In light of deep-sea mining industry development, particularly interested in massive-sulphide deposits enriched in metals with high commercial value, efforts are increasing to better understand potential environmental impacts to local fauna. The aim of this study was to assess the natural background levels of biomarkers in the hydrothermal vent shrimp Rimicaris exoculata and their responses to copper exposure at in situ pressure (30MPa) as well as the effects of depressurization and pressurization of the high-pressure aquarium IPOCAMP. R. exoculata were collected from the chimney walls of the hydrothermal vent site TAG (Mid Atlantic Ridge) at 3630m depth during the BICOSE cruise in 2014. Tissue metal accumulation was quantified in different tissues (gills, hepatopancreas and muscle) and a battery of biomarkers was measured: metal exposure (metallothioneins), oxidative stress (catalase, superoxide dismutase, glutathione-S-transferase and glutathione peroxidase) and oxidative damage (lipid peroxidation). Data show a higher concentration of Cu in the hepatopancreas and a slight increase in the gills after incubations (for both exposed groups). Significant induction of metallothioneins was observed in the gills of shrimps exposed to 4μM of Cu compared to the control group. Moreover, activities of enzymes were detected for the in situ group, showing a background protection against metal toxicity. Results suggest that the proposed method, including a physiologically critical step of pressurizing and depressurizing the test chamber to enable the seawater exchange during exposure to contaminants, is not affecting metal accumulation and biomarkers response and may prove a useful method to assess toxicity of contaminants in deep-sea species.

The deep-sea lantern shark Etmopterus spinax occurs in the northeast Atlantic on or near the bottoms of the outer continental shelves and slopes, and is regularly captured as bycatch in deep-water commercial fisheries. Given the lack of knowledge on the impacts of fisheries on this species, a demographic analysis using age-based Leslie matrices was carried out. Given the uncertainties in the mortality estimates and in the available life history parameters, several different scenarios, some incorporating stochasticity in the life history parameters (using Monte Carlo simulation), were analyzed. If only natural mortality were considered, even after introducing uncertainties in all parameters, the estimated population growth rate (A) suggested an increasing population. However, if fishing mortality from trawl fisheries is considered, the estimates of A either indicated increasing or declining populations. In these latter cases, the uncertainties in the species reproductive cycle seemed to be particularly relevant, as a 2-year reproductive cycle indicated a stable population, while a longer (3-year cycle) indicated a declining population. The estimated matrix elasticities were in general higher for the survivorship parameters of the younger age classes and tended to decrease for the older ages. This highlights the susceptibility of this deep-sea squaloid to increasing fishing mortality, emphasizing that even though this is a small-sized species, it shows population dynamics patterns more typical of the larger-sized and in general more vulnerable species. This study provides the first description of the feeding ecology of the smooth lanternshark Etmopterus pusillus based on stomach contents of specimens caught as bycatch in the Algarve (southern Portugal) with bottom trawling and bottom longline. The diet of E. pusillus consists mainly of fish (dry weight (% W)=87.1%; frequency of occurrence (%FO)=28.6%; number (%N)=30.3%), crustaceans (%W=7.7%; %FO=36.7%; %N=3.4%) and cephalopods (%W=4.7%; %FO=11.3%; %N=11.1%). The diet did not vary between sexes. Ontogenic changes were detected: crustaceans decreased in importance as the sharks increased in size and fish became dominant in the diet of adults. Combining two fishing methods provided broad information on the diet of E. pusillus, as bottom trawling caught smaller specimens and longlines caught larger individuals. E. pusillus feeds mainly on non-commercial species, and therefore does not compete directly with commercial fisheries. Finally, E. pusillus feeds in various parts of the water column and thus it can access a wide range of prey; however, this also means that it can be caught by both gears, making it more vulnerable in terms of conservation.

A new deep-sea fish is reported for the tropical eastern Atlantic. The rare chiasmodontid Kali macrodon was caught for the first time in Cape Verdean waters during an oceanographic survey in September 2009. This record provides the fifth occurrence for the species in the north-eastern Atlantic and represents the third specimen ever caught in tropical waters.

