To date, limited numbers of dental calculus samples have been analyzed by researchers in diverse parts of the world. The combined analyses of these have provided some general guidelines for the analysis of calculus that is non-destructive to archaeological teeth. There is still a need for a quantitative study of large numbers of calculus samples to establish protocols, assess the level of contamination, evaluate the quantity of microfossils in dental calculus, and to compare analysis results with the literature concerning the biology of calculus formation. We analyzed dental calculus from 53 teeth from four Brazilian sambaquis. Sambaquis are the shell-mounds that were established prehistorically along the Brazilian coast. The analysis of sambaqui dental calculi shows that there are relatively high concentrations of microfossils (phytoliths and starch), mineral fragments, and charcoal in dental calculus. Mineral fragments and charcoal are possibly contaminants. The largest dental calculi have the lowest concentrations of microfossils. Biologically, this is explained by individual variation in calculus formation between people. Importantly, starch is ubiquitous in dental calculus. The starch and phytoliths show that certainly Dioscorea (yam) and Araucaria angustifolia (Parana pine) were eaten by sambaqui people. Araceae (arum family), Ipomoea batatas (sweet potato) and Zea mays (maize) were probably in their diet. (C) 2009 Elsevier Ltd. All rights reserved.

Eukaryotic genome expansion/retraction caused by LTR-retrotransposon activity is dependent on the expression of full length copies to trigger efficient transposition and recombination-driven events. The Tnt1 family of retrotransposons has served as a model to evaluate the diversity among closely related elements within Solanaceae species and found that members of the family vary mainly in their U3 region of the long terminal repeats (LTRs). Recovery of a full length genomic copy of Retrosol was performed through a PCR-based approach from wild potato, Solanum oplocense. Further characterization focusing on both LTR sequences of the amplified copy allowed estimating an approximate insertion time at 2 million years ago thus supporting the occurrence of transposition cycles after genus divergence. Copy number of Tnt1-like elements in Solanum species were determined through genomic quantitative PCR whereby results sustain that Retrosol in Solanum species is a low copy number retrotransposon (1-4 copies) while Retrolyc1 has an intermediate copy number (38 copies) in S. peruvianum. Comparative analysis of retrotransposon content revealed no correlation between genome size or ploidy level and Retrosol copy number. The tetraploid cultivated potato with a cellular genome size of 1,715 Mbp harbours similar copy number per monoploid genome than other diploid Solanum species (613-884 Mbp). Conversely, S. peruvianum genome (1,125 Mbp) has a higher copy number. These results point towards a lineage specific dynamic flux regarding the history of amplification/activity of Tnt1-like elements in the genome of Solanum species.

Purple acid phosphatases (PAPs) are a group of metallohydrolases that contain a dinuclear Fe(II)M(II) center (M(II) = Fe, Mn, Zn) in the active site and are able to catalyze the hydrolysis of a variety of phosphoric acid esters. The dinuclear complex [(H(2)O)Fe(III)(mu-OH)Zn(II)(L-H)](CIO(4))(2) (2) with the ligand 2-[N-bis(2-pyridylmethyl)aminomethyl]-4-methyl-6-[N-(2-pyridylmethyl)(2-hydroxybenzyl) aminomethyl]phenol (H(2)L-H) has recently been prepared and is found to closely mimic the coordination environment of the Fe(III)Zn(II) active site found in red kidney bean PAP (Neves et al. J. Am. Chem. Soc. 2007, 129, 7486). The biomimetic shows significant catalytic activity in hydrolytic reactions. By using a variety of structural, spectroscopic, and computational techniques the electronic structure of the Fe(III) center of this biomimetic complex was determined. In the solid state the electronic ground state reflects the rhombically distorted Fe(III)N(2)O(4) octahedron with a dominant tetragonal compression align ad along the mu-OH-Fe-O(phenolate) direction. To probe the role of the Fe-O(phenolate) bond, the phenolate moiety was modified to contain electron-donating or -withdrawing groups (-CH(3), -H, -Br, -NO(2)) in the 5-position. Tie effects of the substituents on the electronic properties of the biomimetic complexes were studied with a range of experimental and computational techniques. This study establishes benchmarks against accurate crystallographic struck ral information using spectroscopic techniques that are not restricted to single crystals. Kinetic studies on the hydrolysis reaction revealed that the phosphodiesterase activity increases in the order -NO(2)<- Br <- H <- CH(3) when 2,4-bis(dinitrophenyl)phosphate (2,4-bdnpp) was used as substrate, and a linear free energy relationship is found when log(k(cat)/k(0)) is plotted against the Hammett parameter a. However, nuclease activity measurements in the cleavage of double stranded DNA showed that the complexes containing the electron-withdrawing -NO(2) and electron-donating CH3 groups are the most active while the cytotoxic activity of the biomimetics on leukemia and lung tumoral cells is highest for complexes with electron-donating groups.

