Esta dissertação de mestrado busca pesquisar a relação estabelecida pelo teólogo Ronald Cole-Turner, entre a engenharia genética e o conceito de co-criação , desenvolvido como uma alternativa para que se pense o papel do ser humano frente aos desenvolvimentos tecnológicos atuais. Tendo em vista o conceito da evolução, da criação contínua e dos potenciais gerados pela engenharia genética, Cole-Turner defende que o ser humano pode participar das atividades criativas de Deus por meio desta tecnologia, desde que priorize os aspectos redentivos que podem ser extraídos de seu uso. Faz isto em demonstrar, por meio do relato Jawista da criação, como o agir criativo de Deus está relacionado ao agir humano, através de suas tecnologias, e em relacionar os milagres de cura operados por Jesus nos evangelhos, como explicitação epistêmica da vontade de Deus, que pode ser participada pelos seres humanos. O objetivo básico desta pesquisa será o de relatar, no primeiro capítulo, o aspecto teórico da engenharia genética, nas suas possibilidades positivas e negativas. Em seguida, no capítulo segundo, se fará uma exposição do pensamento de Cole- Turner, em diálogo com outros interlocutores do tema. Por fim, no último capítulo, se verificará as respostas oferecidas às várias observações críticas que seu pensamento recebe. A hipótese é a de que, se a criação é um projeto contínuo e inacabado, e o destino dos seres humanos se encontra inserido dentro desta continuidade, seria coerente afirmar que estes possam participar das atividades de Deus em tais projetos. Como a evolução biológica é uma dimensão da criação, que ocorre por meio de transformações no capital genético dos organismos vivos, os seres humanos, tendo conhecimento e habilidade para repetir os processos verificados nestas modificações, poderão empreender seus esforços, tanto na área da técnica, como na da ética, para participarem da consecução dos propósitos de Deus.(AU)

O propósito deste estudo foi avaliar a concordância entre a Análise facial subjetiva, proposta por CAPELOZZA FILHO e a Análise Cefalométrica de Tecidos Moles, de ARNETT; McLAUGHLIN. Fotografias de frente e de perfil e telerradiografias em norma lateral padronizadas, de 50 indivíduos, com média de idade de 24 anos e 1 mês foram utilizadas para a avaliação. Verificou-se também nos indivíduos classificados como Padrão I, a correspondência dos valores médios e dos desvios-padrão das medidas obtidas, com os valores normativos da Análise Cefalométrica dos Tecidos Moles para os indivíd uos com harmonia facial. Constatou-se em indivíduos do Padrão I que os lábios sempre se encontram à frente da Linha Vertical Verdadeira, e que apesar de grandes variações do ponto pogônio, ainda é mantido o equilíbrio facial. Os resultados demonstraram que a Análise facial subjetiva eficiente na classificação do padrão facial.

Although photodynamic therapy have been used as a useful tool over the past 30 years in oncology, few clinical trials have been conducted in dentistry. Photodynamic therapy (PDT) uses non - toxic photosensitizers and selective which are administered in target cells followed by local application of visible light, producing reactive oxygen species capable of causing cell death by apoptosis or necrosis, injured the local vasculature, and exert important effects on the im mune system. New generations of photosensitizing agents, such as nanoparticulate phthalocyanines, has shown excellent results in antitumor and antibacterial activity . In this context, the present work constitutes the first clinical protocol of local appli cation of nanoemulsion chloro - aluminum phthalocyanine (AlClFc) followed by irradiation in human gingiva, and analyzed descriptively and comparatively , by means of immunohistochemistry , the expression of RANK , RANKL , OPG and VEGF in a split - mouth model . Eight healthy volunteers with clinical indication for extraction were included in the study . Seven days before the extraction, was injected in the gingiva of participants, 5 μ M of nanoemulsion AlClFc followed by irra diation with diode laser (660nm , 7 J/cm2 ), the contralateral side was used as control. Tissue specimens were removed seven days after the TFD is performed. Tissues sample were divided into two groups (test and con trol groups) for histological and immunohistochemical analysis. Patients were monitored at days, 0, 7, 14 and 30 to assess adverse effects of the therapy. Vascular alterations were seen in gingival samples that received PDT. Areas of edema and vascular con gestion, and intense vascularization were viewed . Additionally, dystrophic calcification in subepithelial region were observed in the test group. The results showed a similar pattern of immunostaining scores of RANK, RANKL and VEGF between the test and co ntrol groups, with no statistically significant difference (p = 0.317, p = 0.777, p = 0 .814, respectively). RANK and RANKL exhibited weak or absent immunostaining in most specimens analyzed. There was n o immunostaining for OPG. VEGF showed moderate to stro ng immunostaining in specimens from the test group. In addition, the clinical study showed that therapy was well tolerated by all patients. Adverse effects were short - time and completely reversible. Taken together, the results presented in this study showe d that PDT mediated by nanoemulsion containing AlClPc is safe for clinical application in gingival tissue and suggests that a strong immunostaining for VEGF after therapy .

