Artemia cysts (of GSL, Utah, USA origin) were produced from the modified traditional solar salt works of Bangladesh during winter months through different feeding/fertilization treatments (T1, T2, T3, T4 ) were analyzed to understand the effects of treatments on their fatty acid profile. Palmitic, Linolenic, Eicosapantaenonic and Docohexaenoic acids (mg/g. DW) were found highest for the cysts in T1 (16.0% ±1 .36), T2 (14.7% ±0.47), T3 (4.7% ±0.40) and T4 (0.7% ±0.06) treatments, respectively. High amount of 18:3(n-3) acids in the cysts of all sources proves to be freshwater type of the cysts. The presence of marine type essential fatty acids in the cysts of all sources were found low for 20:5n-3 (3.7-4.7%) and very low for 22:6n-3 (0.09-0.7%). No significant variation was observed for 16:0 acids within the treatments, but for 18:3(n-3) acid, the variation was found highly significant (P= 0.0052) between T2 and T4 treatments. For 20:5(n-3), only variation between T2 and T4 was found insignificant (P=0.1161), but between other treatments, significant variation was observed between T2 and T4 (P=0.0241), T2 and T4 (P=0.0022) and T1 andT4 (P=0.0161). No significant variation was found in other treatments.

Transmission imaging with an environmental scanning electron microscope (ESEM) (Wet STEM) is a recent development in the field of electron microscopy, combining the simple preparation inherent to ESEM work with an alternate form of contrast available through a STEM detector. Because the technique is relatively new, there is little information available on how best to apply this technique and which samples it is best suited for. This work is a description of the sample preparation and microscopy employed by the authors for imaging bacteria with Wet STEM (scanning transmission electron microscopy). Three different bacterial samples will be presented in this study: first, used as a model system, is Escherichia coli for which the contrast mechanisms of STEM are demonstrated along with the visual effects of a dehydration-induced collapse. This collapse, although clearly in some sense artifactual, is thought to lead to structurally meaningful morphological information. Second, Wet STEM is applied to two distinct bacterial systems to demonstrate the novel types of information accessible by this approach: the plastic-producing Cupriavidus necator along with wild-type and δmreC knockout mutants of Salmonella enterica serovar Typhimurium. Cupriavidus necator is shown to exhibit clear internal differences between bacteria with and without plastic granules, while the δmreC mutant of S. Typhimurium has an internal morphology distinct from that of the wild type. © 2012 Wiley Periodicals, Inc.

The decoupling of CAP payments leads production decisions and resources allocation to be more dependent on market prices and competitive advantages. The objective of this paper is to assess the effects of CAP trends on the montado/dehesa traditional ecosystem of Mediterranean regions in terms of farm income, land, labour and capital. A positive mathematical supply model disaggregated by the montado agro-forestry production systems of the Alentejo region in southern Portugal is developed. The results show that decoupling payments of CAP have negative economic effects on agricultural activities and resource use. Agricultural income increases with single farm payments but the foreseen increases in prices do not compensate the loss of the Agenda 2000 area payments in terms of competitiveness. These results reinforce the need to promote alternative agricultural and non-agricultural activities and policies in Mediterranean rural European areas and regions. European Agenda 2000 considers the integration of environment in CAP and the role farmers can play on natural resources management. Alternative tillage technologies to seed cereals play an important economic and environmental role on the development of a sustainable agriculture and its adoption may depend on the agricultural policy, particularly policies on income support. The results obtained clearly show that the use of alternative soil tillage technologies would be promoted by the change of supports from the first to the second pillar of CAP and we can conclude that, in general, this change will encourage a faster technological adoption. OBJECTIVE: To provide a detailed phenotype/genotype characterization of Bietti crystalline dystrophy (BCD). DESIGN: Observational case series. PARTICIPANTS: Twenty patients from 17 families recruited from a multiethnic British population. METHODS: Patients underwent color fundus photography, near-infrared (NIR) imaging, fundus autofluorescence (FAF) imaging, spectral domain optical coherence tomography (SD-OCT), and electroretinogram (ERG) assessment. The gene CYP4V2 was sequenced. MAIN OUTCOME MEASURES: Clinical, imaging, electrophysiologic, and molecular genetics findings. RESULTS: Patients ranged in age from 19 to 72 years (median, 40 years), with a visual acuity of 6/5 to perception of light (median, 6/12). There was wide intrafamilial and interfamilial variability in clinical severity. The FAF imaging showed well-defined areas of retinal pigment epithelium (RPE) loss that corresponded on SD-OCT to well-demarcated areas of outer retinal atrophy. Retinal crystals were not evident on FAF imaging and were best visualized with NIR imaging. Spectral domain OCT showed them to be principally located on or in the RPE/Bruch's membrane complex. Disappearance of the crystals, revealed by serial recording, was associated with severe disruption and thinning of the RPE/Bruch's membrane complex. Cases with extensive RPE degeneration (N = 5) had ERGs consistent with generalized rod and cone dysfunction, but those with more focal RPE atrophy showed amplitude reduction without delay (N = 3), consistent with restricted loss of function, or that was normal (N = 2). Likely disease-causing variants were identified in 34 chromosomes from 17 families. Seven were novel, including p.Met66Arg, found in all 11 patients from 8 families of South Asian descent. This mutation appears to be associated with earlier onset (median age, 30 years) compared with other substitutions (median age, 41 years). Deletions of exon 7 were associated with more severe disease. CONCLUSIONS: The phenotype is highly variable. Several novel variants are reported, including a highly prevalent substitution in patients of South Asian descent that is associated with earlier-onset disease. Autofluorescence showed sharply demarcated areas of RPE loss that coincided with abrupt edges of outer retinal atrophy on SD-OCT; crystals were generally situated on or in the RPE/Bruch's complex but could disappear over time with associated RPE disruption. These results support a role for the RPE in disease pathogenesis.

