The initial aim of this project was to improve the performance of a chromatographic bioreactor-separator (CBRS). In such a system, a dilute enzyme solution is pumped continuously through a preparative chromatographic column, while pulses of substrate are periodically injected on to the column. Enzymic reaction and separation are therefore performed in a single unit operation. The chromatographic columns used were jacketed glass columns ranging from 1 to 2 metres long with an internal diameter of 1.5 cm. Linking these columns allowed 1, 2, 3 and 4 metre long CBRS systems to be constructed. The hydrolysis of lactose in the presence of β~galactosidase was the reaction of study. From previous work at Aston University, there appeared to be no difficulties in achieving complete lactose hydrolysis in a CBRS. There did, however, appear to be scope for improving the separative performance, so this was adopted as an initial goal. Reducing the particle size of the stationary phase was identified as a way of achieving this improvement. A cation exchange resin was selected which had an average particle size of around half that previously used when studying this reaction. A CBRS system was developed which overcame the operational problems (such as high pressure drop development) associated with use of such a particle size. A significant improvement in separative power was achieved. This was shown by an increase in the number of theoretical plates (N) from about 500 to about 3000 for a 2 metre long CBRS, coupled with higher resolution. A simple experiment with the 1 metre column showed that combined bioreaction and separation was achievable in this system. Having improved the separative performance of the system, the factors affecting enzymic reaction in a CBRS were investigated; including pulse volume and the degree of mixing between enzyme and substrate. The progress of reaction in a CBRS was then studied. This information was related to the interaction of reaction and separation over the reaction zone. The effect of injecting a pulse over a length of time as in CBRS operation was simulated by fed batch experiments. These experiments were performed in parallel with normal batch experiments where the substrate is mixed almost instantly with the enzyme. The batch experiments enabled samples to be taken every minute and revealed that reaction is very rapid. The hydrodynamic characteristics of the two injector configurations used in CBRS construction were studied using Magnetic Resonance Imaging, combined with hydrodynamic calculations. During the optimisation studies, galactooligosaccharides (GOS) were detected as intermediates in the hydrolysis process. GOS are valuable products with potential and existing applications in food manufacture (as nutraceuticals), medicine and drug targeting. The focus of the research was therefore turned to GOS production. A means of controlling reaction to arrest break down of GOS was required. Raising temperature was identified as a possible means of achieving this within a CBRS. Studies were undertaken to optimise the yield of oligosaccharides, culminating in the design, construction and evaluation of a Dithermal Chromatographic Bioreactor-separator.

An HPLC method has been developed and validated for the rapid determination of mercaptopurine and four of its metabolites; thioguanine, thiouric acid, thioxanthine and methylmercaptopurine in plasma and red blood cells. The method involves a simple treatment procedure based on deproteinisation by perchloric acid followed by acid hydrolysis and heating for 45min at 100 degrees C. The developed method was linear over the concentration range studied with a correlation coefficient >0.994 for all compounds in both plasma and erythrocytes. The lower limits of quantification were 13, 14, 3, 2, 95pmol/8 x 10(8) RBCs and 2, 5, 2, 3, 20ng/ml plasma for thioguanine, thiouric acid, mercaptopurine, thioxanthine and methylmercaptopurine, respectively. The method described is selective and sensitive enough to analyse the different metabolites in a single run under isocratic conditions. Furthermore, it has been shown to be applicable for monitoring these metabolites in paediatric patients due to the low volume requirement (200microl of plasma or erythrocytes) and has been successfully applied for investigating population pharmacokinetics, pharmacogenetics and non-adherence to therapy in these patients.

