Although a variety of nanoparticles (NPs) functionalized with amphotericin B, an antifungal agent widely used in the clinic, have been studied in the last years their cytotoxicity profile remains elusive. Here we show that human endothelial cells take up high amounts of silica nanoparticles (SNPs) conjugated with amphotericin B (AmB) (SNP-AmB) (65.4 12.4 pg of Si per cell) through macropinocytosis while human fibroblasts internalize relatively low amounts (2.3 0.4 pg of Si per cell) because of their low capacity for macropinocytosis. We further show that concentrations of SNP-AmB and SNP up to 400 mg/mL do not substantially affect fibroblasts. In contrast, endothelial cells are sensitive to low concentrations of NPs (above 10 mg/mL), in particular to SNP-AmB. This is because of their capacity to internalize high concentration of NPs and high sensitivity of their membrane to the effects of AmB. Low-moderate concentrations of SNP-AmB (up to 100 mg/mL) induce the production of reactive oxygen species (ROS), LDH release, high expression of pro-inflammatory cytokines and chemokines (IL-8, IL-6, G-CSF, CCL4, IL-1b and CSF2) and high expression of heat shock proteins (HSPs) at gene and protein levels. High concentrations of SNP-AmB (above 100 ug/mL) disturb membrane integrity and kill rapidly human cells(60% after 5 h). This effect is higher in SNP-AmB than in SNP.

A dissertação que aqui se apresenta partiu da necessidade de colocar em confronto as bases éticas da conservação e restauro, que assentam na defesa da materialidade histórica, com a progressiva desmaterialização da arte que ocorre a partir da década de 60 em diante. A organização do trabalho implicou, na escrita, a distinção entre duas partes: uma históricoteórica e a outra de carácter mais prático. Na primeira parte procurámos justificar a progressiva tendência histórica para a desmaterialização do suporte artístico bem como a consequente necessidade de um re-pensamento dos aspectos éticos e práticos da conservação, com particular atenção à documentação do ponto de vista autoral. Na segunda parte da dissertação apresentamos um conjunto de oito casos práticos, a partir de obras de oito artistas portugueses – Henrique Ruivo, Lourdes Castro, Noronha da Costa, Ana Vieira, Alberto Carneiro, João Vieira, René Bertholo e Helena Almeida - todos com início de carreira na década de 60. Procurámos reunir, com base numa investigação em torno dos aspectos da criação da obra, mas também da sua recepção, informação detalhada sobre materiais constitutivos, sua origem e semântica, técnicas, processos criativos, bem como a posição de cada autor relativamente ao envelhecimento das obras em análise. Para o efeito procedemos a um conjunto de entrevistas presenciais aos artistas em questão, directores de museus e curadores – com envolvimento com as obras daqueles - bem como ao levantamento da fortuna crítica associada às obras seleccionadas. Um dos aspectos que se revelou decisivo ao longo do trabalho foi o da investigação dos processos criativos, central para obras cuja autenticidade não se pode resumir a qualquer noção de originalidade material, podendo contudo ser alicerçada na investigação e documentação do processo criativo, realizado essencialmente a partir do ponto de vista autoral. O trabalho efectuado teve também como objectivo a compreensão das condições reais de conservação documental das peças. Concluímos: a necessidade de se proceder à documentação(inscrição) como forma de conservação; o papel imprescindível de uma historiografia da arte mais centrada nos aspectos da preservação; a urgência de formar profissionais preparados para desempenhar esta tarefa; bem como a indispensabilidade de ferramentas que permitam a organização da informação e a sua disponibilização, no âmbito da comunidade envolvida na arte contemporânea, e ao público em geral.

According to the latest census (2011) covering the Portuguese housing stock, there is a considerable number of sub-occupied buildings and most of them were built before the first Portuguese Thermal Code (RCCTE). In view of the objectives of the European Union (20-20-20), it is necessary to contribute to the rehabilitation of existing buildings by promoting the densification of oversized houses, while complying with up-to-date comfort standards and codes. In this sense, this research is aimed to analyse this problem at the outskirts of Braga, by identifying the type-morphology of oversized buildings, assessing the degree of satisfaction and expectations of both owners and inhabitants and developing and testing a systematic design tool. Through the development of an intervention kit, wich systematizes and priorizes design strategies and solutions, the design team can have the necessary decision-making support tool to improve the spacial design of the building whilst considering energy efficency, sustainability and improvemente of interior environmental quality.

