Pain perception has evolved as a warning mechanism to alert organisms to tissue damage and dangerous environments. In humans, however, undesirable, excessive or chronic pain is a common and major societal burden for which available medical treatments are currently suboptimal. New therapeutic options have recently been derived from studies of individuals with congenital insensitivity to pain (CIP). Here we identified 10 different homozygous mutations in PRDM12 (encoding PRDI-BF1 and RIZ homology domain-containing protein 12) in subjects with CIP from 11 families. Prdm proteins are a family of epigenetic regulators that control neural specification and neurogenesis. We determined that Prdm12 is expressed in nociceptors and their progenitors and participates in the development of sensory neurons in Xenopus embryos. Moreover, CIP-associated mutants abrogate the histone-modifying potential associated with wild-type Prdm12. Prdm12 emerges as a key factor in the orchestration of sensory neurogenesis and may hold promise as a target for new pain therapeutics.

Six of 7 FXYD proteins have been shown to be tissue-specific modulators of Na,K-ATPase. In this study, we have identified two splice variants of human FXYD3, or Mat-8, in CaCo-2 cells. Short human FXYD3 has 72% sequence identity with mouse FXYD3, whereas long human FXYD3 is identical to short human FXYD3 but has a 26-amino acid insertion after the transmembrane domain. Short and long human FXYD3 RNAs and proteins are differentially expressed during differentiation of CaCo-2 cells. Long human FXYD3 is mainly expressed in nondifferentiated cells and short human FXYD3 in differentiated cells and both FXYD3 variants can be co-immunoprecipitated with a Na,K-ATPase antibody. In contrast to mouse FXYD3, which has two transmembrane domains for lack of cleavage of the signal peptide, human FXYD3 has a cleavable signal peptide and adopts a type I topology. After co-expression in Xenopus oocytes, both human FXYD3 variants associate stably only with Na,K-ATPase isozymes but not with H,K-ATPase or Ca-ATPase. Similar to mouse FXYD3, short human FXYD3 decreases the apparent K(+) and Na(+) affinity of Na,K-ATPase over a large range of membrane potentials. On the other hand, long human FXYD3 decreases the apparent K(+) affinity only at slightly negative and positive membrane potentials and increases the apparent Na(+) affinity of Na,K-ATPase. Finally, both short and long human FXYD3 induce a hyperpolarization activated current, similar to that induced by mouse FXYD3. Thus, we have characterized two human FXYD3 isoforms that are differentially expressed in differentiated and non-differentiated cells and show different functional properties.

Sex chromosome differentiation in Rana temporaria varies strikingly among populations or families: whereas some males display well-differentiated Y haplotypes at microsatellite markers on linkage group 2 (LG2 ), others are genetically undistinguishable from females. We analysed with RADseq markers one family from a Swiss lowland population with no differentiated sex chromosomes, and where sibship analyses had failed to detect any association between the phenotypic sex of progeny and parental haplotypes. Offspring were reared in a common tank in outdoor conditions and sexed at the froglet stage. We could map a total of 2177 SNPs (1123 in the mother, 1054 in the father), recovering in both adults 13 linkage groups (= chromosome pairs) that were strongly syntenic to Xenopus tropicalis despite > 200 My divergence. Sexes differed strikingly in the localization of crossovers, which were uniformly distributed in the female but limited to chromosome ends in the male. None of the 2177 markers showed significant association with offspring sex. Considering the very high power of our analysis, we conclude that sex determination was not genetic in this family; which factors determined sex remain to be investigated.

Identifying homology between sex chromosomes of different species is essential to understanding the evolution of sex determination. Here, we show that the identity of a homomorphic sex chromosome pair can be established using a linkage map, without information on offspring sex. By comparing sex-specific maps of the European tree frog Hyla arborea, we find that the sex chromosome (linkage group 1) shows a threefold difference in marker number between the male and female maps. In contrast, the number of markers on each autosome is similar between the two maps. We also find strongly conserved synteny between H. arborea and Xenopus tropicalis across 200 million years of evolution, suggesting that the rate of chromosomal rearrangement in anurans is low. Finally, we show that recombination in males is greatly reduced at the centers of large chromosomes, consistent with previous cytogenetic findings. Our research shows the importance of high-density linkage maps for studies of recombination, chromosomal rearrangement and the genetic architecture of ecologically or economically important traits.

