The structural characterization of molecules used in the sterilization of blood for transfusions, such as crystal violet (CV), is relevant for understanding the action of these prophylactic drugs. The characterization is feasible by surface enhanced resonance Raman spectroscopy (SERRS) of CV in solution or on surfaces. The limit of detection of CV by SERRS, in the presence of colloidal particles, using 514.5 nm as excitation radiation, was found to be around 1 ppb. The characterization of CV was also made by SERS, by using different active-particles-containing substrates, proving the versatility of this technique for the study of such structures. The results suggest that the controlled production of highly efficient SERS-active substrates may allow qualitative and quantitative analysis, with high sensitivity, with potential applications in medical and environmental fields.

Background: None of the HIV T-cell vaccine candidates that have reached advanced clinical testing have been able to induce protective T cell immunity. A major reason for these failures may have been suboptimal T cell immunogen designs. Methods: To overcome this problem, we used a novel immunogen design approach that is based on functional T cell response data from more than 1,000 HIV-1 clade B and C infected individuals and which aims to direct the T cell response to the most vulnerable sites of HIV-1. Results: Our approach identified 16 regions in Gag, Pol, Vif and Nef that were relatively conserved and predominantly targeted by individuals with reduced viral loads. These regions formed the basis of the HIVACAT T-cell Immunogen (HTI) sequence which is 529 amino acids in length, includes more than 50 optimally defined CD4+ and CD8+ T-cell epitopes restricted by a wide range of HLA class I and II molecules and covers viral sites where mutations led to a dramatic reduction in viral replicative fitness. In both, C57BL/6 mice and Indian rhesus macaques immunized with an HTI-expressing DNA plasmid (DNA.HTI) induced broad and balanced T-cell responses to several segments within Gag, Pol, and Vif. DNA.HTI induced robust CD4+ and CD8+ T cell responses that were increased by a booster vaccination using modified virus Ankara (MVA.HTI), expanding the DNA.HTI induced response to up to 3.2% IFN-γ T-cells in macaques. HTI-specific T cells showed a central and effector memory phenotype with a significant fraction of the IFN-γ+ CD8+ T cells being Granzyme B+ and able to degranulate (CD107a+). Conclusions: These data demonstrate the immunogenicity of a novel HIV-1 T cell vaccine concept that induced broadly balanced responses to vulnerable sites of HIV-1 while avoiding the induction of responses to potential decoy targets that may divert effective T-cell responses towards variable and less protective viral determinants.

Gray mold of roses (Rosa hibrida) caused by Botrytis cinerea requires many management strategies for its control. The effect of pulsing rose cv. Kiss with solutions of citric acid, salicylic acid, sucrose, calcium sulfate, and silver thiosulfate (STS) on disease severity and vase life of the flowers was evaluated. The solutions were applied to cut stems at different stages of harvest, the variation in the opening stage of harvest did not affect the results. Pulsing with STS reduced the values of area under the disease progress curve (AUDPC) and of severity of disease by 15% and 55%, respectively, and increased the vase life of the flowers by 20%. Calcium sulfate consistently reduced AUDPC by 66% and maximum severity by 88%, and increased vase life of the flowers by 37%. Therefore, pulsing rose buds with solutions of STS and calcium sulfate is potentially useful in reducing losses due to gray mold after harvest and in extending the vase life.

Botrytis blight caused by Botrytis cinerea is an important disease of rose (Rosa hybrida) grown in greenhouses in Brazil. As little is known regarding the disease epidemiology under greenhouse conditions, pathogen survival in crop debris and as sclerotia was evaluated. Polyethylene bags with petals, leaves, or stem sections artificially infected with B. cinerea were mixed with crop debris in rose beds, in a commercial plastic greenhouse. High percentage of plant parts with sporulation was detected until 60 days, then sporulation decreased on petals after 120 days, and sharply decreased on stems or leaves after 90 days. Sporulation on petals continued for 360 days, but was not observed on stems after 150 days or leaves after 240 days. Although the fungus survived longer on petals, stems and leaves are also important inoculum sources because high amounts of both are deposited on beds during cultivation. Survival of sclerotia produced on PDA was also quantified. Sclerotia germination was greater than 75% in the initial 210 days and 50% until 360 days. Sclerotia weight gradually declined but they remained viable for 360 days. Sclerotia were produced on the buried petals, mainly after 90 days of burial, but not on leaves or stems. Germination of these sclerotia gradually decreased after 120 days, but lasted until 360 days. Higher weight loss and lower viability were observed on sclerotia produced on petals than on sclerotia produced in vitro

Pedro Abelardo apresenta na sua obra uma teoria da virtude de natureza, à primeira vista, aristotélica. Ao que parece, essa concepção também contém diferentes elementos estoicos, que não se opõem necessariamente à visão do Estagirita. Todavia, o essencial da interpretação da Escola do Pórtico acerca da virtude difere da explicação dada por Aristóteles. No presente estudo, pretendemos examinar, primeiro, a índole da virtude como habitus na obra de lógica de Abelardo. Nesse caso, não há dúvida de que predomina a influência aristotélica. Em seguida, indicamos outras particularidades da virtude encontradas, desta vez, em tratados éticos e teológicos do nosso protagonista, as quais se revelam antes de caráter estoico. Em suma, projetamos identificar as diferentes fontes antigas e os principais intermediários latinos usados por Abelardo para elaborar a sua própria teoria da virtude. Trata-se de deixar clara a influência filosófica dominante em Abelardo sobre essa questão da virtude, já que este lógico não pode harmonizar teses substancialmente inconciliáveis.

O camu-camu (Myrciaria dubia (H.B.K.) McVaugh, Myrtaceae) é uma fruteira nativa que ocorre nas margens de rios e lagos inundáveis da Amazônia, e está sendo domesticada visando o cultivo em terra firme. O alto teor de ácido ascórbico nos frutos, cerca de 2.800 mg.100g-1 de polpa, fortalece a demanda para consumo no país e exportação. No período de 1997 a 1998, investigou-se a biologia floral e fenologia reprodutiva do camu-camu, em plantios da Embrapa Amazônia Oriental, em Belém, e em áreas de ocorrência natural, em Santarém e Oriximiná, no Estado do Pará, norte do Brasil. Observações diretas sobre o comportamento dos visitantes foram acompanhadas da coleta de espécimens para identificação e registro fotográfico. M. dubia apresenta inflorescências com flores brancas, hermafroditas e poliândricas. A antese ocorre entre as 5:00 e 7:00 h. O pólen é seco e facilmente transportado pelo vento ou gravidade, sendo o principal recurso e atrativo floral. Os osmóforos estão localizados no cálice, corola, anteras e estigma. Constatou-se que a fenofase de floração foi do tipo "steady-state", sendo mais expressiva em março e a frutificação teve um pico no mês de julho. Observouse desfolha parcial em todas as plantas estudadas, principalmente em novembro. Os principais visitantes foram Nannotrigona punctata e Trigona pallens (Meliponinae) e pequenos besouros (Chrysomelidae). As abelhas sem ferrão foram consideradas os polinizadores legítimos.
