The GRIP domain is a targeting sequence found in a family of coiled-coil peripheral Golgi proteins. Previously we demonstrated that the GRIP domain of p230/golgin245 is specifically recruited to tubulovesicular structures of the traps-Golgi network (TGN). Here we have characterized two novel Golgi proteins with functional GRIP domains, designated GCC88 and GCC185. GCC88 cDNA encodes a protein of 88 kDa, and GCC185 cDNA encodes a protein of 185 kDa. Both molecules are brefeldin A-sensitive peripheral membrane proteins and are predicted to have extensive coiled-coil regions with the GRIP domain at the C terminus. By immunofluorescence and immunoelectron microscopy GCC88 and GCC185, and the GRIP protein golgin97, are all localized to the TGN of Hela cells. Overexpression of full-length GCC88 leads to the formation of large electron dense structures that extend from the traps-Golgi. These de novo structures contain GCC88 and co-stain for the TGN markers syntaxin 6 and TGN38 but not for alpha2,6-sialyltransferase, beta-COP, or cis-Golgi GM130. The formation of these abnormal structures requires the N-terminal domain of GCC88. TGN38, which recycles between the TGN and plasma membrane, was transported into and out of the GCC88 decorated structures. These data introduce two new GRIP domain proteins and implicate a role for GCC88 in the organization of a specific TGN subcompartment involved with membrane transport.

Ninguém consegue sair ileso de um encontro com o currículo e com a escola, principalmente diante de relações tão assimétricas de poder que não valorizam o que as crianças têm a dizer. Quantas narrativas curriculares sobre as crianças? Quantas narrativas curriculares com as crianças? As narrativas e as fotografias se seguirão nesta pesquisa, porque com elas nos colocamos a pensar nos processos de criação curricular, explorando novas sensibilidades, novas maneiras de ver e falar, a partir das redes de sentidos, forjadas nos contextos vividos, imaginados ou pensados, dentrofora da escola. Também incluímos a forma como as crianças se relacionam com as práticas pedagógicas dos professores e do modo como se relacionam com a escola. Nessa perspectiva, interessa-nos pensar em como os usos (CERTEAU, 1994) da imagem fotográfica são capazes de afetar (e até transformar) as práticas curriculares, traçando algo da potência da/na imagem fotográfica em sua função fabuladora por meio das oficinas de fotografias realizadas com as crianças em virtude da pesquisa. A fotografia se torna potente como um recurso para provocar a invenção tessitura de outros sentidos em currículo não porque em sua materialidade ela está repleta de sentidos de currículo à priori, mas porque pode ou não, ao ser usada (CERTEAU, 1994), ao ser vista pelas crianças, agenciar outros possíveis para o currículo. Assim, o foco da discussão não é a fotografia em si nem a criança em si, ou seja, não há protagonismo nem da criança nem da fotografia. O foco está nas relações, naquilo que nos passa, isto é, na experiência estética (LARROSA, 2004b) que ocorre ao entrarmos em contato, ao vermos, ao compormos com as fotografias! Nesse sentido, pensamos as oficinas de fotografias como um dispositivo de criação e produção de acontecimentos em currículo, considerando-as máquinas de fazer “ver e falar”, o que as justifica como uma estratégia narrativa capaz de produzir acontecimentos na imagem e no mundo. Que sentidos de currículo são produzidos em multiplicidades? Pelas minoridades pretendemos movimentar nosso pensamento: que quer pensar um currículo como fabulação sem dizer o que ele é, mas no que ele vai se transformando com a chegada das crianças. No encontro das imagens com as palavras, em que o currículo vai se transformando? Sob a mesma superfície chamada currículo em extensão com as crianças, co-habitantes, encontrar os modos de olhar esse currículo e de dizê-lo por meio das fotografias, das narrativas, dos cartazes, dos desenhos, das poesias... As conversas com as crianças provocam o real, colocam em desequilíbrio algumas ideias feitas em educação, exigindo reordenações e invenções de outros pensamentos para a educação. É dessa criação de efeitos impensáveis que surge a invenção de currículos possíveis. Aprender a olhar mais (até cansar!) aquilo que não percebemos no dia a dia tem uma dimensão política muito importante; por meio desse gesto (que aprendemos com as crianças) podemos criar um novo pensamento político em educação. A partir daquilo que nos dá a ver, as crianças vão inaugurar sentidos impertinentes, desestabilizadores daquilo que chamamos de currículo e escola. O desafio consiste em falar da força contida na imagem fotográfica sem vontade de interpretá-la ou descrevê-la, mas escrever e pensar pelas fotografias num movimento de criação de sentidos e acontecer por elas.