The volatile oil from the leaves of E. urograndis was analyzed by GC and GC-MS. It was identified 10 compounds in which, orto-cimene (41.4%) and 1,8-cineol (25.8%) were the main constituents. The induction of deleterious effect to aquatic organisms due to the presence of volatile oil lixiviated from E. urograndis leaves was studied using Daphnia laevis and D. similis as bioindicators. Through the results of toxicological tests it was possible to show that the litterbag of E. urograndis represents a risk factor for the aquatic ecosystem of lakes and rivers that are in the surrounding area of large scale Eucalyptus plantations. The method can be used for monitor the quality of these types of aquatic environments.

A composição taxonômica de diatomáceas presentes sobre o molusco Limnoperna fortunei (mexilhão dourado), introduzido no Lago Guaíba através da água de lastro de navios oriundos da Argentina, foi analisada com o objetivo de identificar e avaliar a ocorrência das espécies quanto ao habitat e origem. Os moluscos foram retirados de plataforma flutuante de madeira e de bóias situadas na região norte do Lago Guaíba, no período de agosto a novembro de 2005. As diatomáceas presentes sobre as conchas dos moluscos foram raspadas, analisadas sob microscópio e posteriormente oxidadas para identificação. Os resultados demonstraram que a concha do molusco pode ser considerada um habitat favorável às diatomáceas, tendo em vista a riqueza de espécies. A associação de diatomáceas não demonstrou estar restrita a este habitat, pois as espécies mais frequentes Cocconeis fluviatilis Wall., C. placentula Ehr. var. lineata (Ehr.) Heurck, Encyonema silesiacum (Bleisch) Mann, Eunotia pseudosudetica Metz., Lange-Bert. & Garcia-Rodr., Gomphonema parvulum (Kütz.) Kütz., Hydrosera whampoensis (Schw.) Deby, Luticola goeppertiana (Bleisch) Mann, Melosira varians Ag., Navicula symmetrica Patr., Nitzschia palea (Kütz.) Smith, Pinnularia acrosphaeria Smith, Planothidium frequentissimum (Lange-Bert.) Round & Bukh., Pleurosira laevis (Her.) Comp. e Ulnaria ulna (Nitzsch.) Comp. são também encontradas em substratos rochosos e no plâncton na Bacia Hidrográfica do Lago Guaíba. Entretanto, Eunotia pseudosudetica, Luticola dapalis (Freng.) Mann e Gomphonema mexicanum Grun. são táxons só registrados, até o momento, para o Rio da Prata, Argentina. Estes primeiros registros nas águas do Lago Guaíba suportam a hipótese de uma recente introdução dessas espécies via água de lastro, assim como L. fortunei.

A floristic study of Coscinodiscophyceae and Fragilariophyceae in the Lago dos Tigres hydrologic system is presented. Phytoplankton samples were collected with a plankton net (25 µm mesh) at 11 sampling stations, three in lotic areas and eight in lacustrine zones. Eleven species of Coscinodiscophyceae and eight of Fragilariophyceae were identified; descriptions, comments, their geographic distributions for the midwestern region of Brazil, type of environment (lentic/lotic), and illustrations are provided. Among the taxa found, 13 were recorded for the first time in midwestern Brazil: Aulacoseira alpigena, A. distans, A. granulata var. angustissima, A. granulata var. valida, A. muzzanensis, Discostella stelligera var. stelligera, Fragilaria capucina var. fragilarioides, F. delicatissima var. delicatissima, F. javanica, Pleurosira laevis, Staurosira acutirostrata, Staurosirella crassa, and Ulnaria ulna.

