We characterized sequences from genes encoding cathepsin L-like (CatL-like) cysteine proteases from African and South American isolates of Trypanosoma vivax and T. vivax-like organisms, and evaluated their suitability as genetic markers for population structure analysis and diagnosis. Phylogenetic analysis of sequences corresponding to CatL-like catalytic domains revealed substantial polymorphism, and clades of sequences (TviCatL1-9) were separated by large genetic distances. TviCatL1-4 sequences were from cattle isolates from West Africa (Nigeria and Burkina Faso) and South America (Brazil and Venezuela), which belonged to the same T. vivax genotype. T. vivax-like genotypes from East Africa showed divergent sequences, including TviCatL5-7 for isolates from Mozambique and TviCatL8-9 for an isolate from Kenya. Phylogenetic analysis of CatL-like gene data supported the relationships among trypanosome species reflected in the phylogenies based on the analysis of small subunit (SSU) of ribosomal RNA gene sequence data. The discovery of different CatL-like sequences for each genotype, defined previously by ribosomal DNA data, indicate that these sequences provide useful targets for epidemiological and population genetic studies. Regions in CatL-like sequences shared by all T. vivax genotypes but not by other trypanosomes allowed the establishment of a specific and sensitive diagnostic PCR for epidemiological studies in South America and Africa. To find the most reliable screening method for Trypanosoma cruzi infection in blood banks. Epidemiological data, lymphoproliferation assay, parasitological, conventional serological tests: immunofluorescence, haemagglutination, ELISA with epimastigote and trypomastigote antigens and reference serological tests: trypomastigote excreted-secreted antigens (TESA) blot and chemiluminescent ELISA assay with mucine from trypomastigote forms were applied to individuals with inconclusive serology, non-chagasic individuals and chronic chagasic patients. TESA blot had the best performance when used as a single test in all the groups. In the inconclusive group 20.5% of individuals were positive for TESA blot, 23.3% for either lymphoproliferation or TESA blot, and 17.8% for lymphoproliferation only. Positive lymphoproliferation without detectable antibodies was observed in 5.47% of all inconclusive serology cases. Analysis of six parameters (three serological assays, at least one parasitological test, one lymphoproliferation assay and epidemiological data) in the inconclusive group showed that diagnosis of Chagas` disease was probable in 15 patients who were positive by two or more serological tests or for whom three of those six parameters were positive. TESA blot is a good confirmatory test for Chagas` disease in the inconclusive group. Although lymphoproliferation suggests the diagnosis of Chagas` disease in the absence of antibodies when associated with a high epidemiological risk of acquiring Chagas` disease, the data from this study and the characteristics of the lymphoproliferation assay (which is both laborious and time-consuming) do not support its use as a confirmatory test in blood-bank screening. However, our findings underscore the need to develop alternative methods that are not based on antibody detection to improve the diagnosis when serological tests are inconclusive.

In order to consider the photodynamic therapy (PDT) as a clinical treatment for candidosis, it is necessary to know its cytotoxic effect on normal cells and tissues. Therefore, this study evaluated the toxicity of PDT with PhotogemA (R) associated with red light-emitting diode (LED) on L929 and MDPC-23 cell cultures and healthy rat palatal mucosa. In the in vitro experiment, the cells (30000 cells/cm(2)) were seeded in 24-well plates for 48 h, incubated with PhotogemA (R) (50, 100, or 150 mg/l) and either irradiated or not with a red LED source (630 +/- 3 nm; 75 or 100 J/cm(2); 22 mW/cm(2)). Cell metabolism was evaluated by the MTT assay (ANOVA and Dunnet`s post hoc tests; p < 0.05) and cell morphology was examined by scanning electron microscopy. In the in vivo evaluation, PhotogemA (R) (500 mg/l) was applied to the palatal mucosa of Wistar rats during 30 min and exposed to red LED (630 nm) during 20 min (306 J/cm(2)). The palatal mucosa was photographed for macroscopic analysis at 0, 1, 3, and 7 days posttreatment and subjected to histological analysis after sacrifice of the rats. For both cell lines, there was a statistically significant decrease of the mitochondrial activity (90-97%) for all PhotogemA (R) concentrations associated with red LED regardless of the energy density. However, in the in vivo evaluation, the PDT-treated groups presented intact mucosa with normal characteristics both macroscopically and histologically. From these results, it may be concluded that the association of PhotogemA (R) and red LED caused severe toxic effects on normal cell cultures, characterized by the reduction of mitochondrial activity and morphological alterations, but did not cause damage to the rat palatal mucosa in vivo.

