Difficulties in reproducing the citrus variegated chlorosis (CVC) disease symptoms in expertmental plants have delayed implementation of studies to better understand the essential aspects of this important disease. In an extensive Study, cultivars of sweet orange (Citrus sinensis) were inoculated with Xylella fastidiosa using procedures that included root immersion, and stein absorption, pricking, or infiltration of the inoculum into plants of different ages. Inoculum consisted of 5-day-old cultures or cell suspensions of CVC strain 9a5c diluted in phosphate-buffered saline. Inoculated plants and controls were grown, or transferred just after inoculation, to 5-liter pots or 72-cell foam trays. Approximately 4, 5, 9, and 12 months after inoculation, leaves were collected and processed for polymerase chain reaction analysis or X. fastidiosa isolation on BCYE agar medium. Root immersion and stem inoculation of 4- and 6-month-old plants resulted in low percentages of symptomatic (0 to 7%) and plants positive by isolation (0 to 9%). Pinpricked or injected stems of I-month-old seedlings resulted in high percentages of plants symptomatic (29 and 90% in Pera Rio, 75, 59, and 83% in Valencia, and 77% in Natal) or positive by isolation (26 and 93% in Pera Rio, 98, 96, and 83% in Valencia, and 77% in Natal), In foam trays, the seedlings grew less, the incubation period was shorter. and disease severity was higher than in pots. This system allows testing of higher numbers of plants in a reduced space with a more precise reproduction of the experimental conditions.

Two allopatric species of Coryphasia (Araneae: Salticidae), both described for the first time in this study, have been found associated with Bromeliaceae in distinct phytophysiognomies (e.g., inselbergs, highland forests and restingas) from southeastern Brazil. In this study, we investigated whether these salticids were associated specifically with bromeliads, and whether they used bromeliads of different species and sizes in distinct geographic regions. The Coryphasia species were rarely found outside bromeliads, occupied larger bromeliad species among those available, and were generally more frequent on bromeliads in open areas, such as inselbergs on mountain tops. The two Coophasia species were observed submerging in phytotelmata, possibly as an anti-predatory behavior. The patterns of spatial and microspatial distribution and the submergence behavior of these species were similar to those of other bromeliad-dwelling salticids, which suggests a convergence among spiders that live on bromeliads.

A total of 20 insect species were observed on the extrafloral nectaries (EFNs) of Croton sarcopetalus. The most frequent ant species were Zacryptocerus sp., Crematogaster brevispinosa, C. scelerata, and Linepithema humile. Ants continuously patrolled the plants taking extrafloral nectar. Experimental data showed that there were no significant differences in either the degree of herbivory or in the reproductive output between control stems (with ants) and treated ones (without ants). We found no significant evidence of protection by ants mediated by EFNs in C. sarcopetalus. It is possible that the plant has other mechanisms than ant protection to prevent herbivore damage (e.g., hairs, latex, chemical defense). As this species occupies the southernmost distribution for the genus, the presence of EFNs in this species may be the remnant of a mutualistic interaction with the ants in tropical regions, where the abundance and diversity of ants and herbivores are comparatively higher. To draw accurate patterns in the genus Croton, further investigations in other species from tropical regions of South America are needed.

The objective of this article is to apply the Design of Experiments technique along with the Discrete Events Simulation technique in an automotive process. The benefits of the design of experiments in simulation include the possibility to improve the performance in the simulation process, avoiding trial and error to seek solutions. The methodology of the conjoint use of Design of Experiments and Computer Simulation is presented to assess the effects of the variables and its interactions involved in the process. In this paper, the efficacy of the use of process mapping and design of experiments on the phases of conception and analysis are confirmed.

Mutualistic associations shape the evolution in different organism groups. The association between the leaf-cutter ant Atta sexdens and the basidiomycete fungus Leucoagaricus gongylophorus has enabled them to degrade starch from plant material generating glucose, which is a major food source for both mutualists. Starch degradation is promoted by enzymes contained in the fecal fluid that ants deposit on the fungus culture in cut leaves inside the nests. To understand the dynamics of starch degradation in ant nests, we purified and characterized starch degrading enzymes from the ant fecal fluid and from laboratory cultures of L. gongylophorus and found that the ants intestine positively selects fungal α-amylase and a maltase likely produced by the ants, as a negative selection is imposed to fungal maltase and ant α-amylases. Selected enzymes are more resistant to catabolic repression by glucose and proposed to structure a metabolic pathway in which the fungal α-amylase initiates starch catalysis to generate byproducts which are sequentially degraded by the maltase to produce glucose. The pathway is responsible for effective degradation of starch and proposed to represent a major evolutionary innovation enabling efficient starch assimilation from plant material by leaf-cutters. © 2013 Elsevier Ltd.

