The objective of this work was to study the effect of root and foliar application of two commercial products containing amino acids from plant and animal origin on iron (Fe) nutrition of tomato seedlings cultivated in two nutrient media: lime and normal nutrient solutions. In the foliar-application experiment, each product was sprayed with 0.5 and 0.7 mL L–1 2, 7, 12, and 17 d after transplanting. In the root application experiment, 0.1 and 0.2 mL L–1 of amino acids products were added to the nutrient solutions. In both experiments, untreated control plants were included as well. Foliar and root application of the product containing amino acids from animal origin caused severe plant-growth depression and nonpositive effects on Fe nutrition were found. In contrast, the application of the product from plant origin stimulated plant growth. Furthermore, significantly enhanced root and leaf FeIII-chelate reductase activity, chlorophyll concentration, leaf Fe concentration, and FeII : Fe ratio were found in tomato seedlings treated with the product from plant origin, especially when the amino acids were directly applied to the roots. These effects were more evident in plants developed under lime-induced Fe deficiency. The positive results on Fe uptake may be related to the action of glutamic acid, the most abundant amino acid in the formulation of the product from plant origin.

Elevated jasmonic acid (JA) concentrations in response to herbivory can induce wounded plants to produce defences against herbivores. In laboratory and field experiments we compared the effects of exogenous JA treatment to two closely related cabbage species on the host-searching and oviposition preference of the diamondback moth (DBM), Plutella xylostella. JA-treated Chinese cabbage (Brassica campestris) was less attractive than untreated Chinese cabbage to ovipositing DBM, while JA-treatment of common cabbage (B. oleracea) made plants more attractive than untreated controls for oviposition by this insect. Similar effects were observed when plants of the two species were damaged by DBM larvae. In the absence of insect-feeding, or JA application, Chinese cabbage is much more attractive to DBM than common cabbage. Inducible resistance therefore appears to occur in a more susceptible plant and induced susceptibility appears to occur in a more resistant plant, suggesting a possible balance mechanism between constitutive and inducible defences to a specialist herbivore.

Tomato (Lycopersicon esculentum L.) is the second most important vegetable crop worldwide and a key component in the so-called “Mediterranean diet”. In the Northeastern region of Portugal, local populations still prefer to consume traditional tomato varieties which they find very tasty and healthy, as they are grown using extensive farming techniques. A previous study of our research team described the nutritional value of the round (batateiro), long (comprido), heart (coração) and yellow (amarelo) tomato varieties [1], but the phenolic profile was unknown until now. Thus, the objective of this study was to characterize the phenolic profiles of these four tomato farmers’ varieties by using HPLC-DAD-ESI/MS and evaluate its antioxidant capacity through four in vitro assays based on different reaction mechanisms. A cis p-coumaric acid derivative was the most abundant compound in yellow and round tomato varieties, while 4-O-caffeolyquinic acid was the most abundant in long and heart varieties. The most abundant flavonoid was quercetin pentosylrutinoside in the four tomato varieties. Yellow tomato presented the highest levels of phenolic compounds, including phenolic acids and flavonoids, but the lowest antioxidant activity. In turn, the round tomato gave the best results in all the antioxidant activity assays. This study demonstrated that these tomato farmers’ varieties are a source of phenolic compounds, mainly phenolic acid derivatives [2], and possess high antioxidant capacity [1]; being thus key elements in the diet to prevent chronic degenerative diseases associated to oxidative stress, such as cancer and coronary artery disease.

Tomato is the second most important vegetable crop worldwide and a rich source of industrially interesting antioxidants. Hence, the microwave-assisted extraction of hydrophilic (H) and lipophilic (L) antioxidants from a surplus tomato crop was optimized using response surface methodology. The relevant independent variables were temperature (T), extraction time (t), ethanol concentration (Et) and solid/liquid ratio (S/L). The concentration-time response methods of crocin and β-carotene bleaching were applied, since they are suitable in vitro assays to evaluate the antioxidant activity of H and L matrices, respectively. The optimum operating conditions that maximized the extraction were as follows: t, 2.25 min; T, 149.2 ºC; Et, 99.1 %; and S/L, 45.0 g/L for H antioxidants; and t, 15.4 min; T, 60.0 ºC; Et, 33.0 %; and S/L, 15.0 g/L for L antioxidants. This industrial approach indicated that surplus tomatoes possess a high content of antioxidants, offering an alternative source for obtaining natural value-added compounds. Additionally, by testing the relationship between the polarity of the extraction solvent and the antioxidant activity of the extracts in H and L media (polarity-activity relationship), useful information for the study of complex natural extracts containing components with variable degrees of polarity was obtained.

