INTRODUCTION: Schizophrenia is a chronic mental disorder associated with impairment in social functioning. The most widely used scale to measure social functioning is the GAF (Global Assessment of Functioning), but it has the disadvantage of measuring at the same time symptoms and functioning, as described in its anchors. OBJECTIVES:Translation and cultural adaptation of the PSP, proposing a final version in Portuguese for use in Brazil. METHODS: We performed five steps: 1) translation; 2) back translation; 3) formal assessment of semantic equivalence; 4) debriefing; 5) analysis by experts. Interrater reliability (Intraclass correlation, ICC) between two raters was also measured. RESULTS: The final version was applied by two independent investigators in 18 adults with schizophrenia (DSM-IV-TR). The interrater reliability (ICC) was 0.812 (p < 0.001). CONCLUSION: The translation and adaptation of the PSP had an adequate level of semantic equivalence between the Portuguese version and the original English version. There were no difficulties related to understanding the content expressed in the translated texts and terms. Its application was easy and it showed a good interrater reliability. The PSP is a valid instrument for the measurement of personal and social functioning in schizophrenia.

A Política Nacional do Meio Ambiente tem como objetivo a conciliação do desenvolvimento econômico e social com a qualidade ambiental, e dispõe de vários instrumentos que procuram contemplara capacidade de suporte do meio. Assim, a discussão sobre a localização das atividades é uma etapa importante para a avaliação da viabilidade ambiental de um determinado empreendimento, notadamente em processos de licenciamento ambiental. O trabalho apresenta uma abordagem metodológica para integração de critérios técnicos, ambientais e sociais em estudos de alternativas de localização de aterros sanitários, com especial atenção à interação entre os aspectos ambientais e os elementos fundamentais de um projeto (a saber, requisitos locacionais e concepção tecnológica), aplicado em um estudo de caso em São Carlos (SP). Como resultados principais, destacam-se o maior foco nos impactos significativos e a possibilidade de envolvimento de segmentos importantes da sociedade previamente à elaboração dos estudos ambientais.

Every seismic event produces seismic waves which travel throughout the Earth. Seismology is the science of interpreting measurements to derive information about the structure of the Earth. Seismic tomography is the most powerful tool for determination of 3D structure of deep Earth's interiors. Tomographic models obtained at the global and regional scales are an underlying tool for determination of geodynamical state of the Earth, showing evident correlation with other geophysical and geological characteristics. The global tomographic images of the Earth can be written as a linear combinations of basis functions from a specifically chosen set, defining the model parameterization. A number of different parameterizations are commonly seen in literature: seismic velocities in the Earth have been expressed, for example, as combinations of spherical harmonics or by means of the simpler characteristic functions of discrete cells. With this work we are interested to focus our attention on this aspect, evaluating a new type of parameterization, performed by means of wavelet functions. It is known from the classical Fourier theory that a signal can be expressed as the sum of a, possibly infinite, series of sines and cosines. This sum is often referred as a Fourier expansion. The big disadvantage of a Fourier expansion is that it has only frequency resolution and no time resolution. The Wavelet Analysis (or Wavelet Transform) is probably the most recent solution to overcome the shortcomings of Fourier analysis. The fundamental idea behind this innovative analysis is to study signal according to scale. Wavelets, in fact, are mathematical functions that cut up data into different frequency components, and then study each component with resolution matched to its scale, so they are especially useful in the analysis of non stationary process that contains multi-scale features, discontinuities and sharp strike. Wavelets are essentially used in two ways when they are applied in geophysical process or signals studies: 1) as a basis for representation or characterization of process; 2) as an integration kernel for analysis to extract information about the process. These two types of applications of wavelets in geophysical field, are object of study of this work. At the beginning we use the wavelets as basis to represent and resolve the Tomographic Inverse Problem. After a briefly introduction to seismic tomography theory, we assess the power of wavelet analysis in the representation of two different type of synthetic models; then we apply it to real data, obtaining surface wave phase velocity maps and evaluating its abilities by means of comparison with an other type of parametrization (i.e., block parametrization). For the second type of wavelet application we analyze the ability of Continuous Wavelet Transform in the spectral analysis, starting again with some synthetic tests to evaluate its sensibility and capability and then apply the same analysis to real data to obtain Local Correlation Maps between different model at same depth or between different profiles of the same model.

