O câncer gástrico representa um grave problema de saúde pública mundial. A alta incidência de tumores avançados com baixa sobrevida pelas metástases, sobretudo no norte do país, nos fez realizar o estudo comparativo das linhagens de adenocarcinomas gástricos metastáticos (AGP01) com adenocarcinomas gástricos sem metástases (ACP02) através da avaliação proteômica da via de mobilidade celular, que possam ter relação com a formação dessas metástases. Foi realizado estudo proteômico das linhagens AGP01 e ACP02 através da técnica da cromatografia líquida de alta performance 2D Nanoultra (UPLC) em conjunto com nanoESI-MSE (MudPIT) e análise funcional das proteínas diferencialmente expressas no programa Ingenuity Pathways Analysis (IPA). Observamos 19 proteínas com aumento da expressão na linhagem AGP01 em relação a ACP02, as quais apresentam relação com movimento, organização e morfologia celular, onde podemos sugerir que as proteínas ACTB, ANXA1, LGALS1, IQGAP1, EZR, MSN, MYH9 e S100A11, de acordo com nossos achados e corroborados pela literatura pesquisada, tem associação com a metástase de adenocarcinomas gástricos. Outras proteínas se mostraram em forte expressão em nosso estudo, mas na literatura pesquisada sua expressão tem relação com as vias de disseminação apenas de outros tumores, como: mama (RAB5C), pulmão (PLS1 e CAP1), reto (ACTN1) e GIST (SYNE2). Conflitantes com nosso estudo, as expressões das proteínas CAPZA1, FLNA e FLNC, foram observadas na literatura como um inibidor de avanço tumoral, enquanto que as expressão das proteínas MYL6, MYL6B, e ACTN2, aparecem pela primeira vez como tendo relação com a mobilidade celular, invasão e metástase em câncer.

Several studies of the physiological responses of different organisms exposed to extremely low-frequency electromagnetic fields (ELF-EMF) have been described. In this work, we report the minimal effects of in situ exposure to ELF-EMF on the global protein expression of Chromobacterium violaceum using a gel-based proteomic approach. The protein expression profile was only slightly altered, with five differentially expressed proteins detected in the exposed cultures; two of these proteins (DNA-binding stress protein, Dps, and alcohol dehydrogenase) were identified by MS/MS. The enhanced expression of Dps possibly helped to prevent physical damage to DNA. Although small, the changes in protein expression observed here were probably beneficial in helping the bacteria to adapt to the stress generated by the electromagnetic field.

Purpose Mantle-cell lymphoma (MCL) has a variable natural history but is incurable with current therapies. MicroRNAs (miRs) are useful in prognostic assessment of cancer. We determined an miR signature defining aggressiveness in B-cell non-Hodgkin lymphomas (NHL) and assessed whether this signature aids in MCL prognosis.MethodsWe assessed miR expression in a training set of 43 NHL cases. The miR signature was validated in 44 additional cases and examined on a training set of 119 MCL cases from four institutions in Canada. miRs significantly associated with overall survival were examined in an independent cohort of 114 MCL cases to determine association with patient outcome. miR expression was combined with current clinical prognostic factors to develop an enhanced prognostic model in patients with MCL.ResultsFourteen miRs were differentially expressed between aggressive and indolent NHL; 11 of 14 were validated in an independent set of NHL (excluding MCL). miR-127-3p and miR-615-3p were significantly associated with overall survival in the MCL training set. Their expression was validated in an independent MCL patient set. In comparison with Ki-67, expression of these miRs was more significantly associated with overall survival among patients with MCL. miR-127-3p was combined with Ki-67 to create a new prognostic model for MCL. A similar model was created with miR-615-3p and Mantle Cell Lymphoma International Prognostic Index scores.ConclusionEleven miRs are differentially expressed between aggressive and indolent NHL. Two novel miRs were associated with overall survival in MCL and were combined with clinical prognostic models to generate novel prognostic data for patients with MCL. The length of the post-partum anoestrous interval affects reproductive efficiency in many tropical beef cattle herds. In this study, results from genome-wide association studies (Experiment 1: GWAS) and gene expression (Experiment 2: microarray) were combined in a systems approach to reveal genetic markers, genes and pathways underlying the physiology of post-partum anoestrus in tropically adapted cattle. The microarray study measured the expression of 13,964 genes in the hypothalamus of Brahman cows. A total of 366 genes were differentially expressed (DE) in the post-partum period, when acyclic cows were compared to cows that had resumed ovarian cycles. Associated markers (P < 0.05) from a high density GWAS pointed to 2829 genes that were associated with post-partum anoestrous interval (PPAI) in two populations of beef cattle: Brahman and Tropical composite. Together the experiments provided evidence for 63 genes that are likely to influence the resumption of ovulation post-partum in tropically adapted beef cattle. Functional annotation analysis revealed that some of the 63 genes have known roles in hormonal activity, energy balance and neuronal synapse plasticity. Polymorphisms within candidate genes identified by this systems approach could have biological significance in post-partum anoestrus and help select Zebu (Bos indicus) influenced cattle with genetic potential for shorter post-partum anoestrus.
