This work comprises three different types of unconventional correlated systems.rnChapters 3-5 of this work are about the open shell compounds Rb4O6 and Cs4O6. These mixed valent compounds contain oxygen in two different modifications: the closed-shell peroxide anion is nonmagnetic, whereas the hyperoxide anion contains an unpaired electrons in an antibonding pi*-orbital. Due to this electron magnetic ordering is rendered possible. In contrast to theoretical predictions, which suggested half-metallic ferromagnetism for Rb4O6,rndominating antiferromagnetic interactions were found in the experiment. Besidesrna symmetry reduction due to the mixed valency, strong electronic correlations of this highly molecular system determine its properties; it is a magnetically frustrated insulator. The corresponding Cs4O6 was found to show similar properties.rnChapters 6-9 of this work are about intermetallic Heusler superconductors. rnAll of these superconductors were rationally designed using the van Hove scenario as a working recipe. A saddle point in the energy dispersion curve of a solid leads to a van Hove singularity in the density of states. In the Ni-based and Pd-based Heusler superconductors presented in this work this sort of a valence instability occurs at the high-symmetry L point and coincides or nearly coincides with the Fermi level. The compounds escape the high density of states at the Fermi energy through a transition into the correlated superconducting state.rnChapter 10 of this work is about the tetragonally distorted ferrimagnetic DO22 phase of Mn3Ga. This hard-magnetic modification is technologically useful for spin torque transfer applications. The phase exhibits two different crystallographic sites that are occupied by Mn atoms and can thus be written as Mn2MnGa. The competition between the mainly itinerant moments of the Mn atoms at the Wyckoff position 4d and the localized moments of the Mn atoms at the Wyckoff position 2b leads to magnetic correlations. The antiferromagnetic orientation of these moments determines the compound to exhibit a resulting magnetic moment of approximately 1 µB per formula unit in a partially compensated ferrimagnetic configuration.

A series of imidazolium salts of the type [BocNHCH2CH2ImR]X (Boc = t-Bu carbamates; Im = imidazole) (R = Me, X = I, 1a; R = Bn, X = Br, 1b; R = Trityl, X = Cl, 1c) and [BnImR’]X (R’ = Me, X = Br, 1d; R’ = Bn, X = Br, 1e; R’ = Trityl, X = Cl, 1g; R’ = tBu, X = Br, 1h) bearing increasingly bulky substituents were synthetized and characterized. Subsequently, these precursors were employed in the synthesis of silver(I)-N-heterocyclic (NHC) complexes as transmetallating reagents for the preparation of rhodium(I) complexes [RhX(NBD)(NHC)] (NHC = 1-(2-NHBoc-ethyl)-3-R-imidazolin-2-ylidene; X = Cl; R = Me, 4a; R = Bn, 4b; R = Trityl, 4c; X = I, R = Me, 5a; NHC = 1-Bn-3-R’-imidazolin-2-ylidene; X = Cl; R’ = Me, 4d, R’ = Bn, 4e, R’ = Trityl, 4g; R’ = tBu, 4h). VT NMR studies of these complexes revealed a restricted rotation barriers about the metal-carbene bond. While the rotation barriers calculated for the complexes in which R = Me, Bn (4a,b,d,e and 5a) matched the experimental values, this was not true for the complexes 4c,g, bearing a trityl group for which the values are much smaller than the calculated ones. Energy barriers for 4c,g, derived from a line shape simulation, showed a strong dependence on the temperature while for 4h the rotational energy barrier is stopped at room temperature. The catalytic activity of the new rhodium compounds was investigated in the hydrosilylation of terminal alkynes and in the addition of phenylboronic acid to benzaldehyde. The imidazolium salts 1d,e were also employed in the synthesis of new iron(II)-NHC complexes. Finally, during a six-months stay at the University of York a new ligand derived from Norharman was prepared and employed in palladium-mediated cross-coupling.

