Avian intestinal spirochaetosis (AIS) caused by Brachyspira spp., and notably Brachyspira pilosicoli, is common in layer flocks and reportedly of increasing incidence in broilers and broiler breeders. Disease manifests as diarrhoea,increased feed consumption, reduced growth rates and occasional mortality in broilers and these signs are shown in layers also associated with a delayed onset of lay, reduced egg weights, faecal staining of eggshells and non-productive ovaries. Treatment with Denagard® Tiamulin has been used to protect against B. pilosicoli colonisation, persistence and clinical presentation of AIS in commercial layers, but to date there has been no definitive study validating efficacy. Here, we used a poultry model of B. pilosicoli infection of layers to compare the impact of three doses of Denagard® Tiamulin. Four groups of thirty 17 week old commercial pre-lay birds were all challengedwith B. pilosicoli strain B2904with three oral doses two days apart. All birdswere colonised within 2 days after the final oral challenge and mild onset of clinical signs were observed thereafter. A fifth group that was unchallenged and untreated was also included for comparison as healthy birds. Five days after the final oral Brachypira challenge three groups were given Denagard® Tiamulin in drinking water made up following the manufacturer's recommendations with doses verified as 58.7 ppm, 113 ppm and 225 ppm. Weight gain body condition and the level of diarrhoea of birds infected with B. pilosicoli were improved and shedding of the organism reduced significantly (p = 0.001) following treatment with Denagard® Tiamulin irrespective of dose given. The level and duration of colonisation of organs of birds infected with B. pilosicoli was also reduced. Confirming previous findings we showed that the ileum, caeca, colon, and both liver and spleen were colonised and here we demonstrated that treatment with Denagard® Tiamulin resulted in significant reduction in the numbers of Brachyspira found in each of these sites and dramatic reduction in faecal shedding (p b 0.001) to approaching zero as assessed by culture of cloacal swabs. Although the number of eggs produced per bird and the level of eggshell staining appeared unaffected, egg weights of treated birds were greater than those of untreated birds for a period of approximately two weeks following treatment. These data conclusively demonstrate the effectiveness of Denagard® Tiamulin in reducing B. pilosicoli infection in laying hens.

Analysis of experimental interlocking blocks of concrete with addition of residues of process the tires retreading production. With the population growth in recent years, industry in general has adjusted itself to resulting demand. the industry of tire retreading generates residues that have been discarded without any control. this adds to environmental pollution and promotes the proliferation of vectors harmful to health, aiming to find an application for this type of residues, this study presents experimental results to interlocking concrete block pavements, with addition of residues tires, interlocking blocks were built up and we determined, through laboratory tests, the need to set the mark that provide greater return regarding analyzed characteristics, there are four types of dosage of concrete with residues tires. We accomplished tests of compression strength, water absorption and resistance to impact. Through the preliminary results, we verified that are satisfactory, confirming the possibility of applying this type of interlocking block in environments with low demand, which would bring the economy of natural sources of aggregates, beyond ecological benefits through the reuse of residues from retreading of tires.

The development and application of a functionalized carbon nanotubes paste electrode (CNPE) modified with crosslinked chitosan for determination of Cu(II) in industrial wastewater, natural water and human urine samples by linear scan anodic stripping voltammetry (LSASV) are described. Different electrodes were constructed using chitosan and chitosan crosslinked with glutaraldehyde (CTS-GA) and epichlorohydrin (CTS-ECH). The best voltammetric response for Cu(II) was obtained with a paste composition of 65% (m/m) of functionalized carbon nanotubes, 15% (m/m) of CTS-ECH, and 20% (m/m) of mineral oil using a solution of 0.05 mol L(-1) KNO(3) with pH adjusted to 2.25 with HNO(3), an accumulation potential of 0.3V vs. Ag/AgCl (3.0 mol L(-1) KCl) for 300 s and a scan rate of 100 mV s(-1). Under these optimal experimental conditions, the voltammetric response was linearly dependent on the Cu(II) concentration in the range from 7.90 x 10(-8) to 1.60 x 10(-5) mol L(-1) with a detection limit of 1.00 x 10(-8) mol L(-1). The samples analyses were evaluated using the proposed sensor and a good recovery of Cu(II) was obtained with results in the range from 98.0% to 104%. The analysis of industrial wastewater, natural water and human urine samples obtained using the proposed CNPE modified with CTS-ECH electrode and those obtained using a comparative method are in agreement at the 95% confidence level. We consider methods for estimating causal effects of treatment in the situation where the individuals in the treatment and the control group are self selected, i.e., the selection mechanism is not randomized. In this case, simple comparison of treated and control outcomes will not generally yield valid estimates of casual effects. The propensity score method is frequently used for the evaluation of treatment effect. However, this method is based onsome strong assumptions, which are not directly testable. In this paper, we present an alternative modeling approachto draw causal inference by using share random-effect model and the computational algorithm to draw likelihood based inference with such a model. With small numerical studies and a real data analysis, we show that our approach gives not only more efficient estimates but it is also less sensitive to model misspecifications, which we consider, than the existing methods.

