The study revealed the potential of marine yeasts as a source of single cell protein and immunostimulant for prawns. Prawns fed with the selected marine yeasts were showing more growth compared to the control feed and commercial feed. Yeasts being rich with proteins, vitamins and carbohydrates serve as a growth promoter for prawns as being evidenced in this study. The better performance of marine yeasts, D. hansenii S8 and S100 and C. tropicalis S186 compared to S. cerevisiae S36 as a feed supplement is worth investigating. Besides being a rich nutritional source, yeasts act as immunostimulants by virtue of its high carbohydrate (Beta, 1-3 glucan) and RNA content. Beta, 1-3 glucan, a cell wall component of yeasts /fungi is the most commonly used immunostimulant in aquaculture. The present study shows that even the whole cell yeast could serve as a good immunostimulant when supplied through diet. Extraction of Beta-1,3 glucan results in the removal of nutrients like proteins, vitamins etc. from the cell biomass.Utilization of the yeast biomass as such in the diet would help perform a dual role as nutritional component and immunostimulant for aquaculture applications.

This thesis presents a detailed account of a cost - effective approach towards enhanced production of alkaline protease at profitable levels using different fermentation designs employing cheap agro-industrial residues. It involves the optimisation of process parameters for the production of a thermostable alkaline protease by Vibrio sp. V26 under solid state, submerged and biphasic fermentations, production of the enzyme using cell immobilisation technology and the application of the crude enzyme on the deproteinisation of crustacean waste.The present investigation suggests an economic move towards Improved production of alkaline protease at gainful altitudes employing different fermentation designs utilising inexpensive agro-industrial residues. Moreover, the use of agro-industrial and other solid waste substrates for fermentation helps to provide a substitute in conserving the already dwindling global energy resources. Another alternative for accomplishing economically feasible production is by the use of immobilisation technique. This method avoids the wasteful expense of continually growing microorganisms. The high protease producing potential of the organism under study ascertains their exploitation in the utilisation and management of wastes. However, strain improvement studies for the production of high yielding variants using mutagens or by gene transfer are required before recommending them to Industries.Industries, all over the world, have made several attempts to exploit the microbial diversity of this planet. For sustainable development, it is essential to discover, develop and defend this natural prosperity. The Industrial development of any country is critically dependent on the intellectual and financial investment in this area. The need of the hour is to harness the beneficial uses of microbes for maximum utilisation of natural resources and technological yields. Owing to the multitude of applications in a variety of industrial sectors, there has always been an increasing demand for novel producers and resources of alkaline proteases as well as for innovative methods of production at a commercial altitude. This investigation forms a humble endeavour towards this perspective and bequeaths hope and inspiration for inventions to follow.