Sustainable computer systems require some flexibility to adapt to environmental unpredictable changes. A solution lies in autonomous software agents which can adapt autonomously to their environments. Though autonomy allows agents to decide which behavior to adopt, a disadvantage is a lack of control, and as a side effect even untrustworthiness: we want to keep some control over such autonomous agents. How to control autonomous agents while respecting their autonomy? A solution is to regulate agents’ behavior by norms. The normative paradigm makes it possible to control autonomous agents while respecting their autonomy, limiting untrustworthiness and augmenting system compliance. It can also facilitate the design of the system, for example, by regulating the coordination among agents. However, an autonomous agent will follow norms or violate them in some conditions. What are the conditions in which a norm is binding upon an agent? While autonomy is regarded as the driving force behind the normative paradigm, cognitive agents provide a basis for modeling the bindingness of norms. In order to cope with the complexity of the modeling of cognitive agents and normative bindingness, we adopt an intentional stance. Since agents are embedded into a dynamic environment, things may not pass at the same instant. Accordingly, our cognitive model is extended to account for some temporal aspects. Special attention is given to the temporal peculiarities of the legal domain such as, among others, the time in force and the time in efficacy of provisions. Some types of normative modifications are also discussed in the framework. It is noteworthy that our temporal account of legal reasoning is integrated to our commonsense temporal account of cognition. As our intention is to build sustainable reasoning systems running unpredictable environment, we adopt a declarative representation of knowledge. A declarative representation of norms will make it easier to update their system representation, thus facilitating system maintenance; and to improve system transparency, thus easing system governance. Since agents are bounded and are embedded into unpredictable environments, and since conflicts may appear amongst mental states and norms, agent reasoning has to be defeasible, i.e. new pieces of information can invalidate formerly derivable conclusions. In this dissertation, our model is formalized into a non-monotonic logic, namely into a temporal modal defeasible logic, in order to account for the interactions between normative systems and software cognitive agents.

It is well known that the best grape quality can occur only through the achievement of optimal source/sink ratio. Vine balance is in fact a key parameter in controlling berry sugar, acidity and secondary metabolites content (Howell, 2001; Vanden Heuvel et al., 2004). Despite yield reduction and quality improvement are not always strictly related, cluster thinning is considered a technique which could lead to improvement in grape sugar and anthocyanin composition (Dokoozlian and Hirschfelt, 1995; Guidoni et al., 2002). Among several microclimatic variables which may impact grape composition, the effect of cluster light exposure and temperature, which probably act in synergistic and complex way, has been widely explored showing positive even sometimes contradictory results (Spayd et al., 2001; Tarara et al., 2008). Pre-bloom and véraison defoliation are very efficient techniques in inducing cluster microclimatic modification. Furthermore pre-bloom defoliation inducing a lower berry set percentage On these basis the aim of the first experiment of the thesis was to verify in cv Sangiovese the effects on ripening and berry composition of management techniques which may increase source/sink ratio and /or promote light incidence on berries throughout grape ripening. An integrated agronomic, biochemical and microarray approach, aims to understand which mechanisms are involved in berry composition and may be conditioned in the berries during ripening in vines submitted to three treatments. In particular the treatments compared were: a) cluster thinning (increasing in source/sink ratio) b) leaf removal at véraison (increasing cluster light exposure) c) pre-bloom defoliation (increasing source sink ratio and cluster light exposure). Vine response to leaf removal at véraison was further evaluated in the second experiment on three different varieties (Cabernet Sauvignon, Nero d’Avola, Raboso Piave) chosen for their different genetic traits in terms of anthocyanin amount and composition. The integrated agronomic, biochemical and microarray approach, employed in order to understand those mechanisms involved in berry composition of Sangiovese vines submitted to management techniques which may increase source/sink ratio and induce microclimatic changes, bring to interesting results. This research confirmed the main role of source/sink ratio in conditioning sugars metabolism and revealed also that carbohydrates availability is a crucial issue in triggering anthocyanin biosynthesis. More complex is the situation of pre-bloom defoliation, where source/sink and cluster light increase effects are associated to determine final berry composition. It results that the application of pre-bloom defoliation may be risky, as too much dependent on seasonal conditions (rain and temperature) and physiological vine response (leaf area recovery, photosynthetic compensation, laterals regrowth). Early induced stress conditions could bring cluster at véraison in disadvantage to trigger optimal berry ripening processes compared to untreated vines. This conditions could be maintained until harvest, if no previously described physiological recovery occurs. Certainly, light exposure increase linked to defoliation treatments, showed a positive and solid effect on flavonol biosynthesis, as in our conditions temperature was not so different among treatments. Except the last aspects, that could be confirmed also for véraison defoliation, microclimatic changes by themselves seemed not able to induce any modification in berry composition. Further studies are necessary to understand if the peculiar anthocyanic and flavonols composition detected in véraison defoliation could play important role in both color intensity and stability of wines.

