Assays that assess cellular mediated immune responses performed under Good Clinical Laboratory Practice (GCLP) guidelines are required to provide specific and reproducible results. Defined validation procedures are required to establish the Standard Operating Procedure (SOP), include pass and fail criteria, as well as implement positivity criteria. However, little to no guidance is provided on how to perform longitudinal assessment of the key reagents utilized in the assay. Through the External Quality Assurance Program Oversight Laboratory (EQAPOL), an Interferon-gamma (IFN-γ) Enzyme-linked immunosorbent spot (ELISpot) assay proficiency testing program is administered. A limit of acceptable within site variability was estimated after six rounds of proficiency testing (PT). Previously, a PT send-out specific within site variability limit was calculated based on the dispersion (variance/mean) of the nine replicate wells of data. Now an overall 'dispersion limit' for the ELISpot PT program within site variability has been calculated as a dispersion of 3.3. The utility of this metric was assessed using a control sample to calculate the within (precision) and between (accuracy) experiment variability to determine if the dispersion limit could be applied to bridging studies (studies that assess lot-to-lot variations of key reagents) for comparing the accuracy of results with new lots to results with old lots. Finally, simulations were conducted to explore how this dispersion limit could provide guidance in the number of replicate wells needed for within and between experiment variability and the appropriate donor reactivity (number of antigen-specific cells) to be used for the evaluation of new reagents. Our bridging study simulations indicate using a minimum of six replicate wells of a control donor sample with reactivity of at least 150 spot forming cells per well is optimal. To determine significant lot-to-lot variations use the 3.3 dispersion limit for between and within experiment variability.

Sapropels -organic-matter rich layers- are common in Neogene sediments of the eastern Mediterranean Sea. The formation of these layers has been attributed to climate-related increases in organic-matter production (Calvert et al., 1992, doi:10.1038/359223a0; Rossignol-Strick et al., 1982, doi:10.1038/295105a0; Rohling, 1994, doi:10.1016/0025-3227(94)90202-X) and increased organic-matter preservation due to oxygen depletion in more stagnant bottom waters (Rossignol-Strick et al., 1982, doi:10.1038/295105a0; Rohling, 1994, doi:10.1016/0025-3227(94)90202-X). Here we report that eastern Mediterranean Pliocene sapropels (Emeis et al., 1996, doi:10.2973/odp.proc.ir.160.102.1996) contain molecular fossils of a compound (isorenieratene) known to be synthesized by photosynthetic green sulphur bacteria, suggesting that sulphidic (euxinic) -and therefore anoxic- conditions prevailed in the photic zone of the water column. These sapropels also have a high trace-metal content, which is probably due to the efficient scavenging of these metals by precipitating sulphides in a euxinic water column. The abundance and sulphur-isotope composition of pyrite are consistent with iron sulphide formation in the water column. We conclude that basin-wide water-column euxinia occurred over substantial periods during Pliocene sapropel formation in the eastern Mediterranean Sea, and that the ultimate degradation of the increased organic-matter production was strongly influential in generating and sustaining the euxinic conditions.

The response of the Gulf Stream (GS) system to atmospheric forcing is generally linked either to the basin-scale winds on the subtropical gyre or to the buoyancy forcing from the Labrador Sea. This study presents a multiscale synergistic perspective to describe the low-frequency response of the GS system. The authors identify dominant temporal variability in the North Atlantic Oscillation (NAO), in known indices of the GS path, and in the observed GS latitudes along its path derived from sea surface height (SSH) contours over the period 1993-2013. The analysis suggests that the signature of interannual variability changes along the stream's path from 75 degrees to 55 degrees W. From its separation at Cape Hatteras to the west of 65 degrees W, the variability of the GS is mainly in the near-decadal (7-10 years) band, which is missing to the east of 60 degrees W, where a new interannual (4-5 years) band peaks. The latter peak (4-5 years) was missing to the west of 65 degrees W. The region between 65 degrees and 60 degrees W seems to be a transition region. A 2-3-yr secondary peak was pervasive in all time series, including that for the NAO. This multiscale response of the GS system is supported by results from a basin-scale North Atlantic model. The near-decadal response can be attributed to similar forcing periods in the NAO signal; however, the interannual variability of 4-5 years in the eastern segment of the GS path is as yet unexplained. More numerical and observational studies are warranted to understand such causality.

