随着后基因组时代的到来以及蛋白质组学研究的深入开展,研究蛋白质晶体生长成为生物化学和结构生物学领域一个广泛关注的课题。通过使用原子力显微镜(Atomic Force Microscope,简称AFM)对杜仲抗真菌蛋白(eucommia antifungal protein,简称EAFP)的晶体在有母液存在下原位实时动态地进行了晶面生长观察。研究结果表明:不同过饱和度对EAFP晶体生长形貌的影响较大,较高的过饱和度下生长很快,生长台阶密度高,较高的过饱和度下主要进行各向异性二维台阶的发生、发展,较低的过饱和度下主要采用螺旋位错的生长方式,当过饱和度极低时生长缓慢,且晶体表面有很多小孔存在,晶面生长很不完整;还对不同过饱和度下晶体生长速率进行了定量的测量,也反映了过饱和度对EAFP晶体生长的影响;同时对在AFM观察过程中由探针的扫描速度和方向对表面形貌的影响进行了讨论。

There is a wealth of literature dealing with fish gills (Review, see Hoar & Randall, 1984), yet hardly anything is known about the gills of cephalopods. This is rather surprising considering the commercial importance of the cephalopods. In view of the paucity of information available it was necessary to start by establishing the morphology of the gills. This is covered in the first section of this thesis. Of all the cephalopods, Octopus vulgaris was singled out for more detailed investigation (see chapters 2 & 3) as its physiology is comparatively well understood (Wells, 1978). The gills of cephalopods are the major sites for respiratory gaseous exchange. It follows that their dimensions might be expected to govern their potential for absorbing oxygen. Section two deals with the morphometries of cephalopod gills, and predicted values are compared with physiological measurements of oxygen uptake for four representative The final section describes the physiological experiments I performed on octopuses. These experiments were designed to find out whether the animals could regulate the gills' potential to take up oxygen through changes to the gills themselves.

Paralarval and juvenile cephalopods collected in plankton samples on 21 western North Atlantic cruises were identified and enumerated. The 3731 specimens were assigned to 44 generic and specific taxa. This paper describes their spatial and temporal distributions and their developmental morphology. The smallest paralarvae recognized for a number of species are identified and illustrated. The two most abundant and most frequently collected taxa were identifiable to species based on known systematic characters of young, as well as on distribution of the adults. These were the neritic squids Loligo pealeii and Illex illecebrosus collected north of Cape Hatteras, both valuable fishery resources. Other abundant taxa included two morphotypes of ommastrephids, at least five species of enoploteuthids, two species of onychoteuthids, and unidentified octopods. Most taxa were distributed widely both in time and in space, although some seasonal and mesoscale-spatial patterns were indicated. The taxa that appeared to have distinct seasonal distribution included most of the neritic species and, surprisingly, the young of the bathypelagic cranchiids. In eight seasonal cruises over the continental shelf of the middle U.S. Atlantic states, neritic taxa demonstrated approximately the same seasonal patterns during two consecutive years. Interannual differences in the oceanic taxa collected on the shelf were extreme. The highest abundance and diversity of planktonic cephalopods in the oceanic samples were consistently found in the vicinity of the Gulf Stream. Only eight of the oceanic taxa appeared to have limited areal distributions, compared with twelve taxa that were found throughout the western North Atlantic regions sampled in this study. Many taxa, however, were not collected frequently enough to describe seasonal or spatial patterns. Comparisons with published accounts of other cephalopod surveys indicate both strengths and weaknesses in various sampling techniques for capturing the young of oceanic cephalopods. Enoploteuthids were abundant both in our study and in other studies using midwater trawls in several areas of the North Atlantic. Thus, this family probably is adequately sampled over its developmental range. In contrast, octopoteuthids and chtenopterygiids are rare in collections made by small to medium-sized midwater trawls but are comparatively common in plankton samples. For families that are relatively common in plankton samples, paralarval abundance, derived similarly to the familiar ichthyoplankton surveys of fisheries science, may be the most reliable method of gathering data on distribution and abundance. 固体表面的性质对蛋白质的吸附具有很大的影响作用,蛋白质在固体表面的吸附可分为共价吸附和非共价吸附.非共价吸附主要包括氢键、疏水力和静电力三种相互作用.其中静电相互作用对蛋白质吸附的影响仍未有一致的看法.该文将椭偏显微成像系统与电化学方法联用来研究静电相互作用在蛋白质吸附中的影响.在0.1MKH<,2>PO<,4>-Na<,2>HPO<,4>(pH=7.80)溶液中,金膜表面零电荷电势(PZC)在-0.60V(vs.SCE)左右.开路电位为+0.10V(vs.SCE),大于零电荷电势(PZC),说明金膜表面带正电荷.假设蛋白质在金表面表面吸附后,没有蛋白质吸附区域的单位面积的电容与吸附前时的单位面积的电容相同.计算出在FIB吸附达到饱和后实际蛋白质真正占据的比表面仅为36.7%.
Phage M13Ko7 Detection With Biosensor Based On Imaging Ellipsometry And Afm Microscopic Confirmation

