A água é vital para as plantas. Uma redução na disponibilidade de água pode afetar a transpiração e o crescimento foliar das culturas. O objetivo deste trabalho foi determinar a resposta da transpiração e do crescimento foliar ao conteúdo de água disponível no solo, representado pela fração de água transpirável no solo (FATS), em dois clones de batata (um antigo e um recente) adaptados ao Sul do Brasil. Foram realizados três experimentos em ambiente protegido por abrigo telado em Santa Maria, RS, dois deles na primavera (cultivo de safra), com plantios em 20/08/2008 (E1) e 20/10/2008 (E2), e um experimento no outono (cultivo de safrinha), com plantio em 25/03/2009 (E3), usando-se dois clones de batata: um antigo ('Macaca') e um recente ('SMINIA793101-3'). A água disponível, representada pela FATS, a transpiração e o crescimento foliar foram medidos diariamente durante o período de imposição da deficiência hídrica. A FATS crítica que começa a alterar a transpiração, indicativo do início do fechamento estomático, foi de 0,39, 0,47 e 0,28 no clone 'Macaca' e de 0,47, 0,49 e 0,33 no clone 'SMINIA793101-3', para os experimentos E1, E2 e E3, respectivamente, enquanto o crescimento foliar começou a ser reduzido, com valor de FATS crítica maior para o clone 'Macaca', indicando que o clone 'SMINIA793101-3' é mais tolerante ao déficit hídrico no solo que o clone 'Macaca'.

Amyloidosis is defined as the extracellular deposition of proteins that have the capacity to form beta-pleated sheets and become insoluble. More than 17 types of amyloidosis have been described. Systemic light chain amyloid (AL) and AA amyloid (secondary to chronic inflammatory process) are by far the most frequent forms of amyloidosis. In these systemic forms, organs involved are the kidneys, the heart and the gastrointestinal tract in AL amyloidosis. The diagnostic can be established only by tissue biopsy. Treatment of primary amyloidosis (AL) aims at suppressing the responsible clone whereas treatment of secondary amyloidosis relies on controlling the underlying inflammatory process. Prognosis is globally poor and depends on the extend of organs involvement particularly cardiac and renal. The prognosis is even worse in patients requiring dialysis.

The antiviral response is largely mediated by dendritic cells (DCs), including conventional (c) DCs that function as antigen-presenting cells, and plasmacytoid (p) DCs that produce type I interferons, making them an attractive target for viruses. We find that the Old World arenaviruses lymphocytic choriomeningitis virus clone 13 (LCMV Cl13) and Lassa virus bind pDCs to a greater extent than cDCs. Consistently, LCMV Cl13 targets pDCs early after in vivo infection of its natural murine host and establishes a productive and robust replication cycle. pDCs coproduce type I interferons and proinflammatory cytokines, with the former being induced in both infected and uninfected pDCs, demonstrating a dissociation from intrinsic virus replication. TLR7 globally mediates pDC responses, limits pDC viral load, and promotes rapid innate and adaptive immune cell activation. These early events likely help dictate the outcome of infections with arenaviruses and other DC-replicating viruses and shed light on potential therapeutic targets.

