For esophageal reconstruction in newborns with esophageal atresia, esophageal reunion with an end-to-end anastomosis is the ideal procedure, although it may result in leaks and strictures due to tension on the suture line, mainly in cases with a wide gap between the ends. Circular myotomy (Livaditis' procedure) is the best method to elongate the proximal esophageal pouch and reduce anastomotic tension. This experimental investigation in dogs was undertaken to attempt to verify that circular myotomy decreases the anastomotic leak rate in newborns with wide gap esophageal atresia, and to analyze whether the technique promotes morphologic changes in the anastomotic scar. A pilot study demonstrated that it is necessary to resect more than 8 cm (40% of the total esophageal length) in order to obtain high leak rates. In the experimental project, such resection was performed in dogs divided into two groups (control group, anastomosis only, and experimental group, anastomosis plus circular myotomy in the proximal esophageal segment). The animals were killed in the 14th postoperative day, submitted to autopsy, and were evaluated as to the presence of leaks and strictures, as well as to the features (macroscopic and microscopic aspects) of the anastomosis. Leak rates were the same in both groups. Morphometric analysis revealed that in animals in the experimental group, the anastomotic scar was thinner than the control animals, and the isolated muscular manchette distal to the site of myotomy was replaced by fibrous tissue. Correspondingly, a decreased number of newly formed small vessels were noted in the experimental animals, compared to control animals. We concluded that circular myotomy does not decrease the incidence of anastomotic leaks, and it also promotes deleterious changes in anastomotic healing.

Este estudo tem como objetivo construir um modelo estocástico de alta resolução da morfologia e dos teores em metal, do depósito mineral do Zambujal, Mina de Neves-Corvo. O depósito do Zambujal é um corpo vulcanogénico que se localiza no setor Português, na parte mais a Sul da Faixa Piritosa Ibérica, com sulfuretos maciços no topo e fissurais na base. Para construir um modelo estocástico deste depósito, onde os teores em metal evidenciam zonamento, é importante ter em conta a proporção local de sulfuretos na matriz rochosa. Como esta variável não é quantificada em laboratório, propõe-se a utilização da densidade das amostras como um indicador indireto da proporção de minérios na matriz rochosa. Conhecida esta variável, a modelação dos teores em metal pode ser feita para os chamados teores relativos (teores em metal na fração de sulfuretos). As principais etapas da metodologia proposta são: (a) para cada amostra analisada no laboratório, estimação de soluções para a variável proporção de sulfuretos na matriz rochosa ????(????), tendo em conta a paragénese principal do depósito, os teores e a densidade da rocha; (b) construção de um modelo morfológico 3D de baixa resolução com duas regiões, minérios maciços e minérios fissurais, por digitalização de limites em perfis, interpolação de superfícies e conversão para o modelo de blocos do depósito; (c) construção de um modelo morfológico 3D de alta resolução para todo o depósito da variável ????(????) por Simulação Sequencial Direta (SSD), tendo como informação condicionante os histogramas regionais de ????(????) para minérios maciços e minérios fissurais e as soluções obtidas em (a) para ????(????) na localização das sondagens; (d) SSD dos teores relativos em cobre, zinco e prata; (e) discussão dos resultados e quantificação de recursos. Os resultados foram validados por comparação com os equivalentes obtidos numa estimação por krigagem normal dos teores em metal e mostraram ser da mesma ordem de grandeza. O conjunto de imagens simuladas das variáveis ????(????) e teores permite quantificar a incerteza do conhecimento do depósito relativamente à informação disponível.

PURPOSE: The recovery of a bone fracture is a process that is not yet fully understood. The literature conflicts on the results obtained by the interposition of foreign tissue inside a damaged bone. The objective of the present study was to ascertain the effect of placing muscle tissue between the stumps of a fractured bone. METHOD: The study was carried out on 10 rabbits divided into 2 groups (n = 5): Group 1-partial fracture of the humerus and interposition of muscle tissue; Group 2-complete fracture of the humerus and interposition of muscle tissue. The fractured limb of all animals was immobilized for 8 weeks. At the end of this time, the rabbits were killed and their operated humeri were carefully removed for roentgenological and histological assessment. RESULTS: All humeri of Group 1 recovered their integrity and normal aspect. However, the healing of the humeri of Group 2 was not perfect. Gross angulation of the bone diaphysis occurred in all animals, and immature trabecular bone, osteochondral tissue, and persistence of muscle tissue substituted normal bone. CONCLUSIONS: Interposed muscle does not affect partial bone fracture healing but causes instability in a complete fracture.