The production of natural extracts requires suitable processing conditions to maximize the preservation of the bioactive ingredients. Herein, a microwave-assisted extraction (MAE) process was optimized, by means of response surface methodology (RSM), to maximize the recovery of phenolic acids and flavonoids and obtain antioxidant ingredients from tomato. A 5-level full factorial Box-Behnken design was successfully implemented for MAE optimization, in which the processing time (t), temperature (T), ethanol concentration (Et) and solid/liquid ratio (S/L) were relevant independent variables. The proposed model was validated based on the high values of the adjusted coefficient of determination and on the non-significant differences between experimental and predicted values. The global optimum processing conditions (t=20 min; T=180 ºC; Et=0 %; and S/L=45 g/L) provided tomato extracts with high potential as nutraceuticals or as active ingredients in the design of functional foods. Additionally, the round tomato variety was highlighted as a source of added-value phenolic acids and flavonoids.

Em Latossolo Vermelho-Amarelo textura arenosa, do submedio Sao Francisco, avaliou-se a dispnibilidade de fosforo no solo extraido pelos extratores de Mehlich e Bray 1 e estimaram-se niveis adequados de adubacao fosfatada para o tomateiro rasteiro (Lycopersicum esculentum Mill) no periodo de 1982 a 1983. Diferentes niveis de fosforo no solo foram criados atraves da adicao previa de quantidades crescentes de fosfato sob a forma de superfosfato triplo. Obtiveram-se correlacoes significativas dos teores de fosforo no solo com os niveis de fosfato aplicado e a produtividade da cultura. Os teores de P no solo obtidos pelos extratores de Mehlich e Bray 1 foram classificados, respectivamente, nos seguintes niveis: muito baixo, inferiores a 5 ppm e 7 ppm; baixo, de 5 ppm a 10 ppm e 7 ppm a 12 ppm; medio, de 11 ppm a 16 ppm e 13 ppm a 18 ppm; alto, de 17 ppm a 29 ppm e 19 ppm a 31 ppm; e muito alto, superiores a 19 ppm e 31ppm. As doses estimadas de fosforo a serem aplicadas ao solo em sulco para obter a produtividade otima esperada seriam: 120, 100, 70, 30 e 0 kg/ha de P2O5 para os niveis muito baixo, baixo, medio, alto e muito alto, respectivamente.

Visando obter informações sobre as melhores épocas de plantio e verificar o comportamento de quatro cultivares de tomateiro (Lycopersicon esculentum L.) - Rossol, IPA 1, IPA 2 e IPA 3 -, foram instalados ensaios em solos aluviais da estação experimental do perímetro irrigado de São Gonçalo, em Souza, PB. Os trabalhos de campo foram iniciados em janeiro de 1981 e concluidos em maio de 1982. A cada mês era instalado um enaio. Foram avaliadas, em cada cultivar, nas diferentes épocas de plantio, as seguintes características: produtividade, incidência de podridão estilar, peso médio de frutos e incidência de pragas e doenças. Não houve diferença de produção entre as cultivares, dentro das diferentes épocas de plantio. As melhores épocas de plantio para a região foram os meses de fevereiro, março, abril e maio- As menores produções obtidas foram as dos plantios realizados nos meses de setembro, outubro, novembro, dezembro e janeiro.

A gene encoding an elongation factor LeEF-Tsmt that participates in the protein synthesis process in mitochondria shows strong expression in ripening fruit as compared to other organs. It is strongly up-regulated during the first stages of the ripening process in parallel with the climacteric rise in respiration. LeEF-Tsmt expression is stimulated by ethylene, wounding and high temperature but ethylene-insensitive mutants exhibit normal expression. Transgenic fruit have been generated in which LeEF-Tsmt has been constitutively up- and down-regulated. Surprisingly, altering the expression of the gene by genetic transformation with antisense and sense LeEF-Tsmt constructs did not affect the pattern of respiration and ethylene production during ripening and upon wounding. In addition, expression of the alternative oxidase gene which is known to play an important role in respiratory climacteric was not affected. Possible reasons for the absence of effect on respiration of variations of LeEF-Tsmt gene expression are discussed.

O oídio, causado pelo fungo Oidium neolycopersici, é uma doença comum do tomateiro, sobretudo em condições de cultivo protegido. Para esclarecer a natureza da resistência a oídio avaliou-se o processo de infecção, através da histopatologia em diferentes genótipos de tomateiro: CNPH 416, CNPH 423, CNPH 1287 (Lycopersicon hirsutum), CNPH 0081 (L. esculentum var. cerasiforme), cv. Santa Cruz Kada e cv. Santa Clara (L. esculentum). Para isso, três discos foliares (da 3ª, 4ª e 5ª folha "verdadeira") de cada planta com 5 -7 folhas verdadeiras foram cortados e colocados em placas de Petri contendo ágar-água. Os discos foram inoculados a partir de micélio esporulante fresco desenvolvido em tomateiro suscetível e incubados a 19-22ºC, 4000 lx e fotoperíodo de 12 h. Os discos foram clareados em etanol aquecido e examinados microscopicamente 19 h, 8 e 9 dias após-inoculação para avaliar desenvolvimento de tubo germinativo, esporulação e severidade da doença, respectivamente. A germinação dos conídios sobre o tecido foliar não apresentou diferenças entre genótipos. A formação de hifa secundária, apressórios e haustórios por conídio germinado foram menores nos genótipos CNPH 1287 e 423, que também apresentaram menor esporulação e severidade da doença. Os genótipos de L. esculentum e L. esculentum var. cerasiforme apresentaram maior suscetibilidade ao oídio e CNPH 416 apresentou suscetibilidade intermediária. Assim, observou-se que a resistência a oídio de CNPH 1287 e 423 ficou evidenciada já desde as 19 horas após a inoculação, principalmente pela menor porcentagem de hifa secundária e número de apressórios e haustórios formados quando comparados com os genótipos suscetíveis.

