We have developed a procedure for the rapid diagnosis of plague that also allows the identification of prominent virulence markers of Y. pestis strains. This procedure is based upon the use of a single polymerase chain reaction with multiple pairs of primers directed at genes present in the three virulence plasmids as well as in the chromosomal pathogenicity island of the bacterium. The technique allowed the discrimination of strains which lacked one or more of the known pathogenic loci, using as template total DNA obtained from bacterial cultures and from simulated blood cultures containing diluted concentration of bacteria. It also proved effective in confirming the disease in a blood culture from a plague suspected patient. As the results are obtained in a few hours this technique will be useful in the methodology of the Plague Control Program.

O vírus da hepatite delta (HDV) é o agente etiológico de uma das formas mais graves de hepatite viral e é ainda endémico em diversas regiões do globo, nomeadamente em África, na Amazónia e no Extremo Oriente. O HDV co-infecta ou super-infecta hepatócitos infectados com o vírus da hepatite B (HBV) aumentando em cerca de 10 vezes o risco de cirrose e hepatite fulminante. A associação clínica entre os dois vírus deve-se ao facto do invólucro do HDV ser constituído pelos antigénios de superfície do HBV (HBsAgs) que são necessários para a propagação da infecção. O genoma do HDV é constituído por uma molécula de RNA de cadeia simples, circular, com cerca de 1.7 Kb, que possui cerca de 70% de emparelhamento interno. Foi identificada uma única grelha de leitura aberta (ORF) no RNA viral que codifica para o antigénio delta (HDAg). A ocorrência de um mecanismo de editing do RNA, resulta na expressão de duas formas do HDAg, a pequena (S-HDAg) e a grande (L-HDAg). Várias funções essenciais para a replicação do HDV têm sido atribuídas a ambas as formas do HDAg, sendo a S-HDAg essencial para a acumulação de RNA viral e a L-HDAg responsável pela interacção com os HBsAgs para formar partículas virais. No entanto, dada a simplicidade dos seus componentes, admite-se que a replicação viral depende das interacções estabelecidas entre os HDAgs e factores celulares do hospedeiro. Apesar do número considerável de factores celulares descritos como interactores dos HDAgs ou RNA virais, a importância de muitas destas interacções não foi elucidada e muitas etapas do ciclo de replicação do HDV permanecem pouco claras. Para além disso, dado o número limitado de factores do hospedeiro que estão envolvidos na sua replicação, é muito provável que um número elevado de interactores do HDV permaneça por identificar. Este trabalho teve como objectivo a identificação de proteínas de fígado humano capazes de interagir com os HDAgs, utilizando o sistema yeast Two-Hybrid (YTH). Identificaram-se trinta proteínas com capacidade de interagir com a S-HDAg no sistema YTH, sendo que estas proteínas se encontram envolvidas em diferentes processos celulares. Com base nas características funcionais, foram seleccionadas três destas proteínas e as suas interacções com a S-HDAg foram investigadas com maior detalhe. As três proteínas seleccionadas foram a ribonucleoproteína nuclear heterogénea C (hnRNPC), a embryonic lethal abnormal vision like1 (ELAVL1/HuR) e a proteína 2 de ligação a EBNA1 (EBP2). As duas primeiras são proteínas de ligação a RNA, previamente descritas como envolvidas em processos de replicações de outros vírus com genoma RNA, enquanto a EBP2, é uma proteína de localização preferencialmente nucleolar, tal como por vezes acontece com os HDAgs. As interacções foram analisadas recorrendo a vários ensaios bioquímicos. No caso da hnRNPC e da HuR, após validação no sistema YTH, a capacidade de interacção com a S-HDAg foi confirmada quer in vitro por blot overlay quer in vivo por co-imunoprecipitação em células de hepatoma humano. Nas mesmas células, observou-se uma co-localização considerável entre os HDAgs e os RNAs virais. Finalmente, de modo a investigar a contribuição das proteínas hnRNPC e HuR na replicação do HDV, procedeu-se ao silenciamento destas proteínas pela utilização de short hairpin RNAs (shRNAs) específicos para os mRNAs correspondentes Observou-se que o silenciamento de ambas as proteínas hnRNPC e HuR endógenas, individualmente resultou numa diminuição acentuada nos níveis de expressão dos HDAgs. No que respeita à EBP2, a interacção com a S-HDAg foi confirmada em condições in vitro com recurso a ensaios de blot overlay e de cromatografia de afinidade. A análise por imunofluorescência indirecta e microscopia confocal revelou co-localização elevada entre os HDAgs e a EBP2, principalmente nos nucléolos de células de hepatoma humano. Finalmente, foi ainda utilizado o sistema YTH para estudar os mecanismos de importação dos HDAgs. Assim, este sistema foi utilizado com o propósito de identificar proteínas celulares capazes de interagir com um domínio específico dos HDAgs, o sinal de localização nuclear (NLS). Na pesquisa YTH realizada obtiveram-se 161 clones positivos, sendo que um deles mostrou codificar para a carioferina α4 (KPNA4). A interacção da KPNA4 com a S-HDAg foi reproduzida em condições in vitro através de um ensaio de cromatografia de afinidade tendo sido utilizadas formas recombinantes das duas proteínas. Este trabalho permitiu identificar várias proteínas celulares que interagem com a S-HDAg. Obtiveram-se evidências sugestivas de que algumas das proteínas identificadas podem desempenhar funções importantes no ciclo de replicação do HDV e que abrem novas perspectivas para o estudo do ciclo de replicação do vírus.

