The timely regulation of gonadotropin-releasing hormone (GnRH) secretion requires a GABAergic signal. We hypothesized that GEC1, a protein promoting the transport of GABA(A) receptors, could represent a circadian effector in GnRH neurons. First, we demonstrated that gec1 is co-expressed with the GABA(A) receptor in hypothalamic rat GnRH neurons. We also confirmed that the clock genes per1, cry1 and bmal1 are expressed and oscillate in GnRH secreting GnV-3 cells. Then we could show that gec1 is expressed in GnV-3 cells, and oscillates in a manner temporally related to the oscillations of the clock transcription factors. Furthermore, we could demonstrate that these oscillations depend upon Per1 expression. Finally, we observed that GABA(A) receptor levels at the GnV-3 cell membrane are timely modulated following serum shock. Together, these data demonstrate that gec1 expression is dependent upon the circadian clock machinery in GnRH-expressing neurons, and suggest for the first time that the level of GABA(A) receptor at the cell membrane may be under timely regulation. Overall, they provide a potential mechanism for the circadian regulation of GnRH secretion by GABA, and may also be relevant to the general understanding of circadian rhythms.

Isolated gonadotropin-releasing hormone (GnRH) deficiency is a treatable albeit rare form of reproductive failure that has revealed physiological mechanisms controlling human reproduction, but despite substantial progress in discovering pathogenic single-gene defects, most of the genetic basis of GnRH deficiency remains uncharted. Although unbiased genetic investigations of affected families have identified mutations in previously unsuspected genes as causes of this disease in some cases, their application has been severely limited because of the negative effect of GnRH deficiency on fertility; moreover, relatively few of the many candidate genes nominated because of biological plausibility from in vitro or animal model experiments were subsequently validated in patients. With the advent of exciting technological platforms for sequencing, homozygosity mapping, and detection of structural variation at the whole-genome level, human investigations are again assuming the leading role for gene discovery. Using human GnRH deficiency as a paradigm and presenting original data from the screening of numerous candidate genes, we discuss the emerging model of patient-focused clinical genetic research and its complementarities with basic approaches in the near future.

BACKGROUND: Neurospheres (NS) are colonies of neural stem and precursor cells capable of differentiating into the central nervous system (CNS) cell lineages upon appropriate culture conditions: neurons, and glial cells. NS were originally derived from the embryonic and adult mouse striatum subventricular zone. More recently, experimental evidence substantiated the isolation of NS from almost any region of the CNS, including the hypothalamus. METHODOLOGY/FINDINGS: Here we report a protocol that enables to generate large quantities of NS from both fetal and adult rat hypothalami. We found that either FGF-2 or EGF were capable of inducing NS formation from fetal hypothalamic cultures, but that only FGF-2 is effective in the adult cultures. The hypothalamic-derived NS are capable of differentiating into neurons and glial cells and most notably, as demonstrated by immunocytochemical detection with a specific anti-GnRH antibody, the fetal cultures contain cells that exhibit a GnRH phenotype upon differentiation. CONCLUSIONS/SIGNIFICANCE: This in vitro model should be useful to study the molecular mechanisms involved in GnRH neuronal differentiation.

Se ha estudiado el papel, como inhibidores de la acción feromonal, de determinados fluoroderivados, análogos estructurales del acetato de (Z)-l 1-hexadecenilo, componente principal de la feromona sexual del noctuido Sesamia nonagrioides Lef. Dichos compuestos fueron los análogos fluoroacetato (mono, di y trifluoroacetato de (Z)-l 1-hexadecenilo) y la trifluorometilcetona análoga [(Z)-1,1,1 -trifluoro-14-nonadecen-2-ona]. La acción inhibidora fue evaluada a partir de los resultados de actividad electrofisiológica (pruebas de electroantenograma, EAG), de estudios de comportamiento en pruebas de túnel de viento y de pruebas de capturas en campo. Los acetatos fluorados, especialmente el mono y el trifluoracetato, se mostraron como buenos inhibidores de la acción atrayente de la feromona en los tres tipos de pruebas realizadas, mientras que la trifluorometilcetona análoga mostró una actividad mucho menor.