Protease-producing bacteria are known to play an important role in degrading sedimentary particular organic nitrogen, and yet, their diversity and extracellular proteases remain largely unknown. In this paper, the diversity of the cultivable protease-producing bacteria and their extracellular proteases in the sediments of the South China Sea was investigated. The richness of the cultivable protease-producing bacteria reached 10(6) cells/g in all sediment samples. Analysis of the 16S rRNA gene sequences revealed that the predominant cultivated protease-producing bacteria are Gammaproteobacteria affiliated with the genera Pseudoalteromonas, Alteromonas, Marinobacter, Idiomarina, Halomonas, Vibrio, Shewanella, Pseudomonas, and Rheinheimera, with Alteromonas (34.6%) and Pseudoalteromonas (28.2%) as the predominant groups. Inhibitor analysis showed that nearly all the extracellular proteases from the bacteria are serine proteases or metalloproteases. Moreover, these proteases have different hydrolytic ability to different proteins, reflecting they may belong to different kinds of serine proteases or metalloproteases. To our knowledge, this study represents the first report of the diversity of bacterial proteases in deep-sea sediments.

The Mediterranean Sea constitutes a unique environment to study cold-seep ecosystems due to the presence of different geodynamic settings, from an active margin along the Mediterranean Ridge (MR) to a passive margin in the Nile Deep-Sea Fan (NDSF). We attempted to identify the structure of benthic communities associated with the Napoli and Amsterdam mud volcanoes (MVs) located on the MR and to establish the links between faunal distribution and environmental conditions at different spatial scales. Comparison between the 2 MVs revealed that the faunal distribution seemed to be mainly controlled by the characteristics of the microhabitats. On both geological structures, the variability between the different microhabitats was higher than the variability observed between replicates of the same microhabitat, and the distribution of macro-fauna was apparently linked to gradients in physico-chemical conditions. The peripheral sites from Napoli were generally more oxygenated and harboured lower species richness than the active sites. The reduced sediment microhabitat from Amsterdam presented the highest methane concentrations and was mainly colonised by symbiont-bearing vesicomyid bivalves and heterotrophic dorvilleid polychaetes. Overall, a higher taxonomic diversity was observed on Napoli. Sub-stratum type was hypothesised to be the second factor influencing faunal distribution. The results of this study highlight the high heterogeneity of faunal communities associated with seep ecosystems within this region and the need to pursue investigations at various spatial and temporal scales.

Radiolarian distribution in surface sediments of 104 stations from northern and central South China Sea show that the abundance and diversity of radiolarians increase with the water depth and are related to radiolarian concentrations from the water column, diminished terrigenous input, variability in calcareous shell content and the rate of silica and carbonate dissolution in the deep sea. According to the appearances of individual species in surface sediments at particular depths, seven faunal boundaries distribution are recognized at water depths of 100, 450, 650, 1000, 1200, 1400 and 2500 m. Four radiolarian assemblages in the sediments were identified by applying clustering procedures. Geographic distributions of these four assemblages coincide with present-day hydrologic features of the surface waters in this area.

In this study, marine sponges collected in Irish waters were analysed for their associated microbiota. Of the approximately 240 bacterial isolates obtained from two sponges several showed antimicrobial activity; among them members of genera which have rarely been shown to produce antimicrobial compounds. Differences observed from the sponge-derived groups of isolates in terms of bioactivity suggests that S. carnosus isolates may be a better source of antibacterial compounds, while Leucosolenia sp. isolates appear to be a better source of antifungal compounds. More than 60% of fungal isolates obtained from 12 sponge samples proved to be bioactive. One of the isolates, which was closely related to Fusarium oxysporum and showed activity against bacteria and fungi, was investigated for its secondary metabolite genes. At least 5 different NRPS genes, with a sequence similarity as low as 50 % to known genes, were identified highlighting the likelihood that this isolate may be capable of producing novel secondary metabolites. A Micromonospora sp. was isolated from a Haliclona simulans sample collected in Irish waters. The isolate inhibited the growth of Gram positive bacterial test strains in three different antimicrobial assays. Employing preparative layer chromatography the compound responsible for the bioactivity could be isolated. According to LC-MS andNMR data the bioactive compound could indeed be novel. Finally, two deep water sponges were shown to host a remarkably different bacterial and archaeal diversity by application of 454 Pyrosequencing. The L. diversichela –proteobacterial community was dominated by a single ƴ-proteobacterial bacterium whereas the S. normani sample hosted a largely sponge specific microbial community, even more diverse than has been previously reported for shallow water sponges. Organisms potentially involved in nitrification, sulphate reduction and secondary metabolite production were found to be spatially distributed in the sponge. Furthermore, a deep sea specific population was implied.
