Structural and functional information encoded in DNA combined with unique properties of nanomaterials could be of use for the construction of novel biocomputational circuits and intelligent biomedical nanodevices. However, at present their practical applications are still limited by either low reproducibility of fabrication, modest sensitivity, or complicated handling procedures. Here, we demonstrate the construction of label-free and switchable molecular logic gates that use specific conformation modulation of a guanine- and thymine- rich DNA, while the optical readout is enabled by the tunable alphabetical metamaterials, which serve as a substrate for surface enhanced Raman spectroscopy (MetaSERS). By computational and experimental investigations, we present a comprehensive solution to tailor the plasmonic responses of MetaSERS with respect to the metamaterial geometry, excitation energy, and polarization. Our tunable MetaSERS-based DNA logic is simple to operate, highly reproducible, and can be stimulated by ultra-low concentration of the external inputs, enabling an extremely sensitive detection of mercury ions.

Despite considerable advances in reducing the production of dioxin-like toxicants in recent years, contamination of the food chain still occasionally occurs resulting in huge losses to the agri-food sector and risk to human health through exposure. Dioxin-like toxicity is exhibited by a range of stable and bioaccumulative compounds including polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs), produced by certain types of combustion, and man-made coplanar polychlorinated biphenyls (PCBs), as found in electrical transformer oils. While dioxinergic compounds act by a common mode of action making exposure detection biomarker based techniques a potentially useful tool, the influence of co-contaminating toxicants on such approaches needs to be considered. To assess the impact of possible interactions, the biological responses of H4IIE cells to challenge by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in combination with PCB-52 and benzo-a-pyrene (BaP) were evaluated by a number of methods in this study. Ethoxyresorufin-O-deethylase (EROD) induction in TCDD exposed cells was suppressed by increasing concentrations of PCB-52, PCB-153, or BaP up to 10 mu M. BaP levels below 1 mu M suppressed TCDD stimulated EROD induction, but at higher concentrations, EROD induction was greater than the maximum observed when cells were treated with TCDD alone. A similar biphasic interaction of BaP with TCDD co-exposure was noted in the AlamarBlue assay and to a lesser extent with PCB-52. Surface enhanced laser desorption/ionization-time of flight mass spectrometry (SELDI-TOF) profiling of peptidomic responses of cells exposed to compound combinations was compared. Cells co-exposed to TCDD in the presence of BaP or PCB-52 produced the most differentiated spectra with a substantial number of non-additive interactions observed. These findings suggest that interactions between dioxin and other toxicants create novel, additive, and non-additive effects, which may be more indicative of the types of responses seen in exposed animals than those of single exposures to the individual compounds.

A utilização combinada de espectroscopia vibracional e de cálculos envolvendo a teoria do funcional de densidade (DFT) possibilita o estudo de ligações de hidrogénio em fase condensada, assim como a análise da estrutura molecular dos sistemas em estudo. Por um lado, a espectroscopia vibracional permite a detecção de associações moleculares, enquanto os métodos computacionais auxiliam na obtenção de informação referente aos mecanismos de associação, nomeadamente no que diz respeito à possível estrutura de dímeros e compostos de inclusão em ciclodextrinas e às energias de interacção e de inclusão. O estudo que originou a presente dissertação pretende contribuir para o reforço da aplicação de estudos espectroscópicos e computacionais na elucidação de diversos fenómenos químicos, com especial destaque para o papel desempenhado por interacções intermoleculares fracas na estrutura e propriedades de materiais moleculares. No âmbito desta tese foram investigados os seguintes tópicos: polimorfismo e pseudopolimorfismo em sólidos farmacêuticos, transições de fase em misturas binárias de ácidos gordos, inclusão em ciclodextrinas, interacção de compostos farmacêuticos com superfícies metálicas e formação de agregados de água em materiais híbridos orgânicos-inorgânicos. Os sistemas foram analisados utilizando a espectroscopia vibracional – particularmente a espectroscopia de difusão de Raman – como técnica fundamental. Para uma melhor caracterização de processos envolvendo transições de fase, foram efectuados estudos com variação de temperatura, variação de humidade relativa e substituição isotópica. O estudo da interacção com superfícies metálicas foi realizado por espectroscopia de Raman intensificada à superfície. Dada a complexidade dos sistemas em estudo, a informação obtida por espectroscopia vibracional foi complementada por resultados de cálculos mecânico-quânticos. Em particular, os cálculos DFT foram utilizados para a optimização de geometrias e previsão de frequências vibracionais de moléculas e associações moleculares, permitindo assim a análise e interpretação de espectros vibracionais e a caracterização da estrutura de materiais.