Atreladas a uma estética própria e “efeitos de verdade” (PELLEJERO, 2008), as videografias turísticas acabam por compor linguagens fartamente informativas sobre aquilo que se quer dizer sobre os lugares. Suas cenas são as apontadas para propagandear uma imagem a ser consumida, delas esperam-se o melhor ângulo a ser fotografado, experiências únicas e roteiros alternativos e naturais para se conhecer o lugar. Sendo assim, são as imagens turísticas, na atualidade, linguagens potentes para se entender as narrativas sobre os lugares, suas imaginações espaciais, bem como as construções de ficções sobre determinada realidade. Uma vez envolvidas as produções de ficções hegemônicas, os vídeos turísticos e as imaginações espaciais que temos deles podem promover modos cristalizados de se pensar o espaço; distanciando-se dos propósitos de entender o espaço a partir das suas conexões-desconexões e multiplicidade de trajetórias (MASSEY, 2008). Nesse contexto, essa pesquisa tem como objetivo principal discutir como os vídeos turísticos, em especial dois vídeos da atual campanha da Secretaria de Turismo do Espírito Santo, “Descubra o Espírito Santo”, apresentam uma imaginação espacial. Também seguem como interesse: refletir e analisar a política visual e a estética das videografias turísticas; entender e analisar a produção de uma ficção para construção e mobilização de uma imaginação espacial e estudar autores e produções videográficas que se dedicaram a pensar possibilidades outras de mobilizar e desterritorializar uma imaginação espacial e as estéticas videográficas.

A presente pesquisa aborda o estudo dos movimentos do pensamento nas redes formação com professores no Ensino Fundamental. Procura problematizar a constituição de processos formativos não dogmáticos nos cotidianos escolares, pela articulação de imagens cinema e imagens escola, exibidas, compartilhadas e criadas nas redes de conversações em uma escola municipal de Vitória/ES, entre os anos de 2013 a 2014. Apresenta um campo problemático que se dedica a compreender que movimentos do pensamento são produzidos a partir do uso de imagens cinema nas redes de conversações com professores em formação continuada, a partir de uma cartografia de pesquisa que articula o mapeamento de virtualidades presentes no banco de teses da CAPES e as cinecartografias do cotidiano escolar em duas dobras do tempo. Tece um debate teórico que se compõe pelas linhas de pensamento, principalmente, de Bergson (2006a; 2006b), Carvalho (2008; 2009; 2010; 2012) Deleuze (1988; 1991; 1992; 1995; 1996; 2003; 2006; 2007; 2010; 2011), Foucault (1979; 1999. 2006a; 2006b; 2008), Guéron (2010; 2011), Pelbart (2011), Sauvagnargues (2009), dentre outros. As opções teórico-metodológicas estabelecem uma pesquisa de campo na intercessão entre os estudos com o cotidiano escolar e os da pesquisa cartográfica, tomando o plano de imanência como mundo material para a ocorrência de acontecimentos. Utiliza, para a produção de dados a observação participante, os registros em diário de campo, considerados como uma descrição cristalina, assim como, as narrativas cristalinas que emergiam nas redes de conversações com as personagens-escolas (docentes), que se configuram como imagensnarrativas de pesquisa. Organiza o debate dos dados produzidos em sete capítulos. Apresenta como considerações três principais agrupamentos do movimento do pensamento nas redes de conversações com professores provocados pelas imagens cinema: cinescola-clichês, imagensformação docente e cristaisescolas. Destaca que não há uma realidade que comporte uma só verdade sobre a formação com professores, mas o que existe são cortes, percepções, afecções e produções de verdades que entrelaçam os diferentes modos de existir e reexistir na educação brasileira. As telasescolas, assim como as telas do cinema, apresentam e nos cercam com um mundo que articula um cinescola-clichê, imagensformação docente e cristaisescolas, que para além do pensamento reto e dogmático, exibem entre imagens cinema e imagens escolas, outros movimentos do pensamento com a formação de professores, sendo capazes de potencializar as pesquisas educacionais a não ir em ‘busca de um tempo perdido, mas de um tempo a ser redescoberto’, fazendo crer ainda mais nesse mundo e na educação.

