Obesity is increasing, reaching epidemic levels in many regions of the world. Studies have shown that consumption of peanuts influences on weight control and this influence may be due to the action of trypsin inhibitors sacietogênica that condition increased plasma colescistocinina (CCK). Moreover, the peanut has other health benefits, and these assignments are guaranteed to increase their production and consumption of several of its products, including the paçoca peanut. The aim of this study was to identify the presence of a trypsin inhibitor in paçoca peanut and evaluate its effect on food intake, weight gain and histomorphological changes in swiss mice (n = 8) and Wistar rats (n = 6). Experimental diets were prepared based on the AIN-93G and supplemented with tack or peanut trypsin inhibitor partially purified paçoca peanut (AHTI). After each treatment, the animals were anesthetized and euthanized, their bloods were collected by cardiac puncture for the determination of CCK and other biochemical parameters (glucose, triglycerides, total cholesterol, high density lipoprotein, low density lipoprotein, glutamic-pyruvic transaminase, glutamic oxaloacetic transaminase and albumin) and their pancreas removed for histologic and morphometric analysis. The supplementation with paçoca peanut and the AHTI showed a decrease of body weight gain and food intake in both mice and rats, due to the satiety, since the animals showed no evidence of impairment of nutritional status conditioned by consumption the AHTI. There were also observed biochemical or morphological important when compared with controls. However, AHTI led to increased secretion of CCK, a peptide sacietogênico. Thus, these results indicate that AHTI present in paçoca peanut, is able to enhance the secretion of plasma CCK and thereby reduce the weight gain associated with lower food intake of experimenta animals

The seeds are excellent sources of proteinase inhibitors and have been highlighted owing to various applications. Among these applications are those in effect on food intake and weight gain that stand out because of the increasing number of obese individuals. This study evaluated the effects of trypsin inhibitor present in the seed of tamarind (Tamarindus indica L.) reduction in weight gain, biochemical and morphological alterations in Wistar rats. For this, we partially purified a trypsin inhibitor tamarind seed. This inhibitor, ITT2 at a concentration of 25 mg / kg body weight, over a period of 14 days was able to reduce food intake in rats (n = 6) by approximately 47%, causing a reduction in weight gain approximately 70% when compared with the control group. With the evaluation of the in vivo digestibility was demonstrated that the animals lost weight due to satiety, presented by the reduction of food intake, since there were significant differences between true digestibility for the control group (90.7%) and the group treated with inhibitor (89.88%). Additionally, we checked the deeds of ITT2 on biochemical parameters (glucose, triglycerides, total cholesterol, high-density lipoprotein, low-density lipoprotein, glutamic-pyruvic transaminase, glutamic oxaloacetic transaminase, gamma glutamyl transferase albumin, globulin, total protein and C-reactive protein) and these, when assessed in the study groups showed no statistically significant variations. We also evaluate the histology of some organs, liver, stomach, intestine, and pancreas, and showed no changes. And to evaluate the effect of trypsin inhibitor on food intake due to the satiety is regulated by cholecystokinin (CCK) were measured plasma levels, and it was observed that the levels of CCK in animals receiving ITT2 were significantly higher ( 20 + 1.22) than in animals receiving only solution with casein (10.14 + 2.9) or water (5.92 + 1.15). Thus, the results indicate that the effect caused ITT2 satiety, reducing food intake, which in turn caused a reduction in weight gain in animals without causing morphological and biochemical changes, this effect caused by the elevation of plasma levels CCK