Esta tese descreve diversas estratégias preparativas assim como estudos de caracterização de nanocompósitos e outras nanoestruturas, para a análise em espectroscopia de Raman com intensificação por superfície (SERS). Em particular foi estudada a incorporação de nanopartículas (NPs) metálicas de Ag e Au em matrizes poliméricas visando avaliar o potencial destes materiais como novos substratos para SERS na deteção de moléculas. O primeiro capítulo consiste numa revisão bibliográfica, onde é destacado o desenvolvimento de novos substratos para SERS utilizando nomeadamente nanopartículas de Ag, Au e Cu. Numa primeira fase, esta secção apresenta uma breve descrição sobre as propriedades plasmónicas dos metais utilizados e alguns conceitos básicos de espectroscopia de difusão de Raman. Posteriormente, descreve-se em mais detalhe o efeito de SERS, revendo-se sobretudo a química de materiais descrita em trabalhos recentes tendo em conta a sua utilização como novas plataformas para análise química por SERS. O capítulo 2 descreve a síntese e caracterização de nanocompósitos de poli(acrilato de butilo) obtidos através de polimerização in situ por miniemulsão na presença de nanopartículas de Ag. Os nanocompósitos Ag/PtBA foram investigados como novos substratos de SERS visando o desenvolvimento de novas plataformas versáteis para deteção molecular. Estudos como o efeito da temperatura, pH e pressão foram investigados, visando a compreensão do efeito da matriz polimérica na difusão/adsorção do analito (ácido tiossalicílico) na superfície metálica. No capítulo 3, os nanocompósitos de Ag/PtBA descritos anteriormente foram investigados detalhadamente como substratos para bio-deteção em SERS, usando a adenina como analito modelo. Os nanocompósitos foram submetidos a vários tratamentos pré-analíticos para a bio-deteção da adenina. Foram realizadas experiências nos sistemas Ag/PtBA de modo a obter informação sobre o efeito do pH na deteção deste analito em soluções aquosas diluídas. Os nanocompósitos poliméricos obtidos apresentam a vantagem de poderem ser processados utilizando tecnologia disponível. Pelo que o estudo das suas propriedades térmicas é especialmente relevante. Assim, a influência da incorporação de NPs inorgânicas na temperatura de transição vítrea (Tg) do polímero PtBA foi investigada por meio de calorimetria diferencial de varrimento (DSC) e os resultados são apresentados no capítulo 4. Estes estudos descrevem efeitos na Tg do PtBA quando analisado em nanocompósitos obtidos por diferentes métodos (métodos ex situ e in situ), contendo nanopartículas metálicas com diferentes distribuições de tamanho e presentes em quantidade variável na matriz polimérica. Estes estudos possibilitaram relacionar a influência das NPs metálicas na mobilidade das cadeias poliméricas com as propriedades térmicas observadas. O capítulo 5 descreve a preparação e caracterização de materiais compósitos de base polimérica (PtBA) e NPs de Au com morfologia de bastonetes (NRs). Estes materiais foram investigados como substratos para SERS tendo originado um bom sinal de SERS na análise de 2-2’-ditiodipiridina. Investigouse igualmente o efeito da variação da morfologia das NPs metálicas (esferas e bastonetes), a razão de aspecto (R.A.) dos bastonetes e o tipo de matriz polimérica (PtBA e PnBA) no sinal de SERS. No capítulo 6 é descrita a utilização da técnica de SERS como método alternativo para a monitorização de alterações morfológicas de coloides de NRs de Au. Os NRs de Au foram recolhidos em diferentes fases de oxidação promovida pela presença de K2S2O8 e a sua sensibilidade como substratos de SERS foi avaliada utilizando o anião dietilditiocarbamato (DTC) como analito modelo. Os estudos foram realizados utilizando as linhas de excitação a 1064 nm e 633 nm. Este estudo demonstrou que a sensibilidade dos NRs de Au como substratos de SERS diminui à medida que a sua R.A. diminui devido à competitividade do CTAB (estabilizante) e o DTC pela superfície dos NRs. É de salientar que este processo é acompanhado por um diferente comportamento em termos de adsorção dos dois tautómeros do DTC à superfície do metal. O capítulo 7 introduz um novo tipo de compósitos para SERS, utilizando matrizes biopoliméricas. Assim, descreve-se a preparação e caracterização de nanocompósitos de carragenano e NPs de Ag. Nesta secção é avaliada a utilização destes materiais como novos substratos para a análise em SERS, utilizando a 2-2’-ditiodipiridina como molécula modelo. Descrevem-se estudos pioneiros que procuram relacionar a dependência do sinal de SERS com a força do gel. Para tal, realizou-se um estudo sistemático aos nanobiocompósitos usados como substratos de SERS em diferentes condições analíticas e investigaram-se as suas propriedades reológicas. No capitulo 8 é descrita a investigação de nanocompósitos de Ag/Gelatina como substratos para SERS, utilizando o anião dietilditiocarbamato como analito modelo. Realizaram-se várias experiências para correlacionar a variação da força do gel com o aumento do sinal de SERS bem como a diferente adsorção dos dois tautómeros do DTC à superfície do metal. Ao longo desta dissertação são apresentados metodologias distintas para a preparação e obtenção de nanocompósitos com base em polímeros (sintéticos ou naturais) e NPs metálicas (Ag e Au). Esta investigação não só permitiu a síntese e estudo de novos substratos para SERS mas também a compreensão do efeito matriz/NPs metálicas no sinal de SERS e na formação de “hot spots”. Este trabalho contribui para o enriquecimento na área da Nanociência e Nanotecnologia demonstrando a eficácia e reprodutibilidade de nanocompósitos com base em polímeros como novos substratos para SERS. Embora as propriedades óticas apresentadas por estes materiais serem aqui direcionadas para a deteção molecular pela técnica de SERS, estes materiais podem ser investigados em outras área tecnológicas.