Measurement of lipid peroxidation is a commonly used method of detecting oxidative damage to biological tissues, but the most frequently used methods, including MS, measure breakdown products and are therefore indirect. We have coupled reversed-phase HPLC with positive-ionization electrospray MS (LC-MS) to provide a method for separating and detecting intact oxidized phospholipids in oxidatively stressed mammalian cells without extensive sample preparation. The elution profile of phospholipid hydroperoxides and chlorohydrins was first characterized using individual phospholipids or a defined phospholipid mixture as a model system. The facility of detection of the oxidized species in complex mixtures was greatly improved compared with direct-injection MS analysis, as they eluted earlier than the native lipids, owing to the decrease in hydrophobicity. In U937 and HL60 cells treated in vitro with t-butylhydroperoxide plus Fe2+, lipid oxidation could not be observed by direct injection, but LC-MS allowed the detection of monohydroperoxides of palmitoyl-linoleoyl and stearoyl-linoleoyl phosphatidylcholines. The levels of hydroperoxides observed in U937 cells were found to depend on the duration and severity of the oxidative stress. In cells treated with HOCl, chlorohydrins of palmitoyloleoyl phosphatidylcholine were observed by LC-MS. The method was able to detect very small amounts of oxidized lipids compared with the levels of native lipids present. The membrane-lipid profiles of these cells were found to be quite resistant to damage until high concentrations of oxidants were used. This is the first report of direct detection by LC-MS of intact oxidized phospholipids induced in cultured cells subjected to oxidative stress.

A comprehensive forensic investigation of sensitive ecosystems in the Everglades Area is presented. Assessing the background levels of contamination in these ecosystems represents a vital resource to build up forensic evidence required to enforce future environmental crimes within the studied areas. This investigation presents the development and validation of a fractionation and isolation method for two families of herbicides commonly applied in the vicinity of the study area, including phenoxy acids like 2,4-D, MCPA, and silvex; as well as the most common triazine-based herbicides like atrazine, prometyne, simazine and related metabolites like DIA and DEA. Accelerated solvent extraction (ASE) and solid phase extraction (SPE) were used to isolate the analytes from abiotic matrices containing large amounts of organic material. Atmospheric-pressure ionization (API) with electrospray ionization in negative mode (ESP-), and Chemical Ionization in the positive mode (APCI+) were used to perform the characterization of the herbicides of interest.

We quantified pigment biomarkers by high performance liquid chromatography (HPLC) to obtain a broad taxonomic classification of microphytobenthos (MPB) (i.e. identification of dominant taxa). Three replicate sediment cores were collected at 0, 50 and 100 m along transects 5-9 in Heron Reef lagoon (n=15) (Fig. 1). Transects 1-4 could not be processed because the means to have the samples analysed by HPLC were not available at the time of field data collection. Cores were stored frozen and scrapes taken from the top of each one and placed in cryovials immersed in dry ice. Samples were sent to the laboratory (CSIRO Marine and Atmospheric Research, Hobart, Australia) where pigments were extracted with 100% acetone during fifteen hours at 4°C after vortex mixing (30 seconds) and sonication (15 minutes). Samples were then centrifuged and filtered prior to the analysis of pigment composition with a Waters - Alliance HPLC system equipped with a photo-diode array detector. Pigments were separated using a Zorbax Eclipse XDB-C8 stainless steel 150 mm x 4.6 mm ID column with 3.5 µm particle size (Agilent Technologies) and a binary gradient system with an elevated column temperature following a modified version of the Van Heukelem and Thomas (2001) method. The separated pigments were detected at 436 nm and identified against standard spectra using Waters Empower software. Standards for HPLC system calibration were obtained from Sigma (USA) and DHI (Denmark).