This work consists of the conception, developing and implementation of a Computational Routine CAE which has algorithms suitable for the tension and deformation analysis. The system was integrated to an academic software named as OrtoCAD. The expansion algorithms for the interface CAE genereated by this work were developed in FORTRAN with the objective of increase the applications of two former works of PPGEM-UFRN: project and fabrication of a Electromechanincal reader and Software OrtoCAD. The software OrtoCAD is an interface that, orinally, includes the visualization of prothetic cartridges from the data obtained from a electromechanical reader (LEM). The LEM is basically a tridimensional scanner based on reverse engineering. First, the geometry of a residual limb (i.e., the remaining part of an amputee leg wherein the prothesis is fixed) is obtained from the data generated by LEM by the use of Reverse Engineering concepts. The proposed core FEA uses the Shell's Theory where a 2D surface is generated from a 3D piece form OrtoCAD. The shell's analysis program uses the well-known Finite Elements Method to describe the geometry and the behavior of the material. The program is based square-based Lagragean elements of nine nodes and displacement field of higher order to a better description of the tension field in the thickness. As a result, the new FEA routine provide excellent advantages by providing new features to OrtoCAD: independency of high cost commercial softwares; new routines were added to the OrtoCAD library for more realistic problems by using criteria of fault engineering of composites materials; enhanced the performance of the FEA analysis by using a specific grid element for a higher number of nodes; and finally, it has the advantage of open-source project and offering customized intrinsic versatility and wide possibilities of editing and/or optimization that may be necessary in the future

The inefficiency of chemical pesticides to control phytopathogenic fungi in agriculture and the frequent incidence of human diseases caused by bacteria which are resistant to antibiotics lead to the search for alternative antimicrobial compounds. In this context, plant defensins are a promising tool for the control of both plant and human pathogenic agents. Plant defensins are cationic peptides of about 50 amino acid residues, rich in cysteine and whose tridimensional structure is considerably conserved among different plant species. These antimicrobial molecules represent an important innate component from plant defense response against pathogens and are expressed in various plant tissues, such as leaves, tubers, flowers, pods and seeds. The present work aimed at the evaluation of the antimicrobial activity of two plant defensins against different phytopathogenic fungi and pathogenic bacteria to humans. The defensin Drr230a, whose gene was isolated from pea (Pisum sativum), and the defensin CD1,whose gene was identified within coffee (Coffea arabica) transcriptome, were subcloned in yeast expression vector and expressed in Pichia pastoris. The gene cd1 was subcloned as two different recombinant forms: CD1tC, containing a six-histidine sequence (6xHis) at the peptide C-terminal region and CD1tN, containing 6xHis coding sequence at the N-terminal region. In the case of the defensin Drr230a, the 6xHis coding sequence was inserted only at the N-terminal region. Assays of the antimicrobial activity of the purified recombinant proteins rDrr230a and rCD1 against Phakopsora pachyrhizi, causal agent of soybean Asian rust, were performed to analyze the in vitro spore germination inhibition and disease severity caused by the fungus in planta. Both recombinant defensins were able to inhibit P. pachyrhizi uredospore germination, with no difference between the antimicrobial action of either CD1tC or CD1tN. Moreover, rDrr230a and rCD1 drastically reduced severity of soybean Asian rust, as demonstrated by in planta assays. In spite of the fact that rCD1 was not able to inhibit proliferation of the human pathogenic bacteria Staplylococcus aureus and Klebsiella pneumoniae, rCD1 was able to inhibit growth of the phytopathogenic fungus Fusarium tucumaniae, that causes soybean sudden death syndrome. The obtained results show that these plant defensins are useful candidates to be used in plant genetic engineering programs to control agriculture impacting fungal diseases.