O cultivo comercial do maracujá é afetado por diversos problemas fitossanitários, os quais contribuem para quebras de produção e significativa redução da vida útil dos plantios. Em algumas situações, a incidência de doenças pode inviabilizar o cultivo do maracujá. Fontes de resistência a distintas doenças têm sido identificadas em acessos de espécies de Passiflora. Neste trabalho, buscou-se avaliar a diversidade genética de acessos de oito espécies silvestres (P. setacea, P. nitida, P. serratodigitata, P. caerulea, P. gibertii, P. odontophyla, P. edulis e P. coccinea) e de um híbrido interespecífico (P. setacea x P. coccinea), utilizando marcadores moleculares análogos a genes de resistência (RGAs). Verificou-se uma grande diversidade no perfil eletroforético de RGAs nos acessos de Passiflora, permitindo a anotação de 96 amplicons polimórficos entre, pelo menos, um par de acessos. Os níveis de dissimilaridade genética (calculados exclusivamente com os marcadores RGAs) variaram entre 0,40 e 0,89 nos acessos das espécies de Passiflora avaliadas. A análise de sequência de um subgrupo destes amplicons obtidos com primers RGAs indicou que estas bandas correspondem a regiões genômicas que contêm segmentos (motivos) com identidade aos encontrados em genes de resistência previamente caracterizados em outras espécies vegetais. Desta forma, os dados indicam a existência de um repertório variado de marcadores do tipo RGA em Passiflora que podem ser potencialmente úteis em sistemas de caracterização molecular de germoplasma e em programas de melhoramento genético visando à resistência a doenças nesta cultura.

The synthesis of two new spirostanic analogs of the natural occurring brassinosteroid 6-desoxocastasterone (1) is described. The scheme consists in the formation and elimination of tigogenin mesylate followed by catalytic dihydroxylation of the resulting D2-steroid (3) and acetylation of the 2a, 3a-diol introduced.Treatment diacetate (5) with NaNO2/BF3.Et2O and chromatography in alumina led to a 23-keto (6) which on reduction produced the 23S alcohol (8) as major product. Saponification of the 2a, 3a-diacetoxy-23-keto compound (6) and the 2a,3a-diacetoxy-23-hydroxy compound (8) led to the spirobrasinosteroids (7) and (9).13C NMR and ¹H RMN characteristics derived from substitution at C23 are briefly discussed.

Several compounds related to helminthosporic acid (3) were synthesized via the [3+4] cycloaddition. The reaction of 3-hydroxymethyl-2-methylfuran (12) with 1,1,3,3-tetrabromo-4-methylpentan-2-one (13) resulted in 7-hydroxymethyl-4alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (8) (37%) and 7-hydroxymethyl-2alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (14) (12%), which were converted into 7-formyl-4alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (16) (32% from 8) and 7-formyl-2alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (18) (40% from 14), respectively. Reduction of (8) resulted in 7-hydroxymethyl-4alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6 -en-3alpha-ol (11) (63% from 8) and 7-hydroxymethyl-4alpha-isopropyl-1alpha-methyl-8-oxabicyclo[3.2.1]oct-6-en-3 beta-ol (15) (30% from 8). The 4alpha-isopropyl-1alpha-methyl-3-oxo-8-oxabicyclo[3.2.1]oct-6-en-7-oic acid (19) was obtained by oxidation of (16) (78%). The results of biological tests are described in details. The best result was observed for compound (15) that caused 76% inhibition on the root growth of D. tortuosum.

Two ortho-iodoallyloxybenzoates, methyl 4-O-allyl-2,3-di-O-benzyl-6-O-(2-iodobenzoyl)- alpha-D-glucopyranoside (3) and methyl 4-O-allyl-2,3-di-O-benzyl-6-O-(2-iodobenzoyl)- alpha-D-galactopyranoside (4) were synthesized in seven conventional steps from methyl alpha-D-glucopyranoside and methyl alpha-D-galactopyranoside, respectively. Bu3SnH-mediated aryl radical cyclization of 3 provided exclusively the hydrogenolysis product 12. The reaction of 4 gave the reduced uncyclized product 13 and only traces of 4A, resulting from 11-endo aryl radical cyclization. In previous papers we described that in similar Bu3SnH-mediated radical reaction of ortho-iodoallyloxybenzamides, analogs of 3 and 4, we obtained macrolactams resulting from 11-endo cyclization. An hypothesis to explain the differences is presented. It was assumed that in the aryl radical formed from iodobenzamides there is a suitable conformation to cyclization, which is stabilized by an intramolecular hydrogen bond.

Anatoxin-a and its analogues are azabicyclic alkaloids that represent one of the most powerful nicotinic agonists known for the nicotinic acetylcholine receptor (nAChR). Because of this potent mechanism of action, anatoxin-a and its derivatives represent a target for the discovery of novel drugs. Their syntheses are useful for environmental monitoring and also for pharmacokinetic/pharmacodynamic and toxicological studies. Some strategies for the synthesis of anatoxin-a and analogous compounds are described herein, covering the period from 1996 to the present date. In this review, emphasis is given to the chemical and toxicological aspects of some variants of anatoxin-a, including homoanatoxin-a and anatoxin-a(s).