Intra and interspecific variability was measured in the genus Lycopersicon for the traits: productivity rate (PR, total number of regenerated shoots/total number of cultures), regeneration percentage (%R, number of cultures regenerating shoots or primordia/total number of cultures) and callus percentage (%C, number of cultures only producing callus/total number of cultures). Leaf explants from various genotypes of L. esculentum, L. esculentum var. cerasiforme, L. pimpinellifolium and L. peruvianum were placed on Murashige and Skoog (Physiol. Plant. 15: 473-493, 1962) medium + 0.175 mg/l IAA + 2.25 mg/l BA. Significant differences among species and among genotypes within the same species were found, while genotypes from different species showed similar responses.

Three welding procedures used to rebuild worn shafts in sugar cane mills were analysed: two submerged arc welding processes and one flux cored arc welding (FCAW) process. Sliding wear tests were in accordance with ASTM G 77 standard, using rings of welding material, blocks of bronze SAE 67, and oil as lubricant. The worn surfaces of rings and blocks were analysed by scanning electron microscopy to determine the wear mechanisms. High contact pressure, high operating temperature, and low relative speed were applied in sliding wear tests to match the conditions in sugar cane mills. Transferred material and evidence of adhesive junctions were detected. Additionally, hardened fragments produced abrasive grooves on the worn surfaces. The welding deposits that presented strong adhesion on the worn surface showed higher mass loss than the materials that presented more abrasive characteristics. Plastic mechanical properties were measured and related to the mass loss. The tested materials presented similar hardness but different yield stress and hardening coefficient. A relationship between wear, strain hardening coefficient, and yield stress was found. The welding deposit that presented the highest hardening coefficient showed the highest mass loss, with evidence of severe adhesion on the worn surface.

The release of xylose reductase (XR) from Candida mogii by cell disruption in a glass beads mill was studied using an experimental design. Statistical analysis of the results indicated that XR volumetric activity increases by using lower glass beads diameter and cell concentration, and by increasing the number of agitation pulses. Based on results attained in experimental design, assays were carried out aiming at the maximization of XR release. Under optimized conditions (300 mu m glass beads, 45 g/l of cell concentration and 50 pulses), the XR volumetric activity reach 0.683 U/ml. Disruption with glass beads showed to be the most efficient method for XR release when compared to sonication process. The highest specific activity (0.175 U/mg of protein) was found in extracts obtained by suspension freezing and thawing, which suggests that this method can be used as a selective process of cell disruption for XR release. Biopulping of Eucalyptus grandis wood chips with Phanerochaete chrysosporium RP-78 was evaluated under non-aseptic conditions in laboratory and mill wood-yard. The ability of P. chrysosporium to compete with indigenous fungi present in fresh wood chips was notorious under controlled laboratory experiments. A subsequent step involved an industrial test performed with 10-ton of fresh wood chips inoculated and maintained at 37 +/- 38 degrees C for 39 days in a biopulping pilot plant. Biotreated wood chips were pulped in a chemithermomechanical pulping mill. Net energy consumption during refining was 745 kWh ton(-1) and 610 kWh ton(-1) of processed pulp for control and biotreated wood chips, respectively. Accordingly, 18.5% net energy saving could be achieved. Biopulps contained lower shive content and had improved strength properties compared to control pulps. Tensile index improved from 25 +/- 1 N m g(-1) to 33.6 +/- 0.5 N m g(-1) and delamination strength from 217 +/- 19 kPa to 295 +/- 30 kPa.

Aiming at the optimization of the friction and yield stress coefficients an adaptation algorithm is proposed in this paper. This work studied the radiation resistance of Listeria monocytogenes and Salmonella species and the effect of irradiation on leaf flavonoid content and sensory acceptability of minimally processed arugula. Immersion in ozone-treated water reduced the analyzed microorganisms by 1 log. L. monocytogenes and Salmonella were not isolated from samples. Samples of this vegetable were inoculated with a cocktail of Salmonella spp. and L. monocytogenes and exposed to gamma irradiation. D-10 values for Salmonella ranged from 0.16 to 0.19 kGy and for L. monocytogenes from 0.37 to 0.48 kGy. Kaempferol glycoside levels were 4 and ca. 3 times higher in samples exposed to 1 and 2 kGy, respectively, than in control samples. An increase in quercetin glycoside was also observed mainly in samples exposed to 1 kGy. In sensory evaluation, arugula had good acceptability, even after exposure to 2 and 4 kGy. These results indicate that irradiation has potential as a practical processing step to improve the safety of arugula.
