Abstract Fish sauce belongs to the most important condiments in Southeast Asian cuisine. It is a clear, amber to reddish liquid with an intensive smell. Fish sauce is used instead of salt for nearly each meal. Asian fish sauce is made from anchovies and other small fish. For the traditional process whole fresh fish are mixed with salt in the ratio 1:1 to 6:1 in wooden, clay or concrete tanks at tropical temperatures for 6 to 18 months. The liquefaction of the fish tissue is due to the action of endogenous enzymes in fish and exogenous enzymes from bacteria. During the fermentation amino acids, peptides and a lot of other substances are built, which are responsible for the characteristic aroma and flavour of these sauces. You can buy pure fish sauce, diluted fish sauce and fish sauce made from other types of animals like mussels, prawns and squids. In single Asian countries there are different national standards for the quality of fish sauces. In order to get a general idea of these products we have bought 16 fish and two oyster sauces from the retail trade in Hamburg and analyzed them with physical, chemical, sensory and microbiological methods. Kurzfassung Fischsauce gehört zu den wichtigsten Würzsaucen in der südostasiatischen Küche. Es ist eine klare, bernsteinfarbene bis rötlichbraune, sehr intensiv riechende Flüssigkeit. Sie wird anstelle von Salz verwendet und daher fast zu jedem Essen gereicht. Zur Herstellung von Fischsaucen werden hauptsächlich Anchovis und ähnliche kleine Fische verwendet. Bei der traditionellen Herstellung werden die ganzen Fische mit Meersalz in einem Holzfass, Tongefäß oder Betontank im Verhältnis 1:1 bis 6:1 gemischt. Während der anschließenden 6 – 18 Monate dauernden Lagerung bei tropischen Temperaturen bauen sich die Gewebeproteine durch fischeigene Enzyme und Mikroorganismen ab. Bei diesem mehrmonatigen Fermentationsprozess entstehen die für den Geschmack wichtigen Aminosäuren, Peptide und Aromastoffe. Es gibt neben reiner Fischsauce, auch verdünnte Fischsauce und Fischsaucen aus anderen Tieren wie Muscheln, Garnelen und Tintenfische. In den einzelnen asiatischen Ländern gibt es unterschiedliche nationale Qualitätsstandards. Um diese Produktgruppe näher kennen zu lernen, haben wir 16 Fisch- und 2 Austernsaucen aus dem Einzelhandel (Hamburg) mit physikalischen, chemischen, sensorischen und mikrobiologischen Verfahren untersucht.

ENGLISH: Strong coastal upwelling occurs in the Gulf of Panama regularly each year during the season, from about January through April, when strong northerly winds are blowing offshore. Because of the evident importance of upwelling to the ecology of the Gulf of Panama, we commenced in the fall of 1954 a study of various physical, chemical, and biological phenomena associated therewith. Observations were taken at bi-weekly intervals at a fixed location in the Gulf (approximately 10 miles SE of Taboga Island) to supplement the serial observations of sea level, sea temperature, and winds that have been gathered for many years by the Panama Canal Company. SPANISH: Cado año, en la estación de enero a abril, cuando los vientos del norte soplan vigorosamente frente a la costa, ocurre en el Golfo de Panamá un fuerte afloramiento costanero. Se cree que este afloramiento periódico en el Golfo de Panamá es responsable de la alta productividad biológica que sostiene considerables cantidades de organismos de importancia comercial. Esta región, por ejemplo, es una fuente importante de la especie Cetengraulis mysticetus) pez de carnada para el atún, (Alverson y Shimada, 1957) y mantiene una considerable pesca de camarones llamados langostinos (Burkenroad, Obarrio y Mendoza,1955). ENGLISH: The Nankai Regional Fisheries Research Laboratory of Kochi, Japan conducted a long-line fishery exploration and hydrographic survey in the eastern Pacific Ocean aboard the R/V Shoyo Maru during October 1963- March 1964. An invitation to the Inter-American Tropical Tuna Commission to participate in the cruise gave its investigators the opportunity to make surface biological observations and to preserve water samples for subsequent analyses of nutrients. The result of this survey is a comprehensive body of physical, chemical and biological data covering a large portion of the eastern half of the Pacific Ocean. SPANISH: El Nankai Regional Fisheries Research Laboratory de Rochi, Japón, llevó a cabo una exploración pesquera con palangre y un reconocimiento hidrográfico en el Océano Pacífico oriental, a bordo del barco de investigación Shoyo Maru, desde octubre de 1963 hasta marzo de 1964. Una invitación dirigida a la Comisión Interamericana del Atún Tropical para participar en el crucero, confirió a sus investigadores la oportunidad de hacer observaciones biológicas superficiales y conservar muestras de agua para el subsiguiente análisis de los nutrientes. El resultado de este reconocimiento es un conjunto de datos físicos, químicos y biológicos que abarcan una gran parte del sector medio oriental del Océano Pacífico. (PDF contains 153 pages.)