The research described in this thesis is concerned with the synthesis and stereoselective transformations of 4,5-dihydro-3(2H)-furanones and their 3-hydroxy derivatives. In Chapter 1, a review of synthetic routes to 3-hydroxytetrahydrofurans is presented. This incorporates the wide range of applications for these types of compounds. Preparative routes to and stereoselective transformations of the furanones investigated in this study are discussed in Chapter 2. The bulk of the work centers on stereoselective carbonyl group reductions to generate the 3-hydroxytetrahydrofuran derivatives in racemic form followed by kinetic resolution via lipase mediated esterification, resulting in enantioenriched 3-acetoxy and 3-hydroxytetrahydrofuran derivatives. In many cases, these processes proceed in a highly enantioselective manner. The influence of the lipase species and concentration of enzyme employed on the yield and stereochemical outcome of the reactions is examined in detail. Access to the complementary series of furanone and hydroxytetrahydrofuran derivatives by oxidation or reduction of the enantioenriched compounds was achieved through conventional synthetic methods. Chapter 2 also contains details of a novel synthetic route to a range of 2,3,5-trisubstituted furans from α-hydroxyenones and 4,5-dihydro-3(2H)-furanones. The mechanistic rationale for these transformations and the migratory aptitude of alkyl groups towards the formation of these furans is discussed in detail. Finally, Chapter 2 outlines the synthesis of a series of diarylcyclopentenones that were synthesised as part of our investigations. Chapter 3 contains a description of the synthetic procedures and biotransformations carried out together with key analytical and spectroscopic properties of the compounds studied and where appropriate, their analysis using chiral HPLC analysis.

Angiotensin-converting enzyme (EC3.4.15. I; ACE), isa membrane-bounddipeptidyl carboxypeptidase that mediates the cleavage of the C-terminal dipeptide His-Leu of the decapeptide angiotensin, generating the most powerful endogenous vaso-constricting angiotensin.
Some ACE inhibitors, such as Captopril, have been used as anti-hypertensive drugs. Moreover in recent years, large quantities of ACE inhibitors have been identijied and isolated from peptides derivedfrom food material such as casein, soy protein, jish protein and so on. Functional food with hypotensive effect has been developed on the basis of these works.
Typicalprocedures for screening hypotensive peptides offood origins are separationof products of peptic and tryptic digestion of proteins followed by inhibitory activitydetermination of each fraction. A method developed by Cushman has been the mostwidely used, in which ACE activity is determined by the amount of hippuric acid
generated as a product of enzymatic reaction of ACE with tripeptide of hippuryl-Lhistidyl-L-leucine. Hippuric acid is determined spectrophotometrically at 228 nm after its isolation from the reaction system by ethylacetate extraction, which not only requires alarge quantity of reagent but also results in large error.
An improved method based on Cushman ’s method is proposed in this paper. In this method, an enzymatic reaction system is based on Cushman’s method, while isolation and determination of hippuric acid is performed by medium perjormance gel chromatography on a Toyopearl HW-40s column. Due to the size exclusion nature of the column with somewhat hydrophobic properties, complete separation of four existing fractions in the reaction system is obtained within a smallfraction of the time necessary in Cushman’s method, with ideal reproducibility.

