Cette thèse en électronique moléculaire porte essentiellement sur le développement d’une méthode pour le calcul de la transmission de dispositifs électroniques moléculaires (DEMs), c’est-à-dire des molécules branchées à des contacts qui forment un dispositif électronique de taille moléculaire. D’une part, la méthode développée vise à apporter un point de vue différent de celui provenant des méthodes déjà existantes pour ce type de calculs. D’autre part, elle permet d’intégrer de manière rigoureuse des outils théoriques déjà développés dans le but d’augmenter la qualité des calculs. Les exemples simples présentés dans ce travail permettent de mettre en lumière certains phénomènes, tel que l’interférence destructive dans les dispositifs électroniques moléculaires. Les chapitres proviennent d’articles publiés dans la littérature. Au chapitre 2, nous étudions à l’aide d’un modèle fini avec la méthode de la théorie de la fonctionnelle de la densité de Kohn-Sham un point quantique moléculaire. De plus, nous calculons la conductance du point quantique moléculaire avec une implémentation de la formule de Landauer. Nous trouvons que la structure électronique et la conductance moléculaire dépendent fortement de la fonctionnelle d’échange et de corrélation employée. Au chapitre 3, nous discutons de l’effet de l’ajout d’une chaîne ramifiée à des molécules conductrices sur la probabilité de transmission de dispositifs électroniques moléculaires. Nous trouvons que des interférences destructives apparaissent aux valeurs propres de l’énergie des chaînes ramifiées isolées, si ces valeurs ne correspondent pas à des états localisés éloignés du conducteur moléculaire. Au chapitre 4, nous montrons que les dispositifs électroniques moléculaires contenant une molécule aromatique présentent généralement des courants circulaires qui sont associés aux phénomènes d’interférence destructive dans ces systèmes. Au chapitre 5, nous employons l’approche « source-sink potential » (SSP) pour étudier la transmission de dispositifs électroniques moléculaires. Au lieu de considérer les potentiels de sources et de drains exactement, nous utilisons la théorie des perturbations pour trouver une expression de la probabilité de transmission, T(E) = 1 − |r(E)|2, où r(E) est le coefficient de réflexion qui dépend de l’énergie. Cette expression dépend des propriétés de la molécule isolée, en effet nous montrons que c’est la densité orbitalaire sur les atomes de la molécule qui sont connectés aux contacts qui détermine principalement la transmission du dispositif à une énergie de l’électron incident donnée. Au chapitre 6, nous présentons une extension de l’approche SSP à un canal pour des dispositifs électroniques moléculaires à plusieurs canaux. La méthode à multiples canaux proposée repose sur une description des canaux propres des états conducteurs du dispositif électronique moléculaire (DEM) qui sont obtenus par un algorithme auto-cohérent. Finalement, nous utilisons le modèle développé afin d’étudier la transmission du 1-phényl-1,3-butadiène branché à deux rangées d’atomes couplées agissant comme contacts à gauche et à la droite.

In industrial polymer and synthetic rubber production facilities, workers are exposed to 1,3-butadiene. This compound is converted in vivo to 1,2,3,4-diepoxybutane (DEB) and has been linked to increased incidences of cancer in these individuals. Carcinogenesis has been attributed to formation of DEB induced DNA interstrand cross-links. Previous studies have demonstrated that DEB cross-links deoxyguanosine residues within 5'-GNC sequences in synthetic DNA, in restriction fragments, and in defined sequence nucleosomes. The current study utilized the polymerase chain reaction (PCR) to examine DEB damage frequencies within nuclear genes, found within "open" regions of chromatin, as compared to regions of unexpressed sequence that reside in tightly packed, "closed" chromatin, to more closely model DEB reactivity in vivo. These initial studies have been performed in chicken liver homogenates. Preliminarily, we have found a dose-dependent DEB lesion-forming response within "open" chromatin. DEB appears to have little-to-no effect upon regions of "closed" chromatin.