The present study aimed at the utlisation of microbial organisms for the production of good quality chitin and chitosan. The three strains used for the study were Lactobacillus plantarum, Lactobacililus brevis and Bacillus subtilis. These strains were selected on the basis of their acid producing ability to reduce the pH of the fermenting substrates to prevent spoilage and thus caused demineralisation of the shell. Besides, the proteolytic enzymes in these strains acted on proteinaceous covering of shrimp and thus caused deprotenisation of shrimp shell waste. Thus the two processes involved in chitin production can be affected to certain extent using bacterial fermentation of shrimp shell.Optimization parameters like fermentation period, quantity of inoculum, type of sugar, concentration of sugar etc. for fermentation with three different strains were studied. For these, parameters like pH, Total titrable acidity (TTA), changes in sugar concentration, changes in microbial count, sensory changes etc. were studied.Fermentation study with Lactobacillus plantarum was continued with 20% w/v jaggery broth for 15 days. The inoculum prepared yislded a cell concentration of approximately 108 CFU/ml. In the present study, lactic acid and dilute hydrochloric acid were used for initial pH adjustment because; without adjusting the initial pH, it took more than 5 hours for the lactic acid bacteria to convert glucose to lactic acid and during this delay spoilage occurred due to putrefying enzymes active at neutral or higher pH. During the fermentation study, pH first decreased in correspondence with increase in TTA values. This showed a clear indication of acid production by the strain. This trend continued till their proteolytic activity showed an increasing trend. When the available sugar source started depleting, proteolytic activity also decreased and pH increased. This was clearly reflected in the sensory evaluation results. Lactic acid treated samples showed greater extent of demineralization and deprotenisation at the end of fermentation study than hydrochloric acid treated samples. It can be due to the effect of strong hydrochloric acid on the initial microbial count, which directly affects the fermentation process. At the end of fermentation, about 76.5% of ash was removed in lactic acid treated samples and 71.8% in hydrochloric acid treated samples; 72.8% of proteins in lactic acid treated samples and 70.6% in hydrochloric acid treated samples.The residual protein and ash in the fermented residue were reduced to permissible limit by treatment with 0.8N HCI and 1M NaOH. Characteristics of chitin like chitin content, ash content, protein content, % of N- acetylation etc. were studied. Quality characteristics like viscosity, degree of deacetylation and molecular weight of chitosan prepared were also compared. The chitosan samples prepared from lactic acid treated showed high viscosity than HCI treated samples. But degree of deacetylation is more in HCI treated samples than lactic acid treated ones. Characteristics of protein liquor obtained like its biogenic composition, amino acid composition, total volatile base nitrogen, alpha amino nitrogen etc. also were studied to find out its suitability as animal feed supplement.Optimization of fermentation parameters for Lactobacillus brevis fermentation study was also conducted and parameters were standardized. Then detailed fermentation study was done in 20%wlv jaggery broth for 17 days. Also the effect of two different acid treatments (mild HCI and lactic acid) used for initial pH adjustment on chitin production were also studied. In this study also trend of changes in pH. changes in sugar concentration ,microbial count changes were similar to Lactobacillus plantarum studies. At the end of fermentation, residual protein in the samples were only 32.48% in HCI treated samples and 31.85% in lactic acid treated samples. The residual ash content was about 33.68% in HCI treated ones and 32.52% in lactic acid treated ones. The fermented residue was converted to chitin with good characteristics by treatment with 1.2MNaOH and 1NHCI.Characteristics of chitin samples prepared were studied and extent of Nacetylation was about 84% in HCI treated chitin and 85%in lactic acid treated ones assessed from FTIR spectrum. Chitosan was prepared from these samples by usual chemical method and its extent of solubility, degree of deacetylation, viscosity and molecular weight etc were studied. The values of viscosity and molecular weight of the samples prepared were comparatively less than the chitosan prepared by Lactobacillus plantarum fermentation. Characteristics of protein liquor obtained were analyzed to determine its quality and is suitability as animal feed supplement.Another strain used for the study was Bacillus subtilis and fermentation was carried out in 20%w/v jaggery broth for 15 days. It was found that Bacillus subtilis was more efficient than other Lactobacillus species for deprotenisation and demineralization. This was mainly due to the difference in the proteolytic nature of the strains. About 84% of protein and 72% of ash were removed at the end of fermentation. The thesis entitled "Studies on improved practices of prawn farming for higher production in central Kerala" prepared by the author describes various practices prevailing in the study area in order to elucidate their relative merits. The study on semi-intensive farming at Mundapuram, Kannur was also carried out and included in the thesis for comparison.The author felt it important to make a critical study of the existing culture practices in the central Kerala, a region where it has been existing since time immemorial.Careful analysis of data accrued by the author has helped him to identify strength, weakness, opportunities and threats confronting the shrimp farming. As a result it was possible to evolve an appropriate management technology taking into consideration the various ecological (location specific), social and economical conditions prevalent in the vast study area.

