptadenia moniliformis seeds, a tree of the sub-family Mimosoideae, by TCA precipitation, affinity chromatography on immobilized trypsin-Sepharose, DEAE cellulose (ion exchange) and Superose 12 (molecular exclusion) column FPLC/AKTA. The inhibitor has Mr of 25 kDa by SDS-PAGE and chromatography molecular exclusion. The N-terminal sequence of this inhibitor showed high homology with other family Kunitz inhibitors. This also stable variations in temperature and pH and showed a small decrease in its activity when incubated with DDT in the concentration of 100mM for 120 minutes. The inhibition of trypsin by PmTI was competitive, with Ki of 1.57 x10-11 M. The activity of trypsin was effectively inhibited by percentage of inhibition of 100%, among enzymes tested, was not detected inhibition for the bromelain, was weak inhibitor of pancreatic elastase (3.17% of inhibition) and inhibited by 76.42% elastase of neutrophils, and inhibited in a moderate, chymotrypsin and papain with percentage of inhibition of 42.96% and 23.10% respectively. In vitro assays against digestive proteinases from Lepidoptera, Diptera and Coleoptera pests were carried out. Several degrees of inhibition were found. For Anthonomus grandis and Ceratitis capitata the inhibition was 89.93% and 70.52%, respectively, and the enzymes of Zabrotes subfasciatus and Callosobruchus maculatus were inhibited by 5.96% and 9.41%, respectively, and the enzymes of Plodia. interpunctella and Castnia licus were inhibited by 59.94% and 23.67, respectively. In vivo assays, was observed reduction in the development of larvae in 4rd instar of C. capitata, when PmTI was added to the artificial diet, getting WD50 and LD50 of 0.30% and 0.33%, respectively. 1 Nine synthetic amides similar to natural N-piperidine-3-(4,5-methylenedioxyphenyl)-2-(E)-propenainide and N-pyrrolidine-3-(4,5-methylenedyoxiphenyl)2-(E)-propenamide were synthesized and identified by their spectroscopic data.2 the toxicity of these synthetic amides to the Atta sexdens rubropilosa workers and the antifungal activity against Leticoagaricus gongylophorus, the symbiotic fungus of the leaf-cutting ants, were determined.3 Workers ants that were fed daily on an artificial diet to which these compounds were added had a higher mortality rate than the controls for N-pyrrolidine-3(3',4'-methylenedioxyphenyl)-2-(E)-propenamide and N-benzyl-3-(3',4'-methylenedioxyphenyl)-2-(E)-propenamide at a concentration of 100 mu g/mL.4 the completely inhibition (100%) of the fungal growth was observed with N-piperldine-3-(3',4'-methylenedioxyphenyl)-2-(E)-propenamide and N,N-diethyl-3-(3',4'-methylenedioxyphenyl)-2-(E)-propenamide at concentrations of 50 and 100 mu g/mL and N-pirrolidine-3-(3',4'-methylenedioxyphenyl)-2-(E)-propenamide at a concentration of 100 mu g/mL.5 the possibility of controlling these insects in the future using synthetic piperamides that can simultaneously target both organisms is discussed.

The fall armyworm, Spodoptera frugiperda (J. E. Smith), is one of the most important maize pests in the Americas and particularly in South America. With the adoption of genetically modified plants expressing Bacillus thuringiensis toxins for lepidopterous pest control, there is a need for establishing strategies to delay the development of insect resistance (e.g. refuge areas). Thus, information on target insects' dispersal is essential to improve pest management techniques. The objective of this work was to evaluate the dispersal capacity of S. frugiperda adults using mark-release-recapture techniques. Insects were marked using red oil-soluble dye in the larval artificial diet. Marked adults were released twice in each growing season (dry and wet) in southeastern Brazil in 2006 and 2007. Recapture of marked insects was performed using light and pheromone traps. Males are more attracted to light traps than females and the recapture rate was higher in the dry season than in the rainy season. The most adequate model to explain the relationship between flight distance and number of recaptured insects is y = a(2)/ (1+ (2a(1.8)+ bx))((2.6)), where y is the distance and x is the number captured. The maximum recapture distances were 806 m for males and 608 m for females. Therefore, strategies for establishment of refuges should take such distances into consideration.

O objetivo do trabalho foi avaliar diferentes dietas para lagartas de Diatraea saccharalis, com o intuito de obter informações sobre as características biológicas da praga e consequentemente avaliar a qualidade do parasitoide Cotesia flavipes, visando melhorar tecnicamente sua produção massal. Lagartas de 24 horas de idade foram transferidas para tubos (25 lagartas/tubo) contendo dieta artificial com levedura de cerveja e germe de trigo e outra somente com germe de trigo como fonte de proteína. Depois de aproximadamente 15 dias, as lagartas foram retiradas dos tubos e acondicionadas em placas com dieta, sendo observadas lagartas de D. saccharalis não parasitadas e parasitadas para avaliação das características biológicas de D. saccharalis e C. flavipes.nas diferentes dietas. Além da observação das características biológicas de D. saccharalis, foram realizadas medições nas lagartas de 15 dias de idade. Também, massas de pupas de C. flavipes.produzidas em biofábrica foram classificadas em três diferentes tamanhos, sendo pequenas (1,3 a 2,5 cm de comprimento), médias (2,5 a 3,5 cm de comprimento) e grandes (3,5 a 4,0 cm de comprimento). Após a emergência dos adultos foi realizado o parasitismo em lagartas de D. saccharalis, sendo observados o número de machos e fêmeas, tamanho da massa, razão sexual e número de pupas inviáveis. A dieta artificial que contém somente germe de trigo é a mais indicada para criação massal do hospedeiro D. saccharalis para produção em larga escala de C. flavipes. Massas de pupas classificadas como grandes possuem melhor qualidade em criações massais de C. flavipes.