Eucalyptus Shoot Blight in the Vale do Rio Doce (ESBVRD) is an anomaly that leads to reduced growth and, in more extreme cases, to death of eucalyptus plants. Initially diagnosed in plantations in the region of the Vale do Rio Doce, in the State of Minas Gerais, Brazil, this problem has also been found in plantations in other regions of the country and even in other countries. Although the symptoms of this anomaly are well-known, its causes are not yet understood. The aim of this study was to evaluate the cause-effect relationship between accumulation of manganese (Mn) in eucalyptus clones and ESBVRD. Characterization of the environment in areas of greater occurrence of this problem in regard to soil, climate and fluctuation of the water table was undertaken in eucalyptus plantations of the Celulose Nipo-brasileira S.A. (Cenibra) company in the region of the Vale do Rio Doce. Plant tissues were sampled in two situations. In the first situation, diagnosis occurred in the initial phase of the anomaly in clones with differentiated tolerance to the problem; in the second situation, diagnosis was made in a single clone, considered to be sensitive, in two time periods - in the phase with the strong presence of symptoms and in the recovery phase, in areas of occurrence and in areas of escape from the problem. The most ESBVRD-sensitive clone showed much higher (4.8 times higher) leaf Mn contents than more tolerant clones. In plants with the anomaly, Mn leaf contents were greater than 3,070 mg kg-1, much greater than the quantity found in those without the anomaly (734 mg kg-1). In the period in which the symptoms began to wane, there was a sharp decline in leaf Mn contents, from 2,194 to 847 mg kg-1. Manganese content in the above ground part and plant litter (44.4 g ha-1) in the area of occurrence of the anomaly was three times greater than that found in these same components (14.1 g ha-1) in the area of absence of the symptom. Based on the evidence found, such as the existence of environmental conditions favorable to high Mn availability to the plants in the areas of greatest incidence of ESBVRD, greater uptake of Mn in sensitive clones and in plants with symptoms, and a synchronism between the intensity of symptoms of ESBVRD and leaf Mn contents, it may be inferred that temporary excess of Mn in eucalyptus plants is closely related to ESBVRD.

Like many organisms, the cladoceran Simocephalus vetulus (Müller) continues to grow when reproducing, whereas the optimal strategy is to stop growing at maturity, and to invest all available production into reproduction thereafter. It has been proposed that a size constraint is responsible for the observed strategy (Perrin, Ruedi & Saiah, 1987), by preventing organisms from investing more than a given amount of energy into reproduction. This hypothesis is developed here and the two folowing prediction are derived: (1) the onset of reproduction should be independent of age and (2) the reproductive investement should be size-specific, thus independent of the productin rate. Both predictions are tested by rearing a clone of S.vetulus in a gradient of productivity. The results support the first prediction, but not the second one, so that the size-constraint hypothesis is disproved.

Intercellular Ca(2+) wave propagation between vascular smooth muscle cells (SMCs) is associated with the propagation of contraction along the vessel. Here, we characterize the involvement of gap junctions (GJs) in Ca(2+) wave propagation between SMCs at the cellular level. Gap junctional communication was assessed by the propagation of intercellular Ca(2+) waves and the transfer of Lucifer Yellow in A7r5 cells, primary rat mesenteric SMCs (pSMCs), and 6B5N cells, a clone of A7r5 cells expressing higher connexin43 (Cx43) to Cx40 ratio. Mechanical stimulation induced an intracellular Ca(2+) wave in pSMC and 6B5N cells that propagated to neighboring cells, whereas Ca(2+) waves in A7r5 cells failed to progress to neighboring cells. We demonstrate that Cx43 forms the functional GJs that are involved in mediating intercellular Ca(2+) waves and that co-expression of Cx40 with Cx43, depending on their expression ratio, may interfere with Cx43 GJ formation, thus altering junctional communication.

