Controversy still exists concerning the potential ergogenic benefit of caffeine (CAF) for exercise performance. The purpose of this study was to compare the effects of CAF ingestion on endurance performance during exercise on a bicycle ergometer at two different intensities, i.e., approximately 10% below and 10% above the anaerobic threshold (AT). Eight untrained males, non-regular consumers of CAF, participated in this study. AT, defined as the intensity (watts) corresponding to a lactate concentration of 4 mM, was determined during an incremental exercise test from rest to exhaustion on an electrically braked cycle ergometer. on the basis of these measurements, the subjects were asked to cycle until exhaustion at two different intensities, i.e., approximately 10% below and 10% above AT. Each intensity was performed twice in a double-blind randomized order by ingesting either CAF (5 mg/kg) or a placebo (PLA) 60 min prior to the test. Venous blood was analyzed for free fatty acid, glucose, and lactate, before, during, and immediately after exercise. Rating of perceived exertion and time to exhaustion were also measured during each trial. There were no differences in free fatty acids or lactate levels between CAF and PLA during and immediately after exercise for either intensity. Immediately after exercise glucose increased in the CAF trial at both intensities. Rating of perceived exertion was significantly lower (CAF = 14.1 ± 2.5 vs PLA = 16.6 ± 2.4) and time to exhaustion was significantly higher (CAF = 46.54 ± 8.05 min vs PLA = 32.42 ± 14.81 min) during exercise below AT with CAF. However, there was no effect of CAF treatment on rating of perceived exertion (CAF = 18.0 ± 2.7 vs PLA = 17.6 ± 2.3) and time to exhaustion (CAF = 18.45 ± 7.28 min vs PLA = 19.17 ± 4.37 min) during exercise above AT. We conclude that in untrained subjects caffeine can improve endurance performance during prolonged exercise performed below AT and that the decrease of perceived exertion can be involved in this process

Um biodigestor anaeróbio de duas fases foi utilizado para se analisar a produção de metano com diferentes cargas de entrada de manipueira. A fase acidogênica foi realizada em processo de batelada e a metanogênica em biodigestor anaeróbio de fluxo ascendente e leito fixo com alimentação contínua. As cargas orgânicas de entrada variaram de 0,33 a 8,48 gDQO (Demanda Química de Oxigênio)/L.dia. A maior porcentagem de metano encontrada foi de 80,9%, com carga orgânica de 0,33g e a menor, 56,8%, obtida com 8,49gDQO/L.d. A maior taxa de redução de DQO foi de 88,89%, obtida com carga orgânica de 2,25g e a menor, 54,95%, com 8,48gDQO/L.d. Analisando-se os dados apresentados verificou-se que a biodigestão anaeróbia pode ser conduzida, pelo menos, de duas maneiras, ou seja, para produção de energia (metano) ou para redução de carga orgânica. A carga orgânica de entrada deve ser calculada em função do objetivo a ser alcançado com a biodigestão anaeróbia.

O desempenho animal é a medida mais direta de se avaliar a qualidade dos alimentos. Entretanto, dados de desempenho são insuficientes para se detectar as possíveis interações que possam ocorrer no ambiente ruminal. O objetivo do presente trabalho foi avaliar os possíveis efeitos associativos nas concentrações de ácidos graxos voláteis (AGVs), nitrogênio amoniacal (N-NH3) e pH da fração líquida remanescente da digestão da matéria seca (MS) de volumosos exclusivos (cana-de-açúcar= CN; capim-elefante com 60 dias= CP60 e 180 dias= CP180 de crescimento; e silagem de milho= SIL) e suas combinações (cana-de-açúcar+silagem de milho= CNSIL; cana-de-açúcar+capim-elefante-60d= CNCP60; cana-de-açúcar+capim-elefante-180d= CNCP180; silagem de milho+capim-elefante-60d= SILCP60; silagem de milho+capim-elefante-180d= SILCP180) na proporção de 50% na MS, que levam a resultados de desempenhos positivos ou negativos de bovinos. As concentrações de AGVs, N-NH3 e pH dos tratamentos foram: CN= 56,9 mmol L-1, 50,1 mg dL-1, 5,7; CNSIL= 61,4 mmol L-1, 50,7 mg dL-1, 5,8; CNCP60= 54,7 mmol L-1, 47,6 mg dL-1, 5,8; CNCP180= 45,4 mmol L-1, 49,4 mg dL-1, 6,0; SIL= 57,2 mmol L-1, 54,0 mg dL-1, 5,8; SILCP60= 57,1 mmol L-1, 53,1 mg dL-1, 5,9; SILCP180= 55,9 mmol L-1, 52,3 mg dL-1, 6,0; CP60= 58,1 mmol L-1, 49,4 mg dL-1, 5,9; CP180= 44,0 mmol L-1, 46,4 mg dL-1, 6,1. Os carboidratos não estruturais e amido, aliados à fibra e proteína, contribuíram para que ocorresse o efeito associativo positivo na mistura 50:50 cana/silagem. Isso pode ter propiciado os melhores resultados de desempenho em bovinos devido ao elevado padrão fermentativo.