Obesity is a chronic metabolic disease characterized by adipose tissue formation excess leading to an increase in body fat mass, of multifactorial origin, produced mainly by poor eating habits combined with a sedentary lifestyle. Data consider obesity as a serious disease that affects the world's population, ranking fifth in death rates. Faced with this situation, individuals seek, increasingly, means to lose weight with less physical effort and food. In 2009 and 2010 the drug liraglutide was lauched in order to reduce weight in individuals with diabetes mellitus type 2, thus avoiding the emergence of other diseases. The aggravating factor is that obese nondiabetic individuals are making use of this substance, even if its use is not authorized by ANVISA (Brazilian Health Surveillance Agency). Thus the objective of this research is to evaluate the effect of liraglutide for muscle or fat tissues and biochemical parameters in Swiss mice submitted to cafeteria diet and physical activity. The study was approved by the Ethics Committee on Animal Use - CEUA (nº003 Protocol / 2014). For this study 74 animals (Swiss mice) were used, divided as follows: in the initial phase of this study, we carried out a pilot study (n = 10) divided into a control group (PCON) (n = 5) and cafeteria group (PCAF) (n = 5), in order to evaluate a cafeteria diet which was both attractive to the animals and that could provide an increase in adipose tissue. After the induction of the diet, animals were euthanized and as a result, the animals in the PCAF group showed an intra-abdominal adiposity 0.74 ± 0.05 g, taken as the parameter for increasing fat in animals. Subsequently the study base was conducted for this research where animals were used (n = 64) divided into 2 groups: the Cafeteria Study Base Group (EBCAF) divided as follows: cafeteria + exercise + liraglutide (CEL) (n = 8), cafeteria + exercise + saline (CES) (n = 8), cafeteria + liraglutide (CL) (n = 8) and cafeteria + saline (CS) (n = 8). The Chow Study Base group (EBR) was divided into: exercise + liraglutide (EL) (n = 8), exercise + saline + (ES) (n = 8), liraglutide (L) (n = 8) and saline solution (SS) (n = 8). All animals went through the submission process to the cafeteria diet, followed by exercise protocol through swimming and treatment with the test substance intraperitoneally (200 mg / mL / kg). After the treatments, the animals were euthanized and had the following parameters evaluated: the muscle tissue mass, adipose tissue mass and biochemical parameters. It was observed that the processing done with the exercise-associated liraglutide reduced adipose tissue mass significantly (0.32 ± 0.05 g) compared to the saline group (0.53 ± 0.07 g). There were no changes in the muscle tissue of the group which was treated and exercised (1.39 ± 0.03 g) compared to the saline group (1.33 ± 0.03 g). Regarding biochemical parameters it was evident that there were changes in these parameters. Interesting to note that, although blood glucose values have been changed, the animals did not become diabetic. Thus, it appears that physical activity together with liraglutide is eficcient to the loss of intraabdominal adipose tissue and the maintenance of lean body mass thereby generating a satisfactory result in the pursuit of quality of life and disease prevention.

The gingiva, part of the masticatory mucosa tissue that covers the alveolar process and surrounds the cervical portion of teeth, has a definitive shape and texture associated with the eruption of teeth. Therefore, it must be included in rehabilitative planning, as the absence of papillae induces problems with esthetics, phonetics and food-impaction. The purpose of the present study was to approach the indications, limitations and techniques for making a removable artificial gingiva made of acrylic resin, by reporting on a clinical case. The patient, a 29-year-old woman, with the sequelae of periodontal disease, presented loss of interdental papilla causing “black holes”. She was dissatisfied with the esthetic appearance of her teeth, and also complained of long teeth and air escaping when she spoke. As treatment, prosthetic reconstruction was proposed, by means of a removable artificial gingiva (gingival epithesis) to simulate the presence of interdental papillae, providing better esthetics and phonetics. Gingival epithesis is an easily made, inserted and hygienically cleaned appliance that provides a simple and safe alternative solution for cases in which surgical techniques are limited.