Introduction:¦L'incidence annuelle du cancer de l'ovaire en Suisse est de 600 cas, il touche principalement les femmes âgées de plus de 60 ans. Le cancer de l'ovaire est aujourd'hui la 1ère cause de mortalité par cancer gynécologique chez la femme. Le but de notre recherche, est de créer une base de données de toutes les¦patientes atteintes d'un cancer de l'ovaire et hospitalisées au CHUV pour la prise en charge de leur maladie.¦Cette étude rétrospective monocentrique nous permettra en premier lieu d'analyser les caractéristiques de¦cette tumeur présentées par les patientes, les traitements instaurés pour traiter ce cancer et les taux de¦récidives et de survie des patientes en fonction de ces variables.¦Méthodologie:¦Analyse rétrospective de 147 patientes diagnostiquées d'un cancer de l'ovaire et hospitalisées au CHUV entre¦septembre 2001 et mars 2010 pour la prise en charge de leur tumeur ovarienne. Utilisation du programme informatique ARCHIMED qui contient les dossiers médicaux de toutes les patientes inclues dans l'étude et création de 2 bases des données. La 1ère base de données regroupe l'ensemble des patientes de l'étude y¦compris les tumeurs type borderline, la 2ème base de données concerne uniquement les patientes ayant récidivé de leur tumeur. Les tumeurs bénignes ont été exclues de l'étude.¦Résultats:¦La probabilitéde survie à 1 an chez les patientes avec un cancer de l'ovaire, tous stades FIGO et prises en¦charge confondus, hospitalisées au CHUV est de 88,04% (95% CI = 0.7981-‐0.9306), à 3 ans la probabilité de survie est de 70,4% (95% CI = 0.5864-‐0.7936), et à plus de 5 ans, elle est de 60% (95% CI = 0.4315-‐0.6859).¦Nous avons comparé le taux de survie en fonction du stade FIGO de la tumeur ovarienne et nous avons observé une différence significative de survie entre les stades FIGO précoces et les stades avancés (Pvaleur=¦0.0161).¦En plus d'une intervention chirurgicale, les patientes atteintes d'un cancer de l'ovaire sont normalement traitées par une chimiothérapie. Dans notre étude, 70 patientes ont bénéficié d'une chimiothérapie; un¦traitement adjuvant a été donné dans 78 % des cas (N = 55), un traitement néoadjuvant a été administré chez 22% des patientes (N=15). Le type combiné carboplatine-‐taxane est la chimiothérapie la plus fréquente (75%). Au total sur l'étude, 66 patientes sur les 147 (44%) ont récidivé de leur tumeur. En ce qui¦concerne leur prise en charge, 46% des patientes ont reçu une chimiothérapie unique comme du gemzar, cealyx ou taxotère après leur récidive. Une cytoréduction secondaire a également été effectuée chez 33% de ces patientes ayant une récidive. Nous avons également étudié l'intervalle de temps entre la date de la¦récidive et celle du décès. Parmi les 28 patientes décédées chez les récidives, 10 d'entre-‐elles (36%) ont survécu moins d'un an une fois la récidive diagnostiquée, 8 (28%) patientes ont survécu jusqu'à 2 ans, et¦les 10 (36%) autres patientes ont survécu de 2 à 5 ans. En ce qui concerne le taux de mortalité; 39 patientes sur les 147 étudiées sont décédées pendant la période d'observation, soit 26% des cas. La tumeur¦type borderline, présente une prolifération épithéliale atypique sans invasion dans le stroma et représente¦10 à 20% de toutes les tumeurs ovariennes. Dans notre étude, 41 patientes sont porteuses de cette tumeur¦(28%) et la moyenne d'âge est de 49 ans. En ce qui concerne leur prise en charge, l'intervention chirurgicale¦la plus fréquente, soit 23% des cas, est l'annexectomie unilatérale, qui reste une attitude conservative pour¦ces patientes désirant préserver leur fertilité. 6 patientes présentant ce type de tumeur ont récidivé, soit 14% des cas, avec une progression pelvienne, et 3 de ces 6 patientes sont décédées. Dans notre analyse, on observe que la probabilité de vivre plus longtemps que 1an pour les patientes ayant une tumeur borderline est de 93,8% (95% CI= 0.6323-‐ 0.9910), à 3 ans elle est également de 93,8% (95% CI = 0.6323-‐0.9910) et à 5 ans elle est de 78,1% (95% CI = 0.3171-‐0.9483). Nous n'avons pas observé de¦différence de survie dans notre étude entre les patientes présentant une tumeur borderline et le « non‐borderline ». (Pvaleur=0.3301)