The ultrastructural aspects of the ileum of Melipona quadrifasciata anthidioides, a stingless bee whose workers obtain their protein requirements from pollen, present morphological evidences of digestion and absorption. These evidences are represented by special relationships between food particles and the bacterial nora present and between their assemblage and the ileum wall, mainly in the ileum three anterior fourths. The apical features of the epithelium also are indicative of organic material absorption.

The equilibrium point between blood lactate production and removal (La-min(-)) and the individual anaerobic threshold (IAT) protocols have been used to evaluate exercise. During progressive exercise, blood lactate [La-](b), catecholamine and cortisol concentrations, show exponential increases at upper anaerobic threshold intensities. Since these hormones enhance blood glucose concentrations [Glc](b), this study investigated the [Glc] and [La-](b) responses during incremental tests and the possibility of considering the individual glucose threshold (IGT) and glucose minimum;(Glc(min)) in addition to IAT and La-min(-) in evaluating exercise. A group of 15 male endurance runners ran in four tests on the track 3000 m run (v(3km)); IAT and IGT- 8 x 800 m runs at velocities between 84% and 102% of v(3km); La-min(-) and Glc(min) - after lactic acidosis induced by a 500-m sprint, the subjects ran 8 x 800 m at intensities between 87% and 97% of v(3km); endurance test (ET)- 30 min at the velocity of IAT. Capillary blood (25 mu l) was collected for [La-](b) and [Glc](b) measurements. The TAT and IGT were determined by [La-](b) and [Glc](b) kinetics during the second test. The La-min(-) and Glc(min) were determined considering the lowest [La-] and [Glc](b) during the third test. No differences were observed (P < 0.05) and high correlations were obtained between the velocities at IAT [283 (SD 19) and IGT 281 (SD 21)m. min(-1); r = 0.096; P < 0.001] and between La,, [285 (SD 21)] and Glc(min) [287 (SD 20) m. min(-1) = 0.77; P < 0.05]. During ET, the [La-](b) reached 5.0 (SD 1.1) and 5.3 (SD 1.0) mmol 1(-1) at 20 and 30 min, respectively (P > 0.05). We concluded that for these subjects it was possible to evaluate the aerobic capacity by IGT and Glc(min), as well as by IAT and La-min(-).

A method for the total mercury determination in fish and shrimps employing chronopotentiometric stripping analysis on gold film electrodes is described. Fish and shrimp tissues were digested using a microwave oven equipped with closed vessels. We developed a microwave heating program which decomposed all the samples employing diluted nitric acid and hydrogen peroxide. The proposed method was validated by analyzing a certified reference material and then applied for different fish species from fresh water and seawater acquired in local markets of São Paulo city, Brazil. The Brazilian legislation establishes 0.5 and 1 mg per kilogram of fish as upper limit of mercury for omnivorous and predator species, respectively. Except for blue shark tissues, the mercury content was situated below 0.5 mu g g(-1) for all the analyzed samples. The detection limit of the proposed method was calculated as 5 ng g(-1) of sample utilizing 5 minutes of electrodeposition (+300 mV vs. Ag/AgCl) on the gold electrode. (c) 2006 Elsevier Ltd. The present study was designed to determine the exercise intensity equivalent to the metabolic aerobic/anaerobic transition of alloxan diabetic rats, through lactate minimum test (LMT), and to evaluate the effects of swimming exercise at this intensity (LM) on the glucose and protein metabolism of these animals. Adult male Wistar rats received alloxan (SD, alloxan-injected rats that remained sedentary) intravenously (30 mg kg(-1) body weight) for diabetes induction. As controls (SC, vehicle-injected rats that remained sedentary), vehicle-injected rats were utilized. Two weeks later, the animals were submitted to oral glucose tolerance test (oGTT) and LMT. After the tests, some of the animals were submitted to swimming exercise training [TC (vehicle-injected rats that performed a 6-week exercise program) and TD (alloxan-injected rats that performed a 6-week exercise program)] for I h day(-1), 5 days week(-1), with an overload equivalent to LM determined by LMT, for 6 weeks. At the end of the experiment, the animals were submitted to a second LMT and oGTT, and blood and skeletal muscle assessments (protein synthesis and degradation in the isolated soleus muscle) were made. The overload equivalent to LM at the beginning of the experiment was lower in the SID group than in the SC group. After training, the overload equivalent to LM was higher in the TC and TD groups than in the SC and SD groups. The blood glucose of TD rats during oGTT was lower than that of SD rats. Protein degradation was higher in the SD group than in other groups. We conclude that LMT was sensitive to metabolic and physiologic alterations caused by uncontrolled diabetes. Training at LM intensity improved aerobic condition and the glucose and protein metabolism of alloxan diabetic rats. (C) 2007 Elsevier B.V. All rights reserved.