Host specificity tests on Gynaikothrips ficorum (Marchal) and Gynaikothrips uzeli (Zimmerman) (Thysanoptera: Phlaeothripidae) have shown that under experimental conditions, G. ficorum will induce leaf galls on both Ficus benjamina L. and Ficus microcarpa L. f. (Rosales: Moraceae), but G. uzeli will induce galls only on F. benjamina. A further interesting aspect of the results is that gall induction by G. uzeli on F. benjamina appears to have been suppressed in the presence of F. microcarpa plants in the same cage. Liothrips takahashii (Moulton) (Thysanoptera: Phlaeothripidae), an inquiline in the galls of these Gynaikothrips, is reported for the first time from Australia, mainland China, Malaysia, Costa Rica, and western USA.

The In Situ Analysis System (ISAS) was developed to produce gridded fields of temperature and salinity that preserve as much as possible the time and space sampling capabilities of the Argo network of profiling floats. Since the first global re-analysis performed in 2009, the system has evolved and a careful delayed mode processing of the 2002-2012 dataset has been carried out using version 6 of ISAS and updating the statistics to produce the ISAS13 analysis. This last version is now implemented as the operational analysis tool at the Coriolis data centre. The robustness of the results with respect to the system evolution is explored through global quantities of climatological interest: the Ocean Heat Content and the Steric Height. Estimates of errors consistent with the methodology are computed. This study shows that building reliable statistics on the fields is fundamental to improve the monthly estimates and to determine the absolute error bars. The new mean fields and variances deduced from the ISAS13 re-analysis and dataset show significant changes relative to the previous ISAS estimates, in particular in the southern ocean, justifying the iterative procedure. During the decade covered by Argo, the intermediate waters appear warmer and saltier in the North Atlantic and fresher in the Southern Ocean than in WOA05 long term mean. At inter-annual scale, the impact of ENSO on the Ocean Heat Content and Steric Height is observed during the 2006-2007 and 2009-2010 events captured by the network.

significant amount of Expendable Bathythermograph (XBT) data has been collected in the Mediterranean Sea since 1999 in the framework of operational oceanography activities. The management and storage of such a volume of data poses significant challenges and opportunities. The SeaDataNet project, a pan-European infrastructure for marine data diffusion, provides a convenient way to avoid dispersion of these temperature vertical profiles and to facilitate access to a wider public. The XBT data flow, along with the recent improvements in the quality check procedures and the consistence of the available historical data set are described. The main features of SeaDataNet services and the advantage of using this system for long-term data archiving are presented. Finally, focus on the Ligurian Sea is included in order to provide an example of the kind of information and final products devoted to different users can be easily derived from the SeaDataNet web portal.

An objective of the project is the climatic characterization of this large viticultural region with the participation of ten countries: Argentine, Bolivia, Brazil, Chile, Cuba, Spain, Mexico, Peru, Portugal, and Uruguay.

Phragmites australis (Cav.) Trin. ex Steud. is a hydrophyte particularly resistant to harsh conditions, e.g. drought, high salinity, contaminants, such as heavy metals and toxic molecules, and high nutrients concentrations. These resistances render the plant suitable for water depuration, where its particular metabolism is exploited to remove pollutants and excessive nutrients from the environment. In constructed wetlands, this principle is applied to phyto-purify wastewater with various origins, such as industrial, agricultural and household, with the aim to improve its quality to an extent which would render its reuse possible. In the framework of a pre-existing project of Department of Agricultural and Food Sciences (DiSTAl), this work integrates the knowledge and data relative to an Emilia Romagna (IT) constructed wetland plant, in order to expand the knowledge about this particular facility and of the system in general. By assaying antioxidants, both non- enzymatic and enzymatic, chlorophylls content and net photosynthetic rates, and by measuring the elemental composition of the specimens, the health status and the elemental uptake of the wetland plants sampled in different areas were investigated. The results were compared amongst the examined specimens with the aim to detect areas where there may be a higher stress due to a different wastewater composition, potentially varying along the constructed route. In addition, different parameters regarding the extraction and assay protocols were investigated, in order to optimise the procedure and to select the best conditions to perform the analyses, as well as to integrate information missing in literature or found as contradictory.

