The goal of the present study was to investigate possible differences towards strenght recovery measured by different methods and running economy (RE), after one session of downhill running (DR), in order to induce muscle damage (MD). Our hypothesis is that the strenght recovery measured by jumps is more alike RE markers, due to the measures' specificity. Ten male subjects, ages 18 to 30, apparently healthy, without any experience with strenght training at least 6 months before the tests took place in this study. After going to the lab twice (to familiarize themselves with the protocols, and determine their maximum oxygen consumption/running speeds), they went to the lab five times in the following week, in order to collect all the variables before, immediately after, and 24, 48, 72 and 96 hours in. The variables obtained were: stride frequency (SF), stride lenght (SL), oxygen volume (VO2), carbon dioxide volume (VCO2), ventilation (VE), lactate blood concentration ([Lac]), isometric peak torque (IPT), subjective pain perception (SPP), effort perception (EP), medial-portion circumference (CIR), knee movement extent (EXT), torque development rate (TDR), counter movement jump and squat jump height (HCM and HS), strenght development rate of both jumps (SDCM and SDS), and maximum soil strenght reaction (SRCM and SRS). Changes over time in all variables were verified by one way variancy analysis. Differences between the strenght measures were verified by two way variancy analysis. When significant effects were verified, Tuckey's post-hoc were applied. The significancy level taken on this study was p < 0,05. Of all indirect muscle damage markers, IPT, SPP and TDR were the only ones in which ocurred significant changes. We couldn't find the moments where this happened for TDR with the post-hoc used. Several methods are used towards delayed onset muscle soreness appraisal. This study's goal was to investigate, amongst three commonly adopted methods, which one would be the most effective (the one that shows higher values) in the quantification of this phenomenon. 10 male subjects, of age 22.8 ± 3.1 years old, weight 80 ± 12.4 kg, height 174 ± 0.07 cm, without recent experience with strength training (lower limbs) and/or running took place in this study. All subjects carried out a muscle damage induction protocol, which consisted of 30 minute downhill running (-16% or -9.09º) at 80% speed compared to their maximum oxygen consumption. Muscle damage determinants such as isometric peak torque, knee joint range of motion and circumference of the medial portion of the thight were measured before, during, 24, 48, 72 and 96 hours after downhill running. The subjective pain perception was measured simultaneously with the other determinants through three different tests: sitting on and getting up of a chair; climbing and descending from a 45cm step; and self thigh palpation. After going through all subjective pain perception tests, the subjects filled out a visual analog scale with their perception of pain. Muscle damage changes over time were compared through variance analysis (ANOVA) one way for repeated measures. Subjective pain perception values obtained in all three different tests were compared through two way ANOVAs for repeated numbers. The significance level adopted in this study was z ≤ 0.05. The results showed that the step test was the on which better evaluated the delayed onset muscle soreness. No significant differences were found through the ADM and CIR recovery markers. Downhil running determined reduced of PTI (~22.4%). The sol-gel process is a method for obtaining vitreous or vitro-ceramic materials which, are prepared a sol and by drying the liquid phase. This technique has been used extensively for the preparation of glassy gels, films, fibers and particles from the hydrolysis and polycondensation reactions of metal alkoxides. The usual methods for drying are: evaporation drying (xerogels), freeze drying (criogéis) and via supercritical CO2 extraction (aerogels). In the present work, we studied the preparation of silica gels by the sol-gel process from the hydrolysis of alkoxides tetraethylorthosilicate (TEOS) and 3-glycidoxypropyltrimethoxysilane (GPTS). The hydrolysis was promoted from GPTS and TEOS in proportion (4: 1) under acidic conditions. The hydrolysis reaction was promoted inside a Becker at room temperature. After hydrolyses the prepared sol had pH 2, and kept under mechanical agitation for a period of 1 hour. In order to accelerate the polycondensation reaction, the pH was corrected to a value near 5 by slowly adding NH4OH. Then the sols were leaked in sealed polycarbonate containers and maintained for 20 days at 40°C for gelation. Silica aerogels were prepared via supercritical CO2 extraction of the wet gel at temperature and pressure higher than 31°C and 74 atm, in an autoclave specially developed for the process. The structural characteristics were studied in the dry gel (aerogel). Aerogels were then characterized by nitrogen adsorption and small angle light