Canine Visceral Leishmania (CVL) is an important zoonotic disease that has a world wide distribution and has a large impact on public health on the American Continent, especially in Brazil, where the nature of endemic diseases in humans affects a large part of the nation. The influence of the prevalence of CVL in the increased rate of human cases in endemic areas and in the unleashing of epidemic outbreaks shows the need for a more profound understanding, that would generate significant advances in the current measures used to control the reservoirs of sickness that are practiced by the Programa Nacional de Vigilância e Controle da Leishmaniose Visceral. The present work describes and compares the clinical-laboratorial and histopathological findings of twenty-three dogs that were naturally infected by Leishmania chagasi, from endemic areas in metropolitan Natal, Rio Grande do Norte, Brazil. These animals, that were selected and given physical and serological exams (IFI and ELISA rK-39), were classified according to the degree of clinical severity and had blood samples drawn (whole blood and serum) for a complete hemogram and a coagulogram to be done as well as biochemical tests for kidney and liver function. The confirmation of infection by L. chagasi was done after the euthanasia of the animals, through the direct demonstration of the parasite in the impression of the spleen and liver crowned with GIEMSA and through a cultivation by means of NNN/Schneider. According to the clinical evaluation, the animals were classified as asymptomatic (7), oligosymptomatic (7) and polysymptomatic (9). Among the animals that were chosen to be autopsied, there were 2 asymptomatic, 3 oligosymptomatic and 3 polysymptomatic, for the purpose of studying their histopathology, having collected fragments of the spleen, liver, kidneys and skin and were fixed in 10% tamponed formol. The comparison between the average parameters of the clinical-laboratory tested animals in the groups was done through the Student t test (a<0.05). The main clinical signals observed were lymphadenomegaly, alopecy, dermatitis, exfoliation, cutaneous ulcers, onicogriphosis and emaciation. The main clinical-laboratorial alterations established, mainly in the polysymptomatic group, were anemia, hyperproteinemia, hyperglobulinemia, alterations in the albumin/globulin ratio and increased ALT activity. Renal alterations were not verified (urea and creatinine levels were normal). Thrombocytopenia was observed in three clinical groups. However, the other indicators of coagulation function (TAP and TTPA) did not have abnormal variations. There were inflammatory infiltrations and leishmania amastigotes in the skin of polysymptomatic dogs, however, they were not found in the skin of asymptomatic animals. Hypertrophy and hyperplasia of the phagocyte mononuclear system, leishmania amastigote parasites were found in the macrophages, extramedullary hematopoiesis and degenerative alterations were detected in the spleen and liver of 8 of the animals submitted to histopathological exams. In accord with these results, it was demonstrated that the expected alterations in the hematological and biochemical parameters in function of their viscerotropic nature of CVL are mainly observed in the more advanced stages of the disease. The absence of inflammatory infiltration and parasite load in the skin suggest that infected animals without symptoms may have an importance irrelevant to the infectiousness of the vector

Linseed is an important oilseed consumed raw as nutritional supplement, that although represents a rich source of nutrients, its nutritional value could be impaired due to the presence of antinutritional factors. In this study, protein fractions from raw linseed flour were extracted and isolated being obtained 12% of albumins, 82% of globulins, 5% of glutelins and 1% of prolamins. These proteins were visualized by SDS-PAGE and albumins showed low molecular mass protein bands around 21 kDa and minor bands, similar to that of trypsin inhibitor; Globulins presented protein bands with high molecular masses, which possibly are constituents of multimeric proteins, such as legumins. After determination of the centesimal composition of raw linseed, it was used as exclusive protein source for young rats to evaluate its effect on animal growth. The results showed negative effects on rat growth (weight gain 73% less than the control group) and reduction of intestinal villus (35%), that could be related with in vitro and in vivo globulin digestibility and proteinaceous antinutritional factors (mammalian digestive enzymes inhibitors and lectins) in albumin fraction. Native globulins showed, by SDS-PAGE, low susceptibility in vitro to trypsin and chymotrypsin, however presented high degradation by pancreatin. Thermal treatment of globulins for 5 and 15 minutes at 100ºC improved considerably its digestibility by trypsin and pancreatin. Globulins presented 93.2% in vivo digestibility, similar to the control protein. Albumin fraction had high trypsin inhibition activity (100%) and chymotrypsin inhibition of 28.3%; haemagglutinating activity was not detected. The results of this study indicate the negative action of trypsin inhibitors on animal growth, but can not be discarded its combined action with other antinutritional factors, which could compromise the raw linseed utilization as an alternative food

American visceral leishmaniasis is a zoonosis caused by Leishmania infantum and transmitted by the bite of the sand flies Lutzomia longipalpis.The main domestic reservoir is the dog, while foxes and opposums are the known wild reservoirs. However, identification of natural infections with L. infantum in rodents appears for need of investigating the participation of these rodents how source of infection of the parasite. In the present work the Leishmania infantum infection was investigated in rodents captured in Rio Grande do Norte, aiming at to offer subsidies to the understanding of the epidemic chains of LVA in the State. Thirteen Galea spixii were distributed in four groups, being G1 the group control with four animals and the others, G2, G3 and G4, with three animals each. Those animals were intraperitoneally inoculated with 107 promastigotas of L. infantum and accompanied for, respectively, 30, 90 and 180 days. Weekly the animals were monitored as for the corporal weight and rectal temperature. At the end of each stipulated period the animals were killed. Blood were used for determination of the parameters biochemical and haematological, PCR, ELISA, microscopic examination and cultivation in NNN medium. Liver, spleen and lymph node were used in Giemsa-stained impression and cultivation in NNN medium. Liver and spleen fragments were still used in PCR and histopathological, respectively. At the same time 79 rodents of the species Rattus rattus, Bolomys lasiurus, Oligoryzomys nigripis, Oryzomys subflavus and Trichomys apereoides were captured in the Municipal districts of Brejinho, Campo Grande, Coronel Ezequiel, Passa e Fica and Vázea for identification of natural infection with L. infantum. Evidence of infection was checked by direct examination of Giemsa-stained impression of liver, spleen and blood and culture of these tissues in NNN medium. Antibodies were researched by ELISA. They were not found differences among the weigh corporal final, rectal temperature and biochemical and haematological parameters of the Galea spixii controls and infected. The rectal temperature of the animals varied from 36OC to 40OC. For the first time values of the haematocrit (33,6% to 42,8%), hemoglobin (10,2 to 14,5g/dl), erythrocyts number (4,67x106 to 6,90x106/mm3), total leukocytes (0,9x103 to 9,2x103/mm3), platelets (49x103 to 509x103/mm3) total proteins (1,56 to 6,06 g/dl), albumin (1,34 to 3,05 g/dl) and globulins (0,20 to 3,01 g/dl) of the Galea spixii were determined. The lymphocytes were the most abundant leucocytes. Infection for L. infantum was diagnosed in two animals euthanasied 180 days after the infection. In one of the animals was also identified antibodies anti-Leishmania. The parasite was not found in none of the five other species of rodents captured. Galea spixii are resistant to the infection for L. infantum and they are not good models for the study for visceral leishmaniose, although they can act as infection sources. More studies are necessary to determine the paper of the rodents in the epidemic chain of transmission of the visceral leishmaniose in the State of Rio Grande do Norte

