BACKGROUND. Maternal genetics and feeding before and during pregnancy, different maternal metabolic pathologies, as well as nutrient intakes of newborns in their first months of life may be involved in the obesity aetiology and its long-term consequences. The possible role of these and others factors, the mechanisms and the effects on the metabolism, and the development of this disease need further research. OBJECTIVE. To acquire more knowledge about foetal adipose tissue development and the influence of genetic, dietetic and environmental factors on the risk to suffer from obesity. METHODOLOGY. Four study groups have been established with 30 pregnant women in each one: 1) control group; 2) mothers with glucose intolerance/gestational diabetes; 3) women with low weight gain during pregnancy, and 4) women with overweight/obesity at the beginning of the pregnancy. The magnitudes to be studied are: 1) dietary intake; 2) life-style habits; 3) physical activity; 4) anthropometry and body composition; 5) haematological study; 6) biochemical study (lipid and metabolic biomarkers); 7) immune function profile related to nutritional status; 8) psychological profile; 9) genetic biomarkers, and 10) microbiological markers; all of them in relation to the development of the foetal adipose tissue in the first stages of life and the risk of suffering from obesity in the future. CONCLUSION. This project, coordinated by the Department of Paediatrics of the School of Medicine in the University of Granada, and with the collaboration of well-known and expert research groups, tries to contribute to the knowledge about the obesity aetiology in infancy and its subsequent development in later periods of life.

BACKGROUND Recent advances in characterizing the immune recovery of HIV-1-infected people have highlighted the importance of the thymus for peripheral T-cell diversity and function. The aim of this study was to investigate differences in immune reconstitution profiles after highly active antiretroviral therapy (HAART) between HIV-children and adults. METHODS HIV patients were grouped according to their previous clinical and immunological status: 9 HIV-Reconstituting-adults (HIV-Rec-adults) and 10 HIV-Reconstituting-children (HIV-Rec-children) on HAART with viral load (VL) or=500 cells/microL at least during 6 months before the study and CD4+ dependent peripheral T-cell expansion while in HIV-Rec-children thymic output could play a predominant role in immune reconstitution.

BACKGROUND Severe hypertriglyceridaemia due to chylomicronemia may trigger an acute pancreatitis. However, the basic underlying mechanism is usually not well understood. We decided to analyze some proteins involved in the catabolism of triglyceride-rich lipoproteins in patients with severe hypertriglyceridaemia. METHODS Twenty-four survivors of acute hypertriglyceridaemic pancreatitis (cases) and 31 patients with severe hypertriglyceridaemia (controls) were included. Clinical and anthropometrical data, chylomicronaemia, lipoprotein profile, postheparin lipoprotein lipase mass and activity, hepatic lipase activity, apolipoprotein C II and CIII mass, apo E and A5 polymorphisms were assessed. RESULTS Only five cases were found to have LPL mass and activity deficiency, all of them thin and having the first episode in childhood. No cases had apolipoprotein CII deficiency. No significant differences were found between the non-deficient LPL cases and the controls in terms of obesity, diabetes, alcohol consumption, drug therapy, gender distribution, evidence of fasting chylomicronaemia, lipid levels, LPL activity and mass, hepatic lipase activity, CII and CIII mass or apo E polymorphisms. However, the SNP S19W of apo A5 tended to be more prevalent in cases than controls (40% vs. 23%, NS). CONCLUSION Primary defects in LPL and C-II are rare in survivors of acute hypertriglyceridaemic pancreatitis; lipase activity measurements should be restricted to those having their first episode during childhood.

BACKGROUND It is known that mitochondria play an important role in certain cancers (prostate, renal, breast, or colorectal) and coronary disease. These organelles play an essential role in apoptosis and the production of reactive oxygen species; in addition, mtDNA also reveals the history of populations and ancient human migration. All these events and variations in the mitochondrial genome are thought to cause some cancers, including prostate cancer, and also help us to group individuals into common origin groups. The aim of the present study is to analyze the different haplogroups and variations in the sequence in the mitochondrial genome of a southern European population consisting of subjects affected (n = 239) and non-affected (n = 150) by sporadic prostate cancer. METHODOLOGY AND PRINCIPAL FINDINGS Using primer extension analysis and DNA sequencing, we identified the nine major European haplogroups and CR polymorphisms. The frequencies of the haplogroups did not differ between patients and control cohorts, whereas the CR polymorphism T16356C was significantly higher in patients with PC compared to the controls (p = 0.029). PSA, staging, and Gleason score were associated with none of the nine major European haplogroups. The CR polymorphisms G16129A (p = 0.007) and T16224C (p = 0.022) were significantly associated with Gleason score, whereas T16311C (p = 0.046) was linked with T-stage. CONCLUSIONS AND SIGNIFICANCE Our results do not suggest that mtDNA haplogroups could be involved in sporadic prostate cancer etiology and pathogenesis as previous studies performed in middle Europe population. Although some significant associations have been obtained in studying CR polymorphisms, further studies should be performed to validate these results.