Gas-phase reactions of model carbosulfonium ions (CH3-S+?=?CH2; CH3CH2-S+?=?CH2 and Ph-S+?=?CH2) and an O-analogue carboxonium ion (CH3-O+?=?CH2) with acyclic (isoprene, 1,3-butadiene, methyl vinyl ketone) and cyclic (1,3-cyclohexadiene, thiophene, furan) conjugated dienes were systematically investigated by pentaquadrupole mass spectrometry. As corroborated by B3LYP/6-311?G(d,p) calculations, the carbosulfonium ions first react at large extents with the dienes forming adducts via simple addition. The nascent adducts, depending on their stability and internal energy, react further via two competitive channels: (1) in reactions with acyclic dienes via cyclization that yields formally [4?+?2+] cycloadducts, or (2) in reactions with the cyclic dienes via dissociation by HSR loss that yields methylenation (net CH+ transfer) products. In great contrast to its S-analogues, CH3-O+?=?CH2 (as well as C2H5-O+?=?CH2 and Ph-O+?=?CH2 in reactions with isoprene) forms little or no adduct and proton transfer is the dominant reaction channel. Isomerization to more acidic protonated aldehydes in the course of reaction seems to be the most plausible cause of the contrasting reactivity of carboxonium ions. The CH2?=?CH-O+?=?CH2 ion forms an abundant [4?+?2+] cycloadduct with isoprene, but similar to the behavior of such alpha,beta-unsaturated carboxonium ions in solution, seems to occur across the C?=?C bond. Due to the high price of natural oil and harmful effects of its usage, as the increase in emission of greenhouse gases, the industry focused in searching of sustainable types of the raw materials for production of chemicals. Ethanol, produced by fermentation of sugars, is one of the more interesting renewable materials for chemical manufacturing. There are numerous applications for the conversion of ethanol into commodity chemicals. In particular, the production of 1,3-butadiene whose primary source is ethanol using multifunctional catalysts is attractive. With the 25% of world rubber manufacturers utilizing 1,3-butadiene, there is an exigent need for its sustainable production. In this research, the conversion of ethanol in one-step process to 1,3-butadiene was studied. According to the literature, the mechanisms which were proposed to explain the way ethanol transforms into butadiene require to have both acid and basic sites. But still, there are a lot of debate on this topic. Thus, the aim of this research work is a better understanding of the reaction pathways with all the possible intermediates and products which lead to the formation of butadiene from ethanol. The particular interests represent the catalysts, based on different ratio Mg/Si in comparison to bare magnesia and silica oxides, in order to identify a good combination of acid/basic sites for the adsorption and conversion of ethanol. Usage of spectroscopictechniques are important to extract information that could be helpful for understanding the processes on the molecular level. The diffuse reflectance infrared spectroscopy coupled to mass spectrometry (DRIFT-MS) was used to study the surface composition of the catalysts during the adsorption of ethanol and its transformation during the temperature program. Whereas, mass spectrometry was used to monitor the desorbed products. The set of studied materials include MgO, Mg/Si=0.1, Mg/Si=2, Mg/Si=3, Mg/Si=9 and SiO2 which were also characterized by means of surface area measurements.

A causa delle questioni economiche ed ambientali legate alla sostenibilità dei processi petrolchimici, recentemente l'industria chimica ha focalizzato il proprio interesse nello sviluppo di processi per la produzione di chemicals, che utilizzino materiali di partenza rinnovabili. L'etanolo, prodotto per via fermentativa, sembra essere uno dei bio-building block più promettenti e versatili e può essere utilizzato per numerose applicazioni. È noto da tempo che l’etanolo può reagire su catalizzatori costituiti da ossidi misti con caratteristiche acido-base a dare numerosi composti chimici tra cui acetaldeide, 1,3-butadiene, 1-butanolo e 2-butenale. Nonostante il lungo impiego dell’etanolo nell’industria chimica, il meccanismo di formazione di composti C4 a partire da etanolo è ancora però materia di dibattito. Il meccanismo generalmente accettato si basa sulle seguenti reazioni chiave: deidrogenazione di etanolo ad acetaldeide e condensazione aldolica di due molecole di acetaldeide. Tuttavia in letteratura sono riportate anche altre proposte alternative. In questo lavoro è stato studiato il processo di trasformazione di etanolo su catalizzatori a base di MgO e sistemi misti Mg/SiO, attraverso esperimenti di reattività condotti in un micro-impianto da laboratorio, al fine di fare chiarezza sul meccanismo di formazione di composti C4 a partire da etanolo. In particolare è stato condotto uno studio meccanicistico utilizzando MgO come catalizzatore modello, materiale che possiede esclusivamente proprietà basiche, ritenute essenziali per catalizzare la condensazione di molecole C2. Inoltre, è stata investigata l’influenza delle caratteristiche acido-base del catalizzatore sulla selettività del processo di conversione di etanolo, studiandone la reattività su materiali costituiti da ossidi misti Mg/Si/O, con diverso rapporto atomico tra i due cationi.