O uso de probióticos na alimentação dos peixes pode estimular o sistema imunológico e agir na prevenção de enfermidades. O presente estudo avaliou a fisiologia de pirarucu Arapaima gigas após alimentação por 30 dias com dietas enriquecidas com probiótico Bacillus subtilis em 3 níveis de inclusão: 0 (Controle); 0,02 (BS0,02%) e 0,05% (BS0,05%) (triplicata), sendo realizada biometria e coleta de amostras de sangue. Os grupos foram comparados entre si (P<0,05). O peso, comprimento e o índice hepatossomático (IHS) dos pirarucus BS0,05% foram menores que os do Controle. As análises hematológicas e bioquímicas indicaram que BS0,02% promoveu aumento do hematócrito e da albumina, com relação aos demais grupos; e diminuição da concentração de hemoglobina corpuscular média (CHCM) e da glicose plasmática com relação ao Controle. A hemoglobina corpuscular média (HCM) e atividade respiratória dos leucócitos do BS0,05% diminuíram com relação ao Controle. BS0,02% apresentaram redução do número de leucócitos, com relação ao Controle, que refletiu na diminuição de monócitos, neutrófilos, LG-PAS e eosinófilos. Assim, os níveis de B. subtilis avaliados não promoveram crescimento ou alterações fisiológicas nos pirarucus, que indiquem melhoria do seu sistema imune.

Una strategia promettente per potenziare l’efficacia di un trattamento antitumorale limitandone gli effetti collaterali sistemici e i fenomeni di resistenza, è quella di combinare, all’interno di un nanoveicolo, l’azione sinergica/additiva di diversi trattamenti terapeutici, come ad esempio la terapia fotodinamica (PDT) e l’immunoterapia. In questo lavoro di tesi sono state preparate nanoparticelle multimodali altamente biodegradabili e biocompatibili, a base di albumina umana (HSA) caricate con un pro-farmaco del paclitaxel (PTX), un agente chemioterapeutico approvato per il trattamento di diversi tumori solidi; una molecola fotosensibile, IR780, che illuminata a una specifica lunghezza d'onda, induce la formazione di specie radicaliche dell'ossigeno (ROS) e ossigeno di singoletto (1O2), in grado di uccidere le cellule malate; ed infine con Indoximod (IND), in grado di inibire un checkpoint immunitario, con azione indiretta sull’enzima indoleamine 2,3-dioxygenase (IDO-1). Le nanoparticelle sono poi state rivestite con un guscio di MnO2, per limitare l’ipossia caratteristica dell’ambiente tumorale, potenziando così l’azione citotossica. Le nanoparticelle ottenute sono state caratterizzate in termini di dimensioni, capacità di loading, stabilità in acqua e in ambiente fisiologico. Inoltre, mediante spettroscopia UV-vis, è stata studiata la capacità dell’IR780 di produrre specie reattive all’ossigeno (ROS). In futuro, saranno studiati i fenomeni di rilascio dei diversi agenti dalle nanoparticelle e sarà verificato se la presenza dell’ossido di manganese sia in grado di migliorare l’ossigenazione del microambiente tumorale.