A preocupação com a poluição das águas por agrotóxicos tem aumentado, visto que aumentou o número de detecções de agrotóxicos em águas. A falta de avaliação da qualidade da água consumida pela população de áreas rurais onde não existe o abastecimento público de água potável, deve ser considerada, pois essas águas se encontram próximo a áreas de cultivo, onde há intensa aplicação de agrotóxicos. Nessas regiões, o abastecimento de água para as residências e para a irrigação é feito geralmente através das águas de poços. Neste trabalho, um método para determinação dos agrotóxicos carbofurano, clomazona, 2,4-D e tebuconazol em água subterrânea foi desenvolvido e validado. O método utilizou a Extração em Fase Sólida (SPE) e determinação por Cromatografia Líquida de Alta eficiência com Detecção por Arranjo de Diodos (HPLC-DAD) e confirmação por Cromatografia Líquida tandem Espectrometria de Massas (LC-MS/MS). Para a SPE utilizou-se cartuchos C18 de 200 mg, e eluição com 1 mL de metanol. Após a otimização dos parâmetros de extração e separação dos compostos, o método foi validado avaliando-se curva analítica, linearidade, limites de detecção e quantificação, precisão (repetitividade e precisão intermediária) e exatidão (recuperação). Todas as curvas analíticas apresentaram valores de r maiores que 0,99. Os LOQs para o método, considerando a etapa de pré-concentração de 250 vezes, foram de 0,2 µg L -1 para todos os agrotóxicos por HPLC-DAD e, por LC-MS/MS, 4,0 ng L -1 para clomazona, carbofurano e tebuconazol e de 40,0 ng L -1 para 2,4-D. As recuperações foram entre 60,3 e 107,7% para a repetitividade e entre 67,5 e 115,3% para a precisão intermediária, com RSD de 0,8 a 20,7% para todos os compostos por HPLC-DAD. Para o LC-MS/MS a precisão em termos de repetitividade, variou entre 0,97 e 20,7%, e as recuperações entre 67,0 e 108,9%. O método foi aplicado na determinação de agrotóxicos em amostras de águas subterrâneas durante um ano. Nas amostras foram detectados agrotóxicos em níveis de µg L -1 . Dentro do contexto atual da Química Analítica, de desenvolver métodos mais rápidos, que utilizem menor quantidade de solvente, de amostra e com altos fatores de enriquecimento, foi otimizado um método de extração para os agrotóxicos carbofurano, clomazona e tebuconazol utilizando a Microextração Líquido-Líquido Dispersiva (DLLME) e determinação por LC-MS/MS. Foram otimizados alguns parâmetros que influenciam no processo de extração, como: tipo e volume dos solventes dispersores e extratores, tempo de extração, força iônica e velocidade de centrifugação. Nas condições otimizadas, as recuperações para os níveis de concentração entre 0,02 e 2,0 g L -1 variaram entre 62,7 e 120,0%, com valores de RSD entre 1,9 e 9,1%. O LOQ do método foi de 0,02 µg L -1 para todos os compostos. Quando comparado com a SPE se demonstrou rápido, simples, de baixo custo, além de necessitar de menores volumes de amostra para determinação de agrotóxicos em águas. O método mostrou-se adequado à análise dos agrotóxicos em água subterrânea e todos os parâmetros de validação obtidos estão dentro dos limites sugeridos para validação de métodos cromatográficos

O aumento de consumo de cogumelos tem-se verificado em todo o mundo, não só pelo seu valor nutricional, sabor apurado e textura, mas também pelas suas propriedades medicinais. Existem vários estudos científicos que descrevem os benefícios do consumo de cogumelos, que advêm da sua riqueza em compostos bioativos, tais como micosteróis, em particular, ergosterol. Agaricus bisporus L. é o cogumelo mais consumido em todo o mundo, sendo a sua fração de esteróis constituída essencialmente por ergosterol (90%) [1], tornando a sua extração um tópico de elevado interesse já que esta molécula apresenta elevado valor comercial e inúmeras aplicações nas indústrias alimentar, farmacêutica e cosmética. Segundo a literatura, o teor de ergosterol pode variar entre 3 e 9 mg por g de cogumelo seco. Atualmente, os métodos tradicionais tais como a maceração e a extração em Soxhlet estão a ser substituídos por metodologias emergentes, nomeadamente a extração assistida por microondas, visando diminuir a quantidade de solvente utilizado e o tempo de extração e, naturalmente, aumentar o rendimento da mesma. No presente trabalho, utilizou-se A. bisporus como fonte de ergosterol, tendo-se otimizado as seguintes variáveis relevantes para a sua extração pela tecnologia de microondas (MAE): tempo (0-20 min), temperatura (60-210 ºC) e razão sólido-líquido (1-20 g/L). O solvente utilizado foi o etanol tendo-se aplicado a técnica estatística de superfície de resposta por forma a gerar modelos matemáticos que permitissem maximizar a resposta e otimizar as variáveis que afetam a extração de ergosterol. O conteúdo em ergosterol foi monitorizado por HPLC-UV. Os resultados demonstraram que a técnica MAE é promissora para a extração de ergosterol, tendo-se obtido, para as condições ótimas (20,4 min, 121,5ºC e 1,6 g/L), 569,4 mg ergosterol/100 g de massa seca, valor similar ao obtido com extração convencional por Soxhlet (671,5±0,5 mg/100 g de massa seca). Em síntese, a extração assistida por microondas demonstrou ser uma tecnologia eficiente para maximizar o rendimento de extração em ergosterol.
