A human cDNA encoding an 841-aa guanine nucleotide-exchange protein (GEP) for ADP-ribosylation factors (ARFs), named ARF-GEP100, which contains a Sec7 domain, a pleckstrin homology (PH)-like domain, and an incomplete IQ-motif, was identified. On Northern blot analysis of human tissues, a ≈8-kb mRNA that hybridized with an ARF-GEP100 cDNA was abundant in peripheral blood leukocytes, brain, and spleen. ARF-GEP100 accelerated [35S]GTPγS binding to ARF1 (class I) and ARF5 (class II) 2- to 3-fold, and to ARF6 (class III) ca. 12-fold. The ARF-GEP100 Sec7 domain contains Asp543 and Met555, corresponding to residues associated with sensitivity to the inhibitory effect of the fungal metabolite brefeldin A (BFA) in yeast Sec7, but also Phe535 and Ala536, associated with BFA-insensitivity. The PH-like domain differs greatly from those of other ARF GEPs in regions involved in phospholipid binding. Consistent with its structure, ARF-GEP100 activity was not affected by BFA or phospholipids. After subcellular fractionation of cultured T98G human glioblastoma cells, ARF6 was almost entirely in the crude membrane fraction, whereas ARF-GEP100, a 100-kDa protein detected with antipeptide antibodies, was cytosolic. On immunofluorescence microscopy, both proteins had a punctate pattern of distribution throughout the cells, with apparent colocalization only in peripheral areas. The coarse punctate distribution of EEA-1 in regions nearer the nucleus appeared to coincide with that of ARF-GEP100 in those areas. No similar coincidence of ARF-GEP100 with AP-1, AP-2, catenin, LAMP-1, or 58K was observed. The new human BFA-insensitive GEP may function with ARF6 in specific endocytic processes.

The sedimentary succession drilled at Sites 840 and 841 on the Tonga forearc allows the sedimentary evolution of the active margin to be reconstructed since shortly after the initiation of subduction during the mid Eocene. Sedimentation has been dominated by submarine fan deposits, principally volcaniclastic turbidites and mass-flows derived from the volcanic arc. Volcaniclastic sedimentation occurred against a background of pelagic nannofossil sedimentation. A number of upward-fining cycles are recognized and are correlated to regional tectonic events, such as the rifting of the Lau Basin at 5.6 Ma. Episodes of sedimentation dating from 16.0 and 10.0 Ma also correlate well with major falls in eustatic sea level and may be at least partially caused by the resulting enhanced erosion of the arc edifice. The early stages of rifting of the Lau Basin are marked by the formation of a brief hiatus at Site 840 (Horizon A), probably a result of the uplift of the Tonga Platform. Controversy exists as to the degree and timing of the uplift of Site 840 before Lau Basin rifting, with estimates ranging from 2500 to 300 m. Structural information favors a lower value. Breakup of the Tonga Arc during rifting resulted in deposition of dacite-dominated, volcaniclastic mass flows, probably reflecting a maximum in arc volcanism at this time. A pelagic interval at Site 840 suggests that no volcanic arc was present adjacent to the Tonga Platform from 5.0 to 3.0 Ma. This represents the time between separation of the Lau Ridge from the Tonga Platform and the start of activity on the Tofua Arc at 3.0 Ma. The sedimentary successions at both sites provide a record of the arc volcanism despite the reworked nature of the deposits. Probe analyses of volcanic glass grains from Site 840 indicate a consistent low-K tholeiite chemistry from 7.0 Ma to the present, possibly reflecting sediment sourcing from a single volcanic center over long periods of time. Trace and rare-earth-element (REE) analyses of basaltic glass grains indicate that thinning of the arc lithosphere had begun by 7.0 Ma and was the principle cause of a progressive depletion of the high-field-strength (HFSE), REE, and large-ion-lithophile (LILE) elements within the arc magmas before rifting. Magmatic underplating of the Tofua Arc has reversed this trend since that time. Increasing fluid flux from the subducting slab since basin rifting has caused a progressive enrichment in LILEs. Subduction erosion of the underside of the forearc lithosphere has caused continuous subsidence and tilting toward the trench since 37.0 Ma. Enhanced subsidence occurred during rifting of the South Fiji and Lau basins. Collision of the Louisville Ridge with the trench has caused no change in the nature of the sedimentation, but it may have been responsible for up to 300 m of uplift at Site 840.