Scientific workflows are becoming a valuable tool for scientists to capture and automate e-Science procedures. Their success brings the opportunity to publish, share, reuse and repurpose this explicitly captured knowledge. Within the myGrid project, we have identified key resources that can be shared including complete workflows, fragments of workflows and constituent services. We have examined the alternative ways these can be described by their authors (and subsequent users), and developed a unified descriptive model to support their later discovery. By basing this model on existing standards, we have been able to extend existing Web Service and Semantic Web Service infrastructure whilst still supporting the specific needs of the e-Scientist. myGrid components enable a workflow life-cycle that extends beyond execution, to include discovery of previous relevant designs, reuse of those designs, and subsequent publication. Experience with example groups of scientists indicates that this cycle is valuable. The growing number of workflows and services mean more work is needed to support the user in effective ranking of search results, and to support the repurposing process.

São apresentados os aspectos teóricos, práticos e bibliográficos envolvidos no desenvolvimento da tese de doutorado intitulada Modificação estrutural de bentonitas nacionais: caracterização e estudos de adsorção. O trabalho consistiu no desenvolvimento de um material adsorvente a partir de bentonitas, do tipo montmorilonitas, modificadas estruturalmente com o objetivo de aumentar sua capacidade de adsorção de poluentes, orgânicos e inorgânicos. O estudo visa incrementar o valor agregado deste recurso mineral e insere-se na área de tratamento de efluentes líquidos usando adsorventes não tradicionais, eficientes e de baixo custo em substituição ao carvão ativado ou às resinas de troca iônica. Foram estudadas as propriedades físicas e químicas; distribuição de tamanho de partículas, área superficial, potenciais eletrocinéticos, capacidade de troca catiônica, composição mineralógica, morfologia superficial e espaçamento basal, bem como as propriedades adsorptivas dos argilominerais não tratados e modificados, não modificadas e pilarizadas respectivamente. Também são discutidos os mecanismos de adsorção envolvidos e o desenvolvimento de um reator contínuo (adsorção em flocos) e de separação sólido/líquido. As modificações estruturais dos argilominerais foram realizadas via homoionização com cloreto de cálcio e posterior intercalação com compostos orgânicos com ação quelante de metais. A FENAN, bentonita obtida pela intercalação com Orto Fenantrolina (OF), foi a que apresentou melhor viabilidade técnica em termos de adsorção, adsorção/dessorção, floculação e de acumulação de poluentes na forma floculada e não floculada. Adicionalmente os estudos de reversibilidade da intercalação revelaram a alta estabilidade da OF na FENAN, em soluções fortemente ácidas, onde aproximadamente 90% da OF permanece ligada à superfície da argila. A quantidade de OF adsorvida na forma de unidades micelares foi de 112 mg por grama de bentonita a pH 8,5 ± 0,5. A caracterização das bentonitas, via difração de Raios X, análise térmica, microscopia eletrônica de varredura e por microscopia de força atômica, revelou que as FENAN possuem um comportamento estrutural muito estável ao longo da seqüência de adsorção/dessorção e que após a adsorção de poluentes inorgânicos, o quelato metálico formado apresenta alta estabilidade dentro da estrutura da organobentonita. A capacidade de acumulação alcançada nas FENAN foi de 110 mg de Cu/g de bentonita, valor superior à de diversos materiais adsorventes alternativos propostos em outros trabalhos similares. Os estudos de acumulação das FENAN floculadas – FENANFLOC, indicaram que a presença de floculante, na quantidade utilizada, não afeta significativamente a capacidade de remoção das bentonitas modificadas. Este comportamento apresentado, permitiu o desenvolvimento do Reator Expandido de Flocos Adsorventes (REFA), cujas características e parâmetros operacionais são discutidos em detalhe. Finalmente, os resultados são discutidos em termos dos fenômenos interfaciais envolvidos e dos potenciais práticos deste novo adsorvente e da nova técnica de adsorção em flocos no REFA.