Most cells exchange ions and small metabolites via gap junction channels. These channels are made of two hemichannels (connexons), each formed by the radial arrangement of six connexin (Cx) proteins. Connexins span the bilayer four times (M1-M4) and have both amino- and carboxy-termini (NT, CT) at the cytoplasmic side of the membrane, forming two extracellular loops (E1, E2) and one inner (IL) loop. The channels are regulated by gates that close with cytosolic acidification (e.g., CO2 treatment) or increased calcium concentration, possibly via calmodulin activation. Although gap junction regulation is still unclear, connexin domains involved in gating are being defined. We have recently focused on the CO2 gating sensitivity of Cx32, Cx38 and various mutants and chimeras expressed in Xenopus oocytes and studied by double voltage clamp. Cx32 is weakly sensitive to CO2, whereas Cx38 is highly sensitive. A Cx32 chimera containing the second half of the inner loop (IL2) of Cx38 was as sensitive to CO2 as Cx38, indicating that this domain plays an important role. Deletion of CT by 84% did not affect CO2 sensitivity, but replacement of 5 arginines (R) with sparagines (N) at the beginning of CT (C1) greatly enhanced the CO2 sensitivity of Cx32. This suggests that whereas most of CT is irrelevant, positive charges of C1 maintain the CO2 sensitivity of Cx32 low. As a hypothesis we have proposed a model that involves charge interaction between negative residues of the beginning of IL1 and positive residues of either C1 or IL2. Open and closed channels would result from IL1-C1 and IL1-IL2 interactions, respectively

Water channels or aquaporins (AQPs) have been identified in a large variety of tissues. Nevertheless, their role in the human gastrointestinal tract, where their action is essential for the reabsorption and secretion of water and electrolytes, is still unclear. The purpose of the present study was to investigate the structure and function of water channels expressed in the human colon. A cDNA fragment of about 420 bp with a 98% identity to human AQP3 was amplified from human stomach, small intestine and colon by reverse transcription polymerase chain reaction (RT-PCR) and a transcript of 2.2 kb was expressed more abundantly in colon than in jejunum, ileum and stomach as indicated by Northern blots. Expression of mRNA from the colon of adults and children but not from other gastrointestinal regions in Xenopus oocytes enhanced the osmotic water permeability, and the urea and glycerol transport in a manner sensitive to an antisense AQP3 oligonucleotide, indicating the presence of functional AQP3. Immunocytochemistry and immunofluorescence studies in human colon revealed that the AQP3 protein is restricted to the villus epithelial cells. The immunostaining within these cells was more intense in the apical than in the basolateral membranes. The presence of AQP3 in villus epithelial cells suggests that AQP3 is implicated in water absorption across human colonic surface cells.

Currents mediated by calcium-activated chloride channels (CaCCs), observed for the first time in Xenopus oocytes, have been recorded in many cells and tissues ranging from different types of neurons to epithelial and muscle cells. CaCCs play a role in the regulation of excitability in neurons including sensory receptors. In addition, they are crucial mediators of chloride movements in epithelial cells where their activity regulates electrolyte and fluid transport. The roles of CaCCs, particularly in epithelia, are briefly reviewed with emphasis on their function in secretory epithelia. The recent identification by three independent groups, using different strategies, of TMEM16A as the molecular counterpart of the CaCC is discussed. TMEM16A is part of a family that has 10 other members in mice. The discovery of the potential TMEM16 anion channel activity opens the way for the molecular investigation of the role of these anion channels in specific cells and in organ physiology and pathophysiology. The identification of TMEM16A protein as a CaCC chloride channel molecule represents a great triumph of scientific perseverance and ingenuity. The varied approaches used by the three independent research groups also augur well for the solidity of the discovery.