A case-control study was conducted to identify risk factors for death from tetanus in the State of Pernambuco, Brazil. Information was obtained from medical records of 152 cases and 152 controls, admitted to the tetanus unit in the State University Hospital, in Recife, from 1990 to 1995. Variables were grouped in three different sets. Crude and adjusted odds ratios, p-values and 95% confidence intervals were estimated. Variables selected in the multivariate analysis in each set were controlled for the effect of those selected in the others. All factors related to the disease progression - incubation period, time elapsed between the occurrence of the first tetanus symptom and admission, and period of onset - showed a statistically significant association with death from tetanus. Similarly, signs and/or symptoms occurring on admission or in the following 24 hours (second set): reflex spasms, neck stiffness, respiratory signs/symptoms and respiratory failure requiring artificial ventilation (third set) were associated with death from tetanus even when adjusted for the effect of the others.

More than 70 species of mycobacteria have been defined, and some can cause disease in humans, especially in immunocompromised patients. Species identification in most clinical laboratories is based on phenotypic characteristics and biochemical tests and final results are obtained only after two to four weeks. Quick identification methods, by reducing time for diagnosis, could expedite institution of specific treatment, increasing chances of success. PCR restriction-enzyme analysis (PRA) of the hsp65 gene was used as a rapid method for identification of 103 clinical isolates. Band patterns were interpreted by comparison with published tables and patterns available at an Internet site (http://www.hospvd.ch:8005). Concordant results of PRA and biochemical identification were obtained in 76 out of 83 isolates (91.5%). Results from 20 isolates could not be compared due to inconclusive PRA or biochemical identification. The results of this work showed that PRA could improve identification of mycobacteria in a routine setting because it is accurate, fast, and cheaper than conventional phenotypic identification.

There is no paucity of methods for diagnosing Cryptosporidium spp. infection. The merits of immunoassays notwithstanding, microscopic identification of Cryptosporidium spp. oocysts in fecal samples remains an important diagnostic procedure. It owes the persistence of its use to such characteristics as dispensing with expensive equipment and kits, requiring only basic laboratory facilities, and having a low probability of false positive results when permanent slides are prepared, which can be re-examined in case of doubt. Cryptosporidium spp. oocysts can be readily identified in fecal smears prepared according to a regressive iron hematoxylin staining technique. The number of steps and their duration, as well as costs, were reduced to a minimum without loss of image quality and permanence of the preparations.