Despite stringent requirements for drug development imposed by regulatory agencies, drug-induced liver injury (DILI) is an increasing health problem and a significant cause for failure to approve drugs, market withdrawal of commercialized medications, and adoption of regulatory measures. The pathogenesis is yet undefined, though the rare occurrence of idiosyncratic DILI (1/100,000–1/10,000) and the fact that hepatotoxicity often recurs after re-exposure to the culprit drug under different environmental conditions strongly points toward a major role for genetic variations in the underlying mechanism and susceptibility. Pharmacogenetic studies in DILI have to a large extent focused on genes involved in drug metabolism, as polymorphisms in these genes may generate increased plasma drug concentrations as well as lower clearance rates when treated with standard medication doses. A range of studies have identified a number of genetic variants in drug metabolism Phase I, II, and III genes, including cytochrome P450 (CYP) 2E1, N-acetyltransferase 2, UDP-glucuronosyltransferase 2B7, glutathione S-transferase M1/T1, ABCB11, and ABCC2, that enhance DILI susceptibility (Andrade et al., 2009; Agundez et al., 2011). Several metabolic gene variants, such as CYP2E1c1 and NAT2 slow, have been associated with DILI induced by specific drugs based on individual drug metabolism information. Others, such as GSTM1 and T1 null alleles have been associated with enhanced risk of DILI development induced by a large range of drugs. Hence, these variants appear to have a more general role in DILI susceptibility due to their role in reducing the cell's antioxidative capacity (Lucena et al., 2008). Mitochondrial superoxide dismutase (SOD2) and glutathione peroxidase 1 (GPX1) are two additional enzymes involved in combating oxidative stress, with specific genetic variants shown to enhance the risk of developing DILI

Se han sintetizado nueve alcoholes que incorporan diversos isómeros de los grupos piridil o quinolil y metil-o-carborano u o-carborano, 1-[n′-Piridil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (n′ = 2, R = H (2a); n′ = 3, R = Me (1c) y H (2c)), 1-[2′-6-metil-piridil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (R = Me (1b) y H (2b)), 1-[n′-Quinolil(hidroxi)metil]-2-R-1,2-dicarba-closo-dodecaboranos (n′ = 2, R = Me (1e) y H (2e); n′ = 4, R = Me (1f) y H (2f), mediante la adición de la sal de litio del metil-o-carborano u o-carborano a los aldehídos correspondientes. Los compuestos se han obtenido con rendimientos altos en la mayoría de los casos y han sido caracterizados mediante Resonancia Magnética Nuclear, por Espectroscopía Infrarroja y análisis elemental. Las estructuras cristalinas de siete de los nueve compuestos (1c, 1f, 2a, 2b, 2c, 2e y 2f) se han determinado mediante Cristalografía por Rayos X. Se han analizado las estructuras cristalinas de los todos los compuestos y se han comparado con las estructuras de los compuestos relacionados 1a, 1b, y 2a, obtenidas previamente en el grupo de Síntesis Inorgánica y Catálisis del ICMAB. Todas las estructuras muestran interacciones por puentes de hidrógeno O–H···N moderadamente fuertes. Posteriormente se ha sintetizado el derivado mesilado 4 (derivado del o-carborano) mediante la reacción del alcohol 2a con cloruro de mesitilo a temperatura ambiente, con rendimiento alto. El nuevo mesilato 4 se ha caracterizado mediante Resonancia Magnética Nuclear. El nuevo mesilato resulta un compuesto de partida muy versátil para la síntesis de nuevos derivados mediante la substitución del grupo mesilo por nucleófilos apropiados, como por ejemplo las aminas. El grupo ha publicado recientemente la síntesis de diaminas mediante la reacción de substitución del mesilato relacionado 3 (derivado del metil-o-carborano) con aminas, entre ellas la bencilamina. Se ha realizado por tanto la reacción de substitución del grupo mesilato en 4 por la bencilamina para obtener la diamina correspondiente. Los resultados preliminares de esta reacción muestran que si bien el grupo mesilato en 4 ha sido substituido, el clúster se degrada casi en su totalidad a un derivado tipo nido. Esto contrasta claramente con la reactividad del mesilato análogo 3 frente a la bencilamina, ya que en este caso se obtiene la diamina deseada y el clúster no se degrada apreciablemente bajo las mismas condiciones de reacción. Se han realizado estudios preliminares por RMN que muestran que la bencilamina substituye al grupo mesilo en 4 y posteriormente el clúster closo se degrada a una especie nido que no ha sido posible caracterizar totalmente hasta el momento.