This thesis Entitled Marine actinomycetes as source of antimicrobial compounds and as probiotics and single cell protein for application in penaeid peawn culture systems. Ocean harbours more than 80% of all life on earth and remains our greatest untapped natural resource. The study revealed the potential of marine actinomycetes as a source of antimicrobial compounds. The selected streptomycetes were found to be capable of inhibiting most of the pathogenic vibrios, whichis a major problem both in hatcheries and grow out systems. The bioactive principle can be incorporated with commercial feeds and applied as medicated diet for the control of vibrios in culture systems.The hydrolytic potential inhibitory property against pathogens and non—pathogenicity to penaeid prawns make the selected Streptomycesspp.an effective probioic in aquaculture. Since there is considerably less inhibition to the natural in pond ecosystem the microbial diversityis being maintained and thereby the water quality. Actinomycetes was found to be a good source of single cell protein as an ingredient inaquaculture feed formulations. Large amount of mycelial waste (actinomycete biomassO is produced from antibiotic industries and this nutrient rich waste can be effectively used as a protein source in aquaculture feeds.This study reveals the importance of marine actinomycetes as a source of antimicrobial compounds and as a probiotic and single cell protein for aquaculture applications.

The growing demand for quality prawn seed from the farmers‘ and entrepreneurs, coupled with uncertainity of their availability from nature at the appropriate time in required quantities has prompted‘ research on problems connected with prawn seed production. Endocrine control of reproduction in the penaeid shrimp _P_. monodon has been investigated in detail by adopting a comprehensive approach to the problem. The major aspects of the study included in depth investigations of the cytological details of the reproductive and neuroendocrine organs in correlation with the process of gonadal maturation. Based on the conclusions drawn from such ultrastructural studies various endocrine manipulations were carried out to see their effects on gonadal maturation. Besides that studies on the reproductive quality of male P_. monodon and the cryopreservation of spermatophores form a part of the present investigation. The shrimp 3; Inonodon used in the present study were collected from the offshore waters of Cochin, Madras and Mandapam and from the culture ponds of Vypeen Island near Cochin (Kerala) . The entire investigation on the cytological aspects were carried out using standard histological and electron-microscopic methods. Endocrine manipulations and cryopreservation studies is also carried out using the standard methods.

A study of the reproductive physiology of P. (P). semisulcatus wasundertaken as this information is an essential prerequisite for brood stock development for hatchery operations, and the results are embodied in this thesis. The thesis is presented in seven chapters. The protein, lipid and carbohydrate contents of ovary were estimated for the different vitellogenic phases. The protein, lipid, carbohydrate and cholesterol contents were estimated in testes and spermatophore.The thesis present the results of induced maturation experiments by eyestalk ablation, CNS extract injection and UV rays application on immature female prawns.

The thesis on the"Benthic ecology of selected prawn culture fields and ponds near Cochin” was taken up with a view to provide information on the qualitative and quantitative distribution of benthos and their relationships to prawnproduction of different culture ecosystems and to the physico-chemical parameters influencing their production. A two-year observation was carried out in nine selected prawn culture systems including perennial ponds (stations 1 to 4) seasonal fields (stations 5 to 7) and contiguous canals (stations 8 and 9) during December 1988 to November 1989. All macro- and meiobenthic organisms contributing to the faua were identified and their abundance, distribution, diversity, biomass and trophic relationships between benthos and prawns were studied. The environmental variables studied were temperature pH, salinity, dissolved oxygen, alkalinity, nitrite-nitrogen, nitrate-nitrogen, amonianitrogen, phosphate and silicate of bottom water and organic carbon and texture of the soil The thesis is presented in 4 Chapters. Chapter I presents an’ INTRODUCTION to the topic of study and a review of relevant works to bring an awareness to the present status of research in benthos and benthic ecology. Chapter 11, MATERIALS AND MTHODS, includes the techniques of sampling, preservation of samples and methods of analyses of various physico-chemical factors and area covered under the study is also given in this chapter. Chapter III, HYDROGRAPHY deals with the results of investigation and discussion onthe physico-chemical parameters of water and Chapter IV, SEDIMENT covers the sedimentoloical characteristics of the different culture systems followed by a detailed discussion. Chapter V, BOTTOM FAUNA presents an account on the various aspects of benthos and benthic ecology and the details of prawn production. A discussion on the overall assessment of interrelations between abiotic and biotic factors is given in Chapter VI, DISCUSSION. A critical evaluation of the implication of benthic production on prawn production under culture conditions and trophic relationships are also included in this chapter. An executive SUMMARY of the observations made during this study is presented in the final section of the thesis .