OBJECTIVE: To verify if adaptive left ventricle (LV) characteristics are also present in individuals under 70 years of age with severe aortic stenosis (AS). METHODS: The study comprised 40 consecutive patients under 70 years of age with AS and no associated coronary artery disease, referred for valve surgery. Out of the 40 patients, 22 were men and 18 women, and the mean age was 49.8±14.3 years. Cardiac symptoms, presence of systemic hypertension (SH), functional class according to the New York Heart Association (NYHA), and valve lesion etiology were considered. LV cavity dimensions, ejection fraction (EF), fractional shortening (FS), mass (MS), and relative diastolic thickness (RDT) were examined by Doppler echocardiography. RESULTS: Fourteen (63.6%) men and 11 (61.6%) women were classified as NYHA class III/IV (p=0.70). There was no difference in the frequency of angina, syncope or dyspnea between genders. The incidence of SH was greater in women than in men (10 versus 2, p=0.0044). Women had a smaller LV end-diastolic diameter index (32.1±6.5 x 36.5±5.3mm/m², p=0.027), LV end-systolic diameter index (19.9±5.9 x 26.5±6.4mm/m², p=0.0022) and LV mass index (MS) (211.4±71.1 x 270.9±74.9g/m², p=0.017) when compared with men. EF (66.2±13.4 x 52.0±14.6%, p=0.0032), FS (37.6±10.7 x 27.9±9.6%, p=0.0046) and RDT (0.58±0.22 x 0.44±0.09, p=0.0095) were significantly greater in women than in men. CONCLUSION: It is the patient gender rather than age that influences left ventricular adaptive response to AS.

PURPOSE: To report the result of patients treated with IV methylprednisolone divided into three groups and compare their follow-up during the last 12 years. METHODS: Seventy children with active rheumatic carditis (76 episodes) in heart failure Class III and IV (NYHA) were studied. The diagnosis was based on modified Jones' criteria. After ruling out infections and strongyloidiasis, treatment with IV methylprednisolone bolus was started three times a week until the laboratory tests became negative. Patients were divided into 3 groups, according to the time of hospital admittance: Groups 1, 2 and 3, comprising of 40, 18 and 12 children, respectively. RESULTS: Eighteen children in Group 1 (45%) were in their 1st attack: 2 series of pulsetherapy were used in 10 (25%), 3 in 9 (23%) and 4 in 21 (52%). In Group 2, 14 cases (77%) were in their 1st attack: 2 series were used in 7 (39%), 4 in 9 (50%) and 5 in 2 (11%). The echocardiogram showed a flail mitral valve in 12 (66%) of these patients (1 death occurred after mitral valvoplasty). In Group 3, 6 patients needed 5 or more series of pulsetherapy and a flail mitral valve was present in 5 (41%). One child underwent mitral valve replacement while still in the active phase, after 8 series of pulsetherapy, and another died. The number of patients who needed 5 or more series was significantly higher in Group 3. CONCLUSION: There were variations in the presentation and evolution of the cases during these 12 year. The established pulsetherapy protocol continues to be useful to treat severe cases.

The repellent effect of the molluscicides Niclosamide (Bayluscide WP 70 (R)), Anacardium occidentale and the latex of Euphorbia splendens on Biomphalaria glabrata was observed through the investigation of the occurrence of escape behavior among molluscs that were exposed to dosages lower than the LD 50. The total number of individuals out of water among the surviving snails in the control group provided a "Natural Escape Index". The comparison between this total and the total number of surviving snails in each group exposed to the different dosages of the molluscicides after 24 hr provided the "Molluscicide Escape Index" and the detection of a "Repellency Range" to these snails. The escape indexes for Niclosamide, A. occidentale and E. splendens were 10, 6.22 and 6.44 respectively. Repellency occurred at the following concentration ranges: 0.01, 0.02 and 0.03 ppm Bayluscide, 0.1, 0.2 and 0.3 ppm A. occidentale and 0.05, 0.10, 0.15 and 0.20 ppm E. splendens. The Natural Escape Index obtained in the control group was zero.