scattering. The nitrogen adsorption data were analyzed for the determination of the BET specific surface (SBET), the total pore volume Vp, the pore mean size (lP=4Vp/SBET), the particle mean size (lS) and the pore size distribution (PSD). The purpose of this study was to evaluate the visual outcome of chronic occupational exposure to a mixture of organic solvents by measuring color discrimination, achromatic contrast sensitivity and visual fields in a group of gas station workers. We tested 25 workers (20 males) and 25 controls with no history of chronic exposure to solvents (10 males). All participants had normal ophthalmologic exams. Subjects had worked in gas stations on an average of 9.6 +/- 6.2 years. Color vision was evaluated with the Lanthony D15d and Cambridge Colour Test (CCT). Visual field assessment consisted of white-on-white 24-2 automatic perimetry (Humphrey II-750i). Contrast sensitivity was measured for sinusoidal gratings of 0.2, 0.5, 1.0, 2.0, 5.0, 10.0 and 20.0 cycles per degree (cpd). Results from both groups were compared using the Mann-Whitney U test. The number of errors in the D15d was higher for workers relative to controls (p<0.01). Their CCT color discrimination thresholds were elevated compared to the control group along the protan, deutan and tritan confusion axes (p<0.01), and their ellipse area and ellipticity were higher (p<0.01). Genetic analysis of subjects with very elevated color discrimination thresholds excluded congenital causes for the visual losses. Automated perimetry thresholds showed elevation in the 9 degrees, 15 degrees and 21 degrees of eccentricity (p<0.01) and in MD and PSD indexes (p<0.01). Contrast sensitivity losses were found for all spatial frequencies measured (p<0.01) except for 0.5 cpd. Significant correlation was found between previous working years and deutan axis thresholds (rho = 0.59; p<0.05), indexes of the Lanthony D15d (rho = 0.52; p<0.05), perimetry results in the fovea (rho = -0.51; p<0.05) and at 3, 9 and 15 degrees of eccentricity (rho = -0.46; p<0.05). Extensive and diffuse visual changes were found, suggesting that specific occupational limits should be created.

Pericyte perivascular cells, believed to originate mesenchymal stem cells (MSC), are characterized by their capability to differentiate into various phenotypes and participate in tissue reconstruction of different organs, including the brain. We show that these cells can be induced to differentiation into neural-like phenotypes. For these studies, pericytes were obtained from aorta ex-plants of Sprague-Dawley rats and differentiated into neural cells following induction with trans retinoic acid (RA) in serum-free defined media or differentiation media containing nerve growth and brain-derived neuronal factor, B27, N2, and IBMX. When induced to differentiation with RA, cells express the pluripotency marker protein stage-specific embryonic antigen-1, neural-specific proteins beta 3-tubulin, neurofilament-200, and glial fibrillary acidic protein, suggesting that pericytes undergo differentiation, similar to that of neuroectodermal cells. Differentiated cells respond with intracellular calcium transients to membrane depolarization by KCl indicating the presence of voltage-gated ion channels and express functional N-methyl-D-aspartate receptors, characteristic for functional neurons. The study of neural differentiation of pericytes contributes to the understanding of induction of neuroectodermal differentiation as well as providing a new possible stem-cell source for cell regeneration therapy in the brain. Overstimulation of the glutamatergic system (excitotoxicity) is involved in various acute and chronic brain diseases. Several studies support the hypothesis that guanosine-5'-monophosphate (GMP) can modulate glutamatergic neurotransmission. The aim of this study was to evaluate the effects of chronically administered GMP on brain cortical glutamatergic parameters in mice. Additionally, we investigated the neuroprotective potential of the GMP treatment submitting cortical brain slices to oxygen and glucose deprivation (OGD). Moreover, measurements of the cerebrospinal fluid (CSF) purine levels were performed after the treatment. Mice received an oral administration of saline or GMP during 3 weeks. GMP significantly decreases the cortical brain glutamate binding and uptake. Accordingly, GMP reduced the immunocontent of the glutamate receptors subunits, NR2A/B and GluR1 (NMDA and AMPA receptors, respectively) and glutamate transporters EAAC1 and GLT1. GMP treatment significantly reduced the immunocontent of PSD-95 while did not affect the content of Snap 25, GLAST and GFAP. Moreover, GMP treatment increased the resistance of neocortex to OGD insult. The chronic GMP administration increased the CSF levels of GMP and its metabolites. Altogether, these findings suggest a potential modulatory role of GMP on neocortex glutamatergic system by promoting functional and plastic changes associated to more resistance of mice neocortex against an in vitro excitotoxicity event.