The present work aimed to explore the potential of new nanocomposites based on carbon nanostructures and metal nanoparticles for the detection of biomolecules through surface enhanced Raman scattering (SERS). In a first step, polyvinyl alcohol composites were prepared incorporating silver nanoparticles by two different reduction procedures. At first without introduction of carbon nanostructures. These composites showed good results for the SERS identification of nucleic acids. Next, the synthesis and characterization of graphene oxide was studied to be used in the preparation of silver and gold nanocomposites. The reduction of this nanomaterial with different chemical agents was explored, since its reduction degree may be a determinant factor for the application envisaged (biomolecules interaction). The preparation of the nanocomposites with silver and gold was performed with different reducing agents. The SERS activity of these new nanocomposites was then explored in the presence of different analytes, varying the experimental conditions for Raman spectra acquisition. It was interesting to verify that the silver containing nanocomposites presented the particularity to intensify the graphene D and G bands. It is also important to highlight that a new eco-friendly reducing agent was tested for the synthesis of the graphene oxide composites, an Eucalyptus Globulus extract. Other variable introduced was the preparation of gold nanostars synthesized with hydroxylamine in the presence of graphene oxide, which allowed the preparation of a new nanocomposite with SERS potential. Fibrous membranes were also prepared by electrospinning with the aim to prepare SERS supports with adequate topography and porosity for the formation of nanoparticles agglomerates for the creation of the so-called hot-spots and also to allow the penetration of the analyte molecules. The polymers polyvinyl alcohol and polyacrylonitrile were selected for electrospinning. Using this technique, electrospun mantles with silver and gold nanoparticles and nanocomposites were prepared. Several variables were studied, such as the introduction of the nano-fillers during the electrospinning process, later deposition of the nano-fillers on the simple electrospun polymeric fibres and surface functionalization of the simple polymeric membranes to link the nano-fillers. At last, the potentialities of using carbon nanotubes forests, produced by chemical vapor deposition and coated with gold film by sputtering, as new SERS substrates were explored. It was found that the SERS detection of DNA bases and ADN itself is possible using these substrates.