Amalgamersatz:Neue Wege zur Herstellung von Dentalkompositen mit geringem Polymerisationsschrumpf auf (Meth-)Acrylat-Basis Aufgrund der ästhetischen und gesundheitlichen Bedenken wird seit Jahrzehnten nach einer Alternative für Amalgam als Zahnfüllmaterial gesucht. Der größte Nachteil von organischen Monomeren liegt in der Volumenkontraktion während der Aushärtung, welche sich negativ auf die Materialeigenschaften auswirkt. Aus diesem Grund war das Hauptziel dieser Arbeit, eine Minimierung des Schrumpfes bei der radikalischen Polymerisation zu erreichen. Dazu wurden verschiedene, zum Teil neue, (Meth-)Acrylate synthetisiert und auf ihre Einsetzbarkeit als Bestandteil von Dentalkompositen geprüft.Um die Volumenkontraktion während der Polymerisation zu minimieren, wurde die Beweglichkeit der polymerisierbaren Gruppe eingeschränkt. Im ersten Teil der Arbeit wurden dazu flüssigkristalline Substanzen eingesetzt. Durch Mischen von flüssigkristallinen Diacrylaten konnte eine Mesophase im gewünschten Temperaturintervall von 25 bis 35 °C erhalten werden. Der Einsatz dieser Flüssigkristalle zeigte einen positiven Einfluss auf den Polymerisationsschrumpf. Zudem wurden neue Monomere synthetisiert, deren Methacrylgruppe in direkter Nachbarschaft zum Mesogen angebunden wurde, um die Stabilität der erhaltenen Polymere zu erhöhen.Im zweiten Teil der Arbeit wurde die Beweglichkeit der polymerisierbaren Gruppe durch eine Fixierung an einem starren Kern reduziert. Als Grundkörper wurden Polyphenole, enzymatisch polymerisierte Phenole und ߖCyclodextrin verwendet. Bei den modifizierten Polyphenolen auf Basis von Gallussäure und 3,5-Dihydroxybenzoesäure konnte eine leichte Reduzierung des Polymerisationsschrumpfes erreicht werden. Mit HRP (Horseradish Peroxidase) katalysierten enzymatisch polymerisierten Phenole konnte dagegen nicht photochemisch vernetzt werden, da diese Oligomere in Lösung gefärbt vorlagen. Zudem zeigten die freien, phenolischen Hydroxygruppen eine sehr geringe Reaktivität. Die besten Ergebnisse wurden mit modifizierten ߖCyclodextrinen als Komponente einer Komposite erreicht. Dabei wurde in einem Fall sogar eine leichte Volumenexpansion während der Polymerisation erzielt.

Summary During the infection of Lepidoptera larvae with baculoviruses the horizontal escape of Tc1-like transposons, termed TCl4.7 and TCp3.2, from the genome of the host Cryptophlebia leucotreta and Cydia pomonella into the genome of Cydia pomonella granulovirus was observed. In this study we addressed the question whether the transposon harboring viruses had a replication advantage over the wild-type and became dominant in the virus population or whether the activity of the host transposable elements is stimulated by virus infection. Biological characterization studies demonstrated that the transposon containing viruses killed C. pomonella larvae slower than CpGV-M. In co-infection experiments of C. pomonella larvae using a mixture of CpGV-M and mutant viruses as inoculum, it was shown that the transposon carrying mutants had a significant selection disadvantage compared to CpGV-M. Transcription levels of the transposase gene of TCp3.2 were investigated in virus infected and uninfected larvae. These experiments demonstrated that a higher level of transposase transcription was detectable in CpGV-M infected than in mock infected control larvae. This observation gave strong evidence that CpGV-M infection might trigger the activity of transposon TCp3.2 within the genome of Cydia pomonella. Our results suggest that the horizontal transfer of insect host transposons into baculovirus genomes might be induced by virus infection.