Study Objectives: Interspecific variation in sleep measured in captivity correlates with various physiological and environmental factors, including estimates of predation risk in the wild. However, it remains unclear whether prior comparative studies have been confounded by the captive recording environment. Herein we examine the impact of predation pressure on sleep in sloths living in the wild. Design: Comparison of two closely related sloth species, one exposed to predation and one free from predation. Setting: Panamanian mainland rainforest (predators present) and island mangrove (predators absent). Participants: Mainland (Bradypus variegatus, 5 males and 4 females) and island (Bradypus pygmaeus, 6 males) sloths. Interventions: None. Measurements and Results: EEG and EMG activity were recorded using a miniature data logger. Although both species spent between 9 and 10 hours per day sleeping, the mainland sloths showed a preference for sleeping at night, whereas island sloths showed no preference for sleeping during the day or night. EEG activity during NREM sleep showed lower low-frequency power, and increased spindle and higher frequency power in island sloths when compared to mainland sloths. Conclusions: In sloths sleeping in the wild, predation pressure influenced the timing of sleep, but not the amount of time spent asleep. The preference for sleeping at night in mainland sloths may be a strategy to avoid detection by nocturnal cats. The pronounced differences in the NREM sleep EEG spectrum remain unexplained, but might be related to genetic or environmental factors.

The potential of ochratoxin A (OTA) to damage brain cells was studied by using a three-dimensional cell culture system as model for the developing brain. Aggregating cell cultures of foetal rat telencephalon were tested either during an early developmental period, or during a phase of advanced maturation, over a wide range of OTA concentrations (0.4 nM to 50 microM). By monitoring changes in activities of cell type-specific enzymes (ChAt and GAD, for cholinergic and GABAergic neurones, respectively, GS for astrocytes and CNP for oligodendrocytes), the concentration-dependent toxicity and neurodevelopmental effects of OTA were determined. OTA proved to be highly toxic, since a 10-day treatment at 50 nM caused a general cytotoxicity in both mature and immature cultures. At 10 nM of OTA, cell type-specific effects were observed: in immature cultures, a loss in neuronal and oligodendroglial enzyme activities, and an increase in the activity of the astroglial marker glutamine synthetase were found, Furthermore, at 2 and 10 nM of OTA, a clustering of microglial cells was observed. In mature cultures, OTA was somewhat less potent, but caused a similar pattern of toxic effects. A 24 h-treatment with OTA resulted in a concentration-dependent decrease in protein synthesis, with IC50 values of 25 nM and 33 nM for immature and mature cultures respectively. Acute (24 h) treatment at high OTA concentrations (10 to 50 microM) caused a significant increase in reactive oxygen species formation, as measured by the intracellular oxidation of 2',7'-dichlorofluorescin. These results suggest that OTA has the potential to be a potent toxicant to brain cells, and that its effects at nanomolar concentrations are primarily due to the inhibition of protein synthesis, whereas ROS seem not to be involved in the toxicity mediated by a chronic exposure to OTA at such low concentrations.
