本文采用电子自旋共振ESR方法,结合运用自旋捕捉技术(Spin Trapping-ESR)和时间分辨手段(TRESR),针对某些与生命能量代谢体系电子传递及其化学模拟反应的研究相关的几个重要问题(包括高等植物光系统II颗粒内超氧阴离子自由基(O2-)的产生机制、光合作用模型体系电子传递和跨膜电子传递反应动力学、传统中药有效成分提取物抗氧化分子机理与构效关系),从分子设计、实验方法、分子结构理论、反应机理与动力学分析等几个角度进行了较为系统的探索性研究,并获得以下几点新颖的研究成果: 1.光系统II颗粒内光抑制过程中O2-生成的分子机制 (1).首先,发展了新Spin Trapping-ESR技术,研制一系列性能优良的新型磷酰基取代的吡咯啉类活性氧自旋捕捉剂,并通过对比研究其捕捉性能,证明磷酰基取代的吡咯啉类捕捉剂比常用的DMPO捕捉剂的捕捉能力强、速度快,自由基加合物稳定性高,适合于光系统II体系中活性氧的研究。 (2).在PSII颗粒的光抑制过程中成功地检测到了O2-,并探讨了影响O2-产生的诸多因素。包括氧分子的浓度、1O2增强剂与淬灭剂、pH值效应、电子传递链阻断剂的影响。首次提出了O2-生成的分子机制:PSII颗粒中产生的O2-是光系统II中反应中心产生的1O2与次级电子受体QA形成的质子化半醌自由基反应的产物。此外,设计了一套化学模拟体系,进一步证明了02-的生成的分子机制。 2. 中国传统性中药的酚类提取物抗氧化剂的抗氧化分子机理与构效关系研究 用理论计算与实验结合的手段,研究了酚类抗氧化剂与02的反应。探讨了酚类抗氧化物的分子结构与其抗氧化活性的构效关系,为评价抗氧化剂的抗氧化能力提供了一定的依据。 3.有关光合作用模型体系电子传递和跨膜电子传递反应动力学的探索性基础研究 (1).对原有的电子自旋共振谱仪进行改造,自行设计并研制一套时间分辨ESR装置,时间分辨率达到准微秒级。 (2).利用时间分辨ESR装置,对C60及其环加成衍生物分子间和分子内光诱导电子转移反应的自由基复合过程动力学进行了研究,从分子结构角度分析了影响电荷分离态稳定性的因素。 (3).初步探讨了TPP/DODAC与HA/DODAC两种单层囊泡间的光诱导电子转移反应,获得了长寿命的电荷分离态,为光合作用模拟提供有价值的模型。 (4).通过对比研究mes-卟啉Ⅱ/苯醌/CH。OH的化学诱导动态核自旋态极化( CIDNP)和ESR波谱,提出一个激发态苯醌与质子给体间的光诱导氢转移自由基反应新机理。

Predictions for a 75x205mm surface semi-elliptic defect in the NESC-1 spinning cylinder test have been made using BS PD 6493:1991, the R6 procedure, non-linear cracked body finite element analysis techniques and the local approach to fracture. All the techniques agree in predicting ductile tearing near the inner surface of the cylinder followed by cleavage initiation. However they differ in the amount of ductile tearing, and the exact location and time of any cleavage event. The amount of ductile tearing decreases with increasing sophistication in the analysis, due to the drop in peak crack driving force and more explicit consideration of constraint effects. The local approach predicts a high probability of cleavage in both HAZ and base material after 190s, while the other predictions suggest that cleavage is unlikely in the HAZ due to constraint loss, but likely in the underlying base material. The timing of this event varies from ∼150s for R6 predictions to ∼250-300s using non-linear cracked body analysis.

A novel optical switching matrix measuring 1×2 mm2 in size is fabricated. The switching matrix is composed of waveguides, four 1×4 multimode interference (MMI) splitters, 32 total internal refraction mirrors and four 4×1 MMI combiners with the extremely compact size of 1×2 mm2. This integrated device are assessed and loss contribution measured from test structure is presented.

This paper uses finite element (FE) analysis to examine the residual stresses generated during the TIG welding of aluminium aerospace alloys. It also looks at whether such an approach could be useful for evaluating the effectiveness of various residual stress control techniques. However, such simulations cannot be founded in a vacuum. They require accurate measurements to refine and validate them. The unique aspect of this work is that two powerful engineering techniques are combined: FE modelling and neutron diffraction. Weld trials were performed and the direct measurement of residual strain made using the ENGIN neutron diffraction strain scanning facility. The predicted results show an excellent agreement with experimental values. Finally this model is used to simulate a weld made using a "Low Stress No Distortion" (LSND) technique. Although the stress reduction predicted is only moderate, the study suggests the approach to be a quick and efficient means of optimising such techniques.

Transient test facilities offer the potential for the simultaneous study of turbine aerodynamic performance, unsteady flow phenomena and the heat transfer characteristics of a turbine stage. This paper describes the development of aerodynamic performance measurement techniques in the Oxford Rotor Facility (ORF). The solutions to the technological issues involved with transient testing presented in this paper are expected to achieve levels of precision uncertainty comparable with traditional steady flow test rigs. The theoretical background to the measurement of aerodynamic performance is presented together with a comprehensive pre-test uncertainty analysis. The instrumentation scheme for the measurement of stage mass flow rate is discussed in detail, the measurements of shaft power, total inlet enthalpy, and stage pressure ratio are also outlined. The current working section features a 62% scale, 1-1/2 stage, high-pressure shroudless transonic turbine. The required inlet flow conditions are provided by an Isentropic Light Piston Tunnel (ILPT) with a quasi-steady state run time of approximately 70ms. The testing is conducted at engine representative specific speed, pressure ratio, gas-to-wall temperature ratio, Mach number and Reynolds number.