Le canal calcique de type L, Cav1.2, joue un rôle clé dans le couplage excitation-contraction des myocytes ventriculaires. Il a été montré que la sous-unité Cavα1 était sujette à l’épissage alternatif et que ce phénomène pouvait mener à une protéine tronquée en C-terminal au niveau de l’exon 45 (Liao, Yong et al. 2005). D’autres groupes ont étudié différentes délétions au niveau de l’extrémité C-terminale (De Jongh, Warner et al. 1991; Gao, Cuadra et al. 2001). Les courants mesurés dans la configuration cellule entière, était significativement plus grands que le canal « pleine longueur ». Nous avons décidé de tester certaines de ces délétions (ΔC2030, ΔC1935, ΔC1856, ΔC1733, ΔC1700) en présence ou en absence de la sous-unité auxiliaire Cavβ3, susceptible d’interagir avec l’extrémité C-terminale de la sous-unité Cavα1 par l’intermédiaire de son domaine SH3 (Lao, Kobrinsky et al. 2008). Les résultats obtenus dans les ovocytes de Xénope ont mis en évidence que les sous-unités Cavα1.2 tronquées montraient des courants globaux plus élevés que le canal « pleine longueur » en présence de la sous-unité auxiliaire Cavβ3 et que les sous-unités Cavα1.2 tronquées donnaient des courants en absence de la sous-unité Cavβ3 contrairement à la sous-unité Cavα1.2 « pleine longueur ». Afin de vérifier si l’augmentation des courants macroscopiques était le résultat d’une augmentation du nombre de sous-unités Cavα1.2 à la membrane, nous avons choisi de quantifier la fluorescence spécifiquement due à cette sous-unité en utilisant la méthode de cytométrie de flux (FACS : « Fluorescence Activated Cell Sorting »). L’épitope HA a été inséré dans une région extracellulaire de la sous-unité Cavα1 du canal calcique Cav1.2 et un anticorps anti-HA couplé au FITC (« Fluorescein IsoThioCyanate ») a été utilisé pour observer la fluorescence. Nos résultats confirment que la sous-unité Cavα1-HA du canal calcique Cav1.2, s’exprime à la membrane plasmique en présence de la sous-unité auxiliaire Cavβ3, et qu’en absence de celle-ci, ne s’exprime que peu ou pas à la membrane. Les mêmes résultats ont été obtenus pour les trois délétions testées dans les mêmes conditions soit Cavα1.2-HA ΔC1935, Cavα1.2-HA ΔC1856 et Cavα1.2-HA ΔC1733. Ensemble, ces résultats suggèrent que l’augmentation des courants macroscopiques observés après une délétion partielle du C-terminal n’est pas causée par une augmentation du nombre de protéines Cavα1.2 à la membrane.

La neuropathie sensitive et motrice héréditaire avec agénésie du corps calleux (NSMH/ACC) se traduit par une atteinte neurodégénérative sévère associée à des anomalies développementales dans le système nerveux central et du retard mental. Bien que rare dans le monde, ce désordre autosomique récessif est particulièrement fréquent dans la population Québécoise du Canada Français du fait d’un effet fondateur. L’unique étude réalisée sur la mutation québécoise du gène qui code pour le co-transporteur de potassiumchlore 3 (KCC3) a montré qu’il y a une perte de fonction de la protéine. Cependant, la maladie est également retrouvée hors du Québec et il reste encore à élucider les pathomécanismes mis en jeu. Nous avons donc séquencé les 26 exons du gène KCC3 chez des individus recrutés dans le monde entier et suspectés d’être atteints de la maladie. Nous avons ainsi identifié trois nouvelles mutations. L’étude fonctionnelle de ces mutations nous a confirmé la perte de fonction systématique des co-transporteurs mutés. Puisque l’inactivation de KCC3 se produit majoritairement via l’élimination de segments peptidiques en C-terminus, nous avons concentré notre attention sur l’identification des interactions qui s’y produisent. À l’aide d’approches double hybride, pull-down et immunomarquage, nous avons déterminé que KCC3 interagit avec la créatine kinase CK-B et que cette interaction est perturbée par les mutations tronquantes. De plus, l’utilisation d’un inhibiteur de créatine kinase inactive KCC3, ce qui démontre qu’il existe bien un lien fonctionnel et pathologique entre KCC3 et ses partenaires C-terminaux. Nous avons aussi identifié des anomalies majeures de localisation membranaire des KCC3 mutés. Que KCC3 soit tronqué ou pleine longueur, sa distribution subcellulaire est affectée dans des cellules en culture, dans les ovocytes de Xenopes et dans des échantillons de cerveau de patients. La perte d’interaction entre KCC3 et CK-B et/ou les défauts de transit intracellulaire de KCC3 sont donc les mécanismes pathologiques majeurs de la NSMH/ACC.

Des évidences expérimentales récentes indiquent que les ARN changent de structures au fil du temps, parfois très rapidement, et que ces changements sont nécessaires à leurs activités biochimiques. La structure de ces ARN est donc dynamique. Ces mêmes évidences notent également que les structures clés impliquées sont prédites par le logiciel de prédiction de structure secondaire MC-Fold. En comparant les prédictions de structures du logiciel MC-Fold, nous avons constaté un lien clair entre les structures presque optimales (en termes de stabilité prédites par ce logiciel) et les variations d’activités biochimiques conséquentes à des changements ponctuels dans la séquence. Nous avons comparé les séquences d’ARN du point de vue de leurs structures dynamiques afin d’investiguer la similarité de leurs fonctions biologiques. Ceci a nécessité une accélération notable du logiciel MC-Fold. L’approche algorithmique est décrite au chapitre 1. Au chapitre 2 nous classons les impacts de légères variations de séquences des microARN sur la fonction naturelle de ceux-ci. Au chapitre 3 nous identifions des fenêtres dans de longs ARN dont les structures dynamiques occupent possiblement des rôles dans les désordres du spectre autistique et dans la polarisation des œufs de certains batraciens (Xenopus spp.).