The separator membrane in batteries and fuel cells is of crucial importance for the function of these devices. In lithium ion batteries the separator membrane as well as the polymer matrix of the electrodes consists of polymer electrolytes which are lithium ion conductors. To overcome the disadvantage of currently used polymer electrolytes which are highly swollen with liquids and thus mechanically and electrochemically unstable, the goal of this work is a new generation of solid polymer electrolytes with a rigid backbone and a soft side chain structure. Moreover the novel material should be based on cheap substrates and its synthesis should not be complicated aiming at low overall costs. The new materials are based on hydroxypropylcellulose and oligoethyleneoxide derivatives as starting materials. The grafting of the oligoethyleneoxide side chains onto the cellulose was carried out following two synthetic methods. One is based on a bromide derivative and another based on p-toluolsulfonyl as a leaving group. The side chain reagents were prepared form tri(ethylene glycol) monoethyl ether. In order to improve the mechanical properties the materials were crosslinked. Two different conceptions have been engaged based on either urethane chemistry or photosensitive dimethyl-maleinimide derivatives. PEO - graft - cellulose derivatives with a high degree of substitution between 2,9 and 3,0 were blended with lithium trifluoromethane-sulfonate, lithium bis(trifluorosulfone)imide and lithium tetrafluoroborate. The molar ratios were in the range from 0,02 to 0,2 [Li]/[O]. The products have been characterized with nuclear magnetic resonance (NMR), gel permeation chromatography (GPC) and laserlight scattering (LS) with respect to their degree of substitution and molecular weight. The effect of salt concentration on ionic conductivity, thermal behaviour and morphology has been investiga-ted with impedance spectroscopy, differential scanning calorimetry (DSC) and thermal gravimetric analysis (TGA). The crosslinking reactions were controlled with dynamic mechanical analysis (DMS). The degree of substitution of our products is varying between 2,8 and 3,0 as determined by NMR. PEO - graft - cellulose derivatives are highly viscous liquids at room temperature with glass transition temperatures around 215 K. The glass transition temperature for the Lithium salt complexes of PEO - graft - cellulose deri-vatives increase with increasing salt content. The maximum conductivity at room temperature is about 10-4 and at 100°C around 10-3 Scm-1. The presence of lithium salt decreases the thermal stability of the complexes in comparison to pure PEO - graft - cellulose derivatives. Complexes heated over 140 – 150°C completely lose their ionic conductivity. The temperature dependence of the conductivity presented as Arrhenius-type plots for all samples is similar in shape and follows a VTF behaviour. This proofs that the ionic transport is closely related to the segmental motions of the polymer chains. Novel cellulose derivatives with grafted oligoethylen-oxide side chains with well-defined chemical structure and high side chain grafting density have been synthesized. Cellulose was chosen as stiff, rod like macromolecule for the backbone while oligoethylen-oxides are chosen as flexible side chains. A maximum grafting density of 3.0 have been obtained. The best conductivity reaches 10-3 Scm-1 at 100°C for a Li-triflate salt complex with a [Li]/[O] ratio of 0.8. The cross-linked complexes containing the lithium salts form elastomeric films with convenient mechanical stability. Our method of cellulose modification is based on relatively cheap and commercially available substrates and as such appears to be a promising alternative for industrial applications.