Toxocariasis is a frequent helminthiasis that can cause visceral and ocular damage in humans specially in children. The identification of specific antigens of Toxocara canis is important in order to develop better diagnostic techniques. Ten rabbits were infected orally with a dose of 5000 Toxocara canis embryonated eggs. Rabbits were bled periodically and an ELISA assay was performed to determine levels of specific Toxocara IgG antibodies. ELISA detected antibodies at day 15 after infection. Western blot (WB) assay was performed using excretory/secretory antigens (E/S) of T. canis second stage larvae. Different antigen concentrations were evaluated: 150, 200, 250 and 300 µg/mL. The concentration of 250 µg/mL was retained for analysis. Rabbit sera were diluted 1:100. Secondary antibody was used at a dilution of 1:1000. Results of WB indicated that in the first month after infection specific antibodies against the 200 KDa, 116 KDa, 92 KDa and 35 KDa antigens were detected; antibodies against the 92 KDa, 80 KDa, 66 KDa, 45 KDa, 31 KDa and 28 KDa antigens appeared later. All positive sera in the ELISA test were also positive in WB. Two antigen bands, 92 KDa and 35 KDa, were identified since the beginning and throughout the course of infection. These antigens merit further evaluation as candidates for use in diagnosis.

Salmonella spp. are the etiologic agents of salmonellosis, a worldwide spread zoonoses causing foodborne outbreaks and clinical diseases. By serological identification, Salmonella enterica subsp. enterica serotype 1,4,[5],12:i:- accounted for 8.8% of human and 1.6% of nonhuman Salmonella strains isolated in São Paulo State, during 1991-2000. A total of 28.6% of them amplified a fragment corresponding to H:1,2 (flagellar phase two) through PCR analysis and were further assigned as S. Typhimurium. Antimicrobial resistance was detected in 36.3% of the 369 PCR-negative strains tested, including the multiresistance to ampicillin, chloramphenicol, sulfonamides, tetracycline, and streptomycin.

This work presents a hybrid maneuver for gradient search with multiple AUV's. The mission consists in following a gradient field in order to locate the source of a hydrothermal vent or underwater freshwater source. The formation gradient search exploits the environment structuring by the phenomena to be studied. The ingredients for coordination are the payload data collected by each vehicle and their knowledge of the behaviour of other vehicles and detected formation distortions.

Since October 2001, the Adolfo Lutz Institute has been receiving vesicular fluids and scab specimens of patients from Paraíba Valley region in the São Paulo and Minas Gerais States and from São Patricio Valley, in the Goiás State. Epidemiological data suggested that the outbreaks were caused by Cowpox virus or Vaccinia virus. Most of the patients are dairy milkers that had vesiculo-pustular lesions on the hands, arms, forearms, and some of them, on the face. Virus particles with orthopoxvirus morphology were detected by direct electron microscopy (DEM) in samples of 49 (66.21%) patients of a total of 74 analyzed. Viruses were isolated in Vero cell culture and on chorioallantoic membrane (CAM) of embryonated chicken eggs. Among 21 samples submitted to PCR using primers for hemagglutinin (HA) gene, 19 were positive. Restriction digestion with TaqI resulted in four characteristic Vaccinia virus fragments. HA nucleotide sequences showed 99.9% similarity with Cantagalo virus, described as a strain of Vaccinia virus. The only difference observed was the substitution of one nucleotide in the position 616 leading to change in one amino acid of the protein in the position 206. The phylogenetic analysis showed that the isolates clustered together with Cantagalo virus, other Vaccinia strains and Rabbitpox virus.

Candida dubliniensis is a new, recently described species of yeast. This emerging oral pathogen shares many phenotypic and biochemical characteristics with C. albicans, making it hard to differentiate between them, although they are genotypically distinct. In this study, PCR (Polymerase Chain Reaction) was used to investigate the presence of C. dubliniensis in samples in a culture collection, which had been isolated from HIV-positive and HIV-negative patients with oral erythematous candidiasis. From a total of 37 samples previously identified as C. albicans by the classical method, two samples of C. dubliniensis (5.4%) were found through the use of PCR. This study underscores the presence of C. dubliniensis, whose geographical and epidemiological distribution should be more fully investigated.