La perfusione polmonare extracorporea (EVLP) è una tecnica utilizzata dal 2010 per valutare e migliorare la qualità dell'organo da trapiantare e il danno da ischemia-ripefusione (IRI). Tale perfusione utilizza la soluzione di Steen, la cui composizione è solo parzialmente nota. Lo scopo è quello di identificare gli effetti di T3 su IRI polmonare ex vivo, in un modello di ratto di donatore a cuore non battente. Animali (40) randomizzati in otto gruppi e il protocollo EVLP sono stati standardizzati nel nostro centro. Sono state valutate la funzione polmonare, PEEP, la resistenza vascolare polmonare totale a 45, 60, 120 e 180 minuti di EVLP per eseguire analisi di gas, dosaggio del mediatore di infiammazione, mitocondriale libero DNA, freeT3 e freeT4. Alla fine dei campioni di tessuto polmonare sono stati congelati dal dosaggio ATP, espressione genica, DNA mitocondriale, T3. Non date le concentrazioni del produttore, abbiamo analizzato gli acidi grassi liberi, vitamine, ormoni e composizione della soluzione Steen. Risultati La soluzione di steen contiene albumina umana x2 nel siero umano (7,5-8 g/dl): le concentrazioni di ft4 e ft3 sono x2 quelle nel siero umano e vengono rilasciati dall'albumina. La concentrazione di ft4 e ft3 non è cambiata durante l'EVLP. La Steen ha alta fluorescenza per l'alta concentrazione delle molecole aromatiche (ormoni) mai descritto in precedenza. NADH e mtDNA nel perfused aumenta con danno ischemico e nel gruppo trattato con T3 Conclusione Il modello EVLP è già convalidato nella perfusione nel trapianto polmonare, ma è necessario approfondire l'effetto della Steen in termini di ormoni e analiti. L'effetto sull'IRI dell'EVLP sembra essere influenzato negativamente da un T3 troppo alto in Steen, cosa che descriviamo per la prima volta. L'ulteriore aggiunta di T3 provoca disfunzione mitocondriale e infiammazione.

La mia tesi si concentra sulla sintesi e funzionalizzazione di nanoparticelle d’argento studiandone l’interazione, tramite esperimenti in vitro, con cellule sane di fibroblasti murini NIH-3T3 e cellule tumorali da nodulo al seno MCF7. L’utilizzo di polielettroliti quali PDADMAC, PAH e PSS ha permesso la modifica delle proprietà superficiali delle nanoparticelle. Le nuove proprietà chimico-fisiche sono state caratterizzate tramite Dynamic Light Scattering, potenziale zeta e spettroscopia UV-vis. L’effetto della ricopertura con polielettroliti è stato valutato tramite test di vitalità cellulare somministrando le nanoparticelle funzionalizzate alle cellule sopracitate. Successivamente, è stata ottimizzata la procedura per un’ulteriore ricopertura sulle nanoparticelle cariche con BSA (Bovine Serum Albumin) valutando diversi fattori chiave. Le nanoparticelle ricoperte di albumina sono state caratterizzate e la composizione qualitativa della loro protein corona è stata ottenuta tramite analisi SDS-PAGE. Infine, le nanoparticelle ricoperte di BSA sono state somministrate alle due linee cellulari valutando l’effetto dell’albumina sulla risposta biologica tramite analisi di vitalità cellulare e immunofluorescenza.

As Doenças Crônicas Não Transmissíveis (DCNT) são consideradas como doenças epidêmicas. Estas são a Diabetes Mellitus, Hipertensão Arterial, Doença Cardiovascular e Doença Renal Crônica, estas doenças causam 60% das mortes em todo o mundo. Espera-se que elas sejam a principal causa de morte e incapacidade em 2020. O Brasil apresenta hoje uma maior expectativa de vida e aumento da concentração de pessoas idosas na população em geral, seguindo a tendência mundial. Ocorrem mudanças epidemiológicas com a diminuição das doenças infecto-parasitárias e predomínio das doenças crônicas não transmissíveis, entre elas a Diabetes Mellitus tipo II (DM II), tomando proporções crescentes no que se refere ao aparecimento de novos casos e complicações. Segundo dados da Federação Internacional de Diabetes (FID) até o ano 2030 o número de pessoas com diabetes deverá chegar a 439 milhões. Aqui no Brasil estima-se que haja 12 milhões de diabéticos, 76% deles acometidos pelo tipo II da doença, que é o tipo mais comum, porém o único que é quase totalmente evitável. A Diabetes Mellitus é um dos principais fatores de risco para o desenvolvimento de Doença Renal Crônica(DRC). Pelo menos 50% dos pacientes com diabetes têm microalbuminúria, o primeiro sinal de DRC, caracterizada por excreção de pequenas quantidades de albumina na urina. Destes 50%, cerca de um terço irá desenvolver uma doença grave o suficiente para chegar à fase terminal, e exigem, portanto, diálise e transplante renal. Na verdade, a Diabetes Mellitus é hoje a principal causa de admissão de diálise no mundo. Além disso, foi demonstrado que pacientes com microalbuminúria têm um risco de doenças vasculares tais como infarto agudo do miocárdio ou acidente vascular cerebral, entre duas e três vezes mais elevadas do que os pacientes diabéticos sem microalbuminúria.