Die Selektine initiieren im Verlauf von Entzündungsprozessen einen ersten Zellkontakt zwischen Leukozyten und Endothelzellen und ermöglichen so die Auswanderung der Leukozyten aus den Blutgefäßen in das umliegende Gewebe, wo sie ihre immunologische Wirkung entfalten können. Viele Krankheiten gehen allerdings mit einer übermäßigen, durch Selektine vermittelten Zelladhäsion einher. Daher war es das Ziel dieser Arbeit, Selektininhibitoren zu synthetisieren, die pathologische Zelladhäsionsprozesse, wie man sie z.B. bei rheumatoider Arthritis, bei Erkrankungen der Herzkranzgefäße oder im Verlauf von Tumormetastasierungen findet, unterbinden können. Als Leitstruktur für solche Inhibitoren dient das auf den natürlichen Selektinliganden vorkommende Tetrasaccharid Sialyl-Lewis-X. Sialyl-Lewis-X stellt aber nur einen Teil der natürlichen Selektinliganden dar. Es bindet auch nur im millimolaren Bereich an die Selektine. Die komplexen natürlichen Selektinliganden wie z.B. ESL-1 (E-Selektin-Ligand-1), die an verschiedenen Glycosylierungs-stellen des Glycoproteins Sialyl-Lewis-X präsentieren, binden mit deutlich höherer Affinität an die Selektine. Für eine spezifische Rezeptorbindung sind daher außer dem Tetrasaccharid weitere Partialstrukturen verantwortlich, wobei gezeigt werden konnte, dass ein Anknüpfen von Sialyl-Lewis-X-Derivaten an die Partialsequenz 672-681 des ESL-1 eine Affinitätssteigerung hervorruft. Ein weiterer Nachteil des natürlichen Sialyl-Lewis-X-Tetrasaccharids im Hinblick auf seine pharmakologische Verwendung besteht darin, dass sowohl die fucosidische Bindung als auch die glycosidische Verknüpfung zur Neuraminsäure durch Enzyme leicht gespalten werden, wodurch Sialyl-Lewis-X als potenzielles Anti-Adäsionsmolekül an Wert verliert. Um die Kohlenydratliganden vor einem solchen enzymatischen Abbau zu bewahren, wurden in dieser Arbeit neben der im Sialyl-Lewis-X vorliegenden L-Fucose die im Menschen nicht vorkommenden Kohlenhydrate D-Arabinose und L-Galactose sowie neben der Neuraminsäure die (S)-Cyclohexylmilchsäure zur Herstellung der sechs Glycopeptid-Selektinliganden 1-6 mit der Partialsequenz 672-681 des ESL-1 verknüpft. Die Tetrasaccharide und Tetrasaccharid-Mimetika können aus den geschützten Monosacchariden und der geschützten Cyclohexylmilch-säure in parallelen Synthesen im Gramm-Maßstab hergestellt werden. Die automatisierten Glycopeptid-Festphasensynthesen wurden an einem Peptidsynthesizer nach der Fmoc-Strategie unter Verwendung von mit Asparaginsäure vorbeladenen TentaGel®-Harzen durchgeführt. Die Strukturen aller sechs Glycopeptide 1-6 wurden sowohl durch hoch auflösende massenspektrometrische Analysen als auch durch ein- und zweidimensionale NMR-Experimente belegt. Als Ergebnis dieser Arbeit liegen sechs Sialyl-Lewis-X-Glycopeptide und -Mimetika mit der Partialsequenz 672-681 des ESL-1 vor. Diese werden in Kürze auf ihre Wirksamkeit als Zelladhäsions-inhibitoren für E-Selektin getestet. Daraus sollen sich Erkenntnisse über Struktur-Wirkungs-Beziehungen gewinnen lassen, insbesondere was das kooperative Zusammenwirken von Saccharid- und Peptidteilstrukturen in der Erkennung der Liganden durch das E-Selektin anbetrifft.