Mosquito larvae are believed to be capable of digesting chitin, an insoluble polysaccharide of N-acetylglucosamine, for their nutritional benefit. Studies based on physiological and biochemical assays were conducted in order to detect the presence of chitinase activities in the gut of the detritus-feeding Aedes aegypti larvae. Larvae placed for 24 h in suspensions of chitin azure were able to digest the ingested chitin. Semi-denaturing PAGE using glycol chitin and two fluorogenic substrate analogues showed the presence of two distinct chitinase activities: an endochitinase that catalyzed the hydrolysis of chitin and an endochitinase that cleaved the short substrates [4MU(GlcNAc)(3)] and [4MU(GlcNAc)(2)] that hydrolyzed the chitobioside [4MU(GlcNAc)(2)]. The endochitinase had an extremely broad pH-activity against glycol chitin and chitin azure, pH ranging from 4.0 to 10.0. When the substrate [4MU(GlcNAc)(3)] was used, two activities were observed at pH ranges 4.0-6.0 and 8.0-10.0. Chitinase activity against [4MU(GlcNAc)(3)] was detected throughout the gut with the highest specific activity in the hindgut. The pH of the gut contents was determined by observing color changes in gut after feeding the larvae with color indicator dyes. It was observed a correlation between the pH observed in the gut of feeding larvae (pH 10-6.0) and the optimum pH for gut chitinase activities. In this work, we report that gut chitinases may be involved in the digestion of chitin-containing structures and also in the partial degradation of the chitinous peritrophic matrix in the hindgut. The chemical fractionation and bioleaching of Mn, At, Zn, Cu and Ti in municipal sewage sludge were investigated using Thiobacillus ferrooxidans as leaching microorganism. As a result of the bacterial activity, ORP increase and pH reduction were observed. Metal solubilization was accomplished only in experimental systems supplemented with energy source (Fe(II)). The solubilization efficiency approached similar to80% for Mn and Zn, 24% for Cu, 10% for At and 0.2% for Ti. The chemical fractionation of Mn, At, Zn, Cu and Ti was investigated using a five-step sequential extraction procedure employing KNO3. KF, Na4P2O7, EDTA and HNO3. The results show that the bioleaching process affected the partitioning of Mn and Zn, increasing its percentage of elution in the KNO3 fraction while reducing it in the KF, Na4P2O7 and EDTA fractions. No significant effect was detected on the partitioning of Cu and Al. However, quantitatively the metals Mn, Zn, Cu and At were extracted with higher efficiency after the bacterial activity. Titanium was unaffected by the bioleaching process in both qualitative and quantitative aspects. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

Some snakes have a feeding regime characterized by the infrequent ingestion of relatively large meals, causing impressive increments in post-prandial metabolism. Metabolism remains elevated for many days, while digestion proceeds, resulting in considerable investment of time and energy. Snakes actively adjust thermoregulatory behavior to raise their body temperature during digestion, exhibiting a post-prandial thermophilic response that accelerates digestion at the expense of higher metabolic rates. In the present study, we investigated the possibility that endogenously derived heat, originating as a byproduct of the post-prandial increase in metabolism, could itself contribute to the elevated body temperature during digestion in the South American rattlesnake Crotalus durissus. We assessed heat production, at a constant environmental temperature, by taking infrared (IR) images of snakes during fasting and after being fed meals varying from 10% to 50% of their own body masses. Our results show clearly that digesting rattlesnakes have significantly increased body temperatures, even when precluded from adjusting their thermoregulatory behavior. The feeding-derived thermogenesis caused the surface body temperature of rattlesnakes to increase by 0.9-1.2degreesC, a temperature change that will significantly affect digestive performance. The alterations in body temperature following feeding correlated closely with the temporal profile of changes in post-prandial metabolism. Moreover, the magnitude of the thermogenesis was greater for snakes fed large meals, as was the corresponding metabolic response. Since IR imaging only assesses surface temperatures,, the magnitude of the thermogenesis and the changes in deep core temperature could be even more pronounced than is reported here.