Das Protein Orb2, welches zum Xenopus CPEB homolog ist, erfüllt während der Spermatogenese von Drosophila melanogaster eine wesentliche Funktion. Das teilweise Ausschalten von orb2 führt zu Störungen in der Individualisierung der Spermatiden, Veränderung in der Morphologie und Lokalisation der Spermatidenkerne und damit verbunden zu männlicher Sterilität. Der weit gestreute Phänotyp spricht für eine regulatorische Funktion des Proteins, wie es aufgrund der Homologie zu CPEB zu erwarten ist. Orb2 mutante Weibchen zeigen dagegen keinen Phänotyp. Die Sterilität konnte mit spezifischen Rettungskonstrukten rückgängig gemacht werden, wobei die beiden Proteinformen in ihrer Funktion höchstwahrscheinlich äquivalent sind, da eine größere Menge an kleinem Protein das Fehlen des größeren ausgleichen kann. Beide Proteinformen lokalisieren in fast alle Stadien der Spermatogenese, wobei nur das kleinere auch in reifen Spermien persistiert. Zur Untersuchung der regulatorischen Funktion des Proteins Orb2 wurden zunächst drei mögliche Protein-Interaktionskandidaten analysiert. Obwohl ähnliche mutante Phänotypen in Gap und Cup ausgelöst wurden, lässt sich eine Interaktion bis jetzt mit diesen Kandidaten weder ausschließen noch bestätigen. Daneben zeigte das Protein Tob eine ähnliche Lokalisierung und einen deutlich ähnlicheren mutanten Phänotyp, wie er für Orb2 beschrieben wurde. Besonders auffällig ist die Lokalisation der Tob mRNA an die Spermatidenenden und die Verringerung der Transkriptmenge in der orb2-Mutante. Ob dieser Phänotyp durch den Verlust der regulatorischen Funktion von Orb2 hervorgerufen wird oder durch den späten Zeitpunkt der Transkription bedingt ist, muß in späteren Experimenten geklärt werden. Mit Hilfe eines Co-Immunpräzipitations-Experimentes wurde nach weiteren Proteininteraktionspartnern sowie nach Ziel-mRNAs gesucht, die durch Orb2 reguliert werden könnten. Dabei ergaben die massenspektrometrischen Analysen zwar Proteine, die mit der Translation selbst in Zusammenhang stehen, sowie einige regulatorische RNA-bindende Proteine, wiesen aber auch in Gestalt eines häufig nachgewiesenen Anhangsdrüsenproteins auf deutliche systematische Probleme hin. Auf genetischem Wege war bereits der Nachweis gelungen, dass die Protamine und mst77F, die strukturelle Komponenten der kompaktierten Kern-DNA sind, durch Orb2 in ihrer Translation reprimiert werden. Dieses Ergebnis wurde zum Teil bestätigt durch den Nachweis der Protamin mRNAs in den Eluaten aus dem Co-Immunpräzipitationsexperiment. Damit konnte zum ersten Mal in der Drosophila Spermatogenese das regulatorische Protein zu einer translationskontrollierten mRNA identifiziert werden.

La Fibrosis Quística es la enfermedad autosómica recesiva mas frecuente en caucásicos. En Colombia no se conoce la incidencia de la enfermedad, pero investigaciones del grupo de la Universidad del Rosario indican que podría ser relativamente alta. Objetivo: Determinar la incidencia de afectados por Fibrosis Quística en una muestra de recién nacidos de la ciudad de Bogotá. Metodología: Se analizan 8.297 muestras de sangre de cordón umbilical y se comparan tres protocolos de tamizaje neonatal: TIR/TIR, TIR/DNA y TIR/DNA/TIR. Resultados: El presente trabajo muestra una incidencia de 1 en 8.297 afectados en la muestra analizada. Conclusiones: Dada la relativamente alta incidencia demostrada en Bogotá, se justifica la implementación de Tamizaje Neonatal para Fibrosis Quística en Colombia.