This paper describes the use of a dental amalgam electrode (DAE) to evaluate the electrochemical behaviour and to develop an electroanalytical procedure for determination of diquat herbicide in natural water and potato samples. The work was based on the square wave voltammetry responses of diquat, which presented two well-defined and reversible reduction peaks, at -0.56 V (peak 1) and -1.00V (peak 2). The experimental and voltammetric parameters were optimised, and the analytical curves were constructed and compared to similar curves performed by high performance liquid chromatography coupled to ultraviolet-visible spectrophotometric detector (HPLC/UV-vis). The responses were directly proportional to diquat concentration in a large interval of concentration, and the calculated detection limits were very similar, around 10 mu g L(-1) (10 ppb) for voltammetric and chromatographic experiments. These values were lower than the maximum residue limit established for natural water by the Brazilian Environmental Agency. The recovery percentages in pure electrolyte, natural water and potato samples showed values from 70% to 130%, demonstrating that the voltammetric methodology proposed is suitable for determining any contamination by diquat in different samples, minimising the toxic residues due to the use of liquid mercury or the adsorptive process relative to use of other solid surfaces. The researches on the biological control of nematodes with nematophagous fungi has been intensified in recent years. The knowledge of the ecological conditions for the growth and sporulation of these fungi is a prerequisite for attainment of pure cultures needed to attend the demand for formulation of these organisms. With the objective to evaluate the micelial growth and sporulation of Arthrobotrys musiformis and A. oligospora in two environments (B.O.D at 25 +/- 1 degrees C and the environment of the Laboratory), 20 cultures media prepared with common materials found in the communities and industrialized media such as mycological agar, PDA and CMA were evaluated. The media were tested in Petri dishes, being the micelial growth of the fungi evaluated daily, during six days. The measured sporulation at the end of the experiment was done by estimation of the number of conidia/Petri dish. The experiment was carried out in a random design following a factorial arrangement of 20 x 2 x 2, corresponding to 20 media, two fungi and two environments, with five replicates. The variance analysis of the data evidenced significant statistical difference by the F Test, at 1% probability, among media x fungi x environment interaction. Fifty percent of the tested media provided the adequate micelial growth of A. musiformis and there was no statistical difference among them, namely: cassava meal (FM), sweet starch (PD), "corn meal agar" (CMA), oat in fine flakes (AFF), agar-water + dextrose (AA+D), mycological agar (AM), potato dextrose agar (BDA), meal of maize (FMI), flour of wheat (FT) and wheat for kibble (TK). In relation to A. oligospora, 75% of the tested media promoted the maximum growth of the fungus, which are: AFF, AM, FM, PD, CMA, AA+D, BDA, FT, TK, the water from the decoction of rice (AAZ), rice in grains (AZG), triturated rice (AZT), thread flour (FR), oats flour (FA), oats in thick flakes (AFG) and flour of maize (FU). In relation to the sporulation the media that had better role for A. musiformis, in decreasing order, were: FR, TK, AFG, BDA, FA, AFF, AM, FMI, AZT and FM, varying between 1,01 x 10(6) and 1,4 x 10(4) conidia/Petri dish. For the A. oligospora sporulation, the CMA medium provided the maximum level with an estimated average of 5,7 x 10(6) conidia/Petri dish. In the general, the best media for the micelial growth and sporulation of A. musiformis had also been the best for A. oligospora. However, some that had been the best for the A. oligospora did not had been efficient for the micelial growth or the sporulation of A. musiformis, indicating that the isolate of A. musiformis in case is more demanding than that A. oligospora one. The evidences from the study indicate that, in Jaboticabal, São Paulo state, the growth and the sporulation of these fungi do not demand special chambers. Some adaptations of an environment at the laboratory, enough to obliterate the light are sufficient.