Los problemas relacionados con el peso y la alimentación, incluyendo los trastornos de la conducta alimentaria (TCA) y la obesidad, se asocian con una amplia gama de trastornos físicos y mentales. La tasa de obesidad entre adultos y adolescentes se ha duplicado en los últimos 20 años, mientras que, paralelamente, también se ha registrado un aumento en la prevalencia de TCA. A pesar de que la obesidad y los TCA se tratan y estudian como patologías diferentes, diversos estudios han identificado factores de riesgo y protectores que son comunes entre ambas afectaciones. Sin embargo, pocos son los estudios que analizan conjuntamente factores neuropsicológicos, biológicos y ambientales, de manera a determinar las interacciones existentes entre estos aspectos.Objetivos:1) Explorar las diferencias clínicas (conducta alimentaria, actividad y sueño y personalidad), neuropsicológicas (funciones ejecutivas y de atención) y del sistema olfativo-sensorial entre los dos diferentes subtipos de TCA y la obesidad, al ser comparado entre los grupos; 2) Explorar la asociación entre funcionamiento neuropsicológico y características clínicas (conducta alimentaria y perfil de personalidad), IMC y sistema olfativo-sensorial entre los distintos grupos. Método: Estudio transversal de casos y controles. La muestra será dividida en 4 grupos: 2 de casos (30 bajo peso con TCA y 30 Obesas con TCA), formados por pacientes admitidos, tratados en la Unidad de Trastornos Alimentarios y diagnosticados según el DSM-IV; y 2 de controles (30 bajo peso sin TCA y 30 Obesas sin TCA). CRITERIOS DE INCLUSION: Ser mujer, tener entre 18-45 años, Cumplir criterios diagnósticos DSM-IV (en caso TCA). Instrumentos: 1)Neuropsicologicos: TCI-R, WCST, SCWT; 2)Antropométricos: TANITA MC-180 Multifrecuencia; 3)Olfato-sensoriales: SSOT, Taste Strips; 4)Actividad: ACTIWATCH 7; 5)Calidad del sueño: PSQI ,Escala EPWORTH.Variables socio-demográficas también serán recogidas durante la entrevista clínica. Todos los instrumentos serán aplicados en la misma unidad en una única entrevista exceptuando el ACTIWATCH que los sujetos deberán llevar durante 6 días.

Background: The objective was to investigate the association between BMI and single nucleotide polymorphisms previously identified of obesity-related genes in two Spanish populations. Forty SNPs in 23 obesity-related genes were evaluated in a rural population characterized by a high prevalence of obesity (869 subjects, mean age 46 yr, 62% women, 36% obese) and in an urban population (1425 subjects, mean age 54 yr, 50% women, 19% obese). Genotyping was assessed by using SNPlex and PLINK for the association analysis. Results: Polymorphisms of the FTO were significantly associated with BMI, in the rural population (beta 0.87, p-value <0.001). None of the other SNPs showed significant association after Bonferroni correction in the two populations or in the pooled analysis. A weighted genetic risk score (wGRS) was constructed using the risk alleles of the Tag-SNPs with a positive Beta parameter in both populations. From the first to the fifth quintile of the score, the BMI increased 0.45 kg/m2 in Hortega and 2.0 kg/m2 in Pizarra. Overall, the obesity predictive value was low (less than 1%). Conclusion: The risk associated with polymorphisms is low and the overall effect on BMI or obesity prediction is minimal. A weighted genetic risk score based on genes mainly acting through central nervous system mechanisms was associated with BMI but it yields minimal clinical prediction for the obesity risk in the general population.

Los interruptores moleculares son sistemas moleculares cuyas propiedades pueden ser reversiblemente alternadas entre dos estados diferentes, “On” y “Off”, como respuesta a la aplicación de un estímulo externo. El interruptor molecular fluorescente objetivo de éste trabajo responderá a estímulos fotoquímicos y se detectará por medidas de fluorescencia. En concreto, se ha llevado a cabo la síntesis y caracterización de las tres unidades funcionales que constituyen el interruptor molecular objetivo: dos fluoróforos de perilendiimida y un cromóforo azobencénico.

Actualmente no se disponen de marcadores biológicos específicos para la cáncer de próstata produciéndose en muchas ocasiones biopsias prostáticas innecesarias o un sobretratamientos para cánceres indolentes. Existen cada vez más un número mayor de publicaciones sobre cómo los polimorfismos de nucleótido simple (SNP) se relacionan con la susceptibilidad al cáncer de próstata o predecir con mayor precisión qué grado de agresividad adquiere la enfermedad. Se presenta una revisión bibliográfica de las investigaciones publicadas en PubMed desde el año 2000 hasta el 2012 que ponen de manifiesto la relación de los SNP con el riesgo a padecer cáncer de próstata y con sus características anatomopatológicas.