ABSTRACT Rubber tree (Hevea brasiliensis) crop may accumulate significant amounts of carbon either in biomass or in the soil. However, a comprehensive understanding of the potential of the C stock among different rubber tree clones is still distant, since clones are typically developed to exhibit other traits, such as better yield and disease tolerance. Thus, the aim of this study was to address differences among different areas planted to rubber clones. We hypothesized that different rubber tree clones, developed to adapt to different environmental and biological constrains, diverge in terms of soil and plant biomass C stocks. Clones were compared in respect to soil C stocks at four soil depths and the total depth (0.00-0.05, 0.05-0.10, 0.10-0.20, 0.20-0.40, and 0.00-0.40 m), and in the different compartments of the tree biomass. Five different plantings of rubber clones (FX3864, FDR 5788, PMB 1, MDX 624, and CDC 312) of seven years of age were compared, which were established in a randomized block design in the experimental field in Rio de Janeiro State. No difference was observed among plantings of rubber tree clones in regard to soil C stocks, even considering the total stock from 0.00-0.40 m depth. However, the rubber tree clones were different from each other in terms of total plant C stocks, and this contrast was predominately due to only one component of the total C stock, tree biomass. For biomass C stock, the MDX 624 rubber tree clone was superior to other clones, and the stem was the biomass component which most accounted for total C biomass. The contrast among rubber clones in terms of C stock is mainly due to the biomass C stock; the aboveground (tree biomass) and the belowground (soil) compartments contributed differently to the total C stock, 36.2 and 63.8 %, respectively. Rubber trees did not differ in relation to C stocks in the soil, but the right choice of a rubber clone is a reliable approach for sequestering C from the air in the biomass of trees.

The TCR repertoire of CD8+ T cells specific for Moloney murine leukemia virus (M-MuLV)-associated Ags has been investigated in vitro and in vivo. Analysis of a large panel of established CD8+ CTL clones specific for M-MuLV indicated an overwhelming bias for V beta4 in BALB/c mice and for V beta5.2 in C57BL/6 mice. These V beta biases were already detectable in mixed lymphocyte:tumor cell cultures established from virus-immune spleen cells. Furthermore, direct ex vivo analysis of PBL from BALB/c or C57BL/6 mice immunized with syngeneic M-MuLV-infected tumor cells revealed a dramatic increase in CD8+ cells expressing V beta4 or V beta5.2, respectively. M-MuLV-specific CD8+ cells with an activated (CD62L-) phenotype persisted in blood of immunized mice for at least 2 mo, and exhibited decreased TCR and CD8 levels compared with their naive counterparts. In C57BL/6 mice, most M-MuLV-specific CD8+ CTL clones and immune PBL coexpressed V alpha3.2 in association with V beta5.2. Moreover, these V beta5.2+ V alpha3.2+ cells were shown to recognize the recently described H-2Db-restricted epitope (CCLCLTVFL) encoded in the leader sequence of the M-MuLV gag polyprotein. Collectively, our data demonstrate a highly restricted TCR repertoire in the CD8+ T cell response to M-MuLV-associated Ags in vivo, and suggest the potential utility of flow-microfluorometric analysis of V beta and V alpha expression in the diagnosis and monitoring of viral infections.

Using the Xenopus oocyte expression system, we have previously identified an approximately 4-kb fraction of mRNA from rat liver that expresses sulfobromophthalein-glutathione (BSP-GSH)-insensitive reduced glutathione (GSH) transport (Fernandez-Checa, J., J. R. Yi, C. Garcia-Ruiz, Z. Knezic, S. Tahara, and N. Kaplowitz. 1993. J. Biol. Chem. 268:2324-2328). Starting with a cDNA library constructed from this fraction, we have now isolated a single clone that expresses GSH transporter activity. The cDNA for the rat canalicular GSH transporter (RcGshT) is 4.05 kb with an open reading frame of 2,505 nucleotides encoding for a polypeptide of 835 amino acids (95,785 daltons). No identifiable homologies were found in searching various databases. An approximately 96-kD protein is generated in in vitro translation of cRNA for RcGshT. Northern blot analysis reveals a single 4-kb transcript in liver, kidney, intestine, lung, and brain. The abundance of mRNA for RcGshT in rat liver increased 3, 6, and 12 h after a single dose of phenobarbital. Insensitivity to BSP-GSH and induction by phenobarbital, unique characteristics of canalicular GSH secretion, suggest that RcGshT encodes for the canalicular GSH transporter.