In questo studio ci siamo proposti di investigare i sistemi di modulazione automatica della dose sui sistemi TC Multislice (Automatic Exposure Control – AEC) da tre diversi produttori, aventi differenti indicatori di qualità delle immagini. Il presente lavoro è stato svolto presso il Servizio di Fisica Sanitaria dell’Azienda Ospedaliera Universitaria - Policlinico Sant’Orsola-Malpighi di Bologna e consiste in un’analisi quantitativa della dose impiegata durante esami di Tomografia Computerizzata Multi-Slice (TCMS) e delle rispettive immagini radiologiche. Le immagini sono state acquisite con : GE LightSpeed VCT 64 e GE LightSpeed 16 (AEC AutomA 3D longitudinale e angolare), Siemens Sensation 16 (AEC CARE Dose 4D combinato), Philips Brilliance 16 e iCT 64 (separati in AEC ZDOM longitudinale e AEC DDOM angolare). Le acquisizioni TCMS sono state effettuate a differenti kV e mA di riferimento, al fine di investigarne gli effetti sulla modulazione, impiegando algoritmi di ricostruzione standard presenti su ogni macchina. Due fantocci antropomorfi simulanti la zona del torace e dell’addome sono stati utilizzati per simulare un paziente standard posizionato come in un esame clinico di routine ; a questo proposito, sono stati impiegati protocolli elicoidali standard con e senza modulazione. Sono inoltre stati testati differenti valori di indice di qualità delle immagini. Il profilo dei mA lungo la lunghezza è stato ottenuto utilizzando ImageJ, un programma open source comunemente utilizzato per l’elaborazione di immagini; i mAs sono stati normalizzati ad un fattore che tiene conto delle differenti geometrie e delle filtrazioni dei diversi scanner tomografici analizzati nell’esperienza. Il rumore è stato valutato tramite la scelta di determinate ROI (Region Of Interest) localizzate in aree il più possibili uniformi disponibili lungo i fantocci. Abbiamo registrato che una variazione del Noise Index o dei mAs di riferimento a seconda della tipologia di macchina analizzata risulta in uno shift dei profili di dose; lo stesso si è verificato quando sono stati cambiato kV o mA nella scout di acquisizione. Sistemi AEC longitudinali e combinati hanno mostrato profili di mAs normalizzati simili tra loro, con valori elevati evidenziati nella zona delle spalle e zona pelvi; sono state osservate differenze del 30-40% tra i differenti scanner tomografici. Solo in un caso di macchina analizzata si è verificato un comportamento opposto rispetto alle altre due tipologie di macchina in esame. A dispetto della differente natura dei sistemi AEC, i risultati ottenuti dai protocolli combinati e longitudinali sono simili tra loro. Il rumore presente nelle immagini è aumentato ad un livello accettabile e la sua uniformità lungo la direzione di scan è migliorata.

The research aims at developing a framework for semantic-based digital survey of architectural heritage. Rooted in knowledge-based modeling which extracts mathematical constraints of geometry from architectural treatises, as-built information of architecture obtained from image-based modeling is integrated with the ideal model in BIM platform. The knowledge-based modeling transforms the geometry and parametric relation of architectural components from 2D printings to 3D digital models, and create large amount variations based on shape grammar in real time thanks to parametric modeling. It also provides prior knowledge for semantically segmenting unorganized survey data. The emergence of SfM (Structure from Motion) provides access to reconstruct large complex architectural scenes with high flexibility, low cost and full automation, but low reliability of metric accuracy. We solve this problem by combing photogrammetric approaches which consists of camera configuration, image enhancement, and bundle adjustment, etc. Experiments show the accuracy of image-based modeling following our workflow is comparable to that from range-based modeling. We also demonstrate positive results of our optimized approach in digital reconstruction of portico where low-texture-vault and dramatical transition of illumination bring huge difficulties in the workflow without optimization. Once the as-built model is obtained, it is integrated with the ideal model in BIM platform which allows multiple data enrichment. In spite of its promising prospect in AEC industry, BIM is developed with limited consideration of reverse-engineering from survey data. Besides representing the architectural heritage in parallel ways (ideal model and as-built model) and comparing their difference, we concern how to create as-built model in BIM software which is still an open area to be addressed. The research is supposed to be fundamental for research of architectural history, documentation and conservation of architectural heritage, and renovation of existing buildings.

