There is potential to reduce both operational and embodied greenhouse gas emission from buildings. To date the focus has been on reducing the operational element, although given the urgency of carbon reductions, it may be more beneficial to consider upfront embodied carbon reductions. This paper describes a case study on the whole life carbon cycle of a warehouse building in Swindon, UK. It examines the relationship between embodied carbon (Ec) and operational carbon (Oc), the proportions of Ec from the structural and non-structural elements, carbon benchmarking of the structure, the value of ‘cradle to site’ or ‘cradle to grave’ assessments and the significance of the timing of emissions during the life of the building. The case study indicates that Ec was dominant for the building and that the structure was responsible for more than half of the Ec. Weighting of future emissions appears to be an important factor to consider. The PAS 2050 reduction factors had only a modest effect but weighting to allow for future decarbonisation of the national grid energy supply had a large effect. This suggests that future operational carbon emissions are being overestimated compared to embodied.

This paper examines the significance of seventeen later Bronze Age wells found during construction at Swalecliffe, in north-east Kent. The unusual depth of the features made for exceptional preservation of wooden structural elements, including steps and revetments, demonstrating rare evidence for woodworking and woodmanship. Extensive biological remains facilitated environmental reconstruction, and a lengthy dendrochronological sequence corroborates the internationally important Flag Fen chronology. Dendrochronological and radiocarbon dates demonstrate around 500 years of seemingly continuous use and replacement of wells. Votive deposits and apparatus used for water collection provide glimpses of small-scale ritual and domestic activities. The highly unusual concentration of wells is compared to contemporary sites regionally and elsewhere.

The psychometric properties of scores from the Achievement Goal Questionnaire were examined in samples of Japanese (N = 326) and Canadian (N = 307) post secondary students. Previous research found evidence of a four-factor structure of achievement goals in U.S. samples. Using confirmatory factor-analytic techniques, the authors found strong evidence for the four-factor structure of achievement goals in both the Canadian and Japanese populations. Subsequent multi group structural equation modeling indicated the metric invariance of this four-factor structure across the two populations.

This article examines advocacy of Catholic restorative justice for clerical child sexual abuse from the standpoint of feminist criminological critiques of the use of restorative mediation in sexual offence cases. In particular, it questions the Catholic invocation of grace and forgiveness of survivors of abuse in light of critical feminist concerns about the exploitation of emotions in restorative practices, especially in regard to sexual and other gender-based offences. In the context of sexual abuse, the Catholic appeal to grace has the potential for turning into an extraordinary demand made of victims not only to rehabilitate offenders and the church in the eyes of the community, but also to work towards the spiritual absolution of the abuser. This unique feature of Catholic-oriented restorative justice raises important concerns in terms of feminist critiques of the risk of abuses of power within mediation, and is also incompatible with orthodox restorative justice theory, which, although it advocates a ‘spiritual’ response to crime, is concerned foremost with the rights, needs and experiences of victims.