No presente estudo são apresentados e discutidos os níveis de N, P, H, Ca, Mg, Na e SO4, além da percentagem de carbono orgânico nas partes vegetativas do aría (Calathea allouia (Aubl.) Lindl.). Os dados foram obtidos por espectroscopia de emissão de plasma, análise por injeção em fluído e volumetria.

The weak fixation of biomaterials within the bone structure is one of the major reasons of implants failures. Calcium phosphate (CaP) coatings are used in bone tissue engineering to improve implant osseointegration by enhancing cellular adhesion, proliferation and differentiation, leading to a tight and stable junction between implant and host bone. It has also been observed that materials compatible with bone tissue either have a CaP coating or develop such a calcified surface upon implantation. Thus, the development of bioactive coatings becomes essential for further improvement of integration with the surrounding tissue. However, most of current applied CaP coatings methods (e.g. physical vapor deposition), cannot be applied to complex shapes and porous implants, provide poor structural control over the coating and prevent incorporation of bioactive organic compounds (e.g. antibiotics, growth factors) because of the used harsh processing conditions. Layer-by-layer (LbL) is a versatile technology that permits the building-up of multilayered polyelectrolyte films in mild conditions based on the alternate adsorption of cationic and anionic elements that can integrate bioactive compounds. As it is recognized in natureâ s biomineralization process the presence of an organic template to induce mineral deposition, this work investigate a ion based biomimetic method where all the process is based on LbL methodology made of weak natural-origin polyelectrolytes. A nanostructured multilayer component, with 5 or 10 bilayers, was produced initially using chitosan and chondroitin sulphate polyelectrolyte biopolymers, which possess similarities with the extracellular matrix and good biocompatibility. The multilayers are then rinsed with a sequential passing of solutions containing Ca2+ and PO43- ions. The formation of CaP over the polyelectrolyte multilayers was confirmed by QCM-D, SEM and EDX. The outcomes show that 10 polyelectrolyte bilayer condition behaved as a better site for initiating the formation of CaP as the precipitation occur at earlier stages than in 5 polyelectrolyte bilayers one. This denotes that higher number of bilayers could hold the CaP crystals more efficiently. This work achieved uniform coatings that can be applied to any surface with access to the liquid media in a low-temperature method, which potentiates the manufacture of effective bioactive biomaterials with great potential in orthopedic applications.

Burn wound healing involves a complex set of overlapping processes in an environment conducive to ischemia, inflammation, and infection costing $7.5 billion/year in the US alone, in addition to the morbidity and mortality that occur when the burns are extensive. We previously showed that insulin, when topically applied to skin excision wounds, accelerates re-epithelialization, and stimulates angiogenesis. More recently, we developed an alginate sponge dressing (ASD) containing insulin encapsulated in PLGA microparticles that provides a sustained release of bioactive insulin for >20days in a moist and protective environment. We hypothesized that insulin-containing ASD accelerates burn healing and stimulates a more regenerative, less scarring, healing. Using a heat-induced burn injury in rats, we show that burns treated with dressings containing 0.04mg insulin/cm2, every three days for 9 days, have faster closure, faster rate of disintegration of dead tissue, and decreased oxidative stress.In addition, in insulin-treated wounds the pattern of neutrophil inflammatory response suggests faster clearing of the burn dead tissue. We also observe faster resolution of the pro-inflammatory macrophages. We also found that insulin stimulates collagen deposition and maturation with the fibers organized more like a basket weave (normal skin) than aligned and crosslinked (scar tissue). In summary , application of ASD-containing insulin-loaded PLGA particles on burns every three days stimulates faster and more regenerative healing. These results suggest insulin as a potential therapeutic agent in burn healing and, because of its long history of safe use in humans, insulin could become one of the treatments of choice when repair and regeneration are critical for proper tissue function.