Les liposomes sont des structures sphériques formés par l'auto-assemblage de molécules amphiphiles sous forme d'une bicouche. Cette bicouche sépare le volume intérieur du liposome du milieu extérieur, de la même manière que les membranes cellulaires. Les liposomes sont donc des modèles de membranes cellulaires et sont formulés pour étudier les processus biologiques qui font intervenir la membrane (transport de molécules à travers la membrane, effets des charges en surface, interactions entre la matrice lipidique et d'autres molécules, etc.). Parce qu'ils peuvent encapsuler une solution aqueuse en leur volume intérieur, ils sont aussi utilisés aujourd'hui comme nanovecteurs de principes actifs. Nous avons formulé des liposomes non-phospholipidiques riches en stérol que nous avons appelés stérosomes. Ces stérosomes sont composés d'environ 30 % d'amphiphiles monoalkylés et d'environ 70 % de stérols (cholestérol, Chol, et/ou sulfate de cholestérol, Schol). Quand certaines conditions sont respectées, ces mélanges sont capables de former une phase liquide ordonnée (Lo) pour donner, par extrusion, des vésicules unilamellaires. Certaines de ces nouvelles formulations ont été fonctionnalisées de manière à libérer leur contenu en réponse à un stimulus externe. En incorporant des acides gras dérivés de l’acide palmitique possédant différents pKa, nous avons pu contrôler le pH auquel la libération débute. Un modèle mathématique a été proposé afin de cerner les paramètres régissant leur comportement de libération. En incorporant un amphiphile sensible à la lumière (un dérivé de l’azobenzène), les liposomes formés semblent répondre à une radiation lumineuse. Pour ce système, il serait probablement nécessaire de tracer le diagramme de phase du mélange afin de contrôler la photo-libération de l’agent encapsulé. Nous avons aussi formulé des liposomes contenant un amphiphile cationique (le chlorure de cétylpyridinium). En tant que nanovecteurs, ces stérosomes montrent un potentiel intéressant pour la libération passive ou contrôlée de principes actifs. Pour ces systèmes, nous avons développé un modèle pour déterminer l’orientation des différentes molécules dans la bicouche. La formation de ces nouveaux systèmes a aussi apporté de nouvelles connaissances dans le domaine des interactions détergents-lipides. Aux nombreux effets du cholestérol (Chol) sur les systèmes biologiques, il faut ajouter maintenant que les stérols sont aussi capables de forcer les amphiphiles monoalkylés à former des bicouches. Cette nouvelle propriété peut avoir des répercussions sur notre compréhension du fonctionnement des systèmes biologiques. Enfin, les amphiphiles monoalkylés peuvent interagir avec la membrane et avoir des répercussions importantes sur son fonctionnement. Par exemple, l'effet antibactérien de détergents est supposé être dû à leur insertion dans la membrane. Cette insertion est régie par l'affinité existant entre le détergent et cette dernière. Dans ce cadre, nous avons voulu développer une nouvelle méthode permettant d'étudier ces affinités. Nous avons choisi la spectroscopie Raman exaltée de surface (SERS) pour sa sensibilité. Les hypothèses permettant de déterminer cette constante d’affinité se basent sur l’incapacité du détergent à exalter le signal SERS lorsque le détergent est inséré dans la membrane. Les résultats ont été comparés à ceux obtenus par titration calorimétrique isotherme (ITC). Les résultats ont montré des différences. Ces différences ont été discutées.

Pulsed photoacoustic studies in solutions of C70 in toluene are made using the 532-nm radiation from a frequency-doubled Nd:YAG laser. It is found that contrary to expectation, there is no photoacoustic (PA) signal enhancement in the power-limiting range of laser fluences. Instead, the PA signal tends to saturate during optical power-limiting phenomenon. This could be due to the enhanced optical absorption from the photoexcited state and hence the depletion of the ground-state population. PA measurements also ruled out the possibility of multiphoton absorption in the C70 solution. We demonstrate that the nonlinear absorption leading to optical limiting is mainly due to reverse saturable absorption.

Optical limiting and thermo-optic properties of C60 in toluene are studied using 532 nm, 9 ns pulses from a frequency-doubled Nd:YAG laser. Optical limiting studies in these fullerene molecules lead to the conclusion that reverse saturable absorption is the major mechanism for limiting properties in these molecules. Thermal lensing measurements are also performed in fullerene solutions. The quadratic dependence of thermal lens signal on incident energy confirms that enhanced optical absorption by the sample via excited triplet state absorption may play a leading role in the limiting property.

Vibrational overtone spectra of acetophenone and benzaldehyde in the visible and near-infrared regions are studied by the dual beam thermal lens and the conventional near-infrared absorption techniques. The observed increase in the mechanical frequency of the aryl CH bond from that of benzene is attributed to the decrease in the aryl CH bond length caused by the electron-withdrawing property of the substituents. Overtone spectra also demonstrate that acetophenone contains two types of methyl CH bonds arising from the anisotropic environments created by oxygen lone pair and carbonyl P electrons. The local-mode parameters of the two types of CH bonds are compared with those of acetone and acetaldehyde. The possible factors influencing the methyl CH bonds in acetophenone are discussed.