Oggi il mercato mondiale dell'anidride maleica (AM) è in continua espansione grazie ad un numero sempre crescente di applicazioni finali e sviluppo di nuove tecnologie industriali. La AM viene impiegata nella produzione di resine poliestere insature e resine alchidiche e nella produzione di chemicals a più alto valore aggiunto. Il processo di sintesi è tutt’ora basato sull'ossidazione selettiva del benzene e del n-butano. Con l’aumento delle emissione di gas serra, legate all’impiego di materie di origine fossile e la loro continua diminuzione, si stanno studiando nuovi processi basati su materie derivanti da fonti rinnovali. Tra i vari processi studiati vi è quello per la sintesi di AM, i quali utilizzano come molecola di furfurale, 5-idrossimetilfurfurale e 1-butanolo; tutte queste presentano però il problema di un costo superiore rispetto alle molecole tutt’ora usate. Ad oggi una delle molecole ottenibili da fonti rinnovabili avente un costo competitivo alle materie derivanti da fonti fossili è il bio-etanolo. Essendo nota la possibilità di trasformare dell’etanolo in composti a 4 atomi di carbonio (C4) come 1-butanolo (reazione di Guerbet) e in 1,3- butadiene (processo Lebedev) su ossidi misti Mg/Si e la loro trasformazioni in AM, è’ dunque possibile ipotizzare un processo operante in fase gas che accoppi entrambi i processi. Lo scopo di questo lavoro di tesi è stato quello di effettuare uno studio su sistemi catalitici mediante differenti approcci impiantistici. Il primo ha previsto l’impiego di un sistema detto “a cascata” nel quale è stato accoppiato il sistema misto a base di Mg/Si/O, per la trasformazione dell’etanolo a C4, e il pirofosfato di vanadile (VPP), per l’ossidazione selettiva di quest’ultimi in AM. Il secondo approccio ha previsto l’impiego di un unico sistema multifunzionale in grado di catalizzare tutti gli step necessari. In quest’ultimo caso, i sistemi studiati sono stati il Mg2P2O7 ed un sistema costituito da VPP DuPont sul quale è stato depositato MgO. I catalizzatori sono stati caratterizzati mediante diffrattometria a raggi X, spettroscopia Raman e analisi dell’area superficiale mediante metodo BET, mentre i test catalitici sono stati condotti su un impianto di laboratorio con un reattore assimilabile ad un modello di tipo PFR.

Southeast Texas, including Houston, has a large presence of industrial facilities and has been documented to have poorer air quality and significantly higher cancer rates than the remainder of Texas. Given citizens’ concerns in this 4th largest city in the U.S., Mayor Bill White recently partnered with the UT School of Public Health to determine methods to evaluate the health risks of hazardous air pollutants (HAPs). Sexton et al. (2007) published a report that strongly encouraged analytic studies linking these pollutants with health outcomes. In response, we set out to complete the following aims: 1. determine the optimal exposure assessment strategy to assess the association between childhood cancer rates and increased ambient levels of benzene and 1,3-butadiene (in an ecologic setting) and 2. evaluate whether census tracts with the highest levels of benzene or 1,3-butadiene have higher incidence of childhood lymphohematopoietic cancer compared with census tracts with the lowest levels of benzene or 1,3-butadiene, using Poisson regression. The first aim was achieved by evaluating the usefulness of four data sources: geographic information systems (GIS) to identify proximity to point sources of industrial air pollution, industrial emission data from the U.S. EPA’s Toxic Release Inventory (TRI), routine monitoring data from the U.S. EPA Air Quality System (AQS) from 1999-2000 and modeled ambient air levels from the U.S. EPA’s 1999 National Air Toxic Assessment Project (NATA) ASPEN model. Further, once these four data sources were evaluated, we narrowed them down to two: the routine monitoring data from the AQS for the years 1998-2000 and the 1999 U.S. EPA NATA ASPEN modeled data. We applied kriging (spatial interpolation) methodology to the monitoring data and compared the kriged values to the ASPEN modeled data. Our results indicated poor agreement between the two methods. Relative to the U.S. EPA ASPEN modeled estimates, relying on kriging to classify census tracts into exposure groups would have caused a great deal of misclassification. To address the second aim, we additionally obtained childhood lymphohematopoietic cancer data for 1995-2004 from the Texas Cancer Registry. The U.S. EPA ASPEN modeled data were used to estimate ambient levels of benzene and 1,3-butadiene in separate Poisson regression analyses. All data were analyzed at the census tract