Imaging techniques are the standard method for assessment of fracture healing processes. However, these methods are perhaps not entirely reliable for early detection of complications, the most frequent of these being delayed union and non-union. A prompt diagnosis of such disorders could prevent prolonged patient distress and disability. Efforts should be directed towards the development of new technologies for improving accuracy in diagnosing complications following bone fractures. The variation in the levels of bone turnover markers (BTMs) have been assessed with regard to there ability to predict impaired fracture healing at an early stage, nevertheless the conclusions of some studies are not consensual. In this article the authors have revised the potential of BTMs as early predictors of prognosis in adult patients presenting traumatic bone fractures but who did not suffer from osteopenia or postmenopausal osteoporosis. The available information from the different studies performed in this field was systematized in order to highlight the most promising BTMs for the assessment of fracture healing outcome.

The incorporation of fly ash (FA) in cementitious matrices have been frequently used in order to make the matrix more resistant to the action of chlorides. On the other hand, it is known that Ca (OH)2 existing in the matrix is partially consumed by the pozzolanic reactions, which facilitates the advancement of carbonation. Given that the combined action between carbonation and chloride penetration is a fact little known, we speculate about the behaviour of the matrix in this context. This study investigates the influence of the presence of chlorides on the carbonation in mortars with FA. Samples with 0% and 40% replacement of cement CEM I 42.5 R for FA were molded with water/binder 0.56 and 0.52 respectively. After 90 days of curing the specimens were subjected to cycles of immersion/drying for 56 days. Half of the samples was subjected to the following cycle: two days in a solution containing NaCl (concentration equal to 3.5 %); 12 days in the carbonation chamber (4% of CO2). The other half was: two days in water; 12 days in the carbonation chamber. Then, the development of carbonation was evaluated. The results indicate that the presence of chlorides influences the carbonation. The specimens submitted to the exclusive action of CO2 showed a greater depth of carbonation compared to that presented by the specimens subjected to combined action. This may be related to changes in properties of the matrix which may lead to further refinement of the pores and related to the presence of the salt that can lead to partial filling of the pores and the increase in moisture content.

Long term applications of leguminous green mulch could increase mineralizable nitrogen (N) beneath cupuaçu trees produced on the infertile acidic Ultisols and Oxisols of the Amazon Basin. However, low quality standing cupuaçu litter could interfere with green mulch N release and soil N mineralization. This study compared mineral N, total N, and microbial biomass N beneath cupuaçu trees grown in two different agroforestry systems, north of Manaus, Brazil, following seven years of different green mulch application rates. To test for net interactions between green mulch and cupuaçu litter, dried gliricidia and inga leaves were mixed with senescent cupuaçu leaves, surface applied to an Oxisol soil, and incubated in a greenhouse for 162 days. Leaf decomposition, N release and soil N mineralization were periodically measured in the mixed species litter treatments and compared to single species applications. The effect of legume biomass and cupuaçu litter on soil mineral N was additive implying that recommendations for green mulch applications to cupuaçu trees can be based on N dynamics of individual green mulch species. Results demonstrated that residue quality, not quantity, was the dominant factor affecting the rate of N release from leaves and soil N mineralization in a controlled environment. In the field, complex N cycling and other factors, including soil fauna, roots, and microclimatic effects, had a stronger influence on available soil N than residue quality.

The use of buffers to maintain the pH within a desired range is a very common practice in chemical, biochemical and biological studies. Among them, zwitterionic N-substituted aminosulfonic acids, usually known as Good's buffers, although widely used, can complex metals and interact with biological systems. The present work reviews, discusses and updates the metal complexation characteristics of thirty one commercially available buffers. In addition, their impact on biological systems is also presented. The influences of these buffers on the results obtained in biological, biochemical and environmental studies, with special focus on their interaction with metal ions, are highlighted and critically reviewed. Using chemical speciation simulations, based on the current knowledge of the metal-buffer stability constants, a proposal of the most adequate buffer to employ for a given metal ion is presented.