The radial undistortion model proposed by Fitzgibbon and the radial fundamental matrix were early steps to extend classical epipolar geometry to distorted cameras. Later minimal solvers have been proposed to find relative pose and radial distortion, given point correspondences between images. However, a big drawback of all these approaches is that they require the distortion center to be exactly known. In this paper we show how the distortion center can be absorbed into a new radial fundamental matrix. This new formulation is much more practical in reality as it allows also digital zoom, cropped images and camera-lens systems where the distortion center does not exactly coincide with the image center. In particular we start from the setting where only one of the two images contains radial distortion, analyze the structure of the particular radial fundamental matrix and show that the technique also generalizes to other linear multi-view relationships like trifocal tensor and homography. For the new radial fundamental matrix we propose different estimation algorithms from 9,10 and 11 points. We show how to extract the epipoles and prove the practical applicability on several epipolar geometry image pairs with strong distortion that - to the best of our knowledge - no other existing algorithm can handle properly.

Guimarães hosted the European Capital of Culture (ECOC) during the year of 2012. This study investigates the differences between Portuguese and foreign tourists regarding the main motivations to visit Guimarães and the retained perceived image of the destination. To achieve that purpose a survey was administered to 390 tourists that visited the city during the cultural event. The results show that tourists who visited Guimarães are relatively young, wealthy, employed and well educated. They are touring around the northern part of the country which includes an itinerary beginning in Porto, and extended to other important neighboring cities such as Braga or Viana do Castelo. The main motivations to visit the city, for both Portuguese and foreign tourists, are its historical heritage and the title of ECOC, the associated cultural events and celebrations that take place during 2012. However, these items were more valued by foreigners than Portuguese tourists. Using a factor analysis the tourists’ perceived attributes of Guimarães were described in four dimensions: “material heritage”, “intangible heritage”, “cultural performance”, and “sport and education”. Although foreigners and nationals perceived the tourism attributes of the city differently, the comparison of the mean scores of the four factors across Portuguese and foreigner tourists reveals that the most valued and least valued factors are common to both groups.

Once delighted by the moving image advent as a new method of realistically presenting reality, the viewer has been reposition himself towards the audiovisual contents he consumes, as he is given the opportunity to create and share his own perspective of that reality. We are living in a new technological setting, governed mainly by factors of interactivity, digital systems and technological convergence. The research project that we will present in this paper focuses on the subject of participatory media and the way cultural institutions are increasingly facing the inevitability of a profound revision of their traditional parameters of unidirectional communication, given the increasing availability of tools for audiovisual production as well as the diversity of networked communication contexts. The Serralves Foundation with its Museum of Contemporary Art, in Porto, Portugal, was the subject of a fi rst study of an empirical nature: a series of audiovisual objects were developed, in order to generate material for analysis and proposition. In this new stage of the project, our aim is to identify new procedures and practices that may be effectively implemented within the institutional universe. We intend to propose effi cient audiovisual communication contexts, including the maximizing of the relationship between institutions and audiences regarding dimensions that are traditionally outside the institutional radar: identity, narrative and affection. The project is currently in the process of surveying and categorization, with the aim of producing a map of different vocations and positions of the various institutions in regards to the aforementioned issues, which require participatory communication.

Protein aggregation became a widely accepted marker of many polyQ disorders, including Machado-Joseph disease (MJD), and is often used as readout for disease progression and development of therapeutic strategies. The lack of good platforms to rapidly quantify protein aggregates in a wide range of disease animal models prompted us to generate a novel image processing application that automatically identifies and quantifies the aggregates in a standardized and operator-independent manner. We propose here a novel image processing tool to quantify the protein aggregates in a Caenorhabditis elegans (C. elegans) model of MJD. Confocal mi-croscopy images were obtained from animals of different genetic conditions. The image processing application was developed using MeVisLab as a platform to pro-cess, analyse and visualize the images obtained from those animals. All segmenta-tion algorithms were based on intensity pixel levels.The quantification of area or numbers of aggregates per total body area, as well as the number of aggregates per animal were shown to be reliable and reproducible measures of protein aggrega-tion in C. elegans. The results obtained were consistent with the levels of aggrega-tion observed in the images. In conclusion, this novel imaging processing applica-tion allows the non-biased, reliable and high throughput quantification of protein aggregates in a C. elegans model of MJD, which may contribute to a significant improvement on the prognosis of treatment effectiveness for this group of disor-ders