The anionic complexes [Cu(L(1-))(3)](1-), L(-) = dopasemiquinone or L-dopasemiqui none, were prepared and characterized. The complexes are stable in aqueous solution showing intense absorption bands at ca. 605 nm for Cu(II)-L-dopasemiquinone and at ca. 595 nm for Cu(II)-dopasemiquinone in the UV-vis spectra, that can be assigned to intraligand transitions. Noradrenaline and adrenaline, under the same reaction conditions, did not yield Cu-complexes, despite the bands in the UV region showing that noradrenaline and adrenaline were oxidized during the process. The complexes display a resonance Raman effect, and the most enhanced bands involve ring modes and particularly the vCC + vCO stretching mode at ca. 1384 cm(-1). The free radical nature of the ligands and the oxidation state of the Cu(II) were confirmed by the EPR spectra that display absorptions assigned to organic radicals with g= 2.0005 and g = 2.0923, and for Cu(II) with g = 2.008 and g = 2.0897 for L-dopasemiquinone and dopasemiquinone, respectively. The possibility that dopamine and L-dopa can form stable and aqueous-soluble copper complexes at neutral pH, whereas noradrenaline and adrenaline cannot, may be important in understanding how Cu(II)-dopamine crosses the cellular membrane as proposed in the literature to explain the role of copper in Wilson disease. The SU(3)(L)circle times U(1)(N) electroweak model predicts new Higgs bosons beyond the one of the standard model. In this work we investigate the signature and production of neutral SU(3)(L)circle times U(1)(N) Higgs bosons in the e(-)e(+) Next Linear Collider and in the CERN Linear Collider . We compute the branching ratios of two of the SU(3)(L)circle times U(1)(N) neutral Higgs bosons and study the possibility to detect them and the Z' extra neutral boson of the model.

Radiobiocomplexes are used to obtain images in nuclear medicine and employed in basic research. Blood constituents labeled with technetium-99m (99mTc) have also been employed as radiobiocomplexes and used also experimental model for evaluation of the biological effects of natural or synthetic drugs. The analysis of the morphology and the morphometrics parameters (perimeter/área ratio) can be used to evaluate the effects of drugs upon the structure of the membrane of red blood cells. Cinnamomum zeylanicum (cinnamon) is a spice used as herbal medicine to treat diseases. The aim of this study was to evaluate the effect of in vitro and in vivo treatment with an aqueous cinnamon extract on the labeling of blood constituents with 99mTc and on the morphology of red blood cells from Wistar rats. In the in vitro treatment, isolated blood sample from animals were incubated with cinnamon extract. In the in vivo treatment, blood samples were also withdrawn from animals treated with cinnamon extract. In both cases, the radiolabeling of blood constituents was done. The morphological analysis of red blood cells was also done. As control, blood or animals treated with NaCl 0.9%. The data obtained on the labeling of blood constituents with 99mTc experiments indicated that the in vitro treatment with cinnamon extract was capable to decrease signiicantly (p<0.05) the percentage of radioactivity in cellular compartments and on the fixation of cellular and plasma proteins. These effects were not observed on the in vivo treatment. The results obtained for the morphology of red blood cells suggest that the in vitro and in vivo treatments did not alter the morphology and the perimeter/area ratio. The in vitro treatment with aqueous cinnamon extract could affect the membrane structures related with the oxidation status of the stannous ion pertechnetate ion, altering the labeling of blood constituentes with 99mTc. Com o objetivo de reproduzir sintomas de malformação de raízes, observados em campo, foram estuda dos os efeitos do trifluralin sobre Arachis hypogaea L., cultivar Tatu, em condições de vaso, em Jaboticabal, SP. Quatro formulações de trifluralin foram aplicadas em solo Latossol Roxo - série Jaboticabal, barro argiloso, com 2,7% de matéria orgânica, em dosagem correspondentes a 1,5; 3,0 e 6,0 litros da formulação comercial por ha. Cada vaso contendo 2,5 1 -de solo, recebeu duas sementes pré -germinada s e foi mantido sob condições de casa-de-vegetação durante 29 dias. Foi adotado o delineamento inteiramente casualizado com treze tratamentos e quatro repetições, perfazendo total de 52 parcelas. Aos 29 dias as plantas, foram retiradas e determinadas as seguintes características: peso da matéria seca das raízes, dos caules, dos pecíolos e dos folíolos; número de folíolos; área foliar e comprimento da haste principal. Alterações morfológicas externas e internas foram observadas. Os resultados mostraram que as partes subter rân eas das plantas foram afetadas, com uma correlação linear negativa e significativa entre o aumento da dose dos herbicidas e peso da matéria. As folhas se mostraram pouco sensíveis, sendo que nenhuma das formulações diminuiu o número de foliolos da planta. A área foliar e o peso da matéria seca dos folíolos somente foram afetados pelo aumento da dose de T. Fecotrigo com va lores de F iguais a 9,62* e 7,61*, respectivamente. de modo geral,po de-se concluir que o maior efeito ocorre devido ao incremento nas doses e não devido a possiveis diferenças entre os tipos de formula ções testadas. Quanto a alterações morfológicas observou-se que nas doses mais elevadas houve grande diminuição na quantidade de raízes, com raízes secundárias curtas e grossas. O hipocótilo emitiu raízes até a altura do nó cotiledonar. Na dose de 3,0 1/ha já foram notadas raízes secundárias um pouco mais espessas que na dose de 1,5 1/ha. As lâminas histológicas de amostras da re gião do hipocótilo mostraram parên quima cortical bastante espesso, de vido ao maior numero de camadas de células, porém não se notou anormalidade na forma das células, nem nos demais tecidos da região estudada.