The configuration and the timing of assembly and break-up of Columbia are still matter of debate. In order to improve our knowledge about the Mesoproterozoic evolution of Columbia, a paleomagnetic study was carried out on the 1420 Ma Indiavai mafic intrusive rocks that crosscut the polycyclic Proterozoic basement of the SW Amazonian Craton, in southwestern Mato Grosso State (Brazil). Alternating field and thermal demagnetization revealed south/southwest ChRM directions with downward inclinations for sixteen analyzed sites. These directions are probably carried by SD/PSD magnetite with high coercivities and high unblocking temperatures as indicated by additional rock magnetic tests, including thermomagnetic data, hysteresis data and the progressive acquisition of isothermal remanent magnetization. Different stable magnetization components isolated in host rocks from the basement 10 km NW away to the Indiavai intrusion, further support the primary origin of the ChRM. A mean of the site mean directions was calculated at Dm = 209.8 degrees, Im = 50.7 degrees (alpha(95) = 8.0 degrees, K = 22.1), which yielded a paleomagnetic pole located at 249.7 degrees E, 57.0 degrees S (A(95) = 8.6 degrees). The similarity of this pole with the recently published 1420 Ma pole from the Nova Guarita dykes in northern Mato Grosso State suggests a similar tectonic framework for these two sites located 600 km apart, implying the bulk rigidity of the Rondonian-San Ignacio crust at that time. Furthermore these data provide new insights on the tectonic significance of the 1100-1000 Ma Nova Brasilandia belt-a major EW feature that cuts across the basement rocks of this province, which can now be interpreted as intracratonic, in contrast to previous interpretation. From a global perspective, a new Mesoproterozoic paleogeography of Columbia has been proposed based on comparison of these 1420 Ma poles and a 1780 Ma pole from Amazonia with other paleomagnetic poles of similar age from Baltica and Laurentia, a reconstruction in agreement with geological correlations. The Nuna supercontinent was probably assembled in the Paleoproterozoic, but its paleogeography and the timing for its demise are stills matter of debate. A paleomagnetic and geochronological study carried out on the Mesoproterozoic Nova Guarita dyke swarm (northern Mato Grosso State, SW Amazonian Craton) provides additional constraints on the duration of this supercontinent. Paleomagnetic AF and thermal treatment revealed south/southwest (northeast) magnetic directions with downward (upward) inclinations for 19 analyzed sites. These directions are carried by PSD magnetite with high unblocking temperatures as indicated by additional magnetic tests, including thermomagnetic curves, hysteresis loops and the progressive acquisition of isothermal remanence in selected samples. A positive contact test with the host granite in one of the studied dykes further attests to the primary origin of the characteristic magnetic component. A mean site direction was calculated at D-m = 220.5 degrees, I-m = 45.9 degrees (alpha(95) = 6.5 degrees, K = 27.7), which yielded a paleomagnetic pole located at 245.9 degrees E, 47.9 degrees S (A(95) = 7.0 degrees). Ar-40/Ar-39 dating carried out on biotites from four analyzed dykes yielded well-defined plateau ages with a mean of 1418.5 +/- 3.5 Ma. The Nova Guarita pole precludes a long-lived Nuna configuration in which Laurentia, Baltica, North China, and Amazonia formed a long and continuous block as previously proposed for the Paleoproterozoic. It is nevertheless fully compatible with a SAMBA (Amazonia-Baltica) link at Mesoproterozoic times. (C) 2011 Elsevier B.V. All rights reserved.

PURPOSE: To investigate the possible effect of aspherical or yellow tinted intraocular lens (IOL) on contrast sensitivity and blue-on-yellow perimetry. METHODS: This prospective randomized bilateral double-masked clinical study included 52 patients with visually significant bilateral cataracts divided in two groups; 25 patients (50 eyes) received aspherical intraocular lens in one eye and spherical intraocular lens in the fellow eye; and 27 patients (54 eyes) received ultraviolet and blue light filter (yellow tinted) IOL implantation in one eye and acrylic ultraviolet filter IOL in the fellow eye. The primary outcome measures were contrast sensitivity and blue-on-yellow perimetry values (mean deviation [MD] and pattern standard deviation [PSD]) investigated two years after surgery. The results were compared intra-individually. RESULTS: There was a statistically significant between-group (aspherical and spherical intraocular lens) difference in contrast sensitivity under photopic conditions at 12 cycles per degree and under mesopic conditions at all spatial frequencies. There were no between-group significant differences (yellow tinted and clear intraocular lens) under photopic or mesopic conditions. There was no statistically significant difference between all intraocular lens in MD or PSD. CONCLUSION: Contrast sensitivity was better under mesopic conditions with aspherical intraocular lens. Blue-on-yellow perimetry did not appear to be affected by aspherical or yellow tinted intraocular lens. Further studies with a larger sample should be carried out to confirm or not that hypotheses.