Prawn culture by traditional method forms an important occupation for the people in these areas, especially in the Vypeen island. Though short term studies have been made on various aspects of prawn culture field and its ecology, a study of detailed nature covering perennial, seasonal, fields and canals between coconut plantation is lacking from these areas. This study will also enable to assess the relative productivity of different systems during different seasons and the influence of the environment on the production potentials. Therefore the present study is taken upto throw more light on the ecological characteristics of these fields with special emphasis on its primary, secondary and tertiary production. The present area of investigation includes the prawn culture fields adjacent to Cochin backwater. The Cochin backwater (O9° 58'N 76° 28'E) is a shallow semi-enclosed body of water of tropical estuary. A narrow gut, about 450 M wide forms its main connection with the Arabian sea and this region is subjected to regular tidal influenceertiary production.

The present investigation has been carried out to understand the process and events leading to maturation of the ovary and testis in the Indian white prawn Penaeus indicus. The study includes the classification of the ovarian maturity stages based on its colour, gonadosomatic index, oocyte diameter and morphological changes in the oocyte. Further the process of oogenesis has been investigated using light and electron microscopic techniques. A histochemical study of the ovary has also been carried out to determine the sequence in which yolk substances are synthesized or sequestered in the oocytes and also to elicit the nature of the penaeid yolk material. The process of spermatogenesis and the development of the spermatophore has been studied in detail using light and electron microscopic methods. In addition a brief histochemical study on the testis was also made to understand the nature of the organic reserves in the sperm cells.

Recent studies with prawns indicate that their growth, metamorphosis, maturation and moulting are affected by the typo and level of lipids supplied in the diets. Despite the recognition of the importance of lipids in the diets of prawns there is no information on the essentiality and quantitative lipid requirements of Indian penaeid prawns. Therefore during the present study about 24 laboratory experiments were conducted to determine the essentiality and dietary requirements of total lipids, phoapholipida, fatty acids cholestrol, and to ascertain the nutritional value of natural lipid sources for the larvae, post-larvae tad juveniles of one of the most suitable cultivable species of panaeid prans. All the experiments were conducted in the laboratory following standard procedures, using isonitrogen and approaximately isocaloric purified diets. Changes were made in tha ingridients as required for specific requirements. For the larvae diets of particle size < 37) were fed, For the postlarvae and juveniles pellet feed was given. while data on survival and growth of larvae and pout-larvae 1-10 were recorded, data were collected on the survival, growth, food conversion ratio, protein efficiency ratio and biochemical composition at the body for post-larva 11-25 and juveniles. The influence ot fatty acid pattern of dietary lipid sources on the fatty acids profile of prawns were also studied in the case of juvenile prawns. Analysis of variance andleast significant differences test were employed to determine the significant differences between treatments in the observed parameters with the help of a newlett Packard master computer.

Metapenaeus dobsoni (Miers) is the most abundant species along the coast of Kerala. It is cultured extensively by adoption of traditional farming practices. The geographical location and water source determines the seasonal and annual environmental fluctuations the prawn farming systems experiences. The life cycle of the shrimp includes its migration to the coastal deeper waters for spawning and the immigration of larvae to the estuaries for growth. The survival of the species in such complex ecosystems is thus critical to its life cycle. The animal adapts itself to different environments through a physiological process known as osmoregulation. The present study on osmoregulation in the penaeid prawn Metapenaeus dobsoni was thus undertaken to understand the mechanism adopted by this species to survive in different environments. A number of experimental work have been conducted to understand the effect of salinity on the internal variations. However the effect of the complex environmental conditions as existent in nature on the osmotic variations in this species has not been dealt with in any of the earlier studies.