Mitochondria dynamics is crucial to many biological processes such as mitochondria fusion and fission, which is highly correlated to the mechanics of single mitochondria. However, the mechanobiological coupling of mitochondria has been poorly understood. Here membrane deformability and membrane tension of individual mitochondria isolated from MtDsRed labeled human embryonic T-Rex-293 kidney cells were measured using a micropipette aspiration assay. The results demonstrated that membrane deformation of isolated mitochondria exhibited an elastic transition phase followed by an equilibrium phase, and mitochondrial membrane tension was proportional to the area compressibility. It was also indicated that mitochondrial membrane deformability was significantly affected by physical chemical factors such as osmotic pressure or pH value, and was further correlated to mitochondrial functionality in different respiratory states and Ca2+ regulation. These findings provide a new insight into understanding the mechanical regulation of mitochondrial physiology.

The study of limnology is important to understand ecosystem dynamics and the ecological basis for fish production in the Lake Victoria which is important for fisheries resources use, planning and management. Physical, chemical and biological parameters are important and known to influence fish population production. Energy fixed by primary producers, e.g. algae, is transfered to higher trophic levels, e.g fish. Factors which influence the dynamics of phytoplankton and zooplankton population, e.g nutrient availability and uptake, growth rate, species composition and biomass, ultimately affect fish production. The commercial fisheries of Lake Victoria consists mainly of piscivorous Lates niloticus (L>), algivorous Oreochromis niloticus (L.) and zooplanktivorous Rastrineobola argentea (Pellegrin)

Iterative in situ click chemistry (IISCC) is a robust general technology for development of high throughput, inexpensive protein detection agents. In IISCC, the target protein acts as a template and catalyst, and assembles its own ligand from modular blocks of peptides. This process of ligand discovery is iterated to add peptide arms to develop a multivalent ligand with increased affinity and selectivity. The peptide based protein capture agents (PCC) should ideally have the same degree of selectivity and specificity as a monoclonal antibody, along with improved chemical stability. We had previously reported developing a PCC agent against bovine carbonic anhydrase II (bCAII) that could replace a polyclonal antibody. To further enhance the affinity or specificity of the PCC agent, I explore branching the peptide arms to develop branched PCC agents against bCAII. The developed branched capture agents have two to three fold higher affinities for the target protein. In the second part of my thesis, I describe the epitope targeting strategy, a strategy for directing the development of a peptide ligand against specific region or fragment of the protein. The strategy is successfully demonstrated by developing PCC agents with low nanomolar binding affinities that target the C-terminal hydrophobic motif of Akt2 kinase. One of the developed triligands inhibits the kinase activity of Akt. This suggests that, if targeted against the right epitope, the PCC agents can also influence the functional properties of the protein. The exquisite control of the epitope targeting strategy is further demonstrated by developing a cyclic ligand against Akt2. The cyclic ligand acts as an inhibitor by itself, without any iteration of the ligand discovery process. The epitope targeting strategy is a cornerstone of the IISCC technology and opens up new opportunities, leading to the development of protein detection agents and of modulators of protein functions.

Computational protein design (CPD) is a burgeoning field that uses a physical-chemical or knowledge-based scoring function to create protein variants with new or improved properties. This exciting approach has recently been used to generate proteins with entirely new functions, ones that are not observed in naturally occurring proteins. For example, several enzymes were designed to catalyze reactions that are not in the repertoire of any known natural enzyme. In these designs, novel catalytic activity was built de novo (from scratch) into a previously inert protein scaffold. In addition to de novo enzyme design, the computational design of protein-protein interactions can also be used to create novel functionality, such as neutralization of influenza. Our goal here was to design a protein that can self-assemble with DNA into nanowires. We used computational tools to homodimerize a transcription factor that binds a specific sequence of double-stranded DNA. We arranged the protein-protein and protein-DNA binding sites so that the self-assembly could occur in a linear fashion to generate nanowires. Upon mixing our designed protein homodimer with the double-stranded DNA, the molecules immediately self-assembled into nanowires. This nanowire topology was confirmed using atomic force microscopy. Co-crystal structure showed that the nanowire is assembled via the desired interactions. To the best of our knowledge, this is the first example of a protein-DNA self-assembly that does not rely on covalent interactions. We anticipate that this new material will stimulate further interest in the development of advanced biomaterials.