The transition metal-catalyzed allylic alkylation (Tsuji-Trost type reaction) is a powerful tool for C-C, C-N, and C-O bond formation, which has been widely applied to organic chemistry over the last decades. Typical substrates for this transformation are activated allylic compounds such as halides, esters, carbonates, carbamates, phosphates, and so on. However, use of these substrates is associated with the disadvantage of generating a stoichiometric amount of chemical waste. Furthermore, these starting materials have to be prepared in an extra step from the corresponding allylic alcohol. Thus, ideal substrates would be the allylic alcohols themselves, with water being the only byproduct in this case. However, the scarse propensity of the hydroxyl moiety to act as good leaving group has significantly limited their use so far. During the last decade significant efforts have been made in order to develop more atom-economical and environmentally-friendly allylic alkylation protocols by employing allylic alcohols directly. In this PhD dissertation two main projects addressing this topic are presented. “Project 1” deals with the development of new metal-catalyzed intramolecular Friedel-Crafts (FC) allylic alkylations of electron-rich (PAPER A), as well as challenging electron-poor arenes (PAPER B) with alcohols. In “Project 2”, gold(I)-catalyzed intramolecular and stereoselective allylic alkylation reactions are reported. In particular, a FC alkylation of indole-containing allylic alcohols is presented in PAPER C. While, an O-alkylation of aminol-containing allylic alcohols is reported in PAPER D. To the best of knowledge, these reports represent the first example of gold(I)-catalyzed stereoselective alkylations with alcohols.

The objective of this study is to provide empirical evidence on how ownership structure and owner’s identity affect performance, in the banking industry by using a panel of Indonesia banks over the period 2000–2009. Firstly, we analysed the impact of the presence of multiple blockholders on bank ownership structure and performance. Building on multiple agency and principal-principal theories, we investigated whether the presence and shares dispersion across blockholders with different identities (i.e. central and regional government; families; foreign banks and financial institutions) affected bank performance, in terms of profitability and efficiency. We found that the number of blockholders has a negative effect on banks’ performance, while blockholders’ concentration has a positive effect. Moreover, we observed that the dispersion of ownership across different types of blockholders has a negative effect on banks’ performance. We interpret such results as evidence that, when heterogeneous blockholders are present, the disadvantage from conflicts of interests between blockholders seems to outweigh the advantage of the increase in additional monitoring by additional blockholder. Secondly, we conducted a joint analysis of the static, selection, and dynamic effects of different types of ownership on banks’ performance. We found that regional banks and foreign banks have a higher profitability and efficiency as compared to domestic private banks. In the short-run, foreign acquisitions and domestic M&As reduce the level of overhead costs, while in the long-run they increase the Net Interest Margin (NIM). Further, we analysed NIM determinants, to asses the impact of ownership on bank business orientation. Our findings lend support to our prediction that the NIM determinants differs accordingly to the type of bank ownership. We also observed that banks that experienced changes in ownership, such as foreign-acquired banks, manifest different interest margin determinants with respect to domestic or foreign banks that did not experience ownership rearrangements.

„Risikomaße in der Finanzmathematik“ Der Value-at -Risk (VaR) ist ein Risikomaß, dessen Verwendung von der Bankenaufsicht gefordert wird. Der Vorteil des VaR liegt – als Quantil der Ertrags- oder Verlustverteilung - vor allem in seiner einfachen Interpretierbarkeit. Nachteilig ist, dass der linke Rand der Wahrscheinlichkeitsverteilung nicht beachtet wird. Darüber hinaus ist die Berechnung des VaR schwierig, da Quantile nicht additiv sind. Der größte Nachteil des VaR ist in der fehlenden Subadditivität zu sehen. Deswegen werden Alternativen wie Expected Shortfall untersucht. In dieser Arbeit werden zunächst finanzielle Risikomaße eingeführt und einige ihre grundlegenden Eigenschaften festgehalten. Wir beschäftigen uns mit verschiedenen parametrischen und nichtparametrischen Methoden zur Ermittlung des VaR, unter anderen mit ihren Vorteilen und Nachteilen. Des Weiteren beschäftigen wir uns mit parametrischen und nichtparametrischen Schätzern vom VaR in diskreter Zeit. Wir stellen Portfoliooptimierungsprobleme im Black Scholes Modell mit beschränktem VaR und mit beschränkter Varianz vor. Der Vorteil des erstens Ansatzes gegenüber dem zweiten wird hier erläutert. Wir lösen Nutzenoptimierungsprobleme in Bezug auf das Endvermögen mit beschränktem VaR und mit beschränkter Varianz. VaR sagt nichts über den darüber hinausgehenden Verlust aus, während dieser von Expected Shortfall berücksichtigt wird. Deswegen verwenden wir hier den Expected Shortfall anstelle des von Emmer, Korn und Klüppelberg (2001) betrachteten Risikomaßes VaR für die Optimierung des Portfolios im Black Scholes Modell.