level. We found that census tracts with the highest benzene levels had elevated rates of all leukemia (rate ratio (RR) = 1.37; 95% confidence interval (CI), 1.05-1.78). Among census tracts with the highest 1,3-butadiene levels, we observed RRs of 1.40 (95% CI, 1.07-1.81) for all leukemia. We detected no associations between benzene or 1,3-butadiene levels and childhood lymphoma incidence. This study is the first to examine this association in Harris and surrounding counties in Texas and is among the first to correlate monitored levels of HAPs with childhood lymphohematopoietic cancer incidence, evaluating several analytic methods in an effort to determine the most appropriate approach to test this association. Despite recognized weakness of ecologic analyses, our analysis suggests an association between childhood leukemia and hazardous air pollution.^

An exposure system was constructed to evaluate the performance of a personal organic vapor dosimeter (3520 OVM) at ppb concentrations of nine selected target volatile organic compounds (VOCs). These concentration levels are generally encountered in community air environments, both indoor and outdoor. It was demonstrated that the chamber system could provide closely-controlled conditions of VOC concentrations, temperature and relative humidity (RH) required for the experiments. The target experimental conditions included combinations of three VOC concentrations (10, 20 and 200 $\rm\mu g/m\sp3),$ three temperatures (10, 25 and 40$\sp\circ$C) and three RHs (12, 50 and 90% RH), leading to a total of 27 exposure conditions. No backgrounds of target VOCs were found in the exposure chamber system. In the exposure chamber, the variation of the temperature was controlled within $\pm$1$\sp\circ$C, and the variation of RH was controlled within $\pm$1.5% at 12% RH, $\pm$2% at 50% RH and $\pm$3% at 90% RH. High-emission permeation tubes were utilized to generate the target VOCs. Various patterns of the permeation rates were observed over time. The lifetimes and permeation rates of the tubes differed by compound, length of the tube and manufacturer. By carefully selecting the source and length of the tubes, and closely monitoring tube weight loss over time, the permeation tubes can be used for delivering low and stable concentrations of VOCs during multiple days.^ The results of this study indicate that the performance of the 3520 OVM is compound-specific and depends on concentration, temperature and humidity. With the exception of 1,3-butadiene under most conditions, and styrene and methylene chloride at very high relative humidities, recoveries were generally within $\pm$25% of theory, indicating that the 3520 OVM can be effectively used over the range of concentrations and environmental conditions tested with a 24-hour sampling period. Increasing humidities resulted in increasing negative bias from full recovery. Reverse diffusion conducted at 200 $\rm\mu g/m\sp3$ and five temperature/humidity combinations indicated severe diffusion losses only for 1,3-butadiene, methylene chloride and styrene under increased humidity. Overall, the results of this study do not support the need to employ diffusion samplers with backup sections for the exposure conditions tested. ^

Low molecular weight hydrocarbon (LMWH) distributions were examined in sediments from Sites 1109 and 1115 in the western Woodlark Basin using purge-trap thermal adsorption/desorption gas analysis. A number of different hydrocarbon components >C1, which were not detected during shipboard gas analysis, were detected at both sites using the purge-trap procedure. Concentrations of ethane, propane, and butane remained relatively low (<100 pmol/g) throughout Site 1109 and had no consistent trend with depth. In contrast, the longer-chain components increased in concentration with depth. Hexane concentrations rose to 716 pmol/g at the base of the site with a concomitant increase in both 2-methyl- and 3-methylpentane. At Site 1115, concentrations of ethane, propane, butane, and isobutylene + 1-butene remained low (<60 pmol/g) throughout the site and again had no consistent trend with depth. 2-Methylpentane, 3-methylpentane, and hexane concentrations had a subsurface maximum that coincided with sediments containing abundant plant-rich material. The LMWH downhole profiles plus low in situ temperatures suggest that the LMWH components were formed in situ by low-temperature biological processes. Purge-trap analysis has indicated the presence of some unexpected deep low-temperature bacterial reactions, which demonstrates that further analysis of LMWH may provide valuable information at future Ocean Drilling Program sites.