Phosphofructokinase-1 and -2 (Pfk-1 and Pfk-2, respectively) from Escherichia coli belong to different homologous superfamilies. However, in spite of the lack of a common ancestor, they share the ability to catalyze the same reaction and are inhibited by the substrate MgATP. Pfk-2, an ATP-dependent 6-phosphofructokinase member of the ribokinase-like superfamily, is a homodimer of 66 kDa subunits whose oligomerization state is necessary for catalysis and stability. The presence of MgATP favors the tetrameric form of the enzyme. In this work, we describe the structure of Pfk-2 in its inhibited tetrameric form, with each subunit bound to two ATP molecules and two Mg ions. The present structure indicates that substrate inhibition occurs due to the sequential binding of two MgATP molecules per subunit, the first at the usual site occupied by the nucleotide in homologous enzymes and the second at the allosteric site, making a number of direct and Mg-mediated interactions with the first. Two configurations are observed for the second MgATP, one of which involves interactions with Tyr23 from the adjacent subunit in the dimer and the other making an unusual non-Watson-Crick base pairing with the adenine in the substrate ATP. The oligomeric state observed in the crystal is tetrameric, and some of the structural elements involved in the binding of the Substrate and allosteric ATPs are also participating in the dimer-dimer interface. 1,3-beta-Glucan depolymerizing enzymes have considerable biotechnological applications including biofuel production, feedstock-chemicals and pharmaceuticals. Here we describe a comprehensive functional characterization and low-resolution structure of a hyperthermophilic laminarinase from Thermotoga petrophila (TpLam). We determine TpLam enzymatic mode of operation, which specifically cleaves internal beta-1,3-glucosidic bonds. The enzyme most frequently attacks the bond between the 3rd and 4th residue from the non-reducing end, producing glucose, laminaribiose and laminaritriose as major products. Far-UV circular dichroism demonstrates that TpLam is formed mainly by beta structural elements, and the secondary structure is maintained after incubation at 90 degrees C. The structure resolved by small angle X-ray scattering, reveals a multi-domain structural architecture of a V-shape envelope with a catalytic domain flanked by two carbohydrate-binding modules. We report on the size, shape, structure, and interactions of lysozyme in the ternary system lysozyme/DMSO/water at low protein concentrations. Three structural regimes have been identified, which we term the ""folded"" (0 < phi(DMSO) < 0.7), ""unfolded"" (0.7 <= phi(DMSO) < 0.9), and ""partially collapsed"" (0.9 <= phi(DMSO) < 1.0) regime. Lysozyme resides in a folded conformation with an average radius of gyration of 1.3 +/- 0.1 nm for phi(DMSO) < 0.7 and unfolds (average R(g) of 2.4 +/- 0.1 nm) above phi(DMSO) > 0.7. This drastic change in the protein`s size coincides with a loss of the characteristic tertiary structure. It is preceded by a compaction of the local environment of the tryptophan residues and accompanied by a large increase in the protein`s overall flexibility. In terms of secondary structure, there is a gradual loss of alpha-helix and concomitant increase of beta-sheet structural elements toward phi(DMSO) = 0.7, while an increase in phi(DMSO) at even higher DMSO volume fractions reduces the presence of both a-helix and beta-sheet secondary structural elements. Protein-protein interactions remain overall repulsive for all values of phi(DMSO) An attempt is made to relate these structural changes to the three most important physical mechanisms that underlie them: the DMSO/water microstructure is strongly dependent on the DMSO volume fraction, DMSO acts as a strong H-bond acceptor, and DMSO is a bad solvent for the protein backbone and a number of relatively polar side groups, but a good solvent for relatively apolar side groups, such as tryptophan.

Esta dissertação discute a leitura de textos imagéticos a partir da perspectiva da educação e da semiótica visual, tratando das articulações e relações entre os elementos constitutivos da imagem em livros de literatura infantil sem texto verbal. A investigação busca revelar, na complexidade das relações existentes entre elementos estruturantes das imagens nos livros O caminho do caracol e Cena de rua, o sentido que se inscreve no texto e a possibilidade da sua leitura no meio escolar. As obras literárias para a infância veiculam a linguagem visual em junção com a verbal e propiciam experiências sensíveis e inteligíveis. A partir da teoria semiótica greimasiana, aplicando instrumentos de análise do percurso gerativo de sentido, a pesquisa demonstra que o texto imagético é estruturado por diferentes níveis de complexidade, impondo um modo de ler específico. A identificação, descrição, classificação, bem como as relações entre as categorias, nas suas dimensões cromática, eidética e topológica, levam à constatação que as qualidades plásticas da imagem organizam sistemas de linguagem, reunindo solidariamente o plano da expressão e do conteúdo, criam estruturas e geram efeitos de sentido. A imagem, então, constitui-se como objeto de significação e a ilustração presente no livro de literatura infantil, ao ser tratada como texto lisível, torna-se objeto de leitura. A complexidade desse tipo de texto aponta a necessidade da formação específica de professores para explorar a leitura escolar das linguagens visuais.