The sugarcane borer, Diatraea saccharalis (F.) (Lepidoptera: Crambidae), is a key pest of sugarcane (Saccharum spp.). While damage caused by this pest has increased in the past 20 yr, studies investigating the insect-plant interactions are still lacking. Moreover, there is no information about the consequences of borer damage on the parameters of sugar quality. Therefore, two field experiments were performed during the 2010 and 2011 growing seasons in Brazil to compare the raw material and sugar quality of SP80-3280 sugarcane plants with and without the sugarcane borer. Plants were protected within screen cages and infested weekly during the 2010 and 2011 seasons, using egg masses starting at the second and third internode stage. At harvest, 25.77 and 19.01% of the internodes were bored by larvae (infestation intensity, II) in the first and second seasons, respectively. There was no correlation between the borer gallery total volume and II. The fiber content significantly increased with increasing II. The stalk biometric parameters, such as length, diameter, and yield, were not correlated with II. The sucrose yield significantly decreased with increasing II. Consequently, sugar yield losses were estimated at 8.83 and 19.80% per 1% bored internode for the first and second seasons, respectively. The concentration of phenolic compounds increased, and unclarified juice color quality decreased, with increasing II. Significant differences were detected in the quality of the sugar. These results should be confirmed for other sugarcane cultivars and incorporated into an economic injury level to enhance decision-making strategies for borer management. © 2013 by the American Society of Agronomy, 5585 Guilford Road, Madison, WI 53711. The silverleaf whitefly Bemisia tabaci (Genn.) biotype B (Hemiptera: Aleyrodidae) is an economically important pest of tomatoes Solanum lycopersicum (L.), causing irregular ripening on fruits and transmitting several plant pathogenic geminiviruses. The management of this pest is commonly based on repetitive spraying with synthetic pesticides, causing serious environmental damages and increase of resistance by insect population. In the present study, essential oils from the leaves of Artemisia camphorata Vill., Ageratum conyzoides L., Foeniculum vulgare Mill., Lippia alba (Mill.) N. E. Br., Plectranthus neochilus Schltr., and Tagetes erecta L. were investigated for their possible repellent and oviposition-deterrent effects against B. tabaci biotype B on tomato. In a multi-choice assay, P. neochilus essential oil was the most active repellent and oviposition deterrent. Essential oils of A. conyzoides and T. erecta significantly deterred the female B. tabaci biotype B from laying eggs on treated tomato leaflets compared with the control. (E)-Caryophyllene (30.67 %) and the monoterpenes α-pinene (15.02 %) and α-thujene (11.70 %) were identified as the major constituents of the essential oil of P. neochilus. Our findings demonstrated the potential of essential oil of P. neochilus and other oils in the reduction of settlement and oviposition of B. tabaci biotype B on tomato. © 2012 Springer-Verlag Berlin Heidelberg.

Food webs have been used in order to understand the trophic relationship among organisms within an ecosystem, however the extension by which sampling efficiency could affect food web responses remain poorly understood. Still, there is a lack of long-term sampling data for many insect groups, mainly related to the interactions between herbivores and their host plants. In the first chapter, I describe a source food web based on the Senegalia tenuifolia plant by identifying the associated insect species and the interactions among them and with this host plant. Furthermore, I check for the data robustness from each trophic level and propose a cost-efficiently methodology. The results from this chapter show that the collected dataset and the methodology presented are a good tool for sample most insect richness of a source food web. In total the food web comprises 27 species belonging to four trophic levels. In the second chapter, I demonstrate the temporal variation in the species richness and abundance from each trophic level, as well as the relationship among distinct trophic levels. Moreover, I investigate the diversity patterns of the second and third trophic level by assessing the contribution of alfa and beta-diversity components along the years. This chapter shows that in our system the parasitoid abundance is regulated by the herbivore abundances. Besides, the species richness and abundances of the trophic levels vary temporally. It also shows that alfa-diversity was the diversity component that most contribute to the herbivore species diversity (2nd trophic level), while the contribution of alfa- and beta-diversity changed along the years for parasitoid diversity (3rd level). Overall, this dissertation describes a source food web and bring insights into some food web challenges related to the sampling effort to gather enough species from all trophic levels. It also discuss the relation among communities associated with distinct trophic levels and their temporal variation and diversity patterns. Finally, this dissertation contributes for the world food web database and in understanding the interactions among its trophic levels and each trophic level pattern along time and space