Very little research has been carried out on detrital energetics and pathways in lotic ecosystems. Most investigations have concentrated on the degradation of allochthonous plant litter by fungi, with a glance at heterotrophic bacteria associated with decaying litter. In this short review, the author describes what is known of the detrition of plant litter in lotic waters, which results from the degradative activities of colonising saprophytic fungi and bacteria, and goes on to relate this process to those invertebrates that consume coarse and/or fine particulate detritus, or dissolved organic matter that aggregates into colloidal exopolymer particles. It is clear that many of the key processes involved in the relationships between the physical, chemical, biotic and biochemical elements present in running waters are very complex and poorly understood. Those few aspects for which there are reliable models with predictive power have resulted from data collections made over periods of 20 years or more. Comprehensive research of single catchments would provide a fine opportunity to collect data over a long period.

Soft hierarchical materials often present unique functional properties that are sensitive to the geometry and organization of their micro- and nano-structural features across different lengthscales. Carbon Nanotube (CNT) foams are hierarchical materials with fibrous morphology that are known for their remarkable physical, chemical and electrical properties. Their complex microstructure has led them to exhibit intriguing mechanical responses at different length-scales and in different loading regimes. Even though these materials have been studied for mechanical behavior over the past few years, their response at high-rate finite deformations and the influence of their microstructure on bulk mechanical behavior and energy dissipative characteristics remain elusive.

In this dissertation, we study the response of aligned CNT foams at the high strain-rate regime of 102 - 104 s-1. We investigate their bulk dynamic response and the fundamental deformation mechanisms at different lengthscales, and correlate them to the microstructural characteristics of the foams. We develop an experimental platform, with which to study the mechanics of CNT foams in high-rate deformations, that includes direct measurements of the strain and transmitted forces, and allows for a full field visualization of the sample’s deformation through high-speed microscopy.

We synthesize various CNT foams (e.g., vertically aligned CNT (VACNT) foams, helical CNT foams, micro-architectured VACNT foams and VACNT foams with microscale heterogeneities) and show that the bulk functional properties of these materials are highly tunable either by tailoring their microstructure during synthesis or by designing micro-architectures that exploit the principles of structural mechanics. We also develop numerical models to describe the bulk dynamic response using multiscale mass-spring models and identify the mechanical properties at length scales that are smaller than the sample height.

The ability to control the geometry of microstructural features, and their local interactions, allows the creation of novel hierarchical materials with desired functional properties. The fundamental understanding provided by this work on the key structure-function relations that govern the bulk response of CNT foams can be extended to other fibrous, soft and hierarchical materials. The findings can be used to design materials with tailored properties for different engineering applications, like vibration damping, impact mitigation and packaging.

It is often difficult to define ‘water quality’ with any degree of precision. One approach is that suggested by Battarbee (1997) and is based on the extent to which individual lakes have changed compared with their natural ‘baseline’ status. Defining the base-line status of artificial lakes and reservoirs however, is, very difficult. In ecological terms, the definition of quality must include some consideration of their functional characteristics and the extent to which these characteristics are self-sustaining. The challenge of managing lakes in a sustainable way is particularly acute in semi-arid, Mediterranean countries. Here the quality of the water is strongly influenced by the unpredictability of the rainfall as well as year-to-year variations in the seasonal averages. Wise management requires profound knowledge of how these systems function. Thus a holistic approach must be adopted and the factors influencing the seasonal dynamics of the lakes quantified over a range of spatial and temporal scales. In this article, the authors describe some of the ways in which both long-term and short-term changes in the weather have influenced the seasonal and spatial dynamics of phytoplankton in El Gergal, a water supply reservoir situated in the south of Spain. The quality of the water stored in this reservoir is typically very good but surface blooms of algae commonly appear during warm, calm periods when the water level is low. El Gergal reservoir is managed by the Empresa Municipal de Abastecimiento y Saneamiento (EMASESA) and supplies water for domestic, commercial and industrial use to an area which includes the city of Seville and twelve of its surrounding towns (ca. 1.3 million inhabitants). El Gergal is the last of two reservoirs in a chain of four situated in the Rivera de Huelva basin, a tributary of the Guadalquivir river. It was commissioned by EMASESA in 1979 and since then the company has monitored its main limnological parameters on, at least, a monthly basis and used this information to improve the management of the reservoir. As a consequence of these intensive studies the physical, chemical and biological information acquired during this period makes the El Gergal database one of the most complete in Spain. In this article the authors focus on three ‘weather-related’ effects that have had a significant impact on the composition and distribution of phytoplankton in El Gergal: (i) the changes associated with severe droughts; (ii) the spatial variations produced by short-term changes in the weather; (iii) the impact of water transfers on the seasonal dynamics of the dinoflagellate Ceratium.