Während der Schwangerschaft kommt es häufig zu einer spontanen Verbesserung von klinischen Symptomen der autoimmunen Hepatitis und anderen Th1-vermittelten Autoimmunerkrankungen. Die Gründe hierfür sind bis heute noch nicht vollständig aufgeklärt. Eines der wichtigsten Hormone in der Schwangerschaft ist das humane Choriogonadotropin (hCG), welches schon in der frühen Schwangerschaft eine entscheidende Rolle spielt. Es sorgt für die Stimulation des Corpus luteums, wodurch es zur Ausschüttung von Progesteron kommt und somit die Einnistung der Blastozyte gewährleistet und die Abstoßung des Embryos verhindert wird. In dieser Arbeit wurden Effekt und Signalweg von hCG in primären murinen und humanen Hepatozyten sowie in Mausmodellen mit T-Zell-abhängigem Leberschaden untersucht. hCG führte sowohl bei akuten als auch bei chronischen Leberschäden zu einer drastischen Senkung der Aspartat-Aminotransferase, einem Indikator für Lebererkrankungen. Die Histologie der Leber hCG-behandelter Tiere wies außerdem signifikant weniger apoptotische Zellen und eine deutliche Reduktion infiltrierender CD4+ T-Zellen auf. Die Analyse des hCG-Signalweges zeigte, dass hCG die Langlebigkeitsproteine Foxo3a und Sirt1 reguliert. Die Aktivierung des PI3-Kinase/Akt-Signalweges durch hCG führte zu einem Transport des Transkriptionsfaktors Foxo3a aus dem Zellkern, wodurch die proapoptotischen Zielgene Bim und Puma nicht mehr transkribiert werden können. Eine zusätzliche Hemmung von Foxo3a erfolgte durch die Aktivierung der Deacetylase Sirt1, indem diese phosphoryliert wird und in den Zellkern transloziert. In weiteren Untersuchungen wurde der immunsuppressive Effekt von hCG näher betrachtet. Dabei stellte sich heraus, dass hCG effektiv die proteolytische Aktivität der Caspase-3 in Hepatozyten hemmt, wodurch die Ausschüttung der biologisch aktiven Form von Interleukin-16, einem chemotaktischen Faktor für CD4+ Zellen, herabgesetzt wird. Dadurch wird die Leber erfolgreich vor der Infiltration durch autoaggressive CD4+ Zellen geschützt. IL-16 spielt bei vielen inflammatorischen Krankheiten eine Rolle, was auch in dieser Arbeit durch den Nachweis hoher IL-16-Konzentrationen in Seren von Patienten mit autoimmuner Hepatitis bestätigt werden konnte. Die in dieser Studie beschriebene Wirkung von hCG und die Tatsache, dass hCG ein bereits bewährtes und auf Nebenwirkungen getestetes Medikament bei Infertilität ist, macht es zu einem idealen Kandidaten für immunsuppressive Therapieansätze bei akuten und chronisch entzündlichen Lebererkrankungen.

Una teoria di unificazione ha il notevole compito di fornire un modello in grado di unificare le forze fondamentali della natura in una sola. Storicamente uno dei primi tentativi è rappresentato dal modello di Kaluza, che propone una formulazione unificata di gravità ed elettromagnetismo. In 4 dimensioni il campo gravitazionale e il campo elettromagnetico sono entità nettamente separate. Tale dualismo può essere superato estendendo la teoria della Relatività Generale ad uno spaziotempo a 5 dimensioni. Se alle consuete 4 si aggiunge una quinta dimensione spaziale, allora si dimostra che la gravità e l’elettromagnetismo possono essere visti come la manifestazione di un unico campo di forza, che è interpretabile in termini della geometria dello spaziotempo a 5 dimensioni. Nonostante i suoi intrinseci limiti, il modello di Kaluza rappresenta comunque un punto di partenza per molte altre teorie di campo unificato più moderne e a più di 5 dimensioni. L'obiettivo è di sviluppare le linee fondamentali del modello di Kaluza. Preliminarmente si riportano i risultati principali dell'elettromagnetismo e della Relatività Generale, dato che il modello si formula a partire da questi. Si stabilisce la condizione di cilindro, secondo cui le quantità fisiche non subiscono variazioni nella quinta dimensione. Si ipotizza un ansatz per il tensore metrico 5D e si scrivono le equazioni di campo unitario e della geodetica, come estensioni a 5 dimensioni di quelle in 4. Si dimostra che il campo unitario in 4 dimensioni si separa nel campo scalare costante, nel campo elettromagnetico e nel campo gravitazionale. Le componenti quadridimensionali della geodetica 5D riconducono a quella 4D e alle leggi del moto 4D in presenza dei campi gravitazionale ed elettromagnetico. Inoltre si interpreta la carica elettrica come la quinta componente della velocità covariante 5D.