I Medicane sono rari cicloni che si sviluppano sul Mar Mediterraneo e presentano caratteristiche dei cicloni tropicali, come la forma a spirale delle bande di nubi, un occhio privo di venti e nubi, venti intensi nella banda che circonda l’occhio e la presenza di un nucleo caldo. Nel presente lavoro, è stata compiuta un’analisi del Medicane Numa, verificatosi nel novembre del 2017, utilizzando gli output della simulazione del modello RAMS-ISAC. L’obiettivo di questa tesi è l’identificazione e la descrizione delle caratteristiche tropicali di Numa, focalizzandosi sulla descrizione dei diversi stadi di sviluppo del ciclone. Il sistema di bassa pressione è stato identificato utilizzando la pressione sul livello del mare, mentre l’anomalia termica e il vento orizzontale hanno permesso una descrizione della struttura del Medicane e del suo nucleo caldo. Un’illustrazione della struttura a spirale delle bande di nubi e dell’occhio è stata ottenuta con il grafico dei rapporti di mescolanza delle idrometeore di nubi e pioggia. Questi parametri hanno consentito di ricavare il diametro dell’occhio, pari a 75 km, mentre il diametro del Medicane è risultato 230 km. Numa ha registrato una velocità massima del vento in superficie di 20 m/s nella banda adiacente all’occhio del ciclone. Il diagramma di Hart dello spazio delle fasi del ciclone ha confermato la natura simil-tropicale di Numa e ne ha descritto l’evoluzione, identificando la transizione da sistema a caratteri tropicali a sistema ibrido. La traiettoria nello spazio delle fasi ha consentito l’identificazione delle sottofasi dell’evoluzione di Numa, confermate dai grafici dell’evoluzione temporale dei parametri menzionati in precedenza. L’analisi ha mostrato il ruolo cruciale della presenza di una struttura organizzata nel determinare l’intensità e la durata delle caratteristiche tropicali. Tutti i parametri hanno evidenziato la simmetria della struttura durante la persistente fase matura di Numa.

The mesophotic zone is frequently defined as ranging between 30-40 and 150 m depth. However, these borders are necessarily imprecise due to variations in the penetration of light along the water column related to local factors. Moreover, density of data on mesophotic ecosystems vary along geographical distance, with temperate latitudes largely less explored than tropical situations. This is the case of the Mediterranean Sea, where information on mesophotic ecosystems is largely lower with respect to tropical situations. The lack of a clear definition of the borders of the mesophotic zone may represent a problem when information must be transferred to the policy that requires a coherent spatial definition to plan proper management and conservation measures. The present thesis aims at providing information on the spatial definition of the mesophotic zone in the Mediterranean Sea, its biodiversity and distribution of its ecosystems. The first chapter analyzes information on mesophotic ecosystems in the Mediterranean Sea to identify gaps in the literature and map the mesophotic zone in the Mediterranean Sea using light penetration estimated from satellite data. In the second chapter, different visual techniques to study mesophotic ecosystems are compared to identify the best analytical method to estimate diversity and habitat extension. In the third chapter, a set of Remotely Operated vehicles (ROV) surveys performed on mesophotic assemblages in the Mediterranean Sea are analyzed to describe their taxonomic and functional diversity and environmental factors influencing their structure. A Habitat Suitability Model is run in the fourth chapter to map the distribution of areas suitable for the presence of deep-water oyster reefs in the Adriatic-Ionian area. The fifth chapter explores the mesophotic zone in the northern Gulf of Mexico providing its spatial and vertical extension of the mesophotic zone and information on the diversity associated with mesophotic ecosystems.

This thesis investigates the morphological variations of fibular extremities in humans and non-human hominids using a 3D Geometric Morphometric approach. The study has three objectives: (1) to assess the shape, form, and size variations of fibular epiphyses within the human species, highlighting sexually dimorphic features; (2) to explore interpopulation variability of fibular extremities from the Upper Paleolithic to the 20th century, comparing subsistence, mobility, and lifestyles; and (3) to examine interspecific variations in fibular ends, testing potential associations with locomotor and positional behavior among extant hominid taxa. In terms of intraspecific variations, sex-related differences in fibular form and size were observed, suggesting distinct functional requirements for the lower limb between sexes. Interpopulation variations revealed a decline in activity level over time, influenced by terrain and footwear use. Hunter-gatherer groups exhibited greater joint mobility, loading, and range of motion compared to sedentary pre- and post-industrial populations. Interspecific variations demonstrated significant morphological differences among hominid taxa, indicating functional implications related to both phylogeny and specific loading patterns on the lower limb. The study identified features indicative of bipedalism in humans, as well as shared characteristics among non-human great apes. Furthermore, distinguishing features were found between Asian and African apes, along with unique morphological signals associated with distinct positional behavior in each hominid taxa. By comprehensively analyzing fibular morphology, this research sheds light on the importance of this bone in knee support, ankle stabilization, and overall locomotor function. The findings contribute to our understanding of the evolutionary and functional aspects of the fibula across human populations and non-human hominids throughout history.