Electronic applications are currently developed under the reuse-based paradigm. This design methodology presents several advantages for the reduction of the design complexity, but brings new challenges for the test of the final circuit. The access to embedded cores, the integration of several test methods, and the optimization of the several cost factors are just a few of the several problems that need to be tackled during test planning. Within this context, this thesis proposes two test planning approaches that aim at reducing the test costs of a core-based system by means of hardware reuse and integration of the test planning into the design flow. The first approach considers systems whose cores are connected directly or through a functional bus. The test planning method consists of a comprehensive model that includes the definition of a multi-mode access mechanism inside the chip and a search algorithm for the exploration of the design space. The access mechanism model considers the reuse of functional connections as well as partial test buses, cores transparency, and other bypass modes. The test schedule is defined in conjunction with the access mechanism so that good trade-offs among the costs of pins, area, and test time can be sought. Furthermore, system power constraints are also considered. This expansion of concerns makes it possible an efficient, yet fine-grained search, in the huge design space of a reuse-based environment. Experimental results clearly show the variety of trade-offs that can be explored using the proposed model, and its effectiveness on optimizing the system test plan. Networks-on-chip are likely to become the main communication platform of systemson- chip. Thus, the second approach presented in this work proposes the reuse of the on-chip network for the test of the cores embedded into the systems that use this communication platform. A power-aware test scheduling algorithm aiming at exploiting the network characteristics to minimize the system test time is presented. The reuse strategy is evaluated considering a number of system configurations, such as different positions of the cores in the network, power consumption constraints and number of interfaces with the tester. Experimental results show that the parallelization capability of the network can be exploited to reduce the system test time, whereas area and pin overhead are strongly minimized. In this manuscript, the main problems of the test of core-based systems are firstly identified and the current solutions are discussed. The problems being tackled by this thesis are then listed and the test planning approaches are detailed. Both test planning techniques are validated for the recently released ITC’02 SoC Test Benchmarks, and further compared to other test planning methods of the literature. This comparison confirms the efficiency of the proposed methods.

A crescente produção de resíduos sólidos urbanos e a escassez de áreas para uma destinação final tecnicamente adequada, via implantação de aterros sanitários, faz com que tome importância a técnica de tratamento de resíduos sólidos orgânicos através das compostagem/vermicompostagem. Nesta pesquisa, foi realizada a avaliação destes processos, tendo sido observadas principalmente a influência da aeração e da umidade no desempenho destas técnicas de tratamento, em leiras de pequenos e grandes volumes. Nos experimentos, com leiras de pequenos volumes, realizados no IPH (Instituto de Pesquisas Hidráulicas) da UFRGS, utilizou-se resíduos sólidos de poda (resíduos verdes, com alta concentração de carbono) codispostos (misturados em peso) com resíduos vegetais da CEASA (Companhia Estadual de Abastecimento Sociedade Anônima) e lodos provenientes de estações de tratamento de esgotos sanitários. Nos experimentos, com leiras de grandes volumes, realizados na UTC – Unidade de Triagem e Compostagem de Porto Alegre, utilizou-se resíduos orgânicos domiciliares codispostos com os mesmos resíduos utilizados nos primeiros experimentos. Paralelamente aos experimentos de compostagem, avaliou-se os lixiviados produzidos nos sistemas. Na compostagem de grandes volumes, também foi observado o desempenho de banhados construídos de fluxo subsuperficial para o tratamento desses efluentes líquidos. com teores de resíduos verdes menores que 35 %, o que se justifica pela maior concentração de resíduos de característica facilmente biodegradável e de maior palatabilidade para os vermes. Em todos os experimentos de compostagem, verificou-se que o controle efetivo do processo pode ser realizado através da avaliação sistemática das temperatura e umidade das leiras. Evidenciou-se também que para regiões de clima similar ao de Porto Alegre, com elevadas precipitações principalmente no inverno, é necessário adotar dimensões adequadas ao sistema de compostagem “windrow” para áreas descobertas. Comprovou-se também a necessidade de manutenção da umidade na faixa entre 50% e 70 %, inclusive com reposição desta, mesmo na situação de inverno. A avaliação dos lixiviados da compostagem demonstrou que, com os substratos utilizados nos experimentos, as concentrações de DBO5, DQO e de outras variáveis são elevadas ao início do processo de decomposição, devido à solubilização dos compostos orgânicos e inorgânicos presentes na matéria orgânica. Os lixiviados da compostagem possuem baixas concentrações de condutividade, DBO5, NH4 +, entre outros, quando comparados aos lixiviados de aterros sanitários. tratamento destes efluentes. Os resultados obtidos nos banhados construídos para baixas cargas hidráulicas (1cm/d) e concentrações de DBO5 do afluente abaixo de 150 mg/L apontaram uma eficiência média de remoção de 52,02%. No que se refere a nitrogênio (todas as formas), fósforo, metais, potencial redox e sólidos totais, as eficiências foram variáveis, com melhores resultados para nitrogênio amoniacal e fósforo. A compostagem pode ser considerada uma alternativa viável de tratamento de resíduos orgânicos, utilizando-se o processo “windrow” com revolvimento mecânico, mesmo em pátios descobertos em climas subtropicais. Neste caso, sugere-se que os lixiviados gerados nos primeiros dias de compostagem (30 dias aproximandamente) sejam recirculados e o excedente tratado em ETE. Pode-se, também, utilizar o sistema de banhados construídos como complementação, principalmente para a remoção de nitrogênio e fósforo.