Two shock-capturing methods are considered. One is based on a standard conservative Roe scheme with van Leer's MUSCL variable extrapolation method applied to characteristic variables and a Runge-Kutta time stepping scheme. The other is based on the novel CABARET space-time scheme, which uses two sets of staggered variables, one for the conservation step and the other for characteristic splitting into local Riemann invariants. The parameters a and b of the length-weight relationship of Sepia pharaonis of the form of W=a.L was determined. Sex separated size fequency data collected from Karachi fish Harbour was analysed the length-weight equations, separable by male, female and sex combined. The apparent difference in paired values of exponents b1, b2 for any combination i.e. male versus female and male, female versus sex combined was tested for their significant difference. No significant difference was observed for any combination, this indicated no sex specific variation in length-weigh relationship of Sepia pharaonis.

Energy Piles present an efficient solution for long-term carbon emission reduction and sustainable construction. However, they have received only partial acceptance by the industry, because of concerns regarding the impact of cyclic thermal changes on the serviceability of energy pile foundations. This paper investigates the applicability of the hybrid load transfer approach to load-settlement analysis of single piles behavior during thermal energy exchange processes. Back-analysis results in terms of the thermal and mechanical response of energy piles show good agreement with field test results from Lambeth College in London. © ASCE 2011.

Latex beads were sensitized with monoclonal antibodies (MAb) rose against VP28 of WSSV. The optimum concentration of MAb required to sensitize the latex beads was 125 µg/ml. The sensitized latex beads were used to detect WSSV from PCR-positive stomach tissue homogenates obtained from infected shrimp. Stomach tissue homogenates from WSSV-infected shrimp agglutinated the sensitized latex beads within 10 minutes, while uninfected samples did not produce any agglutination, although non-specific agglutinations were observed in some samples. The analytical sensitivity, analytical specificity, diagnostic sensitivity and diagnostic specificity of the (LAT) agglutination test were assessed. The analytical sensitivity of the test was 40 ng of purified WSSV (2 µg/ml). The sensitized latex beads did not agglutinate with normal shrimp tissue or MBV-infected tissue homogenate. The test has a diagnostic sensitivity of 70 and 45%, respectively, compared to single-step and nested PCR. The diagnostic specificity of the test was 82%. This test is a simple and rapid on-farm test which can be used to corroborate clinical signs for the detection of WSSV in grow-out ponds.

It is observed that the freezing and thawing of fish leads to increase in the total activity of aspartate aminotransferase (AAT) in tissue fluid due to the release of the bound form of mitochondrial enzyme. Electrophoresis of the tissue fluid of fresh unfrozen fish shows only a single fast-moving band of AAT in the anodic region whereas frozen and thawed fish shows an additional slow-moving band corresponding to mitochondrial AAT in the cathodic region. The method can be adopted to distinguish fresh fish from frozen and thawed fish.

A host of methods and tools to support designing are being developed in Cambridge EDC. These range from tools for design management to those for the generation and selection of design ideas, layouts, materials and production processes. A project, to develop a device to improve arm mobility of muscular dystrophy sufferers, is undertaken as a test-bed to evaluate and improve these methods and tools as well as to observe and modify its design and management processes. This paper presents the difficulties and advantages of using design methods and tools within this rehabilitation design context, with special focus on the evolution of the designs, tools, and management processes.

The effect of vitamins C and E on some of growth factors of cultured Acipenser ruthenus was studied in this thesis. For this purpose diets supplemented with a combination of 100 and 400 mg/kg vitamin C, L-ascorbyl-2-polyphosphate and 100 and 400 mg/kg vitamin E, D-alpha-tocopherol,were each fed to sterlet in 2 replications for 15 weeks. Fifteen fish with average weight of 350.92±14.28 gr were distributed to each of 18 tanks after adaptation with experimental diet. After 5 weeks, there were significant differences in RBC, ESR, HCT and differential counting of white blood cells among the treatments (P<0.05), but there was no significant difference in the amount of WBC among the treatments (P>0.05). After 10 weeks, there were significant differences in the amounts of Monocytes, Lymphocytes and Eosinophils (P<0.05), but there was no significant differences in the amount of other hematologic factors (P>0.05). At the end of the experiment (15th week) only WBC, RBC, Monocyte, lymphocyte and neutrophil showed significant differences between the treatments (P<0.05) and other hematologic factors did not show any significant differences between the treatments (P>0.05). The results of biochemical indices analysis showed significant differences (P<0.05) among treatments for all of the parameters for 5th weeks, total protein and glucose for 10th week and only cholesterol for 15th week. The carcass analysis at the end of experiment showed that only the amount of carbohydrate, protein and ash were significantly difference between the treatments (P<0.05). The results of growth parameters at the end of 3th, 9th, 12th and 15th week showed significant differences between the treatments (P<0.05), but at the end of 6th week only GR was significantly different between the treatments (P<0.05). After concerning acute stress test including reduce water volume, cutting the aeration for 30 minutes at the end of experiment, cortisol and glucose significantly increased (P<0.05) compared with prestress period, but the lowest response to the stressor was observed in fish fed by E400 C 400 mg/kg. On the other hand the survival was 100% during the experiment and no mortality was occurred during this period. Results of this study indicate that Vitamins C and E can have remarkable effects on hematological, biochemical and growth indices in different growth periods. So regarding the effects of vitamin C and E on some growth indices, it seems that the diet containing E400 C 100 mg/kg can be considered as the optimum diet in the rearing condition for this weigth range of fish.