CpGV-MCp5 is a natural mutant of the Cydia pomonella Granulovirus (Mexican isolate) (CpGV-M) that harbors an insect host transposon termed TCl4.7 in its genome. TCl4.7 is located between the open reading frames Cp15 and Cp16 and separates two homologous regions hr3 and hr4, which have been recently shown to be origins of replication of CpGV-M. The MCp5 has a significant replication disadvantage in the presence of the wild-type CpGV-M. In this study, the possible effects of TCl4.7 transposon insertion on the genome function of its insertion site has been analysed. The role of Cp15 and Cp16 in the context of the virus infection cycle was examined by generating a CpGV-Bacmid (CpBAC) and Cp15 knock-out (CpBACCp15KO) and Cp16 knock-out (CpBACCp16KO) mutants. The mutant CpBACCp15KO was not able to replicate in CM larvae suggesting that Cp15 was essential for virus replication. In contrast, the mutant CpBACCp16KO infected CM larvae and produced viable occlusion bodies (OBs) demonstrating that Cp16 is a non-essential gene for virus in vivo infection of C. pomonella. The temporal transcription of Cp15 and Cp16, as well as of Cp31 (F protein) as a control, was analysed using RT-PCR and quantitative real-time PCR. It suggested a general delay or reduction of gene transcription of MCp5 compared to the parental CpGV-M. Western blot analyses using anti-Cp15 and anti-Cp16 polyclonal antibodies, however, did not show any immuno-reactive response. Thus, a direct influence of TCl4.7 on the expression of Cp15 and Cp16 could not be substantiated. To investigate whether the interruption of hr3 and hr4 palindromes affects the virus replication, two mutant bacmids with a deletion of hr3 and hr4 (CpBAChr3/hr4-KO) and another with an insertion of a Kanamycin resistance cassette between hr3 and hr4 (CpBAChr3-kan-hr4) were generated. Both mutant bacmids replicated and produced infectious virus OBs, which did not significantly differ in their median lethal concentration (LC50) and median survival time (ST50) compared to the parental CpBAC. Interestingly, the mutant CpBAChr3-kan-hr4 was very effectively out-competed by parental CpBAC, when CM larvae were co-infected with known ratios of OBs of CpBAC and the mutant CpBAChr3-kan-hr4. These observations suggested a functional co-operation between hr3 and hr4 which was interrupted by the KanR insertion in CpBAChr3-kan-hr4 and possibly by TCl4.7 transposon insertion in the mutant MCp5. This hypothesis may explain the observed replication disadvantage of the mutants MCp5 and CpBAChr3-kan-hr4 in the presence of the parental viruses CpGV-M and CpBAC, respectively.

The aim of this thesis was the formulation of new chitosan based delivery systems for transmucosal drug administration. Transmucosal routes, such as buccal, vaginal and nasal routes, allow the circumvention of the hepatic first pass metabolism and avoid the gastrointestinal chemical and enzymatic degradations. Moreover, transmucosal drug administration can allow to avoid pain or discomfort caused by injections, when drugs are administered through parenteral routes, thus increasing patient compliance. On the other side, the major disadvantage of transmucosal drug administration is represented by the presence of biological fluids and mucus that can remove drug systems from the application site, thus reducing the contact time between drug and mucosa and consequently, decreasing drug bioavailability. For this reason, in this study, the investigation of chitosan delivery systems as mucoadhesive formulations able to increase drugs residence time and to improve their bioavailability, was taken into account. In the paper 1, buccal films based on chitosan-gelatin complexes were prepared and loaded with propranolol hydrochloride. The complexes were characterized and studied in order to evaluate their physical- chemical properties and their ability to release the drug and to allow its permeation through buccal mucosa. In the paper 2, vaginal inserts based on chitosan/alginate complexes were formulated for local delivery of chlorhexidine digluconate. Tests to evaluate the interaction between the polymers and to study drug release properties were performed, as well as the determination of antimicrobial activity against the patogens responsible of vaginitis and candidosis. In the project 3, chitosan based nanoparticles containing cyclodextrin and other excipients, with the capacity to modify insulin bioavailabity were formulated for insulin nasal delivery. Nanoparticles were characterized in terms of size, stability and drug release. Moreover, in vivo tests were performed in order to study the hypoglycemic reduction in rats blood samples.