BACKGROUND Understanding of the genetic basis of type 2 diabetes (T2D) has progressed rapidly, but the interactions between common genetic variants and lifestyle risk factors have not been systematically investigated in studies with adequate statistical power. Therefore, we aimed to quantify the combined effects of genetic and lifestyle factors on risk of T2D in order to inform strategies for prevention. METHODS AND FINDINGS The InterAct study includes 12,403 incident T2D cases and a representative sub-cohort of 16,154 individuals from a cohort of 340,234 European participants with 3.99 million person-years of follow-up. We studied the combined effects of an additive genetic T2D risk score and modifiable and non-modifiable risk factors using Prentice-weighted Cox regression and random effects meta-analysis methods. The effect of the genetic score was significantly greater in younger individuals (p for interaction  = 1.20×10-4). Relative genetic risk (per standard deviation [4.4 risk alleles]) was also larger in participants who were leaner, both in terms of body mass index (p for interaction  = 1.50×10-3) and waist circumference (p for interaction  = 7.49×10-9). Examination of absolute risks by strata showed the importance of obesity for T2D risk. The 10-y cumulative incidence of T2D rose from 0.25% to 0.89% across extreme quartiles of the genetic score in normal weight individuals, compared to 4.22% to 7.99% in obese individuals. We detected no significant interactions between the genetic score and sex, diabetes family history, physical activity, or dietary habits assessed by a Mediterranean diet score. CONCLUSIONS The relative effect of a T2D genetic risk score is greater in younger and leaner participants. However, this sub-group is at low absolute risk and would not be a logical target for preventive interventions. The high absolute risk associated with obesity at any level of genetic risk highlights the importance of universal rather than targeted approaches to lifestyle intervention.

Durant diverses dècades les relacions entre la Unió Europea (UE) i Amèrica Llatina es caracteritzaven pel model tradicional de "ajuda sense comerç" fins els anys 1995, que amb acords de quarta generació impulsar el començament del nou instrument. Els acords d'associació (AdA) introdueixen com una novetat tres pilars (diàleg polític, de cooperació i comercial) i centren l'atenció en les relacions comercials, liberalitzen el comerç i estableixen un nou model d'associació biregional. Aquest treball de recerca analitza els AdA de la UE amb països o grups de països llatinoamericans i el seu impacte en les relacions comercials entre la UE i Amèrica Llatina.

BACKGROUND Multiple sclerosis (MS) is a neurodegenerative, autoimmune disease of the central nervous system. Genome-wide association studies (GWAS) have identified over hundred polymorphisms with modest individual effects in MS susceptibility and they have confirmed the main individual effect of the Major Histocompatibility Complex. Additional risk loci with immunologically relevant genes were found significantly overrepresented. Nonetheless, it is accepted that most of the genetic architecture underlying susceptibility to the disease remains to be defined. Candidate association studies of the leukocyte immunoglobulin-like receptor LILRA3 gene in MS have been repeatedly reported with inconsistent results. OBJECTIVES In an attempt to shed some light on these controversial findings, a combined analysis was performed including the previously published datasets and three newly genotyped cohorts. Both wild-type and deleted LILRA3 alleles were discriminated in a single-tube PCR amplification and the resulting products were visualized by their different electrophoretic mobilities. RESULTS AND CONCLUSION Overall, this meta-analysis involved 3200 MS patients and 3069 matched healthy controls and it did not evidence significant association of the LILRA3 deletion [carriers of LILRA3 deletion: p = 0.25, OR (95% CI) = 1.07 (0.95-1.19)], even after stratification by gender and the HLA-DRB1*15:01 risk allele.

Hasta el 2005 no hubo demasiados avances, más allá de los constantes contactos que han ido manteniendo las Conferencias Ministeriales UE-África y la elaboración de una Comunicación por parte de la Comisión al respecto de dicho diálogo. Dicha Comunicación pretendía atenuar los efectos de la anulación de la Cumbre sobre el recién nacido diálogo político, ofreciendo una imagen de continuidad, así como analizar los avances del diálogo en los ocho temas prioritarios establecidos por el Plan de El Cairo. De entre estos temas destacaba, dentro del apartado dedicado a la integración regional, el inicio de las negociaciones sobre los polémicos Acuerdos de Asociación Económica (AAE) entre la UE y las diversas agrupaciones regionales de los países ACP (unos acuerdos que entrarán en vigor a partir del 2008 y que han suscitado críticas por parte de algunos sectores en base a los perniciosos efectos para las economías africanas que puede conllevar la liberalización comercial total con Europa).