Considering the broad spectrum of biological activity of gamma-butyrolactone derivatives, we presented the synthesis of 3,4-dihalo-5-arylidenefuran-2(5H)-ones (17-21) and analogues (24-28) of the natural product nostoclide (7,8). Furanones 17-21 were synthesized from the condensation of aromatic aldehydes with lactones 14 and 15, that were obtained from mucobromic and mucochloric acids. Lactone 15 was converted into the intermediate 23 in 36% overall yield. Compound 23 was then transformed into the nostoclide analogues 24-28. Some of the compounds prepared showed antimicrobial activities against Escherichia coli, Staphylococcus aureus and Bacillus cereus comparable to commercial antibiotics.

We report the single-step derivatization reaction of a biopolymer based onL -lysine with D -biotin analogs:Co -poly(L -lysine)-graft-(ε-N -[X-D-biotinyl]-L -lysine) (PLL-X-Biotin). The valeric acid carboxylate of D -biotin is activated to an NHS ester for direct modification of amine groups in proteins and other macromolecules. NHS esters react by nucleophilic attack of an amine in the carbonyl group, releasing the NHS group, and forming a stable amide linkage. NHS-X-Biotin is the simplest biotinylation reagent commercially available. In contrast withD -biotin, it has a longer spacer arm off the valeric acid side chain allowing better binding potential for avidin or streptavidin probes. Derivatization of poly(L -lysine) (PLL) with NHS-X-Biotin led to a copolymer PLL-X-Biotin. UV-Visible, IR-FT and 1H NMR characteristics derived from synthesis are briefly discussed.

O maior grupo de genes de resistência de plantas já clonados codifica para proteínas com um sítio de ligação a nucleotídios (NBS) na região N-terminal, e um domínio rico em repetições de leucina (LRR) na região C-terminal. Genes desta classe conferem resistência a diversos patógenos incluindo vírus, bactérias, fungos e nematóides. Para diferentes espécies do gênero Arachis, primers de "polymerase chain reaction" (PCR) degenerados foram construídos para a região NBS, e o produto de tradução putativo indicou similaridade com proteínas de resistência conhecidas sendo denominados análogos a genes de resistência (RGAs). Doze destes RGAs foram utilizados para o desenvolvimento de marcadores moleculares baseados em seus padrões de hibridização com DNA de Arachis spp. digerido com enzimas de restrição. Inicialmente, avaliou-se o polimorfismo de cada RGA como sonda nos parentais de uma população de mapeamento, contrastantes quanto à resistência as manchas foliares e nematóides das galhas, e no híbrido F1. Os RGAs, mesmo isolados de espécies diferentes do gênero Arachis apresentaram homologia com o DNA das espécies testadas, além de apresentarem múltiplas cópias e alto polimorfismo na progênie F2. Todas estas características tornam estes RGAs marcadores moleculares altamente informativos, sendo que alguns apresentaram segregação em "clusters" na F2, indicando que seus locos estão ligados. Estes marcadores serão incluídos em um mapa genético de Arachis spp., o que será de grande utilidade para os programas de melhoramento do amendoim (Arachis hypogaea) cultivado.

OBJETIVO: verificar, com o emprego da ultra-sonografia pélvica, a existência de mudanças na genitália interna de meninas com puberdade precoce central submetidas a tratamento com análogo do hormônio liberador de gonadotrofinas (GnRH). MÉTODOS: a ultra-sonografia pélvica foi realizada em 18 meninas com diagnóstico de puberdade precoce central idiopática, antes e três meses após o inicio do tratamento com análogo de GnRH, para avaliar o impacto da terapia na genitália interna feminina. Foram avaliados os volumes uterino e ovariano, o diâmetro longitudinal do útero, a relação entre os diâmetros longitudinais do corpo e colo uterinos, a relação entre os diâmetros ântero-posteriores do corpo e colo uterinos e o eco endometrial. Para a análise estatística foi aplicado o teste de Shapiro-Willks para verificação da normalidade dos dados. Para os dados em que a normalidade foi satisfeita, foi aplicado o teste t de Student. Para os dados cuja distribuição não foi normal aplicou-se o teste não paramétrico (teste do sinal). RESULTADOS: após o tratamento houve redução estatisticamente significante da média dos volumes uterino (de 5,4 para 3,0 cm³, p<0,001) e ovariano (de 2,2 para 1,1 cm³, p=0,004), da média do diâmetro longitudinal do útero (de 4,2 para 3,4 cm, p=0,001) e da média do eco endometrial (de 1,8 para 0,6 mm, p=0,018). CONCLUSÃO: em meninas com puberdade precoce a ultra-sonografia pélvica é útil para avaliar a eficácia do tratamento com análogo de GnRH. Os principais parâmetros de resposta à terapia são as diminuições dos volumes uterino e ovariano, a redução do diâmetro longitudinal do útero e a atrofia ou ausência do eco endometrial.
