O presente trabalho objetivou estudar a distribuição da reserva de K e Mg nas diferentes classes da fração areia de solos do Triângulo Mineiro, destacando-se o potencial de liberação de formas não-trocáveis e estruturais destes nutrientes para as plantas. A seleção do local de amostragem foi definida com base nas diferenças entre os materiais de origem (Grupo Bauru, Araxá, São Bento e Coberturas Detrítico-Lateríticas Terciárias), estádio de desenvolvimento dos solos e representação espacial das litologias. Amostras da fração areia dos solos foram separadas em areia leve (AL) e pesada (AP) com uso do bromofórmio (densidade 2,8 g cm-3) e, por meio de um conjunto de peneiras, a fração AL foi separada em cinco classes diferentes: areia muito grossa (AMG), areia grossa (AG), areia média (AM), areia fina (AF) e areia muito fina (AMF). Os teores totais e não-trocáveis de K e Mg foram obtidos após digestão das amostras com HF, HNO3 e H2SO4 concentrados e com HNO3 1 mol L-1 fervente, respectivamente. Na caracterização mineralógica da fração areia, utilizou-se a difração de raios-X. A fração areia dos solos é composta basicamente por quartzo, com predomínio de minerais leves e de menor tamanho (AF e AMF). Em geral, os difratogramas de raios-X da fração areia apresentaram apenas reflexões discretas de minerais primários fontes de nutrientes (MPFN), como mica e feldspato. Dentro da AL, os maiores teores de K e Mg totais foram advindos das frações mais finas (AF e AMF), tendo a fração AM apresentado a menor liberação de formas não-trocáveis destes nutrientes. Observou-se que solos com teores totais elevados de K e Mg na fração areia, geralmente, apresentaram maior capacidade de liberação de parte desses nutrientes para a solução do solo, atribuída à maior presença de MPFN. A fração areia dos solos originados de arenito da Formação Uberaba e de migmatito/micaxisto do Grupo Araxá apresentaram as maiores reservas e liberação de K e Mg.

Com vistas em avaliar o comportamento da linha do solo para diferentes classes de solos, este trabalho foi realizado, utilizando-se dados de reflectância por meio de um espectrorradiômetro em laboratório. Amostras representativas dos horizontes superficiais e subsuperficiais de 18 classes coletadas em uma área piloto no Município de Rafard, sudoeste de São Paulo, foram secas, peneiradas e acondicionadas para leitura espectral. Os dados foram tabulados, para simularem as bandas 3 e 4 do TM-Landsat e construção dos gráficos. Os resultados demonstraram que as linhas de solo obtidas para as amostras de ambas as camadas comportaram-se de maneira semelhante, estando dispostas ortogonalmente aos eixos de um gráfico, com valores de R² próximos a 1,0 para todas as classes avaliadas. Não se observou uma única linha de solo, embora os dados revelem que cada solo apresenta uma linha individual e característica. De modo complementar, destacou-se a utilização deste índice como método auxiliar na discriminação de classes de solos, uma vez que os solos com textura arenosa e com menores teores de Fe total discriminaram-se daqueles mais argilosos e com maiores teores de Fe, além de apresentarem valores de reflectância mais elevados. Por fim, constatou-se que a matéria orgânica não constituiu fator determinante no comportamento espectral em relação à linha do solo.

BACKGROUND: Diffuse large B-cell lymphomas (DLBCLs) arising in specific extranodal sites have peculiar clinicopathologic features. PATIENTS AND METHODS: We analyzed a cohort of 187 primary Waldeyer's ring (WR) DLBCLs retrieved from GELA protocols using anthracyclin-based polychemotherapy. RESULTS: Most patients (92%) had stage I-II disease. A germinal center B-cell-like (GCB) immunophenotype was observed in 61%, and BCL2 expression in 55%, of WR DLBCLs. BCL2, BCL6, IRF4 and MYC breakpoints were observed in, respectively, 3 of 42 (7%), 9 of 36 (25%), 2 of 26 (8%) and 4 of 40 (10%) contributive cases. A variable follicular pattern was evidenced in 30 of 68 (44%) large biopsy specimens. The 5-year progression-free survival (PFS) and the overall survival (OS) of 153 WR DLBCL patients with survival information were 69.5% and 77.8%, respectively. The GCB immunophenotype correlated with a better OS (P = 0.0015), while BCL2 expression predicted a worse OS (P = 0.037), an effect overcome by the GCB/non-GCB classification. Compared with matched nodal DLBCLs, WR DLBCLs with no age-adjusted international prognostic index factor disclosed a better 5-year PFS rate (77.5% versus 70.7%; P = 0.03). CONCLUSIONS: WR DLBCLs display distinct clinicopathologic features compared with conventional DLBCLs, with usual localized-stage disease, common follicular features and a high frequency of GCB immunophenotype contrasting with a low rate of BCL2 rearrangements. In addition, they seem to be associated with a better outcome than their nodal counterpart.