Avaliou-se a produção de palmito do estipe principal e do primeiro perfilho da pupunheira (Bactris gasipaes Kunth), utilizando diferentes fontes e formas de adubação (orgânica e mineral) num LATOSSOLO AMARELO na Amazônia Central. O delineamento experimental foi blocos casualizados com três repetições, utilizando esquema fatorial 2 x 7, sendo os fatores: plantas com e sem espinhos no estipe oriundas de Yurimaguas, Peru; e diferentes formas de adubação (testemunha sem adubo; esterco de galinha de postura em cova (25 t ha-1); adubo mineral em cova (225-90-180 kg.ha-1 de N-P2O5-K2O); esterco de galinha de postura em cobertura (25 t ha-1); adubo mineral em cobertura (225-90-180 kg.ha-1); esterco de galinha de postura na cova (12,5 t ha-1) + adubo mineral em cobertura (112,5-45-90 kg.ha-1) e adubo mineral parcelado em 3 vezes (na cova 75 kg ha-1 de N, 90 kg ha-1 de P2O5 e 60 kg.ha-1 de K2O + 2 aplicações iguais de 75 kg.ha-1 de N e 60 kg.ha-1 de K2O), todas as formas de adubação foram repetidas no 2º e 3º ano em cobertura. A aplicação de fertilizantes orgânico e mineral elevou o pH e a concentração de nutrientes disponíveis no solo. A produção de palmito no primeiro perfilho com espinhos (1407 kg.ha-1) foi maior que nas plantas sem espinhos (1037 kg.ha-1). A produção de palmito liquido foi maior nos tratamentos com esterco em cobertura (estipe principal 1551kg.ha-1 e perfilhos 3004kg.ha-1) e "esterco 50% na cova + adubo mineral em cobertura 50%" (planta principal 1545 kg ha-1 e perfilhos 2986 kg ha-1). A testemunha não atingiu altura de corte até aos 40 meses após o plantio.

Buruli Ulcer (BU) is a necrotizing skin disease caused by Mycobacterium ulcerans infection. BU is characterized by a wide range of clinical forms, including non-ulcerative cutaneous lesions that can evolve into severe ulcers if left untreated. Nevertheless, spontaneous healing has been reported to occur, although knowledge on this process is scarce both in naturally infected humans and experimental models of infection. Animal models are useful since they mimic different spectrums of human BU disease and have the potential to elucidate the pathogenic/protective pathway(s) involved in disease/healing. In this time-lapsed study, we characterized the guinea pig, an animal model of resistance to M. ulcerans, focusing on the macroscopic, microbiological and histological evolution throughout the entire experimental infectious process. Subcutaneous infection of guinea pigs with a virulent strain of M. ulcerans led to early localized swelling, which evolved into small well defined ulcers. These macroscopic observations correlated with the presence of necrosis, acute inflammatory infiltrate and an abundant bacterial load. By the end of the infectious process when ulcerative lesions healed, M. ulcerans viability decreased and the subcutaneous tissue organization returned to its normal state after a process of continuous healing characterized by tissue granulation and reepethelialization. In conclusion, we show that the experimental M. ulcerans infection of the guinea pig mimics the process of spontaneous healing described in BU patients, displaying the potential to uncover correlates of protection against BU, which can ultimately contribute to the development of new prophylactic and therapeutic strategies.

A pimenta-longa (Piper hispidinervum C. DC.) é uma das principais fontes alternativas para a produção de safrol empregado como matéria prima na fabricação de inseticidas naturais e aromatizantes. O objetivo da pesquisa foi avaliar o efeito da omissão dos macronutrientes e micronutrientes, sobre a composição mineral de plantas de pimenta-longa, e caracterizar os sintomas de deficiências decorrentes dessa limitação, utilizando-se da técnica do elemento faltante. O experimento foi conduzido em casa de vegetação, com quatro repetições e doze tratamentos, completo e omissão individual de N, P, K, Ca, Mg, S, B, Cu, Fe, Mn e Zn em delineamento inteiramente ao acaso. Os valores dos teores foliares nos tratamentos com omissão dos nutrientes foram inferiores àqueles obtidos no tratamento completo. As omissões individuais dos nutrientes promoveram alterações na composição mineral de macro e micronutrientes. Os teores de macronutrientes (g kg-1) e micronutrientes (mg kg-1) obtidos nas folhas sem (completo) e com sintomas de deficiências (omissões) foram, respectivamente: N = 18,32; P = 7,02; K = 22,17; Ca = 15,75; Mg = 8,25; S = 5,12; B = 42,25; Fe = 325,00; Mn = 100; Zn = 61,50, com deficiência: N = 8,98; P = 2,52; K = 8,57; Ca = 10,20; Mg = 1,85; S = 0,90; B = 15,50; Fe = 234,00; Mn = 55; Zn = 53.