Die mittlere Überlebenszeit nach Erkennung eines Glioblastoms ohne Behandlung liegt bei 3 Monaten und kann durch die Behandlung mit Temozolomid (TMZ) auf etwa 15 Monate gesteigert werden. Neben TMZ sind die chlorethylierenden Nitrosoharnstoffe die meistversprechendsten und am häufigsten eingesetzten Chemotherapeutika in der Gliomtherapie. Hier liegt die mittlere Überlebenszeit bei 17,3 Monaten. Um die Therapie des Glioblastoms noch effektiver zu gestalten und Resistenzen zu begegnen, werden unterschiedlichste Ansätze untersucht. Eine zentrale Rolle spielen hierbei das activator protein 1 (AP-1) und die mitogen aktivierten Proteinkinasen (MAPK), deren Funktion in bisherigen Arbeiten noch unzureichend beleuchtet wurde.rnBesonders mit der Rolle des AP-1-bildenden Proteins FRA-1 in der Therapie des Glioblastoms haben sich bisher nur wenige Arbeiten beschäftigt, weshalb im ersten Teil der vorliegenden Arbeit dessen Funktion in der Regulation der Chemosensitivität gegenüber dem chlorethylierenden Agenz ACNU genauer untersucht wurde. Es konnte gezeigt werden, dass die FRA 1-Expression durch Behandlung mit ACNU induziert wird. Die Induktion erfolgte über die beiden MAPKs ERK1/2 und p38K. JNK hatte keinen Einfluss auf die Induktion. Durch die Herunterregulation der FRA-1-Expression mit Hilfe von siRNA und eines shRNA exprimierenden Plasmids kam es zu einer signifikanten Sensitivierung gegenüber ACNU. Dabei konnte gezeigt werden, dass die Herunterregulation der FRA-1-Expression in einer verminderten AP 1-Bildung, bedingt durch eine reduzierte Menge an FRA-1 im AP-1-Komplex resultiert. Die Sensitivierung gegenüber ACNU ist weder durch eine Veränderung in der DNA-Reparatur, noch in der Modulation der FAS-Ligand- bzw. FAS-Rezeptor-Expression bedingt. Auch die hier untersuchten BCL 2-Familienmitglieder wiesen keine Unterschiede in der Expression durch Modulation der FRA 1-Expression auf. Allerdings kam es durch die verminderte FRA-1-Expression zu einer Reduktion der Zellzahl in der G2/M-Phase nach Behandlung mit ACNU. Diese ging einher mit einer reduzierten Menge an phosphoryliertem und unphosphoryliertem CHK1, weshalb davon auszugehen ist, dass FRA 1 nach ACNU-Behandlung in Gliomzellen vor der Apoptose schützt, indem es modulierend auf die Zellzykluskontrolle einwirkt.rnIm zweiten Teil dieser Arbeit wurde die Regulation der apoptotischen Antwort nach Behandlung mit ACNU und TMZ genauer beleuchtet, wobei ein spezielles Augen¬merk auf AP 1 und die MAPKs gelegt wurde. Hier konnte gezeigt werden, dass die Apoptose nach Behandlung mit ACNU bzw. TMZ sowohl durch Spaltung von Pro-Caspase 8, als auch Pro-Caspase 9 eingeleitet wird. Dabei akkumulierte in beiden Fällen p53 vermehrt im Zellkern. Eine Inhibierung der transkriptionellen Aktivität von p53 führte nach ACNU-Behandlung zu einer Sensitivierung der Zellen, nach TMZ-Behandlung kam es zu einem leichten Anstieg in der Vitälität. Der FAS-Rezeptor wurde nach ACNU- und nach TMZ-Behandlung aktiviert und auch die DNA-Reparaturproteine DDB2 und XPC wurden in beiden Fällen vermehrt exprimiert. Für die MAPKs JNK und ERK1/2 konnte gezeigt werden, dass diese pro-apoptotisch wirken. Die AP-1-Bildung nach ACNU-Behandlung erfolgte bereits nach 24 h und war von langer Dauer, wohingegen nach TMZ-Behandlung nur eine transiente AP 1-Bildung zu relativ späten Zeitpunkten detektiert werden konnte. Ebenso konnte für das AP-1-Zielgen FAS-Ligand nach ACNU-Behandlung eine relativ schnelle, lang anhaltende Aktivierung detektiert werden, wohingegen nach TMZ-Behandlung zu einem späten Zeitpunkt ein kurzer Anstieg im Signal zu verzeichnen war. In späteren Experimenten konnte gezeigt werden, dass das BCL-2-Familienmitglied BIM eine zentrale Rolle in der Regulation des intrinsischen Apoptosesignalweges nach Behandlung mit ACNU und TMZ spielt. Die hier entstanden Ergebnisse tragen entscheidend zum Verständnis der durch diese beiden Agenzien gesteuerten, apoptotischen Signalwege bei und bieten eine fundierte Grundlage für weitere Untersuchungen.rn