A região noroeste do Estado de Mato Grosso compreende paisagens de grande complexidade para amostragem de solo. Isso ocorre devido à contínua atividade geomorfológica, que contribuiu para a existência de diferentes classes pedológicas em pequenas distâncias. O trabalho foi desenvolvido em Juruena (MT), com o objetivo de identificar as classes pedológicas em microbacias sob floresta não-perturbada, por meio do estudo da variabilidade espacial da textura (relação textural) e cor (índice de avermelhamento) do solo, considerando-se ainda a altitude da paisagem. A variabilidade espacial da textura e da cor do solo foi determinada em amostras de 185 pontos georreferenciados em malha sistemática regular com espaçamento de 20 x 20 m, abrangendo quatro microbacias. Usando profundidades de 0-20 e 40-60 cm, foi possível distinguir e mapear as principais classes de solo ocorrentes na área de estudo até o segundo nível categórico do Sistema Brasileiro de Classificação de Solos, associadas ao fator relevo da paisagem. Também foi estabelecida uma correspondência bastante satisfatória entre os intervalos do índice de avermelhamento do horizonte diagnóstico e as cores das classes de solo. As microbacias apresentaram as classes Plintossolos e Argissolos (com caráter plíntico) em altitudes inferiores a 280 m e Latossolos nas cotas superiores. Dessa forma, ficou evidenciada a possibilidade do uso da geoestatística para separar classes homogêneas de solo, além de estimar a precisão de mapeamento. Contudo, são indispensáveis o conhecimento pedológico e o trabalho posterior de campo para a aplicação e o ajuste dessa modelagem.