O presente trabalho tem por finalidade estudar a Justiça Restaurativa como uma alternativa a pena de prisão e sua utilização pelo Poder Judiciário. O trabalho foi realizado pelo modelo plan francês, desenvolvido em duas partes, uma parte teórica e uma prática; cada uma das partes foi dividida em dois capítulos. Na parte teórica, no primeiro capítulo foi estudada a justiça restaurativa, seus conceitos e peculiaridades, além de sua contextualização no universo jurídico. No segundo capítulo foram estudados os institutos da mediação e da conciliação, bem como à aproximação dos referidos institutos da justiça restaurativa. Na segunda parte, a prática; primeiramente abordamos o Projeto de Lei nº 7006/2006, que visa introduzir a Justiça Restaurativa em nosso sistema penal de forma institucionalizada e a Resolução nº 125 do Conselho Nacional de Justiça (CNJ). No segundo capítulo analisamos os programas de justiça restaurativa existentes no Brasil, a criação dos Núcleos de Mediação pelo Tribunal de Justiça do Estado do Rio de Janeiro; mapeamos a justiça restaurativa no Estado do Rio de Janeiro e não identificamos nenhum projeto em andamento. E por fim analisamos os indícios da Justiça Restaurativa existentes no Juizado Especial Criminal (JECRIM) da Barra da Tijuca, onde esse tipo de procedimento não existe de forma estruturada. Não existe um programa que dê aporte a prática, sendo utilizada, quando possível, como mais um instrumento para resolução dos conflitos, por meio da mediação penal.

Entre os desafios para a implementação da política de desenvolvimento social do Brasil, muito tem sido discutido acerca das dificuldades de capacitação dos profissionais que diariamente executam ações que traduzem esta política. Parte da dificuldade é decorrente das transformações ocorridas nas últimas décadas nas Políticas Públicas de Assistência Social no país e no entendimento destas novas demandas. O Brasil está transformando a sua antiga política assistencialista na do direito social. A capacitação e a garantia da educação continuada e da educação permanente são elementos fundamentais e estruturantes para a consolidação deste novo projeto de política social. Neste sentido, a criação de uma escola de governo no Estado de São Paulo, com a perspectiva de capacitação e educação continuada dos profissionais envolvidos na prestação de serviços socioassistenciais, vem colaborar com a formação dos atores sociais (servidores e prestadores de serviço), capazes de executar e aprimorar as políticas sociais regionais e do SUAS, de forma transversal e multidisciplinar, visando garantir o direito e o acesso a bens e serviços aos cidadãos e grupos em situação de vulnerabilidade e risco social e pessoal. Este trabalho busca resgatar o contexto e as motivações que levaram à criação da Escola de Desenvolvimento Social do Estado de São Paulo (EDESP), sua história e as expectativas em torno dela, permitindo, a partir de experiências em outras escolas de governo e referenciais de boas práticas, elencar alguns pontos para reflexão.

As most current studies, reinforced plastics have been, in recent years, a viable alternative in building structural elements of medium and large, since the lightness accompanied by high performance possible. The design of hybrid polymer composites (combination of different types of reinforcements) may enable structural applications thereof, facing the most severe service conditions. Within this class of composite materials, reinforced the underlying tissues hybrid high performance are taking space when your application requires high load bearing and high rigidity. The objective of this research work is to study the challenges in designing these fabrics bring these materials as to its mechanical characterization and fracture mechanisms involved. Some parameters associated with the process and / or form of hybridization stand out as influential factors in the final performance of the material such as the presence of anisotropy, so the fabric weave, the process of making the same, normative geometry of the specimens, among others. This sense, four laminates were developed based hybrid reinforcement fabrics involving AS4 carbon fiber, kevlar and glass 49-E as the matrix epoxy vinyl ester resin (DERAKANE 411-350). All laminates were formed each with four layers of reinforcements. Depending on the hybrid fabric, all the influencing factors mentioned above have been studied for laminates. All laminates were manufactured industrially used being the lamination process manual (hand-lay-up). All mechanical characterization and study of the mechanism of fracture (fracture mechanics) was developed for laminates subjected to uniaxial tensile test, bending in three and uniaxial compression. The venom of Crotalus durissus terrificus snakes presents various substances, including a serine protease with thrombin-like activity, called gyroxin, that clots plasmatic fibrinogen and promote the fibrin formation. The aim of this study was to purify and structurally characterize the gyroxin enzyme from Crotalus durissus terrificus venom. For isolation and purification, the following methods were employed: gel filtration on Sephadex G75 column and affinity chromatography on benzamidine Sepharose 6B; 12% SDS-PAGE under reducing conditions; N-terminal sequence analysis; cDNA cloning and expression through RT-PCR and crystallization tests. Theoretical molecular modeling was performed using bioinformatics tools based on comparative analysis of other serine proteases deposited in the NCBI (National Center for Biotechnology Information) database. Protein N-terminal sequencing produced a single chain with a molecular mass of similar to 30 kDa while its full-length cDNA had 714 bp which encoded a mature protein containing 238 amino acids. Crystals were obtained from the solutions 2 and 5 of the Crystal Screen Kit (R), two and one respectively, that reveal the protein constitution of the sample. For multiple sequence alignments of gyroxin-like B2.1 with six other serine proteases obtained from snake venoms (SVSPs), the preservation of cysteine residues and their main structural elements (alpha-helices, beta-barrel and loops) was indicated. The localization of the catalytic triad in His57, Asp102 and Ser198 as well as S1 and S2 specific activity sites in Thr193 and Gli215 amino acids was pointed. The area of recognition and cleavage of fibrinogen in SVSPs for modeling gyroxin B2.1 sequence was located at Arg60, Arg72, Gln75, Arg81, Arg82, Lis85, Glu86 and Lis87 residues. Theoretical modeling of gyroxin fraction generated a classical structure consisting of two alpha-helices, two beta-barrel structures, five disulfide bridges and loops in positions 37, 60, 70, 99, 148, 174 and 218. These results provided information about the functional structure of gyroxin allowing its application in the design of new drugs.