Assessment of brain connectivity among different brain areas during cognitive or motor tasks is a crucial problem in neuroscience today. Aim of this research study is to use neural mass models to assess the effect of various connectivity patterns in cortical EEG power spectral density (PSD), and investigate the possibility to derive connectivity circuits from EEG data. To this end, two different models have been built. In the first model an individual region of interest (ROI) has been built as the parallel arrangement of three populations, each one exhibiting a unimodal spectrum, at low, medium or high frequency. Connectivity among ROIs includes three parameters, which specify the strength of connection in the different frequency bands. Subsequent studies demonstrated that a single population can exhibit many different simultaneous rhythms, provided that some of these come from external sources (for instance, from remote regions). For this reason in the second model an individual ROI is simulated only with a single population. Both models have been validated by comparing the simulated power spectral density with that computed in some cortical regions during cognitive and motor tasks. Another research study is focused on multisensory integration of tactile and visual stimuli in the representation of the near space around the body (peripersonal space). This work describes an original neural network to simulate representation of the peripersonal space around the hands, in basal conditions and after training with a tool used to reach the far space. The model is composed of three areas for each hand, two unimodal areas (visual and tactile) connected to a third bimodal area (visual-tactile), which is activated only when a stimulus falls within the peripersonal space. Results show that the peripersonal space, which includes just a small visual space around the hand in normal conditions, becomes elongated in the direction of the tool after training, thanks to a reinforcement of synapses.

Charakterisierung synapsenassoziierter Proteine des Haushuhns(Gallus gallus domesticus) Die Familie der synapsenassoziierten Proteine (SAP) umfaßt bei Säugern vier Proteine: SAP90 (=PSD-95), SAP97, SAP102 (=PSD-93) und Chapsyn110. Die Proteine enthalten charakteristischerweise drei PDZ-Domänen, eine SH3-Domäne und eine GK-Domäne über die sie mit anderen Proteinen interagieren können. SAP können so Verbindungen zwischen Neurotransmitterrezeptoren und Signaltransduktionsmolekülen sowie dem Zytoskelett herstellen.In dieser Arbeit wurden die synapsenassoziierten Proteine des Huhns charakterisiert. Die cDNAs von SAP90, SAP97 und Chapsyn110 wurden sequenziert. Die cDNA von SAP102 wurde teilweise sequenziert. Die Analyse genomischer DNA durch PCR ergab, daß die SAP90- und SAP97-mRNA von einem Gen transkribiert werden. Die mRNA-Verteilung von SAP90, SAP97 und Chapsyn110 im Gehirn einen Tag alter Küken wurde mit in situ Hybridisierung untersucht. Die Verteilung der SAP90-mRNA und von NMDA-Rezeptoren im Gehirn des Huhns ist sehr ähnlich. Weiterhin wurde bei Küken untersucht, inwieweit SAP bei der Prägung eine Rolle spielen. Der relative mRNA-Gehalt von SAP90, SAP97 und Chapsyn110 wurde 30 Minuten, 5 Stunden und 10 Stunden nach einer akustische Prägung der Küken gemessen. Fünf Stunden nach akustischer Prägung war der Gehalt der SAP90-mRNA, im anterioren lateralen Hyperstriatum ventrale um 13% erhöht. Der mRNA-Gehalt in anderen Regionen und der anderen SAP-Gene war unverändert.