The need for sustainable energy production motivates the study of photovoltaic materials, which convert energy from sunlight directly into electricity. This work has focused on the development of Cu2O as an earth-abundant solar absorber due to the abundance of its constituent elements in the earth's crust, its suitable band gap, and its potential for low cost processing. Crystalline wafers of Cu2O with minority carrier diffusion lengths on the order of microns can be manufactured in a uniquely simple fashion — directly from copper foils by thermal oxidation. Furthermore, Cu2O has an optical band gap of 1.9 eV, which gives it a detailed balance energy conversion efficiency of 24.7% and the possibility for an independently connected Si/Cu2O dual junction with a detailed balance efficiency of 44.3%.

However, the highest energy conversion efficiency achieved in a photovoltaic device with a Cu2O absorber layer is currently only 5.38% despite the favorable optical and electronic properties listed above. There are several challenges to making a Cu2O photovoltaic device, including an inability to dope the material, its relatively low chemical stability compared to other oxides, and a lack of suitable heterojunction partners due to an unusually small electron affinity. We have addressed the low chemical stability, namely the fact that Cu2O is an especially reactive oxide due to its low enthalpy of formation (ΔHf0 = -168.7 kJ/mol), by developing a novel surface preparation technique. We have addressed the lack of suitable heterojunction partners by investigating the heterojunction band alignment of several Zn-VI materials with Cu2O. Finally, We have addressed the typically high series resistance of Cu2O wafers by developing methods to make very thin, bulk Cu2O, including devices on Cu2O wafers as thin as 20 microns. Using these methods we have been able to achieve photovoltages over 1 V, and have demonstrated the potential of a new heterojunction material, Zn(O,S).

A possibilidade de utilização de várias matérias graxas na produção do biodiesel nacional é um fator diferencial em relação aos outros países e requer um conhecimento mais profundo das particularidades do biodiesel obtido a partir de matérias-primas tão distintas. Neste contexto, se insere a presente pesquisa, em que se buscou estabelecer correlações entre algumas propriedades do biodiesel e a fonte oleaginosa que lhe deu origem. O biodiesel foi obtido a partir da reação de transesterificação etílica de dez diferentes matérias-primas graxas (óleos refinados de soja, arroz, canola, girassol, milho, oliva, cyclus (mistura contendo óleos de canola, milho e girassol); óleos brutos de mamona e murumuru, além de resíduo de fritura), utilizando etanolato de potássio como agente catalítico, em um reator com sistema de refluxo a 70C durante uma hora. Após purificação, foram determinadas algumas propriedades do biodiesel (massa específica, viscosidade, índice de acidez e índice de iodo) e da matéria-prima (massa específica, viscosidade, índice de acidez, índice de iodo e composição) que lhe deu origem. Os resultados obtidos nas caracterizações geraram gráficos de correlações entre os diversos parâmetros e os dados foram analisados estatisticamente pelo método de correlações canônicas. Para todas as amostras de biodiesel, os valores obtidos nas caracterizações foram compatíveis com as especificações estabelecidas pela ANP para o produto, com exceção do biodiesel de murumuru (índice de acidez elevado e viscosidade baixa) e do biodiesel de mamona (viscosidade e massa específica elevadas). Os dados estatísticos demonstram altas correlações entre o biodiesel (massa específica (87,4%), viscosidade (98,5%) e índice de acidez (82,8%)) e as matérias-primas (massa específica (92,1%) e viscosidade (99,3%)). Além disso, existe uma grande correlação entre o índice de iodo (84,5%) do biodiesel e o índice de iodo (77,2%) e a massa molar dos ésteres na faixa de C16-C18 (MMTG) presentes na matéria-prima (89,1%). From research carried, out on a section of the Levriere, concretions (granules, nodules, which were sometimes joined together) partly covering the river ”bottom” were observed. The authors propose to make besides a petrographic examination of the calcareous precipitations and to see if their origin is connected to a biological activity, or if it is purely a case of a physical-chemical precipitation. The hydrological background of the Levriere, a small river of the Normandy Vexin, is given and conditions of the formation of the concretions studied.

To be able to carry out physical, chemical and biological investigations on a lake, one needs a thorough knowledge of the volume of water and the shape of the lake basin. Little is known about the about the morphology of the lakes in Schleswig-Holstein and its ecological consequences. For this research a 30 KHz echo sounder with sediment transceiver was used to carry out profile determinations (echo soundings). This apparatus continuously records on paper the different reflexions and absorptions of the sediment and water body in the corresponding depth. By this, acoustically noticeable layers and different densities in the sediment and 'scattering layers' in the water body due to physical, chemical and biological reasons (e.g. plans and single fish) are made visible. Result are summarised here regarding the echo soundings in Blunker See.