Photothermal spectroscopy is a group of high sensitivity methods used to measure optical absorption and thermal characteristics of a sample.The basis of photothermal spectroscopy is a photo-induced change in the thermal state of the sample.Light energy absorbed and not lost by subsequent emission results in sample heating.This heating results in a temperature change as well as changes in thermodynamic parameters of the sample which are related to temperature.Measurements of the temperature,pressure,or density changes that occur due to optical absorption are ultimately the basis for the photothermal spectroscopic methods.This is a more direct measure of optical absorption than optical transmission based spectroscopies.Sample heating is a direct consequence of optical absorption and so photothermal spectroscopy signals are directly dependent on light absorption.Scattering and reflection losses do not produce photothermal signals.Subsequently,photothermal spectroscopy more accurately measures optical absorption in scattering solutions,in solids,and at interfaces.This aspect makes it particularly attractive for application to surface and solid absorption studies,and studies in scattering media.

Near-infrared spectroscopy can be a workhorse technique for materials analysis in industries such as agriculture, pharmaceuticals, chemicals and polymers. A near-infrared spectrum represents combination bands and overtone bands that are harmonics of absorption frequencies in the mid-infrared. Near-infrared absorption includes a combination-band region immediately adjacent to the mid-infrared and three overtone regions. All four near-infrared regions contain "echoes" of the fundamental mid-infrared absorptions. For example, vibrations in the mid-infrared due to the C-H stretches will produce four distinct bands in each of the overtone and combination regions. As the bands become more removed from the fundamental frequencies they become more widely separated from their neighbors, more broadened and are dramatically reduced in intensity. Because near-infrared bands are much less intense, more of the sample can be used to produce a spectra and with near-infrared, sample preparation activities are greatly reduced or eliminated so more of the sample can be utilized. In addition, long path lengths and the ability to sample through glass in the near-infrared allows samples to be measured in common media such as culture tubes, cuvettes and reaction bottles. This is unlike mid-infrared where very small amounts of a sample produce a strong spectrum; thus sample preparation techniques must be employed to limit the amount of the sample that interacts with the beam. In the present work we describe the successful the fabrication and calibration of a linear high resolution linear spectrometer using tunable diode laser and a 36 m path length cell and meuurement of a highly resolved structure of OH group in methanol in the transition region A v =3. We then analyse the NIR spectrum of certain aromatic molecules and study the substituent effects using local mode theory

This work presents the developement of an chemically stable and easy to produce in situ sensor for fast and reliable detection of polycyclic aromatic hydrocarbons (PAH) in low nanomolar concentrations. Metallic nanoparticles on dielectric substrates werde used for the rst time with surface enhanced Raman spectroscopy (SERS) in combination with shifted excitation Raman difference spectroscopy (SERDS). The preparation of the metallic nanoparticle ensembles with Volmer-Webergrowth is described first. The nanoparticles are characterized with both, optical spectroscopy and atomic force microscopy. The morphological properties of the nanoparticle ensembles are de ned by the mean axial ratio (a/b) and the mean equivalent radius (R Äq), respectively. The prepared and characterized nanoparticles were then used for intensive Raman spectroscopy measurements. Two sophisticated diode laser systems were used in cooperation with the TU Berlin, to carry out these experiments. The first step was to establish the ideal combination of excitation wavelength of the diode laser and the maximum of the surface plasmon resonance of the nanoparticle ensembles. From these results it was deduced, that for an optimum Raman signal the plasmon resonance maximum of the nanoparticle ensemble has to be red-shifted a few nanometeres in respect to the excitation wavelength. Different PAHs werde detected in concentrations of only 2 and 0.5 nmol/, respectively. Furthermore, the obtained results show an excellent reproducability. In addition the time dependence of the Raman signal intensity was investigated. The results of these measurements show, that only 2 minutes after placing the substrates in the molecular solution, a detectable Raman signal was generated. The maximum Raman signal, i.e. the time in which the molecular adsorption process is finished, was determined to about 10 minutes. In summary it was shown, that the used metallic nanoparticle ensembles are highly usable as substrates for SERS in combination with SERDS to detect PAHs in low nanomolar concentrations.