Die Luftverschmutzung, die globale Erwärmung sowie die Verknappung der endlichen Ressourcen sind die größten Bedenken der vergangenen Jahrzehnte. Die Nachfrage nach jeglicher Mobilität steigt rapide. Dementsprechend bemüht ist die Automobilindustrie Lösungen für Mobilität unter dem Aspekt der Nachhaltigkeit und dem Umweltschutz anzubieten. Die Elektrifizierung hat sich hierbei als der beste Weg herausgestellt, um die Umweltprobleme sowie die Abhängigkeit von fossilen Brennstoffen zu lösen. Diese Arbeit soll einen Einblick über die Umweltauswirkungen des Hybridfahrzeuges Toyota Prius geben. Hierbei findet eine Gliederung in vier verschiedene Lebensphasen statt. Im Anschluss bietet die Sachbilanz die Möglichkeit die Umweltauswirkungen mit verschiedenen Antriebsmöglichkeiten und Brennstoffen zu vergleichen. Das Modell hat gezeigt, dass der Toyota Prius während der Nutzung einen hohen Einfluss auf das Treibhauspotenzial aufweist. Durch die Nutzung anderer Brennstoffe, wie beispielsweise Ethanol oder Methanol lassen sich die Auswirkungen am Treibhauspotenzial sowie der Verbrauch an abiotischen Ressourcen reduzieren. Vergleicht man die Elektromobilität mit der konventionellen, so ist festzustellen, dass diese Art der Mobilität die derzeit beste Möglichkeit zur Reduzierung der Umweltbelastungen bietet. Die Auswirkungen der Elektromobilität sind im hohen Maße abhängig von der Art des verwendeten Strommixes.

DNA amplification techniques are being used increasingly in clinical laboratories to confirm the identity of medically important bacteria. A PCR-based identification method has been in use in our centre for 10 years for Burkholderia pseudomallei and was used to confirm the identity of bacteria isolated from cases of melioidosis in Ceará since 2003. This particular method has been used as a reference standard for less discriminatory methods. In this study we evaluated three PCR-based methods of B. pseudomallei identification and used DNA sequencing to resolve discrepancies between PCR-based results and phenotypic identification methods. The established semi-nested PCR protocol for B. pseudomallei 16-23s spacer region produced a consistent negative result for one of our 100 test isolates (BCC #99), but correctly identified all 71 other B. pseudomallei isolates tested. Anomalous sequence variation was detected at the inner, reverse primer binding site for this method. PCR methods were developed for detection of two other B. pseudomallei bacterial metabolic genes. The conventional lpxO PCR protocol had a sensitivity of 0.89 and a specificity of 1.00, while a real-time lpxO protocol performed even better with sensitivity and specificity of 1.00, and 1.00. This method identified all B. pseudomallei isolates including the PCR-negative discrepant isolate. The phaC PCR protocol detected the gene in all B. pseudomallei and all but three B. cepacia isolates, making this method unsuitable for PCR-based identification of B. pseudomallei. This experience with PCR-based B. pseudomallei identification methods indicates that single PCR targets should be used with caution for identification of these bacteria, and need to be interpreted alongside phenotypic and alternative molecular methods such as gene sequencing.

Cryptosporidium isolates identified in fourteen stool samples, collected from five HIV-infected patients and nine immunocompetent children, living in the Sate of São Paulo, Brazil, were submitted to a molecular analysis using a nested PCR followed of restriction fragment length polymorphism (RFLP), for genetic characterization. The analysis was based on digestion with RsaI restriction enzyme of a DNA fragment amplified from the Cryptosporidium oocyst wall protein (COWP) gene. Based on this analysis, four samples were identified as Cryptosporidium parvum, eight as Cryptosporidium hominis and two presented a profile that correspondedto Cryptosporidium meleagridis when compared to the standards used in the analysis. The use of molecular methods can be helpful to identify source of infections and risk factors related to Cryptosporidium infection in our communities.