Erythrocyte concentrates (ECs) are the major labile blood product being transfused worldwide, aiming at curing anemia of diverse origins. In Switzerland, ECs are stored at 4 °C up to 42 days in saline-adenine-glucose-mannitol (SAGM). Such storage induces cellular lesions, altering red blood cells (RBCs) metabolism, protein content and rheological properties. A hot debate exists regarding the impact of the storage lesions, thus the age of ECs on transfusion-related clinical adverse outcomes. Several studies tend to show that poorer outcomes occur in patients receiving older blood products. However, no clear association was demonstrated up to date. While metabolism and early rheological changes are reversible through transfusion of the blood units, oxidized proteins cannot be repaired, and it is likely such irreversible damages would affect the quality of the blood product and the efficiency of the transfusion. In vivo, RBCs are constantly exposed to oxygen fluxes, and are thus well equipped to deal with oxidative challenges. Moreover, functional 20S proteasome complexes allow for recognition and proteolysis of fairly oxidized protein, and some proteins can be eliminated from RBCs by the release of microvesicles. The present PhD thesis is involved in a global research project which goal is to characterize the effect of processing and storage on the quality of ECs. Assessing protein oxidative damages during RBC storage is of major importance to understand the mechanisms of aging of stored RBCs. To this purpose, redox proteomic-based investigations were conducted here. In a first part, cysteine oxidation and protein carbonylation were addressed via 2D-DIGE and derivatization-driven immunodetection approaches, respectively. Then, the oxidized sub- proteomes were characterized through LC-MS/MS identification of proteins in spots of interest (cysteine oxidation) or affinity-purified carbonylated proteins. Gene ontology annotation allowed classifying targets of oxidation according to their molecular functions. In a third part, the P20S activity was evaluated throughout the storage period of ECs, and its susceptibility to highly oxidized environment was investigated. The potential defensive role of microvesiculation was also addressed through the quantification of eliminated carbonylated proteins. We highlighted distinct protein groups differentially affected by cysteine oxidation, either reversibly or irreversibly. In addition, soluble extracts showed a decrease in carbonylation at the beginning of the storage and membrane extracts revealed increasing carbonylation after 4 weeks of storage. Engaged molecular functions revealed that antioxidant (AO) are rather reversibly oxidized at their cysteine residue(s), but are irreversibly oxidized through carbonylation. In the meantime, the 20S proteasome activity is decreased by around 40 % at the end of the storage period. Incubation of fresh RBCs extracts with exogenous oxidized proteins showed a dose-dependent and protein-dependent inhibitory effect. Finally, we proved that the release of microvesicles allows the elimination of increasing quantities of carbonylated proteins. Taken together, these results revealed an oxidative pathway model of RBCs storage, on which further investigation towards improved storage conditions will be based. -- Les concentrés érythrocytaires (CE) sont le produit sanguin le plus délivré au monde, permettant de traiter différentes formes d'anémies. En Suisse, les CE sont stocké à 4 °C pendant 42 jours dans une solution saline d'adénine, glucose et mannitol (SAGM). Une telle conservation induit des lésions de stockage qui altèrent le métabolisme, les protéines et les propriétés rhéologique du globule rouge (GR). Un débat important concerne l'impact du temps de stockage des CE sur les risques de réaction transfusionnelles, certaines études tentant de démontrer que des transfusions de sang vieux réduiraient l'espérance de vie des patients. Cependant, aucune association concrète n'a été prouvée à ce jour. Alors que les modifications du métabolisme et changement précoces des propriétés rhéologiques sont réversibles suite à la transfusion du CE, les protéines oxydées ne peuvent être réparées, et il est probable que de telles lésions affectent la qualité et l'efficacité des produits sanguins. In vivo, les GR sont constamment exposés à l'oxygène, et sont donc bien équipés pour résister aux lésions oxydatives. De plus, les complexes fonctionnels de proteasome 20S reconnaissent et dégradent les protéines modérément oxydées, et certaines protéines peuvent être éliminées par les microparticules. Cette thèse de doctorat est imbriquée dans un projet de recherche global ayant pour objectif la caractérisation des effets de la préparation et du stockage sur la qualité des GR. Evaluer les dommages oxydatifs du GR pendant le stockage est primordial pour comprendre les mécanismes de vieillissement des produits sanguin. Dans ce but, des recherches orientées redoxomique ont été conduites. Dans une première partie, l'oxydation des cystéines et la carbonylation des protéines sont évaluées par électrophorèse bidimensionnelle différentielle et par immunodétection de protéines dérivatisées. Ensuite, les protéines d'intérêt ainsi que les protéines carbonylées, purifiées par affinité, sont identifiées par spectrométrie de masse en tandem. Les protéines cibles de l'oxydation sont classées selon leur fonction moléculaire. Dans une troisième partie, l'activité protéolytique du protéasome 20S est suivie durant la période de stockage. L'impact du stress oxydant sur cette activité a été évalué en utilisant des protéines exogènes oxydées in vitro. Le potentiel rôle défensif de la microvesiculation a également été étudié par la quantification des protéines carbonylées éliminées. Dans ce travail, nous avons observé que différents groupes de protéines sont affectés par l'oxydation réversible ou irréversible de leurs cystéines. De plus, une diminution de la carbonylation en début de stockage dans les extraits solubles et une augmentation de la carbonylation après 4 semaines dans les extraits membranaires ont été montrées. Les fonctions moléculaires engagées par les protéines altérées montrent que les défenses antioxydantes sont oxydées de façon réversible sur leurs résidus cystéines, mais sont également irréversiblement carbonylées. Pendant ce temps, l'activité protéolytique du protéasome 20S décroit de 40 % en fin de stockage. L'incubation d'extraits de GR en début de stockage avec des protéines oxydées exogènes montre un effet inhibiteur « dose-dépendant » et « protéine-dépendant ». Enfin, les microvésicules s'avèrent éliminer des quantités croissantes de protéines carbonylées. La synthèse de ces résultats permet de modéliser une voie oxydative du stockage des GRs, à partir de laquelle de futures recherches seront menées avec pour but l'amélioration des conditions de stockage.