Esta Tesis Doctoral tiene como principal objetivo el obtener una cadena de tratamientos seguros de aguas seriados que nos permita asegurar la calidad de las aguas para consumo humano en caso de emergencias, de tal forma que se minimicen los efectos de acciones hostiles, como sabotajes o actos terroristas, desastres naturales, etc y buscar soluciones adecuadas para garantizar en este caso la salud. Las plantas de tratamientos de aguas existentes comercialmente no aseguran dicha calidad y la documentación sobre el tema presenta vacíos de conocimiento, contradicciones entre resultados de investigaciones o insostenibilidad de conclusiones de las mismas. Estas carencias nos permiten determinar los aspectos a tratar durante la investigación. Por ello, este objetivo se concretó en tres acciones: Investigar sobre rendimientos de plantas convencionales en eliminación de microorganismos y productos tóxicos y peligrosos. Introducir mejoras que garanticen el rendimiento de las plantas convencionales. Investigar sobre la conveniencia de complementar las instalaciones existentes buscando seguridad y garantía sanitaria. Y se desarrollaron tres líneas de investigación: LI 1 “Inorgánicos”: Investigación sobre la eliminación de los metales boro, cobre y molibdeno mediante procesos de intercambio iónico y de coagulaciónfloculación- decantación. LI 2 “Compuestos Orgánicos Volátiles”: Investigación sobre la eliminación de los compuestos orgánicos 1,1 dicloroetano, 1,2 dicloroetano, clorobenceno, 1,3 dicloropropeno y hexacloro 1,3 butadieno mediante procesos de carbón activo granular y de oxidación avanzada. LI 3 “Plantas portátiles”: Investigación sobre plantas existentes portátiles para verificar su rendimiento teórico y proponer mejoras. Estas líneas de investigación se desarrollaron tanto en el nivel teórico como en el empírico, bien sea en laboratorio como en campo. A lo largo del documento se demuestra que las principales fuentes de contaminación, salvo la degradación de yacimientos naturales, proceden de la actividad humana (efluentes industriales y agrícolas, aguas residuales y actividades beligerantes) que provocan un amplio espectro de enfermedades por lo que dificultan tanto la definición de la fuente como la anticipada detección de la enfermedad. Las principales conclusiones que se obtuvieron están relacionadas con el rendimiento de eliminación de los parámetros tras la aplicación de los procesos y plantas de tratamiento de aguas anteriormente reseñadas. Sin embargo, el verdadero elemento designador de originalidad de esta Tesis Doctoral, tal como se ha reseñado arriba, radica en la definición de un sistema seriado de procesos de tratamiento de aguas que asegura la calidad en caso de emergencia. Éste se define en el siguiente orden: pretratamiento, oxidación, coagulación-floculación-decantación, filtración por arena, intercambio iónico, carbón activo granular, microfiltración, radiación UV, ósmosis inversa, radiación UV y cloración final. The main objective of this Thesis is to obtain a chain of stepwise safe water treatments that allow us to ensure the quality of water for human consumption in case of emergencies, so that the effects of hostile actions, such as sabotage or terrorism, natural disasters, etc. and seek appropriate solutions in this case to ensure health. The existing commercial water treatment plants do not ensure quality, and the documentation on the subject presents knowledge gaps or contradictions. These gaps allow us to determine the issues to be discussed during the investigation. Therefore, this objective was manifested in three actions: Researching yields in commercial plants and microorganisms, or toxic and dangerous products removal. Improvements to ensure the performance of conventional plants. Inquire about the advisability of implementing existing facilities for safety and health guarantee. And three lines of research are developed: LI 1 “Inorganic elements”: Research removing metals iron, copper and molybdenum by ion exchange processes and coagulation-flocculation-decantation. LI 2 “Volatile Organic Compounds”: Research removing organic compounds 1,1 dichloroethane, 1,2 dichloroethane, chlorobenzene, 1,3-dichloropropene and 1,3-butadiene hexachloro through processes of granular activated carbon and advanced oxidation. LI 3 “Compact Water Treatment Plants”: Research on existing packaged plants to verify theoretical performance and suggest improvements. These lines of research are developed both theoretically and empirically, both in the laboratory and in the field. Throughout the document, it is evident that the main sources of pollution, other than the degradation of natural deposits, come from human activity (industrial and agricultural effluents, sewage and belligerent activities) which cause a broad spectrum of diseases which hamper both the definition of the source and the early detection of the disease. The main conclusions drawn are related to both the removal efficiency parameters after application of processes and treatment plants outlined above water. However, the real designator of originality of this thesis, such as outlined above, lies in the definition of a serial system water treatment processes assuring quality in case of emergency. This is defined in the following order: pretreatment, oxidation, coagulation-flocculation-sedimentation, sand filtration, ion exchange, granular activated carbon, microfiltration, UV radiation, reverse osmosis, UV radiation and final chlorination.