Görgeyite, K2Ca5(SO4)6··H2O, is a very rare monoclinic double salt found in evaporites related to the slightly more common mineral syngenite. At 1 atmosphere with increasing external temperature from 25 to 150 °C, the following succession of minerals was formed: first gypsum and K2O, followed at 100 °C by görgeyite. Changes in concentration at 150 °C due to evaporation resulted in the formation of syngenite and finally arcanite. Under hydrothermal conditions, the succession is syngenite at 50 °C, followed by görgyeite at 100 and 150 °C. Increasing the synthesis time at 100 °C and 1 atmosphere showed that initially gypsum was formed, later being replaced by görgeyite. Finally görgeyite was replaced by syngenite, indicating that görgeyite is a metastable phase under these conditions. Under hydrothermal conditions, syngenite plus a small amount of gypsum was formed, after two days being replaced by görgeyite. No further changes were observed with increasing time. Pure görgeyite showed elongated crystals approximately 500 to 1000 µ m in length. The infrared and Raman spectra are mainly showing the vibrational modes of the sulfate groups and the crystal water (structural water). Water is characterized by OH-stretching modes at 3526 and 3577 cm–1 , OH-bending modes at 1615 and 1647 cm–1 , and an OH-libration mode at 876 cm–1 . The sulfate 1 mode is weak in the infrared but showed strong bands at 1005 and 1013 cm–1 in the Raman spectrum. The 2 mode also showed strong bands in the Raman spectrum at 433, 440, 457, and 480 cm–1 . The 3 mode is characterized by a complex set of bands in both infrared and Raman spectra around 1150 cm–1 , whereas 4 is found at 650 cm–1.

The use of polycaprolactone (PCL) as a biomaterial, especially in the fields of drug delivery and tissue engineering, has enjoyed significant growth. Understanding how such a device or scaffold eventually degrades in vivo is paramount as the defect site regenerates and remodels. Degradation studies of three-dimensional PCL and PCL-based composite scaffolds were conducted in vitro (in phosphate buffered saline) and in vivo (rabbit model). Results up to 6 months are reported. All samples recorded virtually no molecular weight changes after 6 months, with a maximum mass loss of only about 7% from the PCL-composite scaffolds degraded in vivo, and a minimum of 1% from PCL scaffolds. Overall, crystallinity increased slightly because of the effects of polymer recrystallization. This was also a contributory factor for the observed stiffness increment in some of the samples, while only the PCL-composite scaffold registered a decrease. Histological examination of the in vivo samples revealed good biocompatibility, with no adverse host tissue reactions up to 6 months. Preliminary results of medical-grade PCL scaffolds, which were implanted for 2 years in a critical-sized rabbit calvarial defect site, are also reported here and support our scaffold design goal for gradual and late molecular weight decreases combined with excellent long-term biocompatibility and bone regeneration. Having flexible notions of the unit (e.g., 26 ones can be thought of as 2.6 tens, 1 ten 16 ones, 260 tenths, etc.) should be a major focus of elementary mathematics education. However, often these powerful notions are relegated to computations where the major emphasis is on "getting the right answer" thus procedural knowledge rather than conceptual knowledge becomes the primary focus. This paper reports on 22 high-performing students' reunitising processes ascertained from individual interviews on tasks requiring unitising, reunitising and regrouping; errors were categorised to depict particular thinking strategies. The results show that, even for high-performing students, regrouping is a cognitively complex task. This paper analyses this complexity and draws inferences for teaching.