Background and Purpose: Precise needle puncture of the kidney is a challenging and essential step for successful percutaneous nephrolithotomy (PCNL). Many devices and surgical techniques have been developed to easily achieve suitable renal access. This article presents a critical review to address the methodologies and techniques for conducting kidney targeting and the puncture step during PCNL. Based on this study, research paths are also provided for PCNL procedure improvement. Methods: Most relevant works concerning PCNL puncture were identified by a search of Medline/PubMed, ISI Web of Science, and Scopus databases from 2007 to December 2012. Two authors independently reviewed the studies. Results: A total of 911 abstracts and 346 full-text articles were assessed and discussed; 52 were included in this review as a summary of the main contributions to kidney targeting and puncturing. Conclusions: Multiple paths and technologic advances have been proposed in the field of urology and minimally invasive surgery to improve PCNL puncture. The most relevant contributions, however, have been provided by the applicationofmedical imaging guidance, newsurgical tools,motion tracking systems, robotics, andimage processing and computer graphics. Despite the multiple research paths for PCNL puncture guidance, no widely acceptable solution has yet been reached, and it remains an active and challenging research field. Future developments should focus on real-time methods, robust and accurate algorithms, and radiation free imaging techniques
Do desenho tradicional ao desenho Digitalum: caso de estudo “Wreck this Journal” vs “Wreck this App”

Tendo como ponto de partida um caso de estudo que compara dois livros de actividades (um livro tradicional no suporte físico, “Wreck this Journal”, adaptado para um livro digital, “Wreck this App”) da autora Keri Smith, propomo-nos analisar fundamentalmente as possibilidades do desenho de representação em ambos os suportes. Abordaremos ainda outros temas paralelos segundo parâmetros previamente definidos, que irão permitir comparar e analisar as propriedades de cada objecto, enquanto potencial espaço de representação. Este trabalho procura, assim, demonstrar de que forma estes dois suportes se complementam, livro impresso e livro digital, e de que forma podemos beneficiar com a introdução de novas técnicas e da possível transferência de conhecimentos adquiridos num objecto de estudo para o outro. Independentemente de permitirem diferentes experiências de interacção e de registo, torna-se relevante a forma com que estimulam a criatividade e a procura de novas formas de representação.

Background: Regulating mechanisms of branching morphogenesis of fetal lung rat explants have been an essential tool for molecular research. This work presents a new methodology to accurately quantify the epithelial, outer contour and peripheral airway buds of lung explants during cellular development from microscopic images. Methods: The outer contour was defined using an adaptive and multi-scale threshold algorithm whose level was automatically calculated based on an entropy maximization criterion. The inner lung epithelial was defined by a clustering procedure that groups small image regions according to the minimum description length principle and local statistical properties. Finally, the number of peripheral buds were counted as the skeleton branched ends from a skeletonized image of the lung inner epithelial. Results: The time for lung branching morphometric analysis was reduced in 98% in contrast to the manual method. Best results were obtained in the first two days of cellular development, with lesser standard deviations. Non-significant differences were found between the automatic and manual results in all culture days. Conclusions: The proposed method introduces a series of advantages related to its intuitive use and accuracy, making the technique suitable to images with different lightning characteristics and allowing a reliable comparison between different researchers.

In a time of fierce competition between regions, an image serve as a basis to develop a strong sense of community, which fosters trust and cooperation that can be mobilized for regional growth. A positive image and reputation could be used in the promotional activities of the region benefiting all the stakeholders as a whole. Mega cultural events are frequently used to attract tourists and investments to a region, but also to enhance the city’s image. This study adopts a marketing/communication perspective of city’s image, and intends to explain how the image of the city is perceived by their residents. Specifically, we intend to compare the perceptions of residents that effectively participated in the Guimarães European Capital of Culture (ECOC) 2012 (engaged residents), and the residents that only assisted to the event (attendees). Several significant findings are reported and their implications for event managers and public policy administrators presented, along with the limitations of the study.

The purpose of this study is to characterise the environmental management systems (EMS) certification process (International Organization for Standardization (ISO) 14001) in Portuguese small and medium enterprises (SMEs) following quality management system (QMS) certification (ISO 9001). The study is based on a sample from Portuguese SMEs which characterise the local reality in terms of companies certified in accordance with ISO 14001 after ISO 9001 certification. Some Portuguese SMEs have the EMS implemented but not certified, mainly given the lack of investment support and because it is considered merely a form of marketing. As such, they do not feel motivated to certificate an EMS in the company since they consider that it is a form of advertising and not a way to protect the environment. Nonetheless, it is already evident form other Portuguese SMEs that gained EMS certification that gains supersede marketing benefits and allow for evermore enduring benefits such as prevention of environmental risks, environment protection, improved company image, compliance with legislation and efficient use of natural resources. This paper also presented the main difficulties in achieving an EMS certification, including high certification costs, human resources, motivation issues and difficulties in changing the company’s culture.