Dalla raffigurazione artistica fino alla modellazione digitale, passando per il disegno tecnico, la rappresentazione del progetto d’architettura ha conosciuto nel tempo evoluzioni significative che solo di recente hanno raggiunto l’apice nell’utilizzo di modelli cognitivi in grado di collezionare ed organizzare il patrimonio di informazioni che gravitano attorno all’intero processo edilizio. L’impiego sempre più diffuso dello strumento informatico, insieme al coordinamento delle specializzazioni nelle molte discipline coinvolte nel progetto, ha favorito negli ultimi anni l’adozione del Building Information Modeling un processo che permette di rivoluzionare il mondo delle costruzioni, coprendo molteplici aspetti del ciclo di vita per un manufatto edilizio. Questa Tesi intende presentare in maniera specifica le tappe che hanno consentito il formarsi del BIM. La migliore capacità di gestione, un linguaggio comune tra i progettisti, un’ottimizzazione di risorse e costi, unito ad un controllo convincente ed accurato delle fasi di lavoro, sono alcune delle potenzialità non ancora completamente espresse dal Building Information Modeling che è destinato a divenire una consapevolezza strategica nel bagaglio culturale del professionista contemporaneo.

Soilborne wheat mosaic virus (SBWMV) is one of the most important winter wheat pathogens worldwide. To identify genes for resistance to the virus in U.S. winter wheat, association study was conducted using a selected panel of 205 elite experimental lines and cultivars from U.S. hard and soft winter wheat breeding programs. Virus symptoms were evaluated twice in virus-infected fields for the panel at Manhattan, KS in spring 2010 and 2011 and for a subpanel of 137 hard winter wheat accessions at Stillwater, OK in spring 2008. At the two locations, 69.8 and 79.5% of cultivars were resistant or moderately resistant to the disease, respectively. After 282 simple-sequence repeat markers covering all wheat chromosome arms were scanned for association in the panel, marker Xgwm469 on the long arm of chromosome 5D (5DL) showed a significant association with the disease rating. Three alleles (Xgwm469-165bp, -167bp, and -169bp) were associated with resistance and the null allele was associated with susceptibility. Correlations between the marker and the disease rating were highly significant (0.80 in Manhattan at P < 0.0001 and 0.63 in Stillwater at P < 0.0001). The alleles Xgwm469-165bp and Xgwm469-169bp were present mainly in the hard winter wheat group, whereas allele Xgwm469-167bp was predominant in the soft winter wheat. The 169 bp allele can be traced back to 'Newton', and the 165 bp allele to Aegilops tauschii. In addition, a novel locus on the short arm of chromosome 4D (4DS) was also identified to associate with the disease rating. Marker Xgwm469-5DL is closely linked to SBWMV resistance and highly polymorphic across the winter wheat accessions sampled in the study and, thus, should be useful in marker-assisted selection in U.S. winter wheat.