Why mating types exist at all is subject to much debate. Among hypotheses, mating types evolved to control organelle transmission during sexual reproduction, or to prevent inbreeding or same-clone mating. Here I review data from a diversity of taxa (including ciliates, algae, slime molds, ascomycetes, and basidiomycetes) to show that the structure and function of mating types run counter the above hypotheses. I argue instead for a key role in triggering developmental switches. Genomes must fulfill a diversity of alternative programs along the sexual cycle. As a haploid gametophyte, an individual may grow vegetatively (through haploid mitoses), or initiate gametogenesis and mating. As a diploid sporophyte, similarly, it may grow vegetatively (through diploid mitoses) or initiate meiosis and sporulation. Only diploid sporophytes (and not haploid gametophytes) should switch on the meiotic program. Similarly, only haploid gametophytes (not sporophytes) should switch on gametogenesis and mating. And they should only do so when other gametophytes are ready to do the same in the neighborhood. As argued here, mating types have evolved primarily to switch on the right program at the right moment.

To study sensitisation to minor histocompatibility antigens (mHag) before and after BMT, we measured antidonor CTL activity in five patients who had rejected their graft, and in a control group of 10 leukemic patients who engrafted without complications. All patients were transplanted with marrow from an HLA-identical sibling. Fourteen patients were conditioned with cyclophosphamide (120 mg/kg) and TBI (1350 cGy) and received a T cell-depleted graft, while one patient with aplastic anaemia received cyclophosphamide alone and unmanipulated marrow. Before transplantation, anti-donor CTL activity was detected in two of the 15 patients. These patients rejected their grafts at days 21 and 58, respectively. In the other three patients who rejected their grafts at days 41, 60 and 250, CTL activity was found only after transplantation. In contrast, no anti-donor CTLs could be detected at any time in the 10 patients who engrafted permanently. We have identified some of the mHags recognised during graft rejection by cloning and subsequent specificity analysis of the recipient CTLs. In the patient who rejected at day 41 without detectable immunisation before BMT, the response was directed against HA-1, a minor antigen known to play a role in GVHD. In the other combinations, a significant part of the CTL activity was directed against the male antigen H-Y. In the patient who rejected the marrow of her HLA-identical brother at day 250, two clones recognised H-Y, while five others recognised at least three distinct autosomal mHags. This patient had an HLA-identical sister who expressed only one autosomal mHag that had been recognised by one single T cell clone. After re-transplantation with the marrow of this second donor, the CTL activity could no longer be detected and the patient engrafted without further complications.

In this short review we report the basic notions needed for understanding the population genetics of clonal diploids. We focus on the consequences of clonality on the distribution of genetic diversity within individuals, between individuals and between populations. We then summarise how to detect clonality in mainly sexual populations, conversely, how to detect sexuality in mainly clonal populations and also how genetic differentiation between populations is affected by clonality in diploids. This information is then used for building recipes on how to analyse and interpret genetic polymorphism data in molecular epidemiology studies of clonal diploids.

Halecium petrosum and Halecium pusillum on the alga Halimeda tuna from Tossa de Mar, northeastern Spain, were studied. Asexual reproduction of H. petrosum, by stolonisation, occurred throughout the year except for July and August. Asexual reproduction of H. pusillum, by planktonic propagules, occurred throughout the year. Sexual reproduction was limited to the autumn in H. petrosum and spring in H. pusillum. The growth rates of colonies of both species were rapid but declined with increased size. Mean colony size over two consecutive two week periods increased approximately five-fold and three-fold for H. petrosum, and six-fold and four-fold for H. pusillum. Mortality was estimated to be high for both species, especially in summer. The maximum life span of colonies (ramets) of both species was estimated to be only eight weeks. Consequently most colonies do not reproduce sexually. The absence of reproduction of H. petrosum in summer, when the turnover of algal thalli was greatest, probably contributed to the summer decline in its abundance. In both species the genet (clone) survives for unknown, possibly very long, periods by asexual reproduction which facilites colonisation of other substrata.