Photodynamic therapy (PDT) consists of a non-toxic photosensitizing agent (FS) administration followed by a laser source resulting in a sequence of photochemical and photobiological processes that generate reactive oxygen species (ROS) that damaging cells. The present work evaluated the effects of PDT nanoemulsion-aluminum chloride phthalocyanine (AlClFc) mediated on malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD) and glutathione peroxidase (GPx) levels, which represent indicators involved in oxidative stress and antioxidant defenses. For this purpose, this study used 120 female rats of the Rattus norvegicus species, Wistar race, divided into 5 groups: Healthy (H), with periodontal disease (PD), with periodontal disease and treatment with FS (F), with periodontal disease and treatment with the laser (L); and periodontal disease and treatment with PDT (FL). An experimental model for represent periodontal disease (PD) was induced by ligature (split-mouth). Seven days later the induction of PD, the treatments were instituted according to the groups. In the group treated with PDT was applied 40μl FS (5μM) followed by laser irradiation diode InGaAlP (660nm, 100J / cm2). The rats were sacrificed on the 7th and 28th day after treatment and tissue specimens were removed and subjected to histological, immunohistochemical methods and enzymatic colorimetric measurements with detection by UV / VIS spectroscopy. Inflammatory changes, connective tissue disorganization and alveolar bone loss were displaying in groups with PD induced. The enzyme dosages showed that MDA levels were higher in PD induced groups, with no statistically significant differences (p> 0.05). High levels of GSH were found in groups L (p = 0.028) and FL (p = 0.028) compared with PD group, with statistically significant differences. Immunohistochemistry for SOD showed higher immunostaining in L and FL groups, compared to the PD group without statistically significant differences (p> 0.05). GPx showed lower immunoreactivity in the DP group when compared to the other groups and statistically significant differences were observed between the DPxL groups (p <0.05). TFD administered in this experiment did not induce elevation of MDA levels significantly increased the GSH levels and showed intense immunostaining pada SOD and GPx, showing that this therapy does not accentuated lipid peroxidation, however, it was able to induce effects on the antioxidant defenses processes. The LBI therapy appeared to show photomodulatory promoting effects reduction of the MDA levels, increasing GSH levels and with intense immunostaining for SOD and GPx, demonstrating that laser therapy induced antioxidant effects.

Os ecossistemas marinhos estão continuamente a ser sobrecarregados com contaminantes derivados de atividades humanas resultando numa dimunuição dos recursos marinhos. A exposição crónica a contaminantes, como metais pesados e poluentes orgânicos persistentes (POPs), pode afetar negativamente o ambiente marinho e, eventualmente, também os seres humanos. Grandes predadores pelágicos, como tubarões, são particularmente afetados pela poluição, principalmente através de processos de bioacumulação e biomagnificação. A fim de resolver o problema acima mencionado, são necessários estudos de avaliação de risco ambiental para prever os padrões de contaminação e evitar efeitos adversos que muitas vezes só são visíveis quando é tarde demais para tomar ações preventivas. Análises de concentração de químicos fornecem-nos informações sobre o nível de contaminação no ambiente; no entanto, podem não ser suficientes para entender como os organismos estão a ser afetados e há uma necessidade de relacionar essas quantificações com parâmetros biológicos. A avaliação de parâmetros bioquímicos, como a atividade enzimática, pode fornecer uma visão mais sensível e precisa sobre os níveis de contaminação. Tubarões como Prionace glauca são predadores de topo e portanto extremamente importantes nos ecossistemas marinhos. A sua grande distribuição, juntamente com o fácil acesso a amostras, fornecidas por barcos de pesca comercial, tornou-as um alvo favorável para utilização em ensaios toxicológicos. Este estudo teve como objetivo avaliar o potencial de P. glauca como uma espécie sentinela para pesquisas de monitorização de poluição, através do desenvolvimento e da aplicação de biomarcadores apropriados. As amostras de tecidos foram recolhidas de vinte tintureiras na costa de Portugal, a bordo de um barco comercial de pesca de espadarte. Níveis de POPs, assim como parâmetros bioquímicos relacionados com destoxificação, stress oxidativo e funções neuronais, foram medidos. A caracterização prévia da atividade das colinesterases no músculo e cérebro de P. glauca foi feita, já que não havia dados disponíveis sobre esta matéria. Esta caracterização foi essencial devido à existência de três classes de ChE conhecidas em peixes, acetilcolinesterase (AChE), butirilcolinesterase (BChE) e propionilcolinesterase (PChE), todas bastante suscetíveis a agentes anticolinérgicos, e outros contaminantes, tornando-as biomarcadores relevantes em estudos de monitorização de poluição. Os resultados obtidos indicaram que o cérebro de P. glauca aparenta possuir ChEs atípicas, revelando propriedades mistas de AChE e BChE e, que o músculo aparentemente possui maioritariamente AChE. A exposição in vitro a chloropyrifos-oxon provocou inibição de ChE das tintureiras em ambos os tecidos, com o cérebro sendo o tecido mais sensível e, por isso, o mais adequado para a detecção de compostos anticolinérgicos no ambiente. Este estudo indica que a actividade de ChE em tintureiras tem potencial para ser usada como um biomarcador sensível e fiável em programas de biomonitorização marinha. O fígado apresentou níveis mais elevados de POP, quando comparado com músculo. Foram encontradas correlações positivas e negativas entre os parâmetros de contaminação e de stresse oxidativo. Este estudo destaca a importância da caracterização de Che antes de a usar como um biomarcador em estudos ecotoxicológicos, e demonstra o grande potencial de P. glauca como espécie modelo e como sentinela de poluição marinha, através do uso de biomarcadores adequados.