Es wurde ein für bodengebundene Feldmessungen geeignetes System zur digital-holographischen Abbildung luftgetragener Objekte entwickelt und konstruiert. Es ist, abhängig von der Tiefenposition, geeignet zur direkten Bestimmung der Größe luftgetragener Objekte oberhalb von ca. 20 µm, sowie ihrer Form bei Größen oberhalb von ca. 100µm bis in den Millimeterbereich. Die Entwicklung umfaßte zusätzlich einen Algorithmus zur automatisierten Verbesserung der Hologrammqualität und zur semiautomatischen Entfernungsbestimmung großer Objekte entwickelt. Eine Möglichkeit zur intrinsischen Effizienzsteigerung der Bestimmung der Tiefenposition durch die Berechnung winkelgemittelter Profile wurde vorgestellt. Es wurde weiterhin ein Verfahren entwickelt, das mithilfe eines iterativen Ansatzes für isolierte Objekte die Rückgewinnung der Phaseninformation und damit die Beseitigung des Zwillingsbildes erlaubt. Weiterhin wurden mithilfe von Simulationen die Auswirkungen verschiedener Beschränkungen der digitalen Holographie wie der endlichen Pixelgröße untersucht und diskutiert. Die geeignete Darstellung der dreidimensionalen Ortsinformation stellt in der digitalen Holographie ein besonderes Problem dar, da das dreidimensionale Lichtfeld nicht physikalisch rekonstruiert wird. Es wurde ein Verfahren entwickelt und implementiert, das durch Konstruktion einer stereoskopischen Repräsentation des numerisch rekonstruierten Meßvolumens eine quasi-dreidimensionale, vergrößerte Betrachtung erlaubt. Es wurden ausgewählte, während Feldversuchen auf dem Jungfraujoch aufgenommene digitale Hologramme rekonstruiert. Dabei ergab sich teilweise ein sehr hoher Anteil an irregulären Kristallformen, insbesondere infolge massiver Bereifung. Es wurden auch in Zeiträumen mit formal eisuntersättigten Bedingungen Objekte bis hinunter in den Bereich ≤20µm beobachtet. Weiterhin konnte in Anwendung der hier entwickelten Theorie des ”Phasenrandeffektes“ ein Objekt von nur ca. 40µm Größe als Eisplättchen identifiziert werden. Größter Nachteil digitaler Holographie gegenüber herkömmlichen photographisch abbildenden Verfahren ist die Notwendigkeit der aufwendigen numerischen Rekonstruktion. Es ergibt sich ein hoher rechnerischer Aufwand zum Erreichen eines einer Photographie vergleichbaren Ergebnisses. Andererseits weist die digitale Holographie Alleinstellungsmerkmale auf. Der Zugang zur dreidimensionalen Ortsinformation kann der lokalen Untersuchung der relativen Objektabstände dienen. Allerdings zeigte sich, dass die Gegebenheiten der digitalen Holographie die Beobachtung hinreichend großer Mengen von Objekten auf der Grundlage einzelner Hologramm gegenwärtig erschweren. Es wurde demonstriert, dass vollständige Objektgrenzen auch dann rekonstruiert werden konnten, wenn ein Objekt sich teilweise oder ganz außerhalb des geometrischen Meßvolumens befand. Weiterhin wurde die zunächst in Simulationen demonstrierte Sub-Bildelementrekonstruktion auf reale Hologramme angewandt. Dabei konnte gezeigt werden, dass z.T. quasi-punktförmige Objekte mit Sub-Pixelgenauigkeit lokalisiert, aber auch bei ausgedehnten Objekten zusätzliche Informationen gewonnen werden konnten. Schließlich wurden auf rekonstruierten Eiskristallen Interferenzmuster beobachtet und teilweise zeitlich verfolgt. Gegenwärtig erscheinen sowohl kristallinterne Reflexion als auch die Existenz einer (quasi-)flüssigen Schicht als Erklärung möglich, wobei teilweise in Richtung der letztgenannten Möglichkeit argumentiert werden konnte.