We used mitochondrial cyt b sequences to investigate the phylogenetic relationships of Crocidura russula (sensu lato) populations across the Strait of Gibraltar, western Europe, Maghreb, and the Mediterranean and Atlantic islands. This revealed very low genetic divergence between European and Moroccan populations. The application of a molecular clock previously calibrated for shrews suggested that the separation of European from Moroccan lineages occurred less than 60 000 bp, which is at least 5 million years (Myr) after the reopening of the Strait of Gibraltar. This means that an overwater dispersal event was responsible for the observed phylogeographical structure. In contrast, genetic analyses revealed that Moroccan populations were highly distinct from Tunisian ones. According to the molecular clock, these populations separated about 2.2 million years ago (Ma), a time marked by sharp alternations of dry and humid climates in the Maghreb. The populations of the Mediterranean islands Ibiza, Pantelleria, and Sardinia were founded from Tunisian populations by overwater dispersal. In conclusion, overwater dispersal across the Strait of Gibraltar, probably assisted by humans, is possible for small terrestrial vertebrates. Moreover, as in Europe, Quaternary climatic fluctuations had a major effect on the phylogeographical structure of the Maghreb biota.

The atmosphere is a global influence on the movement of heat and humidity between the continents, and thus significantly affects climate variability. Information about atmospheric circulation are of major importance for the understanding of different climatic conditions. Dust deposits from maar lakes and dry maars from the Eifel Volcanic Field (Germany) are therefore used as proxy data for the reconstruction of past aeolian dynamics.rnrnIn this thesis past two sediment cores from the Eifel region are examined: the core SM3 from Lake Schalkenmehren and the core DE3 from the Dehner dry maar. Both cores contain the tephra of the Laacher See eruption, which is dated to 12,900 before present. Taken together the cores cover the last 60,000 years: SM3 the Holocene and DE3 the marine isotope stages MIS-3 and MIS-2, respectively. The frequencies of glacial dust storm events and their paleo wind direction are detected by high resolution grain size and provenance analysis of the lake sediments. Therefore two different methods are applied: geochemical measurements of the sediment using µXRF-scanning and the particle analysis method RADIUS (rapid particle analysis of digital images by ultra-high-resolution scanning of thin sections).rnIt is shown that single dust layers in the lake sediment are characterized by an increased content of aeolian transported carbonate particles. The limestone-bearing Eifel-North-South zone is the most likely source for the carbonate rich aeolian dust in the lake sediments of the Dehner dry maar. The dry maar is located on the western side of the Eifel-North-South zone. Thus, carbonate rich aeolian sediment is most likely to be transported towards the Dehner dry maar within easterly winds. A methodology is developed which limits the detection to the aeolian transported carbonate particles in the sediment, the RADIUS-carbonate module.rnrnIn summary, during the marine isotope stage MIS-3 the storm frequency and the east wind frequency are both increased in comparison to MIS-2. These results leads to the suggestion that atmospheric circulation was affected by more turbulent conditions during MIS-3 in comparison to the more stable atmospheric circulation during the full glacial conditions of MIS-2.rnThe results of the investigations of the dust records are finally evaluated in relation a study of atmospheric general circulation models for a comprehensive interpretation. Here, AGCM experiments (ECHAM3 and ECHAM4) with different prescribed SST patterns are used to develop a synoptic interpretation of long-persisting east wind conditions and of east wind storm events, which are suggested to lead to an enhanced accumulation of sediment being transported by easterly winds to the proxy site of the Dehner dry maar.rnrnThe basic observations made on the proxy record are also illustrated in the 10 m-wind vectors in the different model experiments under glacial conditions with different prescribed sea surface temperature patterns. Furthermore, the analysis of long-persisting east wind conditions in the AGCM data shows a stronger seasonality under glacial conditions: all the different experiments are characterized by an increase of the relative importance of the LEWIC during spring and summer. The different glacial experiments consistently show a shift from a long-lasting high over the Baltic Sea towards the NW, directly above the Scandinavian Ice Sheet, together with contemporary enhanced westerly circulation over the North Atlantic.rnrnThis thesis is a comprehensive analysis of atmospheric circulation patterns during the last glacial period. It has been possible to reconstruct important elements of the glacial paleo climate in Central Europe. While the proxy data from sediment cores lead to a binary signal of the wind direction changes (east versus west wind), a synoptic interpretation using atmospheric circulation models is successful. This shows a possible distribution of high and low pressure areas and thus the direction and strength of wind fields which have the capacity to transport dust. In conclusion, the combination of numerical models, to enhance understanding of processes in the climate system, with proxy data from the environmental record is the key to a comprehensive approach to paleo climatic reconstruction.rn