O teor e a forma dos minerais da fração argila são determinantes na definição da morfologia dos agregados do solo. Objetivando estudar a mineralogia da fração argila e as propriedades químicas de diferentes classes de agregados de Latossolos (Latossolo Bruno Ácrico húmico - LBd e Latossolo Vermelho Distroférrico húmico - LVdf) originados de rochas basálticas no Estado do Paraná, coletaram-se amostras indeformadas em diferentes profundidades (horizontes Bw1 e Bw2) em perfis de solos localizados em duas toposseqüências (quatro perfis no LBd e três no LVdf). Após secagem e separação das amostras indeformadas em seis classes de agregados (2-4; 1-2; 0,5-1; 0,25-0,5; 0,105-0,25; < 0,105 mm) determinaram-se os teores de Si solúvel em ácido acético 0,5 mol L-1 e de K, Ca, Mg e Al trocáveis. A fração argila das diferentes classes de agregados também foi estudada por diferentes técnicas: difratometria de raios X, análise térmica e análises químicas. Verificou-se homogeneidade nos teores trocáveis de elementos entre as classes de tamanho de agregados dos horizontes Bw1 e Bw2 dos perfis dos Latossolos Bruno e Vermelho. A intensa e contínua pedogênese dos Latossolos não foi suficiente para homogeneizar a mineralogia da fração argila (característica mais estável que a dos teores trocáveis) dos agregados. A maior variação nos teores de minerais, em função da classe de tamanho dos agregados, foi para o perfil localizado na posição mais alta da toposseqüência do LBd: variação de 35 % nos teores de gibbsita no horizonte Bw1 e 44 % nos teores de caulinita no horizonte Bw2. Considerando, ainda, o efeito da classe de tamanho dos agregados sobre a mineralogia dos horizontes Bw1 e Bw2 do LBd e LVdf, verificaram-se variações em algumas características cristalográficos da goethita e hematita - diâmetro médio do cristal (DMC) e intensidade de substituição isomórfica de Fe por Al - e da caulinita (DMC).

Phytochromes phyB and phyA mediate a remarkable developmental switch whereby, early upon seed imbibition, canopy light prevents phyB-dependent germination, whereas later on, it stimulates phyA-dependent germination. Using a seed coat bedding assay where the growth of dissected embryos is monitored under the influence of dissected endosperm, allowing combinatorial use of mutant embryos and endosperm, we show that canopy light specifically inactivates phyB activity in the endosperm to override phyA-dependent signaling in the embryo. This interference involves abscisic acid (ABA) release from the endosperm and distinct spatial activities of phytochrome signaling components. Under the canopy, endospermic ABA opposes phyA signaling through the transcription factor (TF) ABI5, which shares with the TF PIF1 several target genes that negatively regulate germination in the embryo. ABI5 enhances the expression of phytochrome signaling genes PIF1, SOMNUS, GAI, and RGA, but also of ABA and gibberellic acid (GA) metabolic genes. Over time, weaker ABA-dependent responses eventually enable phyA-dependent germination, a distinct type of germination driven solely by embryonic growth.

Clin Microbiol Infect ABSTRACT: The aetiological diagnosis of community-acquired pneumonia (CAP) is challenging in children, and serological markers would be useful surrogates for epidemiological studies of pneumococcal CAP. We compared the use of anti-pneumolysin (Ply) antibody alone or with four additional pneumococcal surface proteins (PSPs) (pneumococcal histidine triad D (PhtD), pneumococcal histidine triad E (PhtE), LytB, and pneumococcal choline-binding protein A (PcpA)) as serological probes in children hospitalized with CAP. Recent pneumococcal exposure (positive blood culture for Streptococcus pneumoniae, Ply(+) blood PCR finding, and PSP seroresponse) was predefined as supporting the diagnosis of presumed pneumococcal CAP (P-CAP). Twenty-three of 75 (31%) children with CAP (mean age 33.7 months) had a Ply(+) PCR finding and/or a ≥2-fold increase of antibodies. Adding seroresponses to four PSPs identified 12 additional patients (35/75, 45%), increasing the sensitivity of the diagnosis of P-CAP from 0.44 (Ply alone) to 0.94. Convalescent anti-Ply and anti-PhtD antibody titres were significantly higher in P-CAP than in non P-CAP patients (446 vs. 169 ELISA Units (EU)/mL, p 0.031, and 189 vs. 66 EU/mL, p 0.044), confirming recent exposure. Acute anti-PcpA titres were three-fold lower (71 vs. 286 EU/mL, p <0.001) in P-CAP children. Regression analyses confirmed a low level of acute PcpA antibodies as the only independent predictor (p 0.002) of P-CAP. Novel PSPs facilitate the demonstration of recent pneumococcal exposure in CAP children. Low anti-PcpA antibody titres at admission distinguished children with P-CAP from those with CAP with a non-pneumococcal origin.