The objective of this work was to evaluate biological aspects of Diatraea saccharalis fed on artificial diet containing different concentrations of the Sudan B Red dye and the possibility to mark the parasitoid Cotesia flavipes, when submitted to the parasitism of dyed caterpillars. For that, were added to the artificial diet four concentrations of Sudan Red B dye (100, 200, 300 and 400 ppm) and control (no dye addition). It was evaluated larval and pupal period, larval and pupal viability, longevity, sex rate, pupal weigh, eggs per female, eggs per day, number of eggs per egg mass, egg viability and embrionary period; besides same were accomplished measurements in the caterpillars (bioassay I). Caterpillars of 17 days old (30) of each treatment were removed from the tubes and exposed to the parasitism of C. flavipes (bioassay 2). The egg-pupae period, sex rate, pupal period and viability, number of females, males, total of emerged adults and longevity were evaluated. The data were submitted to the multivariate analisys methods: cluster analysis, two-way and principal component analysis. Based on analysis, it was observed that the treatment of 100 ppm was the least harmful to the biology of the sugar cane borer larvae by groping to the control and did not influence negatively its biological aspects. The concentration of 400 ppm affected negatively the biology of C. flavipes. The Sudan Red B it is ended doses marked the caterpillars and the adults, however the concentration of 100 ppm is the most suitable to dye D. saccharalis. None of the tested concentration marked adults of C. flavipes, despite to affect negatively its biology. It is unviable to increase the concentration seeking futures tests, for that dye to be harmful to the biological aspects of D. saccharalis and C. flavipes.

Este trabalho teve por objetivo verificar a viabilidade do uso de digitalizador de imagens manual, acoplado a um microcomputador, para a avaliação do consumo de folhas de soja, por lagartas de 5o instar de Anticarsia gemmatalis Hübner (Lep.: Noctuidae), em comparação com o método de pesagem e do planímetro, baseando-se na eficiência dos métodos e no tempo gasto para a avaliação. Os testes foram realizados utilizando-se folhas de soja `IAC 8' e lagartas criadas em dieta artificial. Foram realizados 2 tipos de teste: 1o) oferecimento de folíolos inteiros de soja às lagartas e, 2o) oferecimento de disco de folhas de área conhecida. No 1o teste comparou-se o método de pesagem com o digitalizador de imagens (scanner); no 2o experimento foram comparados o método do planímetro com o digitalizador de imagens que emprega o programa PCXAREA. Os resultados obtidos demonstraram que não existem diferenças nas medições de folíolos e discos de soja consumidos por A. gemmatalis quando comparados os métodos tradicionais (planímetro e pesagem) e o de digitalização de imagens. A medição com o digitalizador reduziu o tempo de avaliação em 88,5% e 87%, em relação ao planímetro e método de pesagem, respectivamente, sendo plenamente viável a sua utilização.

A broca do cacho do coqueiro, Homalinotus coriaceus (Gyllenhal), é uma praga limitante da produção de coco no Brasil, sendo que tanto as larvas como os adultos provocam a queda das flores femininas e dos frutos imaturos, pela interceptação do fluxo de seiva ou pela alimentação direta nas estruturas reprodutivas. em virtude da escassez de informações sobre sua biologia, realizou-se esse trabalho com o objetivo de desenvolver uma metodologia mais adequada para a criação da praga em laboratório. Foram utilizados os parâmetros biológicos para avaliação e comparação dos sistemas de criação estudados.Toletes de cana-de-açúcar foram utilizados como substrato para alimentação dos adultos coletados no campo e obtenção dos ovos. As larvas foram criadas em três substratos alimentares no Laboratório de Entomologia da Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA)-CPATC (Centro de Pesquisa Agropecuária dos Tabuleiros Costeiros), em Aracaju, SE. Os substratos alimentares estudados foram: o mesocarpo do coco, dieta para criação da broca dos citros e dieta para criação da broca do olho do coqueiro, sendo esta a que proporcionou o melhor desenvolvimento larval num menor tempo, com boa viabilidade, maior facilidade no preparo e manutenção.

O gênero Prochilodus é amplamente distribuído pela América do Sul, incluindo várias espécies que apresenta certo grau de endemismo nas diferentes bacias. Essas espécies alimentam-se basicamente de detritos orgânicos, organismos do benton, e alimentações artificiais. O objetivo deste estudo foi testar três densidades de estocagem (0,5, 0,75, e 1 larva/litro) e dietas contendo dois níveis diferentes de proteína bruta (35 e 40% PB) nos parâmetros do crescimento e na taxa da sobrevivência do curimbatá, Prochilodus scrofa. As larvas foram estocadas em aquários experimentais de 100 litros, mantidos com volume de 60 litros, com o fluxo de água contínuo, proveniente de poço semi-artesiano. A maior média foi proporcionada pela ração com 40% de PB (1,66g ± 0,21). As diferenças foram significativas para os efeitos dos teores de PB sobre o ganho de peso e densidades de estocagem, na sobrevivência das larvas. Para o ganho de peso, as melhores médias foram obtidas com nível de 40% de PB (2,50g ± 0,40) e para a sobrevivência, as melhores taxas foram 94,45% ± 19,32 e 78,87% ± 19,32 nas densidades de 0,5 e 0,75 larvas/litro, respectivamente.