The immune system exhibits an enormous complexity. High throughput methods such as the "-omic'' technologies generate vast amounts of data that facilitate dissection of immunological processes at ever finer resolution. Using high-resolution data-driven systems analysis, causal relationships between complex molecular processes and particular immunological phenotypes can be constructed. However, processes in tissues, organs, and the organism itself (so-called higher level processes) also control and regulate the molecular (lower level) processes. Reverse systems engineering approaches, which focus on the examination of the structure, dynamics and control of the immune system, can help to understand the construction principles of the immune system. Such integrative mechanistic models can properly describe, explain, and predict the behavior of the immune system in health and disease by combining both higher and lower level processes. Moving from molecular and cellular levels to a multiscale systems understanding requires the development of methodologies that integrate data from different biological levels into multiscale mechanistic models. In particular, 3D imaging techniques and 4D modeling of the spatiotemporal dynamics of immune processes within lymphoid tissues are central for such integrative approaches. Both dynamic and global organ imaging technologies will be instrumental in facilitating comprehensive multiscale systems immunology analyses as discussed in this review.

The uptake of radiolabeled somatostatin analogs by tumor cells through receptor-mediated internalization is a critical process for the in vivo targeting of tumoral somatostatin receptors. In the present study, the somatostatin receptor internalization induced by a variety of somatostatin analogs was measured with new immunocytochemical methods that allow characterization of trafficking of the somatostatin receptor subtype 2 (sst2), somatostatin receptor subtype 3 (sst3), and somatostatin receptor subtype 5 (sst5) in vitro at the protein level. METHODS: Human embryonic kidney 293 (HEK293) cells expressing the sst2, sst3, or the sst5 were used in a morphologic immunocytochemical internalization assay using specific sst2, sst3 and sst5 antibodies to qualitatively and quantitatively determine the capability of somatostatin agonists or antagonists to induce somatostatin receptor internalization. In addition, the internalization properties of a selection of these agonists have been compared and quantified in sst2-expressing CHO-K1 cells using an ELISA. RESULTS: Agonists with a high sst2-binding affinity were able to induce sst2 internalization in the HEK293 and CHO-K1 cell lines. New sst2 agonists, such as Y-DOTA-TATE, Y-DOTA-NOC, Lu-DOTA-BOC-ATE (where DOTA is 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid; TATE is [Tyr3, Thr8]-octreotide; NOC is [1-NaI3]-octreotide; and BOC-ATE is [BzThi3, Thr8]-octreotide), iodinated sugar-containing octreotide analogs, or BIM-23244 were considerably more potent in internalizing sst2 than was DTPA-octreotide (where DTPA is diethylenetriaminepentaacetic acid). Similarly, compounds with high sst3 affinity such as KE108 were able to induce sst3 internalization. In sst2- or sst3-expressing cell lines, agonist-induced receptor internalization was efficiently abolished by sst2- or sst3-selective antagonists, respectively. Antagonists alone had no effect on sst2 or sst3 internalization. We also showed that somatostatin-28 and somatostatin-14 can induce sst5 internalization. Unexpectedly, however, potent sst5 agonists such as KE108, BIM-23244, and L-817,818 were not able to induce sst5 internalization under the same conditions. CONCLUSION: Using sensitive and reproducible immunocytochemical methods, the ability of various somatostatin analogs to induce sst2, sst3, and sst5 internalization has been qualitatively and quantitatively determined. Whereas all agonists triggered sst2 and sst3 internalization, sst5 internalization was induced by natural somatostatin peptides but not by synthetic high-affinity sst5 agonists. Such assays will be of considerable help for the future characterization of ligands foreseen for nuclear medicine applications.