RESUME La dissémination extramédullaire des cellules blastiques est une complication majeure des leucémies myéloïdes (LMA) ou lymphoïdes aiguës (LLA). La migration des cellules blastiques dépend de mécanismes semblables à ceux qui régulent la migration des leucocytes dans un site d'inflammation. Parmi ceux-ci, les oligosaccharides fucosylés décorant les ligands des sélectines jouent un rôle clé en interagissant avec les sélectines. PSGL-1 (P-Selectin Glycoprotein Ligand-1) est une protéine de 240 kD, exprimée à la surface des leucocytes, permettant de soutenir le roulement leucocytaire sur les sélectines, le long de la paroi vasculaire. L'interaction de PSGL-1 avec les sélectines nécessite des modifications post-traductionnelles de type sialylation, sulfatation , N et 0-glycosylation. Parmi les enzymes impliqués, les α1,3-fucosyltransférases jouent un rôle important dans la biosynthèse d'oligosaccharides fucosylés, ligands des sélectines (sLex, Lex, VIM-2, CLA). Comme l'expression des α1,3-fucosyltransférases par les cellules blastiques leucémiques n'a pas été étudiée précédemment, nous l'avons recherchée dans 120 cas de leucémies aiguës. Les ARNm des FucT-IV et -VII ont été détectés, par RT-PCR, dans tous les cas testés. L'ARNm de la FucT-IX n'a été observé que dans 40% des leucémies aiguës (48/120). L'ARNm de la FucT-IX est détecté dans 65% des LMA (47/72) et, moins fréquemment, dans 26% des LLA (11/42). A noter que les cas de LLA exprimant la FucT-IX correspondent essentiellement à des LLA secondaires à la transformation d'une leucémie myéloïde chronique ou des LLA de la lignée B de type leucémie/lymphome de Burkitt. L'expression de PSGL-1 et des oligosaccharides fucosylés par les blastes varie significativement parmi les LMA et les LLA : Lex, VIM-2 et sLex étant exprimés plus fréquemment par les myéloblastes que par les lymphoblastes. Le rôle des FucT-IV, -VII et -IX dans la synthèse des Lex, VIM-2, CLA et sLex a été examiné en exprimant l'ADNc de chaque FucT dans des cellules CHO. L'immunophénotypisation des transfectants indique que la FucT-VII synthétise sLex et CLA, mais pas Lex et VIM-2. Lex et VIM-2 sont générés par la FucT-IV. La FucT-IX ne participe qu'à la synthèse de Lex, sa capacité de synthèse de VIM-2 dans les cellules CHO est très faible. Le rôle de la FucT-IX dans la régulation du roulement cellulaire dépendant des sélectines a été testé dans des conditions de flux. Les vitesses de roulement des cellules CHO co-exprimant la FucT-LX, la core-2 01,6-N-acetylglucosaminyltransferase et PSGL-1 sont très élevées sur la P-sélectine (médiane : 497.95 µm/s, n=96) alors qu'elles sont beaucoup plus lentes sur la E-sélectine (médiane 7 µm/s, n=64). Les recrutements sur la E-sélectine des cellules CHO-C2F9PSGL¬1 et des CHO-C2F7PSGL-1 sont similaires (moyenne ± SEM : 127.44 ± 4.38 vs. 151.16 ± 3.16 cellules/min/mm2, n=5). Celui des cellules CHO-C2F4PSGL-1 est par contre plus faible (54.20 ± 2.13 cellules/min/mm2, n=5). Ces résultats indiquent que la FucT-IX est impliquée dans la biosynthèse de Lex, VIM-2 et CLA et qu'elle régule l'interaction des cellules CHO avec la E-sélectine. Contrairement aux FucT-IV et -VII, la FucT-IX ne joue qu'un rôle mineur dans la régulation du roulement cellulaire sur la L- et la P-sélectine. L'expression fréquente de la FucT-IX par les myéloblastes suggère qu'elle pourrait participer avec les FucT-IV et -VII à la régulation de la migration cellulaire dépendant de la E-sélectine. Finalement, ce travail de thèse a été étendu à l'identification des protéines cytoplasmiques qui interagissent avec le domaine cytoplasmique de PSGL-1 et qui pourraient être impliquées dans la transmission de signaux