Die vorliegende Arbeit beschäftigt sich vorwiegend mit Detektionsproblemen, die bei Experimenten zur Chemie der Transactiniden mit dem schnellen Flüssig-Flüssig-Extraktionssystem SISAK auftraten. Bei diesen Experimenten wird als Detektionsmethode die Flüssigszintillationsspektroskopie (LSC) eingesetzt. Es werden Szintillationspulse registriert, die für das verursachende Teilchen charakteristische Formen zeigen, die unterschieden werden müssen. Am Beispiel der Auswertung des SISAK-Experimentes zur Chemie des Rutherfordiums vom November 1998 wurde gezeigt, dass es mit den herkömmlichen Verfahren zur Pulsformdiskriminierung nicht möglich ist, die aus dem Zerfall der Transactiniden stammenden alpha-Ereignisse herauszufiltern. Ursache dafür ist ein hoher Untergrund, der in erster Linie von beta/gamma-Teilchen, Spaltfragmenten und pile ups verursacht wird. Durch die Verfügbarkeit von Transientenrecordern ergeben sich neue Möglichkeiten für eine digitale Pulsformdiskriminierung. In dieser Arbeit wird erstmals die Methode der digitalen Pulsformdiskriminierung mit künstlichen neuronalen Netzen (PSD-NN) vorgestellt. Es wurde im Zuge der Auswertung des SISAK-Experimentes vom Februar 2000 gezeigt, dass neuronale Netze in der Lage sind, Pulsformen automatisch richtig zu klassifizieren. Es ergeben sich nahezu untergrundfreie alpha-Flüssigszintillationsspektren. Es werden Vor- und Nachteile der neuen Methode diskutiert. Es ist dadurch möglich geworden, in SISAK-Experimenten Transactinidenatome anhand ihres Zerfalls eindeutig zu charakterisieren. Das SISAK-System kann somit bei Experimenten zum Studium des chemischen Verhaltens von Transactiniden in flüssiger Phase eingesetzt werden.____

Das Protein Cytochrom c Oxidase (CcO) ist ein Enzym der mitochondrialen Atmungskette. Als letzter Komplex (Komplex IV) einer Elektronentransportkette katalysiert sie die Reduktion von molekularem Sauerstoff zu Wasser. Hierbei werden Elektronen von Cytochrom c (Cc) in das Enzym geleitet. Die durch den Redoxprozess freiwerdende freie Enthalpie wird dazu genutzt, einen Protonengradienten über die innere Mitochondrien-Membran aufzubauen. Die zurückwandernden Protonen treiben in der ATP-Synthase die Produktion von Adenosintriphosphat (ATP) an, dem universellen Energieträger in lebenden Organismen. Gegenstand dieser Dissertation sind zeitaufgelöste ATR-FTIR-Messungen des direkten Elektronentransfers in die CcO. Das Protein wird hierzu orientiert auf einer Goldelektrode immobilisiert und in eine künstliche Membran rekonstituiert (Protein-tethered Bilayer Lipid Membrane, ptBLM). Das ptBLM-System wird hinsichtlich einer möglichst hohen Protein-Aktivität optimiert. Elektronen werden durch elektrochemische Anregung von der Elektrode in die CcO injiziert. Die Goldoberfläche wird auf die reflektierende Oberfläche eines Silizium-ATR-Kristalls aufgebracht. Durch die Präparation einer rauen Oberfläche (RMS-Rauigkeit ca. 5 nm) wird eine Verstärkung der IR-Absorption erreicht. Die mit den Ladungstransferprozessen einhergehenden Konformationsänderungen der die Redoxzentren umgebenden Gruppen (CONH-Gerüst und Aminosäure-Seitenketten) können durch Infrarot-Spektroskopie nachgewiesen werden. Phasensensitive Detektion (PSD) wird zur Rauschminderung eingesetzt, um Geschwindigkeitskonstanten für die Redox-Übergänge zu bestimmen. Im Bereich der Amid-I-Bande werden etliche Peaks identifiziert, die sich mit dem Redoxzustand des Proteins ändern. Für das CuA-Zentrum, welches als erstes der vier Redoxzentren der CcO reduziert wird, wird die schnellste Geschwindigkeitskonstante ks=4870/s ermittelt. Für das Häm a3-Zentrum wird eine Geschwindigkeitskonstante von ks=13,8/s ermittelt. Die Ergebnisse sind konsistent zu elektrochemischen und Raman-Spektroskopie-Experimenten, welche ebenfalls in unserer Gruppe durchgeführt wurden. Weitere Themen dieser Dissertation sind der Nachweis der Anwendbarkeit des ptBLM-Systems für andere Membranproteine (Beispiel: bakterielles photosynthetisches Reaktionszentrum) und der Einsatz des ATR-FTIR-Setups für verschiedene künstliche Membransysteme (Aktivitätsnachweis des OR5-Geruchsrezeptors in einer peptidgestützten Membran, Eigenschaften eines Oligoethylenglycol-Spacers).