A região dos Campos de Cima da Serra, RS, apresenta solos com elevados teores de matéria orgânica (MOS), sendo freqüente teores altos de Al trocável. A proposta deste trabalho foi investigar a qualidade da MOS de amostras de camadas superficiais (0 a 10 cm) de oito solos representativos da região (dois Neossolos, dois Gleissolos, três Cambissolos e um Latossolo) sob pastagem nativa, com os seguintes objetivos: avaliar seu comportamento diante da desmineralização da amostra com solução de HF; determinar a sua composição química; e investigar a influência do Al trocável e dos óxidos de Fe na qualidade da MOS. A matéria orgânica foi concentrada com solução de HF 10 cL L-1 e posteriormente avaliada por análise elementar e espectroscopias de FTIR e de RMN 13C CP/MAS. A fração da massa recuperada (5,8 a 10 %) após tratamento com HF correlacionou-se com teor de C original do solo, enquanto a proporção de C recuperado (47 a 86 %) correlacionouse inversamente com o teor de óxidos de Fe. Esse resultado indica que parte da MOS encontra-se associada a esses minerais nesses ambientes. Os teores de estruturas C O-alquil totais (52 a 59 %) foram elevados e os valores de índice I1630/I2920(FTIR) foram baixos, evidenciando baixo grau de decomposição e de humificação da MOS desses solos.

The protective effect of cations, especially Ca and Mg, against aluminum (Al) rhizotoxicity has been extensively investigated in the last decades. The mechanisms by which the process occurs are however only beginning to be elucidated. Six experiments were carried out here to characterize the protective effect of Mg application in relation to timing, location and crop specificity: Experiment 1 - Protective effect of Mg compared to Ca; Experiment 2 - Protective effect of Mg on distinct root classes of 15 soybean genotypes; Experiment 3 - Effect of timing of Mg supply on the response of soybean cvs. to Al; Experiment 4 - Investigating whether the Mg protective effect is apoplastic or simplastic using a split-root system; Experiment 5 - Protective effect of Mg supplied in solution or foliar spraying, and Experiment 6 - Protective effect of Mg on Al rhizotoxicity in other crops. It was found that the addition of 50 mmol L-1 Mg to solutions containing toxic Al increased Al tolerance in 15 soybean cultivars. This caused soybean cultivars known as Al-sensitive to behave as if they were tolerant. The protective action of Mg seems to require constant Mg supply in the external medium. Supplying Mg up to 6 h after root exposition to Al was sufficient to maintain normal soybean root growth, but root growth was not recovered by Mg addition 12 h after Al treatments. Mg application to half of the root system not exposed to Al was not sufficient to prevent Al toxicity on the other half exposed to Al without Mg in rooting medium, indicating the existence of an external protection mechanism of Mg. Foliar spraying with Mg also failed to decrease Al toxicity, indicating a possible apoplastic role of Mg. The protective effect of Mg appeared to be soybean-specific since Mg supply did not substantially improve root elongation in sorghum, wheat, corn, cotton, rice, or snap bean when grown in the presence of toxic Al concentrations.

Polymorphonuclear neutrophils (PMN) are key components of the inflammatory response contributing to the development of pathogen-specific immune responses. Following infection with Leishmania major, neutrophils are recruited within hours to the site of parasite inoculation. C57BL/6 mice are resistant to infection, and BALB/c mice are susceptible to infection, developing unhealing, inflammatory lesions. In this report, we investigated the expression of cell surface integrins, TLRs, and the secretion of immunomodulatory cytokines by PMN of both strains of mice, in response to infection with L. major. The parasite was shown to induce CD49d expression in BALB/c-inflammatory PMN, and expression of CD49d remained at basal levels in C57BL/6 PMN. Equally high levels of CD11b were expressed on PMN from both strains. In response to L. major infection, the levels of TLR2, TLR7, and TLR9 mRNA were significantly higher in C57BL/6 than in BALB/c PMN. C57BL/6 PMN secreted biologically active IL-12p70 and IL-10. In contrast, L. major-infected BALB/c PMN transcribed and secreted high levels of IL-12p40 but did not secrete biologically active IL-12p70. Furthermore, IL-12p40 was shown not to associate with IL-23 p19 but formed IL-12p40 homodimers with inhibitory activity. No IL-10 was secreted by BALB/c PMN. Thus, following infection with L. major, in C57BL/6 mice, PMN could constitute one of the earliest sources of IL-12, and in BALB/c mice, secretion of IL-12p40 could contribute to impaired, early IL-12 signaling. These distinct PMN phenotypes may thus influence the development of L. major-specific immune response.