intracellulaires. Les ligands intracellulaires de PSGL-1 seront identifiés par la technique du double hybride qui nous a déjà permis de confirmer que syk et la N-moésine se lient au domaine cytoplasmique de PSGL-1. Des ligands supplémentaires seront identifiés employant une librairie provenant des cellules souches hématopoïétiques comme proie. ABSTRACT Blast cell dissemination is a major complication of acute myeloblastic (AML) and lymphoblastic leukemia (ALL). Blast cell migration is dependent on mechanisms that are similar to those which regulate leukocyte migration into inflammatory lesions. Among them, fticosylated oligosaccharides that decorate selectin ligands play a key role by interacting with selectins. PSGL-1 (P-Selectin Glycoprotein Ligand-1) is a 240 kD glycoprotein constitutively expressed on leucocytes and which supports leukocyte rolling on selectins. PSGL-1 interaction with selectins is dependent on post-translational modifications such as sialylation, sulfation, N- and 0-glycosylation. Among the involved enzymes, the α1,3-fucosyltransferases (FucT) play a major role in generating cell surface glycoconjugates carrying fucosylated oligosaccharides which interact with selectins (sLex, Lex, VIM-2, CLA). Since no information is available on the expression of α1,3-fucosyltransferases by leukemic blast cells, we examined it in 120 cases of acute leukemia. FucT-IV and -VII mRNAs were detected, by RT-PCR, in all tested cases. In contrast, the presence of FucT-IX mRNA was shown in only 40% of patients with acute leukemia (48/120). FucT-IX mRNA was detected in 65% of AML (47/72) and, less frequently, in 26% of ALL (11/42). Importantly, all ALL cases expressing FucT-IX were either secondary leukemia resulting from the transformation of chronic myelocytic leukemia in acute lymphoblastic leukemia or mature B-ALL (FAB L3 subtype or Burkitt lymphoma/leukemia according to WHO classification). FucT-IX was not detected in precursor B or T-ALL. The expression of PSGL-1 and fucosylated epitopes was significantly different among AML and ALL, Lex, VIM-2 and sLex being more frequently expressed by myeloblasts than by lymphoblasts. The role of FucT-IV, -VII and -IX in the biosynthesis of Lex, VIM-2, CLA and sLex was examined by expressing the cDNA of each α1,3-FucT in CHO cells. Immunophenotypic analysis of CHO transfectants indicated that FucT-VII synthesizes sLex and CLA but not Lex or VIM-2. Lex and CLA were generated by both FucT-IV and -IX. FucT-IV and FucT-IX differed in their ability to synthesize VIM-2, FucT-IX being less efficient than FucT-IV. The role of FucT-IX in regulating selectin-dependent rolling was assessed under hydrodynamic flow conditions. P-selectin-dependent interactions were transient and occurred at high velocities (median: 497.95 1,µm/s, n=96). In contrast, much slower rolling velocities were observed on E-selectin (median: 7 µm/s, n=64). The recruitment of CHO-C2F9PSGL-1 and CHO-C2F7PSGL-1 cells was similar on E-selectin (mean ± SEM: 127.44 ± 4.38, n=5 vs 151.16 ± 3.16 cells/min/mm2, n=5). In the other hand, CHO-C2F4PSGL-1 cells were less efficiently recruited on E-selectin (54.20 ± 2.13 cells/min/mm2, n=5). This results indicate that FucT-IX is involved in the biosynthesis of Lex, VIM-2 and CLA and that it confers E-selectin binding activity to CHO cells. The frequent expression of FucT-IX in myeloblasts suggests that it may participate with FucT-IV and -VII in regulating E-selectin-dependent cell migration into tissues. Finally, this thesis work was extended to the identification of the cytoplasmic proteins interacting with cytoplasmic domain of PSGL-1 that may be involved in transducing intracellular