Isoprene synthase is the enzyme responsible for the foliar emission of the hydrocarbon isoprene (2-methyl-1,3-butadiene) from many C3 plants. Previously, thylakoid-bound and soluble forms of isoprene synthase had been isolated separately, each from different plant species using different procedures. Here we describe the isolation of thylakoid-bound and soluble isoprene synthases from a single willow (Salix discolor L.) leaf-fractionation protocol. Willow leaf isoprene synthase appears to be plastidic, with whole-leaf and intact chloroplast fractionations yielding approximately equal soluble (i.e. stromal) and thylakoid-bound isoprene synthase activities. Although thylakoid-bound isoprene synthase is tightly bound to the thylakoid membrane (M.C. Wildermuth, R. Fall [1996] Plant Physiol 112: 171–182), it can be solubilized by pH 10.0 treatment. The solubilized thylakoid-bound and stromal isoprene synthases exhibit similar catalytic properties, and contain essential cysteine, histidine, and arginine residues, as do other isoprenoid synthases. In addition, two regulators of foliar isoprene emission, leaf age and light, do not alter the percentage of isoprene synthase activity in the bound or soluble form. The relationship between the isoprene synthase isoforms and the implications for function and regulation of isoprene production are discussed.

Riassunto I biomarcatori o “marcatori biologici” svolgono un ruolo fondamentale nel monitoraggio biologico. In questo lavoro ci siamo soffermati sullo studio di biomarcatori di effetto e di esposizione a xenobiotici ambientali. Nel primo caso abbiamo valutato i micro RNA (miRNA) da utilizzare per la diagnosi precoce del tumore al polmone in matrici di facile accesso, quale il CAE e il plasma, utilizzando il miRNA-21, oncogeno, e il miRNA-486, oncosoppressore. I risultati evidenziano una loro capacità di distinguere correttamente i soggetti con tumore polmonare dai soggetti sani, ipotizzando un loro utilizzo a scopo diagnostico. Nella seconda parte del lavoro di tesi sono stati studiati i biomarcatori di esposizione a benzene per valutare gli effetti dell’esposizione a concentrazioni ambientali su bambini residenti in città e a diverso livello di urbanizzazione. Lo studio ha evidenziato una correlazione dose-effetto fra le concentrazioni di benzene e dei suoi metaboliti urinari e un danno ossidativo a livello degli acidi nucleici. Tuttavia, le concentrazioni di benzene urinario non sono influenzate dal grado di industrializzazione, a differenza dell’S-PMA e degli indicatori di stress ossidativo (8-oxodGuo e 8-oxoGuo) che sembrano risentire sia della residenza che del momento del campionamento. Infine abbiamo ricercato possibili biomarcatori di esposizione a vinilcicloesene (VCH), sottoprodotto industriale nella polimerizzazione del 1,3-butadiene, poiché non sono ancora stati proposti BEI di riferimento nonostante i bassi valori di TLV-TWA (0.1 ppm) proposti dall’ACGIH. Nella prima fase del lavoro abbiamo studiato i meccanismi di tossicità del VCH tramite modelli in vitro, testando varie linee cellulari. I risultati evidenziano come la dose reale di VCH sia di molto inferiore a quella nominale per effetto dell’evaporazione. Inoltre, nelle linee cellulari più sensibili si sono evidenziati effetti citostatici, con alterazioni del ciclo cellulare, a differenza dell’esposizione agli epossidi del VCH, il VCD e l’1,2-VCHME, che determinano lisi cellulare con IC50 di 3 ordini di grandezza inferiori a quelli del VCH. La quantificazione dei metaboliti di I fase e di II fase del VCH nelle linee cellulari epatiche ha evidenziato concentrazioni di circa 1000 volte inferiori a quelle del VCH confermando come la sua tossicità sia principalmente dovuta alla produzione degli intermedi epossidici. La trasformazione nei metaboliti di II fase conferma inoltre l’effetto detossificante del metabolismo. La trasferibilità dei risultati ottenuti in vitro su sistemi in vivo fornirà le basi per poter identificare possibili metaboliti da proporre per il monitoraggio biologico di lavoratori esposti a VCH. This