Granzyme (Gzm) B-deficient mice obtained by gene targeting were used to assess the role of Gzm B in the mechanisms used by natural killer (NK) and lymphokine-activated killer (LAK) cells to destroy target cells. Gzm B-/- NK cells, LAK cells, and cytotoxic T lymphocytes (CTL) all are defective in their ability to rapidly induce DNA fragmentation/apoptosis in susceptible target cells. This defect can be partially corrected with long incubation times of effector and target cells. Moreover, Gzm B-/- NK cells (but not CTL or LAK cells) exhibit a defect in 51Cr release from susceptible target cells. This 51Cr release defect in Gzm B-deficient NK cells is also not overcome by prolonged incubation times or high effector-to-target cell ratios. We conclude that Gzm B plays a critical and nonredundant role in the rapid induction of DNA fragmentation/apoptosis by NK cells, LAK cells, and CTL. Gzm B may have an additional role in NK cells (but not in CTL or LAK cells) for mediating 51Cr release.

Este trabajo, hermenéutico interpretativo, profundo y emotivo, que elegí para optar mi título doctoral, ha sido un desafío, que retó mi capacidad académica y me ha permitido “dar la palabra” a mujeres racializadas, sujetas a teorías y debates, que las dimensionan desde periféricos rincones de la sinrazón social, política y académica de una América Latina que se jacta de su Multiculturalidad y Pluriétnia, pero que no termina de encajar el tema de la Negritud, como un asunto político que demanda acciones integrales para lograr el reconocimiento “real” de esa declaración derechos humanos que proclama que: “Todos los seres humanos nacen libres e iguales en dignidad y derechos y, dotados como están de razón y conciencia, deben comportarse fraternalmente los unos con los otros” Colombia, ha mostrado su deseo de incorporar en su noción de Estado social de Derecho la premisa de que “Todos los seres humanos nacen libres e iguales en dignidad y derechos” y así lo recogió en su Carta Magna, en la reforma constitucional del año 1991, al declarar que “El Estado reconoce y protege la diversidad étnica y cultural de la Nación colombiana”. (Art, 7) y facultar a esas diversidades étnicas, como lo es la Afrodescendiente (en ese momento autodenominada Comunidades negras de Colombia) a proyectar una ley que les permitiera establecer mecanismos para la protección de la identidad cultural y de los derechos de las comunidades negras de Colombia como grupo étnico, y el fomento de su desarrollo económico y social, con el fin de garantizar que estas comunidades obtengan condiciones reales de igualdad de oportunidades frente al resto de la sociedad colombiana (Art 1, Ley de Comunidades Negras, 70 de 1993)...

El alarmante descenso del número de alumnos que estudian química en bachillerato hace necesaria la búsqueda de herramientas para recuperar los niveles de finales de la pasada década. Los autores proponen algunas estrategias, aplicables en todos los niveles de enseñanza no universitaria, que van desde experiencias de laboratorio para alumnos de primaria, hasta la creación de una serie de personajes de ficción, que intervienen en los enunciados de los problemas de química, poner a disposición de los alumnos colecciones de problemas resueltos y la participación en pruebas como las olimpiadas de química.

Electronic books (e-book) are an interesting option compared to classic paper books. Most e-reading devices of the first generation were based on e-ink technology. With the appearance of the Apple iPad on the market, TFT-LCDs became important in the field of e-reading. Both technologies have advantages and disadvantages but the question remains whether one or the other technology is better for reading. In the present study we analyzed and compared reading behavior when reading on e-inkreader (e-ink displays) and on tablets (TFT-LCDs) as measured by eye-tracking. The results suggest that the reading behavior on tablets is indeed very similar to the reading behavior on e-ink-reader. Participants showed no difference in fixation duration. Significant differences in reading speed and in the proportion of regressive saccades suggest that tablets, under special artificial light conditions, may even provide better legibility.

We improved upper Eocene to Oligocene deep-sea chronostratigraphic control by integrating isotope (87Sr/86Sr, delta18O, delta13C) stratigraphy and magnetostratigraphy. Most previous attempts to establish the timing of isotope fluctuations have relied upon biostratigraphic age estimates which have uncertainties of 0.5 to over 4.0 m.y. Deep Sea Drilling Project (DSDP) Site 522 contains the best available upper Eocene to Oligocene magnetostratigraphic record which allows first-order correlations of isotope records (87Sr/86Sr, delta18O, delta13C) to the Geomagnetic Polarity Time Scale (GPTS). Empirical calibrations between the 87Sr/86Sr of foraminifera and magnetochronology at Site 522 allow more precise correlation of ,unknown' samples with the GPTS. For example, shallow water and high-latitude sections may be tied into the deep-sea record. Sr-isotope stratigraphic resolution for the latest Eocene to Oligocene is approximately 2 m.y.

Deep brain stimulation has shown remarkable potential in alleviating otherwise treatment-resistant chronic pain, but little is currently known about the underlying neural mechanisms. Here for the first time, we used noninvasive neuroimaging by magnetoencephalography to map changes in neural activity induced by deep brain stimulation in a patient with severe phantom limb pain. When the stimulator was turned off, the patient reported significant increases in subjective pain. Corresponding significant changes in neural activity were found in a network including the mid-anterior orbitofrontal and subgenual cingulate cortices; these areas are known to be involved in pain relief. Hence, they could potentially serve as future surgical targets to relieve chronic pain. © 2007 Lippincott Williams & Wilkins, Inc.

In many areas of northern India, salinity renders groundwater unsuitable for drinking and even for irrigation. Though membrane treatment can be used to remove the salt, there are some drawbacks to this approach e.g. (1) depletion of the groundwater due to over-abstraction, (2) saline contamination of surface water and soil caused by concentrate disposal and (3) high electricity usage. To address these issues, a system is proposed in which a photovoltaic-powered reverse osmosis (RO) system is used to irrigate a greenhouse (GH) in a stand-alone arrangement. The concentrate from the RO is supplied to an evaporative cooling system, thus reducing the volume of the concentrate so that finally it can be evaporated in a pond to solid for safe disposal. Based on typical meteorological data for Delhi, calculations based on mass and energy balance are presented to assess the sizing and cost of the system. It is shown that solar radiation, freshwater output and evapotranspiration demand are readily matched due to the approximately linear relation among these variables. The demand for concentrate varies independently, however, thus favouring the use of a variable recovery arrangement. Though enough water may be harvested from the GH roof to provide year-round irrigation, this would require considerable storage. Some practical options for storage tanks are discussed. An alternative use of rainwater is in misting to reduce peak temperatures in the summer. An example optimised design provides internal temperatures below 30EC (monthly average daily maxima) for 8 months of the year and costs about €36,000 for the whole system with GH floor area of 1000 m2 . Further work is needed to assess technical risks relating to scale-deposition in the membrane and evaporative pads, and to develop a business model that will allow such a project to succeed in the Indian rural context.