The synchronization of dynamic multileaf collimator (DMLC) response with respiratory motion is critical to ensure the accuracy of DMLC-based four dimensional (4D) radiation delivery. In practice, however, a finite time delay (response time) between the acquisition of tumor position and multileaf collimator response necessitates predictive models of respiratory tumor motion to synchronize radiation delivery. Predicting a complex process such as respiratory motion introduces geometric errors, which have been reported in several publications. However, the dosimetric effect of such errors on 4D radiation delivery has not yet been investigated. Thus, our aim in this work was to quantify the dosimetric effects of geometric error due to prediction under several different conditions. Conformal and intensity modulated radiation therapy (IMRT) plans for a lung patient were generated for anterior-posterior/posterior-anterior (AP/PA) beam arrangements at 6 and 18 MV energies to provide planned dose distributions. Respiratory motion data was obtained from 60 diaphragm-motion fluoroscopy recordings from five patients. A linear adaptive filter was employed to predict the tumor position. The geometric error of prediction was defined as the absolute difference between predicted and actual positions at each diaphragm position. Distributions of geometric error of prediction were obtained for all of the respiratory motion data. Planned dose distributions were then convolved with distributions for the geometric error of prediction to obtain convolved dose distributions. The dosimetric effect of such geometric errors was determined as a function of several variables: response time (0-0.6 s), beam energy (6/18 MV), treatment delivery (3D/4D), treatment type (conformal/IMRT), beam direction (AP/PA), and breathing training type (free breathing/audio instruction/visual feedback). Dose difference and distance-to-agreement analysis was employed to quantify results. Based on our data, the dosimetric impact of prediction (a) increased with response time, (b) was larger for 3D radiation therapy as compared with 4D radiation therapy, (c) was relatively insensitive to change in beam energy and beam direction, (d) was greater for IMRT distributions as compared with conformal distributions, (e) was smaller than the dosimetric impact of latency, and (f) was greatest for respiration motion with audio instructions, followed by visual feedback and free breathing. Geometric errors of prediction that occur during 4D radiation delivery introduce dosimetric errors that are dependent on several factors, such as response time, treatment-delivery type, and beam energy. Even for relatively small response times of 0.6 s into the future, dosimetric errors due to prediction could approach delivery errors when respiratory motion is not accounted for at all. To reduce the dosimetric impact, better predictive models and/or shorter response times are required.

Information management is a key aspect of successful construction projects. Having inaccurate measurements and conflicting data can lead to costly mistakes, and vague quantities can ruin estimates and schedules. Building information modeling (BIM) augments a 3D model with a wide variety of information, which reduces many sources of error and can detect conflicts before they occur. Because new technology is often more complex, it can be difficult to effectively integrate it with existing business practices. In this paper, we will answer two questions: How can BIM add value to construction projects? and What lessons can be learned from other companies that use BIM or other similar technology? Previous research focused on the technology as if it were simply a tool, observing problems that occurred while integrating new technology into existing practices. Our research instead looks at the flow of information through a company and its network, seeing all the actors as part of an ecosystem. Building upon this idea, we proposed the metaphor of an information supply chain to illustrate how BIM can add value to a construction project. This paper then concludes with two case studies. The first case study illustrates a failure in the flow of information that could have prevented by using BIM. The second case study profiles a leading design firm that has used BIM products for many years and shows the real benefits of using this program.

BACKGROUND: Mast cells activation through FcepsilonRI cross-linking has a pivotal role in the initiation of allergic reactions. The influence of this activation on programmed cell death of human mast cells has not yet been clarified. This study evaluates the influence of IgE-dependent activation alone and in synergy with TRAIL on the expression of molecules involved in the apoptotic signal transduction. METHODS: Human cord blood derived mast cells (CBMC) were cultured with myeloma IgE followed by activation with anti-human IgE. The expression of proteins involved in apoptotic signal transduction was assessed by immunoblot analysis. To test the effect of activation on a pro-apoptotic stimulus, activated, IgE-treated and resting CBMC were incubated with TRAIL, or in a medium with suboptimal concentrations of stem cell factor (SCF). RESULTS: In accordance with a previous study of ours, it was found that IgE-dependent activation increased TRAIL-induced caspase-8 and caspase-3 cleavage. However, it did not have a significant influence on CBMC death induced by SCF withdrawal. IgE-dependent activation increased the expression of FLIP and myeloid cell leukemia 1 (MCL-1) anti-apoptotic molecules as well as the pro-apoptotic one, BIM. In addition, a decrease in BID expression was observed. TRAIL could reverse the increase in FLIP but did not influence the upregulation of MCL-1 and of BIM. CONCLUSIONS: These findings suggest that IgE-dependent activation of human mast cells induces an increase in both pro-survival and pro-apoptotic molecules. We therefore hypothesized that IgE-dependent activation may regulate human mast cell apoptosis by fine-tuning anti-apoptotic and pro-apoptotic factors.