A large number of evidences correlate elevated levels of homocysteine (Hcys) with a higher cardiovascular diseases (CVDs) risk, especially, atherosclerosis. Similarly, abnormal low levels of the vitamins B6, B9 and B12 are associated to an instability in the methionine cycle with an over production of Hcys. Thus, biomedical sciences are looking forward for a cheaper, faster, precise and accurate analytical methodology to quantify these compounds in a suitable format for the clinical environment. Therefore the objective of this study was the development of a simple, inexpensive and appropriate methodology to use at the clinical level. To achieve this goal, a procedure integrating a digitally controlled (eVol®) microextraction by packed sorbent (MEPS) and an ultra performance liquid chromatography (UPLC) coupled to a photodiode array detector (PDA) was developed to identify and quantify Hcys vitamins B6, B9 and B12. Although different conditions were assayed, we were not able to combine Hcys with the vitamins in the same analytical procedure, and so we proceeded to the optimization of two methods differing only in the composition of the gradient of the mobile phase and the injected volume. It was found that MEPS did not bring any benefit to the quantification of the Hcys in the plasma. Therefore, we developed and validate an alternative method that uses the direct injection of treated plasma (reduced and precipitated). This same method was evaluated in terms of selectivity, linearity, limit of detection (LOD), limit of quantification (LOQ), matrix effect and precision (intra-and inter-day) and applied to the determination of Hcys in a group composed by patients presenting augmented CVD risk. Good results in terms of selectivity and linearity (R2> 0.9968) were obtained, being the values of LOD and LOQ 0.007 and 0.21 mol / L, respectively. The intra-day precision (1.23-3.32%), inter-day precision (5.43-6.99%) and the recovery rate (82.5 to 93.1%) of this method were satisfactory. The matrix effect (>120%) was, however, higher than we were waiting for. Using this methodology it was possible to determine the amount of Hcys in real plasma samples from individuals presenting augmented CVD risk. Regarding the methodology developed for vitamins, despite the optimization of the extraction technique and the chromatographic conditions, it was found that the levels usually present in plasma are far below the sensitivity we obtained. Therefore, further optimizations of the methodology developed are needed. As conclusion, part of the objectives of this study was achieved with the development of a quick, simple and cheaper method for the quantification of Hcys.

The WTP produce many kinds of residue on your treatment stages, but the sludge is the more problematic from the final disposition point view. The actual rate of residue production deriving from technological evolution and the crescent population needs prevents the subtle equilibrium generation between consumption and recycling/reuse, creating problems of pollution resulting from inappropriate management of residues. Thus, is necessary achieve a new equilibrium between the grow from raw materials and energy and the residue generation. This equilibrium should be achieved by technical and economic feasibility of environmental supported models through recycling and reuse. The red ceramic industry stand out in residue absorption question as raw material due their clay mass heterogeneity, constituted by clay minerals and non-clay minerals with wide mineralogical variation, allowing residue inclusion which act like plastic or non-plastic materials, contributing to retain heavy metals contained in residues in the vitreous mass formed during the burning of the ceramic bodies. This work propose the study of the influence of incorporation of 25 wt.% sludge from wastewater treatment plant, according preliminary results, in the mass to produce ceramic bodies. The raw materials was characterized through chemical composition analyses by XRF, mineralogical analyses by XRD, thermal analyses by TG and DTA, Atterberg limits and thermodilatometry. Subsequently was composed the mass with 75 wt.% of clay and 25 wt.% of dried wastewater sludge from UFRN WWTP. Samples with 6,0 x 2,0 x 0,5 cm was produced with unidirectional compacting under pressure of 20MPa and burned in temperatures between 950 and 1,200ºC. After fired, the ceramic bodies have been submitted to physical and mechanical analyses through the measure of firing shrinkage, water absorption, density, apparent porosity and flexural strength; crystallographic analyses through XRD and microstructure analyses by SEM. The technological properties obtained was satisfactory to production of roof tiles with 25 wt.% at 1,200 ºC, but the production of others products at lower temperatures was not feasible