As plantas são organismos sésseis, incapazes de se movimentar de modo a procurar melhores condições ambientais ou nutricionais. Desenvolveram, assim mecanismos que lhes permitem adaptar-se e sobreviver em condição de stress. O stress parece ser parcialmente descodificado num sinal de défice de energia que desencadeia uma resposta, que envolve a indução da expressão de genes relacionados com processos catabólicos e a repressão de genes envolvidos em processos anabólicos. As proteínas quinases e fosfatases desempenham um papel fundamental na regulação das vias de sinalização de stress e, em particular as quinases da superfamília das SnRK encontram-se envolvidas em vários processos da resposta a stress, principalmente abióticos. Enquanto as SnRK2 e SnRK3 estão sobretudo envolvidas na resposta a ABA e a stress hídrico e salino, as SnRK1 têm sido descritas como reguladores chave da resposta a défice energético. No entanto, um número crescente de estudos tem evidenciado a interligação entre estas duas vias de sinalização. Apesar da importância de SnRK1 na regulação da resposta ao stress e na regulação do crescimento e desenvolvimento em plantas, os mecanismos moleculares envolvidos são ainda pouco conhecidos. Com o objetivo de identificar proteínas que interagem com SnRK1 e que poderão estar envolvidas na sua via de sinalização, foi efetuado um rastreio, pelo método Y2H, utilizando uma biblioteca comercial normalizada construída a partir de mRNA extraído de onze tecidos de Arabidopsis. Foram identificadas 32 proteínas que potencialmente interagem com SnRK1.1, entre as quais MARD1 e NDF4. O estudo destas interações permitiu verificar que MARD1 medeia a interação entre SnRK1.1 e RAPTOR1B, sugerindo que, de forma semelhante à que ocorre em mamíferos, esta interação pode interligar a resposta ao défice energético envolvendo os complexos SnRK1 e TOR. Curiosamente, verificou-se que MARD1 medeia igualmente a interação entre SnRK1.1 e várias das MAPKs de Arabidopsis, o que poderá indicar que estas duas vias de sinalização estão igualmente interligadas. Foi também verificado que, no sistema de Y2H, SnRK1.1 interage, em alguns casos de forma depende de NDF4, com as proteínas DELLA, componentes essências da via de sinalização de giberelinas, o que pode sugerir uma interligação entre estas duas vias de sinalização e, desta forma, explicar parcialmente o papel de SnRK1 no crescimento e desenvolvimento das plantas. Um novo mecanismo de interligação entre as vias de sinalização de ABA e energia é sugerida pelos resultados obtidos em ensaios de Y2H mostrando que SnRK1.1 interage com SnRK2.3 e, pela observação de que em plantas que não expressam SnRK1.1/2, a expressão de genes de resposta a ABA é fortemente comprometida, sugerindo que SnRK1 poderá ativar as SnRK2 e, deste modo, ativar a resposta a ABA. No seu conjunto, estes dados evidenciam o papel de SnRK1 como regulador central da resposta ao défice energético em plantas e sugerem alguns dos mecanismos moleculares que poderão estar envidos, nomeadamente através da interação com várias outras vias de sinalização como o complexo TOR (interagindo com RAPTOR1B), as MAPKs, a via de sinalização de ABA (através da interação com SnRK2) e a via de sinalização de giberelinas (através da interação com proteínas DELLA).

This study addresses the question of attraction and retention of talent in companies that produce engineering projects in the area of oil and natural gas in the city of Natal. The objectives were to identify the mechanisms that these companies use to attract and retain talented professionals and what the relationship between these practices and performance of these organizations in the market. This is a case study of a qualitative nature which were included in the fullness of companies that work in that class in the capital Potiguar. Have been applied to the managers of these companies structured questionnaires with eleven issues orientativas based on theoretical reference adopted. The research finds that managers understand the word "talent", recognize the importance of the appreciation of its employees and the development of their innate abilities to better organizational performance, much due to the fact they are acting in a market of fierce competition. His companies - though not submit the formal procedures related to the subject in question - have mechanisms that can be characterized as the attraction and retention of talent. The relationships identified in this study are consistent with the results found in other studies and put the information here can serve as the basis for that other managers, including other areas, to reach excellence in their respective industries