An in vitro protocol for Ficus carica cv. 'Roxo de Valinhos' was optimized. Nodal explants containing two buds were excised from field-grown mature plants, and transferred to different proliferation media consisting of combinations of distinct concentrations of activated charcoal with benzyladenine (BA), kinetin with gibberellic acid (GA(3)), and WPM (woody plant medium) with kinetin. The regular strength of WPM in combination with 0.5 mg l(-1) kinetin was the best condition for shoot proliferation of Ficus carica 'Roxo de Valinhos' plants. The addition of activated charcoal in the medium completely inhibited shoot proliferation. The inclusion of BA in the medium induced excessive callus formation as well as small and vitrified shoots, while GA(3) induced excessive elongation associated with vitrification, chlorosis, and tip-burned shoots.

'Paluma' guavas, after internal quality evaluation using magnetic resonance tomography, were used to produce fresh-cut product. Fruits were peeled or not, cut in halves and seed removed, and they were packaged in polystyrene trays covered with PVC film or in a PET container with a lid. These packages were stored for 12 days at 5 degrees C, 10 degrees C and ambient temperature (22.6 degrees C). Tomography evaluation verified that impacts produced internal bruising with loss of cellular integrity and liquefication of the placenta tissues. Compression was more evident on the pericarp and cutting promoted superficial deformation. Storage temperature affected the weight loss, with fruit packaged in the polystyrene tray having a greater weight loss. The peeling did not influence weight loss. Product stored at 5 degrees C and 10 degrees C for 8 days had low microbial growth (< 10(3) UFC.g(-1)) and no coliforms. Rapid spoilage and a short shelf life (3-4 days) occurred when the product was stored at ambient temperature. Peeling reduced ascorbic acid concentration and total soluble solids. Use of calcium to protect fresh-cut products was not efficient. Calcium absorption capacity of 'Pedro Sato' guava was tested using Ca-45. Fruits treated with 2% CaCl2, with or without the radioisotope, were divided in four layers (epicarp, mesocarp, endocarp and seed) and analyzed for the total and Ca-45 calcium. It was observed that the applied calcium remained in superficial layers of fruits, which was confirmed by autoradiography. Internal layers did not contain Ca-45, indicating that calcium was not distributed into different parts of the fruit.

Instalou-se sobre um latossolo vermelho-escuro textura média, um experimento cujo objetivo foi avaliar a influência de modos e épocas de aplicação de gesso e de calcário sobre algumas características nutricionais e tecnológicas da cultura do feijoeiro irrigado. Utilizou-se no experimento o delineamento em blocos casualizados em esquema fatorial (4x3)+2 com quatro repetições, correspondendo a 4 modos de aplicação de gesso e/ou calcário, 3 doses de gesso e 2 tratamentos adicionais. Os resultados mostraram que: o uso do gesso proporcionou maiores teores de Ca nos grãos e consequentemente maior tempo para o seu cozimento. A associação gesso e calcário propiciou aumento no teor de S das folhas e também no peso de 100 grãos, quando comparado à utilização de apenas gesso. A relação de hidratação diminuiu com o aumento das doses de gesso quando utilizou-se os modos de aplicação M1 (gesso aplicado no solo 45 dias antes da semeadura) e M2 (gesso e calcário aplicados conjuntamente aos 45 dias antes da semeadura).