No Estado de São Paulo, a batata-doce é cultivada em escala comercial como produto hortigranjeiro. em vista das dificuldades para o controle manual e o mecanizado das plantas infestantes da cultura, há grande interesse pelo método químico desse controle. Com essa finalidade, tanto pelo baixo custo relativo como pelas facilidades técnicas de utilização, o trifluralin desperta a atenção dos agricultores. Para avaliar a seletividade do herbicida para essa cultura, foi realizado um experimento em vasos, com doses crescentes do produto, nas seguintes concentrações: 0,00; 0,11; 0,22; 0,44; 0,88; 1,76 e 3,52 mg.kg-1 de i.a. em peso. Os resultados mostraram que, a partir da dose de 0,22 mg.kg-1 de i.a., o trifluralin interferiu negativamente e em escala exponencial sobre os parâmetros: comprimento das ramas, número de folhas, peso da massa seca da parte aérea e das raízes, avaliados aos 30 dias, revelando-se não seletivo para a batata-doce quando incorporado ao solo, em profundidades semelhantes às da zona de enraizamento das plantas.

Avaliou-se a resistência de genótipos de batata aos afídeos Myzus persicae (Sulz.), Macrosiphum euphorbiae (Thomas) e Aphis gossypii (Glover), e a influência desses genótipos sobre parasitóides, em dois ensaios em plantios de inverno (1996 e 1997), em condições de campo, em Jaboticabal, SP. Os genótipos plantados no primeiro ano foram: Achat, Baronesa, Bintje, BR-2, Contenda, Monalisa, N 140-201 e NYL 235-4, distribuídos em quatro blocos ao acaso. Dois levantamentos foram efetuados, aos 70 e 85 dias após o plantio, avaliando-se o número de pulgões/folha e pulgões parasitados, nos terços superior, médio e inferior da planta. em 1997 plantaram-se os mesmos genótipos, excetuando-se o N 140-201, com levantamentos aos 72 e 80 dias. O genótipo NYL 235-4 mostrou-se suscetível a M. euphorbiae, sob infestações médias de 2,90 (1996) e 1,19 pulgões/folha (1997); dentre as cultivares comerciais, Baronesa foi a menos atacada por M. persicae (0,12 e 0,10 pulgões/folha em 1996 e 1997), enquanto BR-2 foi bem suscetível (1,10 e 0,66 pulgões/folha). Os afídeos ocorreram em maior população nas partes média e inferior das plantas; maior número de pulgões mumificados por Aphidius ervi Hal. e Diaretiella rapae (McIntosh), com parasitismo variando de 15 a 36%, foi observado em NYL 235-4, que foi o genótipo mais infestado pelos afídeos.

Avaliou-se a preferência alimentar de adultos de Diabrotica speciosa (Germar), pelos genótipos de batata Achat, Baronesa, Monalisa, N 140-201 e NYL 235-4. Foram realizados ensaios com e sem chance de escolha, em laboratórios e em casa de vegetação, na FCAV/UNESP, Jaboticabal, SP. em dois testes utilizaram-se discos de folíolos provenientes de plantios em vaso (plantas com 40 dias); em outros seis semelhantes (exceção ao genótipo N 140-201) foram utilizados discos de folíolos provenientes de plantio de campo (plantas com 30, 50 e 70 dias). Nas placas de Petri (repetições) foram liberados dois adultos/disco. Dois outros ensaios foram efetuados em gaiolas (sem chance de escolha), no interior de casa de vegetação, com plantas com 15 e 45 dias de idade, liberando-se três e 10 adultos/planta respectivamente, avaliando-se o consumo de área foliar quatro e cinco dias após. Nos testes em laboratório com material de vaso, o genótipo NYL 235-4 (com e sem chance) foi o menos consumido (1,08 a 1,12 mg de peso seco). Com material de campo (com e sem chance), o peso de matéria fresca consumida pela praga foi menor em NYL 235-4 (médias de 26,2 e 66,1 mg) e Baronesa (30,2 e 57,9 mg). Nos ensaios em gaiolas (sem chance) também verificou-se menor consumo em NYL 235-4 (3,35 cm² e 16,70 cm²/planta). Esses resultados revelam que o genótipo NYL 235-4 apresenta resistência do tipo não-preferência para alimentação em relação a D. speciosa.