A rapid detection and identification of pathogens is important for minimizing transfer and spread of disease. A label-free and multiplex biosensor based on imaging ellipsometry (BIE) had been developed for the detection of phage M13KO7. The surface of silicon wafer is modified with aldehyde, and proteins can be patterned homogeneously and simultaneously on the surface of silicon wafer in an array format by a microfluidic system. Avidin is immobilized on the surface for biotin-anti-M13 immobilization by means of interaction between avidin and biotin, which will serve as ligand against phage M13KO7. Phages M13KO7 are specifically captured by the ligand when phage M13KO7 solution passes over the surface, resulting in a significant increase of mass surface concentration of the anti-M13 binding phage M13KO7 layer, which could be detected by imaging ellipsometry with a sensitivity of 10(9) pfu/ml. Moreover, atomic force microscopy is also used to confirm the fact that phage M13KO7 has been directly captured by ligands on the surface. It indicates that BIE is competent for direct detection of phage M13KO7 and has potential in the field of virus detection. (C) 2008 Elsevier B.V. All rights reserved.

Collector-type experiments have been conducted to investigate two different aspects of sputtering induced by keV ions. The first study looked for possible ejection mechanisms related to the primary charge state of the projectile. Targets of CsI and LiNbO_3 were bombarded with 48 keV Ar^(q+), and a Au target was bombarded with 60 keV Ar^(q+), for q = 4, 8, and 11. The collectors were analyzed using heavy-ion Rutherford backscattering spectroscopy to determine the differential angular sputtering yields; these and the corresponding total yields were examined for variations as a function of projectile charge state. For the Au target, no significant changes were seen, but for the insulating targets slight (~10%) enhancements were observed in the total yields as the projectile charge state was increased from 4+ to 11+.
In the second investigation, artificial ^(92)Mo/^(100)Mo targets were bombarded with 5 and 10 keV beams of Ar^+ and Xe^+ to study the isotopic fractionation of sputtered neutrals as a function of emission angle and projectile fluence. Using secondary ion mass spectroscopy to measure the isotope ratio on the collectors, material ejected into normal directions at low bombarding fluences (~ 10^(15) ions cm^(-2)) was found to be enriched in the light isotope by as much as ~70‰ compared to steady state. Similar results were found for secondary Mo ions sputtered by 14.5 keV O^-. For low-fluence 5 keV Xe^+ bombardment, the light-isotope enrichment at oblique angles was ~20‰ less than the corresponding enrichment in the normal direction. No angular dependence could be resolved for 5 keV Ar^+ projectiles at the lowest fluence. The above fractionation decreased to steady-state values after bombarding fluences of a few times 10^(16) ions cm^(-2) , with the angular dependence becoming more pronounced. The fractionation and total sputtering yield were found to be strongly correlated, indicating that the above effects may have been related to the presence of a modified target surface layer. The observed effects are consistent with other secondary ion measurements and multiple-interaction computer simulations, and are considerably larger than predicted by existing analytic theory.

Sputtering yields for uranium metal under bombardment by 13 - 120 keV protons and by 20 - 120 keV He+ are presented. Angular distributions of the material sputtered by these ions are also given. Sputtering yields for 40 and 80 keV Ar+ were measured as well.
The technique employed to make these measurements was the detection of fission tracks in mica produced by ^(235)U sputtered onto collector foils which were subsequently exposed to a high fluence of thermal neutrons. The technique is extremely sensitive and allowed the measurement of sputtering yields less than 10^(-4) atoms per ion. It also made possible a detailed study of the emission of chunks from the uranium targets during sputtering. Mass distributions of chunks emitted during bombardment by 40 - 120 keV protons and by 80 keV argon are presented.
Comparisons are made between the experimental results and those predicted by the Sigmund theory of sputtering.

In this paper, some observations are made following a flash-flood that occurred in Stake Clough, a small tributary of the River Goyt, during the evening of 6 August 1996. The site was visited eight times between 8 August - 30 October 1996 to take samples and make observations on the stream. The flood scoured the bed of Stake Clough but more significantly, caused it to change course along the middle part of the floodplain. Initially after the flood, the numbers of insects in all stretches of the stream channel were low (100-200 m super(2)), but then gradually rose to population densities approaching ten times this figure. The benthos was dominated by the Chironomidae and also leuctrid stoneflies (Leuctra nigra, L. hippopus and L. inermis). On 8th August 1996, 12 mesh bags, each containing oak leaves, were placed in the stream and collected after 24 hours. These were also dominated by chironomids, and contained relatively high numbers of the caddis, Potamophylax cingulatus.

Part I. Novel composite polyelectrolyte materials were developed that exhibit desirable charge propagation and ion-retention properties. The morphology of electrode coatings cast from these materials was shown to be more important for its electrochemical behavior than its chemical composition.
Part II. The Wilhelmy plate technique for measuring dynamic surface tension was extended to electrified liquid-liquid interphases. The dynamical response of the aqueous NaF-mercury electrified interphase was examined by concomitant measurement of surface tension, current, and applied electrostatic potential. Observations of the surface tension response to linear sweep voltammetry and to step function perturbations in the applied electrostatic potential (e.g., chronotensiometry) provided strong evidence that relaxation processes proceed for time-periods that are at least an order of magnitude longer than the time periods necessary to establish diffusion equilibrium. The dynamical response of the surface tension is analyzed within the context of non-equilibrium thermodynamics and a kinetic model that requires three simultaneous first order processes.