thesis is primarily concerned with the synthesis and polymerization of 5-methyl-1;3, 2-dioxathiolan-4-one-2-oxide (lactic acid anhydrosulphite (LAAS)) using anionic initiators under various conditions. Poly(lactic acid) is a biodegradable polymer which finds many uses in biomedical applications such as drug-delivery and wound-support systems. For such applications it is desirable to produce polymers having predictable molecular weight distributions and crystallinity, The use of anionic initiators offers a potential route to the creation of living polymers. The synthesis of LAAS was achieved by means of an established route though the procedure was modified to some extent and a new method of purification of the monomer using copper oxides was introduced, Chromatographic purification methods were also examined but found to be ineffective. An unusual impurity was discovered in some syntheses and this was identified by means of 1H and 13C NMR, elemental analysis and GC-MS. Since poly-α-esters having hydroxyl-bearing substituents might be expected to have high equilibrium water contents and hence low surface tension characteristics which might aid bio-compatibility, synthesis of gluconic acid anhydrosulphite was also attempted and the product characterised by 1H and 13C NMR. The kinetics of the decomposition of lactic acid anhydrosulphite by lithium tert-butoxide in nitrobenzene has been examined by means of gas evolution measurements. The kinetics of the reaction with potassium tert-butoxide (and also sec-butyl lithium) in tetrahydrofuran has been studied using calorimetric techniques. LAAS was block co-polymerized with styrene and also with 1,3-butadiene in tetrahydrofuran (in the latter case a statistical co-polymer was also produced).

The burden of chronic diseases such as cancer is increasing in low and middle income countries around the globe. Nepal, one of the world’s poorest countries, is no exception to this trend, with lung cancer as the leading causes of cancer deaths. Despite this, limited data is available on the environmental and behavioral risk factors that contribute to the lung cancer etiology in Nepal. The objectives of this dissertation are to: 1) investigate the ethnic differences in consumption of local tobacco products and their role in lung cancer risk in Nepal; 2) evaluate urinary metabolite of 1,3-butadiene as a biomarker of exposure to combustion related household air pollution (CRHAP); 3) investigate the association between CRHAP exposure and lung cancer risk using urinary metabolite of 1,3-butadiene as a biomarker of exposure; 4) investigate the association between CRHAP exposure and lung cancer risk using questionnaire based measure of exposure. Lung cancer cases (n=606) and frequency matched controls (N=606) were recruited from B.P. Koirala Memorial Cancer Hospital. We obtained biological samples and information on lifestyles including cooking habits and type of fuels used. We used liquid chromatograph tandem mass spectrometer (LC-MS/MS) to quantify urinary metabolites of 1,3-butadiene in urine samples. We employed a combination of logistic and linear regression models to detect any exposure-disease associations while controlling for known confounding variables. Overall, we found that ethnic groups in Nepal use different tobacco products that have different differing cancer potency -we observed the highest odds ratios for the traditional tobacco products. The biomarker analysis showed strong evidence that monohydroxybutyl mercapturic acid is associated with biomass fuel use among participants. However, we did not find significant association between urinary MHMBA and lung cancer risk. When we used questionnaire based measure of exposure to household air pollution, we observed significant, dose-response associations between CRHAP exposure and lung cancer risk, particularly among never-smokers. Our results show that important role of local tobacco products in lung cancer risk in Nepal. Furthermore, we demonstrate that CRHAP exposure is a risk factor for lung cancer risk, independent of tobacco smoking.