signals. We planned to identify these intracellular ligands of PSGL-1 by using the double hybrid technique and already confirmed that syk and N-moesin bind to the cytoplasmic domain of PSGL-1. Additional PSGL-1 ligands will be sought by the same technique using a CD34+ stem cell library as pray. RESUME DESTINE A UN LARGE PUBLIC : L'adhésion et la migration leucocytaire sont nécessaires à de nombreux processus cellulaires comme la régulation de l'hématopoïèse, mais aussi dans la pathogenèse de l'artériosclérose, des maladies inflammatoires et de la métastatisation des cellules cancéreuses. Les molécules impliquées constituent depuis peu des cibles pour la thérapie du cancer. La migration leucocytaire vers un site d'inflammation dépend de mécanismes complexes, se déroulant en plusieurs étapes, nécessitant l'interaction séquentielle de molécules d'adhésion leucocytaires et endothéliales. Ainsi, chronologiquement, suite à un stimulus inflammatoire, les leucocytes « roulent » sur les cellules endothéliales, sont activées, s'arrêtent et traversent la paroi endothéliale (diapédèse) pour migrer dans les tissus environnants inflammés selon un gradient chimiotactique. La première étape de roulement met en jeu deux molécules principales : PSGL-1 (P-Sélectine Glycoprotéine Ligand-1) du coté des leucocytes et les sélectines du coté de l'endothélium de la paroi vasculaire. L'interaction entre ces deux molécules nécessite des décorations de ces protéines par des sucres, des résidus sulfates et des acides sialiques. Le sucre essentiel à la liaison demeure le fucose qui est attaché aux protéines grâce à des enzymes de la famille des fucosyltransferases. Actuellement, neuf fucosyltransférases humaines ont été identifiées et désignées sous FucT-I à IX. La FucT-IX, dernière fucosyltransférase clonée, a un faible degré d'homologie avec les autres fucosyltransférases mais sa séquence est extrêmement conservée entre les espèces. Ceci traduit son importance par une forte résistance à la pression évolutive. L'examen de son expression au sein de 120 cas de leucémies aiguës a mis en évidence son comportement atypique. En effet, alors que les autres FucTs sont toujours présentes, la FucT¬IX ne s'exprime que dans un cas sur deux en moyenne avec une préférence plus importante pour les leucémies myéloïdes. Ainsi, une étude plus approfondie de cet enzyme à mis en évidence sa capacité à induire une interaction cellulaire plus spécifique de la E-sélectine. Elle décore non seulement des protéines de surface, mais aussi certainement les glycolipides constituant la membrane cellulaire.

Thymocytes and class I major histocompatibility complex (MHC)-restricted cytotoxic T lymphocytes express predominantly heterodimeric alpha/beta CD8. By interacting with non-polymorphic regions of MHC class I molecules CD8 can mediate adhesion or by binding the same MHC molecules that interact with the T-cell antigen receptor (TCR) function as coreceptor in TCR-ligand binding and T-cell activation. Using TCR photoaffinity labelling with a soluble, monomeric photoreactive H-2Kd-peptide derivative complex, we report here that the avidity of TCR-ligand interactions on cloned cytotoxic T cells is very greatly strengthened by CD8. This is primarily explained by coordinate binding of ligand molecules by CD8 and TCR, because substitution of Asp 227 of Kd with Lys severely impaired the TCR-ligand binding on CD8+, but not CD8- cells. Kinetic studies on CD8+ and CD8- cells further showed that CD8 imposes distinct dynamics and a remarkable temperature dependence on TCR-ligand interactions. We propose that the ability of CD8 to act as coreceptor can be modulated by CD8-TCR interactions.