Diante do atual modelo penal e processual penal não atender aos reclamos das partes interessadas, gerando um descrédito na Justiça de um modo geral, surge a Justiça Restaurativa como uma alternativa para solucionar tais problemas e como elemento de concretização do Estado Democrático Constitucional. A Constituição Federal de 1988 representa o símbolo maior do processo de democratização e de constitucionalização nacional. O Princípio da Dignidade da Pessoa contida no texto constitucional consiste num dos principais fundamentos da República Federativa do Brasil, funcionando como respaldo aos direitos e garantias fundamentais do cidadão, sobretudo na seara criminal. A partir do processo de constitucionalização nacional, ocorre uma releitura das legislações infra-constitucionais, que passam a ser interpretadas de acordo com o texto constitucional. Atualmente, a conjuntura jurídico-penal pátria está associada à ideia de garantismo, ligada ao conceito de Estado Democrático Constitucional. Apresenta-se a Justiça Restaurativa como um novo modelo de Justiça Penal, mais flexível e humanizado, visando além da aplicação da pena imposta pelo Estado, superar uma situação de conflito, na busca por resultados positivos no combate e redução da criminalidade, a satisfação da vítima e a mudança da cultura de violência, compatível com as diretrizes do Estado Democrático Constitucional. A partir da análise do direito internacional e de projetos e legislações nacionais envolvendo a Justiça Restaurativa, percebe-se a eficácia das medidas restaurativas na solução de conflitos dentro do Processo Penal, além da satisfação da vítima, do infrator e de familiares na participação dos encontros restaurativos, constituindo ferramenta de satisfação da dignidade humana, dentro de uma perspectiva humanista e garantista

The concrete for centuries constituted an essential structural element in the construction industry due to its relative ease of forming, before the weather durability, low cost, its lower maintenance compared to other materials such as steel. However, when the concrete is exposed to high temperatures tends to lose its mechanical characteristics, and may even result in loss of section, which undermines the stability and mechanical strength of structural elements. The pathologies resulting from exposure to elevated temperatures ranging from cracks, pops up chipping explosives (spalling). Recently, the technology of concrete is closely related to the study of its microstructure. The use of fibers added to concrete has been revealed as a solution to increase the mechanical strength of the concrete, it acts directly on the distribution of efforts to act in the play within the microstructure. In this work we used recycled PET fibers embedded in concrete with 15x2mm fck = 30MPa, water/cement ratio of 0.46, in works made for verification of mechanical strength of this mixture submitted to high temperature. The specimens of concrete with addition of PET fibers were tested after exposure to temperatures: ambient (30ºC), 100°C, 200°C, 300°C, 400°C, 600°C and 900°C. It was found that the concrete loses significant strength when exposed to temperatures above 300°C, however the use of fiber PET may delay the risk of collapse of structures for the formation of a network of channels that facilitate the escape of vapor 'water, reducing the pore pressure inside the structural element