Estudou-se o crescimento micelial de dez isolados de Diaporthe citri, utilizando-se seis meios de cultura (aveia-ágar, maltose-peptona-ágar, batata-dextrose-ágar, folha de laranja-dextrose-ágar, folha de limão-dextrose-ágar, milho-ágar) à temperatura de 22 ± 2 °C e fotoperíodo de 12 h claro/12 h escuro. O cultivo em meio de batata-dextrose-ágar (BDA) foi conduzido em cinco temperaturas diferentes (10, 15, 20, 25 e 30 °C). Três diferentes regimes de luminosidade (12 h claro/12 h escuro, claro contínuo, escuro contínuo) foram utilizados para verificar o crescimento do fungo. Foram observadas variações na produção de picnídios e de massa micelial nos diferentes meios de cultura, temperaturas e regimes de luminosidade testados, sendo que, para a maioria dos isolados, o meio de cultura de aveia-ágar, a faixa de 20 a 25 °C e o regime de claro contínuo induziram maior crescimento micelial. A produção de picnídios foi maior para o regime de luz contínua. O teste de patogenicidade foi feito por inoculação de discos de micélio de 5 mm de diâmetro em ferimentos em ramos e caule de limão 'Feminelo' (Citrus limon) enxertado em citrumelo 'Swingle'(Poncirus trifoliolata x Citrus paradisi) e plantas de limão 'Cravo' (C. limonia) enxertados com laranja 'Valência' (C. sinensis). Após sete dias, houve o aparecimento de exsudação de goma nas plantas inoculadas com os isolados, mas não na testemunha. Todos os isolados mostraram-se patogênicos, sendo os isolados PC2 e PC5, os que causaram comprimento de lesão maior nas plantas.

The aerial spraying of plant ripeners on sugar cane (Saccharum officinarum L.) crops causes often the contamination of neighboring areas, which subsidizes formal complaints from the neighbors. These contaminations are due to spraying taking place during inadequate environmental conditions or from technical mistakes during the application. One of the most important causes of this contamination is the susceptibility of the species being cultivated surrounding sugar cane. In order to evaluate the effects of sugar cane plant ripeners trinexapac-ethyl and sulfometuron-methyl on peanuts, cotton, potato, coffee, citrus, beans, sunflower, cassava, rubber, soybean, and grapes, eleven experiments - one for each species - were carried out from May 2009 to Jan. 2010. The field experiment was set according to a completely random design with five treatments and four replications. Just before or during flowering, a single treatment of trinexapac-ethyl at 100 or 200 g ha-1 and sulfometuron-methyl at 7.5 or 15 g ha-1 was applied to plants. A control treatment (plants not treated) for each species was part of each experiment. Trinexapac, at the doses of 100 and 200 g ha-1, showed selectivity to peanuts, cotton, potato, coffee, citrus, sunflower, cassava, rubber, soybean, and grape. At the lowest dose (100 g ha-1), it was selective for bean. Sulfometuron, at the dose of 7.5 g ha-1, was selective for peanuts and, at the two studied doses (7.5 and 15 g ha-1), it was selective for coffee, citrus, cassava, and rubber.

Foram estudadas 40 cultivares de batata-doce Ipomoea batatas (L.) Lamarck, objetivando avaliar a resistência ao ataque da broca Euscepes postfasciatus Fairmaire. Numa primeira fase da pesquisa foi medida a atratividade, a não-preferência para alimentação e oviposição em ramas e raízes. Dos resultados foram escolhidas oito cultivares resistentes e duas suscetíveis. Numa segunda fase utilizaram-se raízes das cultivares escolhidas para estudar a resistência a E. postfasciatus. Ramas e raízes foram oferecidas a adultos da broca em arenas com livre chance de escolha, com seis repetições. As avaliações foram feitas aos 5, 10, 30, 60 min., 3h e 24h após a liberação. O melhor tamanho de rama, entre os testados, a ser utilizado foi o de 35 cm. Os adultos da broca ao serem liberados nas arenas, dirigiram-se rapidamente para o material e atacaram as mais atrativas, prioritariamente nos ramos mais grossos ou na inserção das folhas. As cultivares com ramas menos atrativas foram: Rocha da Paz, Mineira, Lagartixa e Bom Nome, sendo que os tubérculos das cultivares Lagartixa e Bom Nome também foram menos danificados e juntamente com Caboatã, não preferidas para oviposição. As raízes e ramas das cultivares Granfina e UFRPE foram as mais atrativas e as mais danificadas portanto devem ser evitadas, enquanto que as cultivares Lagartixa e Bom Nome devem ser recomendadas pelo seu melhore desempenho.
