Introduction: Lupus erythematosus panniculitis (LEP) or lupus erythematosus profundus is a rare form of chronic cutaneous manifestation affecting both adults and pediatric patients. The prevalence of this manifestation was seldom reported in juvenile systemic lupus erythematosus (JSLE). Case reports: From January 1983 to December 2010, 5,506 patients were followed at the Pediatric Rheumatology Unit of our University Hospital and 278 (5%) of them met the American College of Rheumatology classification criteria for JSLE. Two (0.7%) of them had LEP at JSLE onset. These two cases had tender deep inflammatory subcutaneous nodules or plaques at the time of diagnosis, and the histopathologic pattern evidenced lobular or mixed panniculitis with lymphocytic inflammatory cells of the fat lobule. Treatments for LEP included mainly antimalarials, systemic corticosteroids and sunscreen protection. One male patient required thalidomide and immunosuppressive drugs, including mycophenolate mofetil, cyclosporin and intravenous cyclophosphamide. However, skin lesions improved only after rituximab treatment. Discussion: LEP was rarely observed in our cohort of JSLE patients as the first lupus manifestation. Anti-CD20 monoclonal antibody therapy may be an option for refractory LEP treatment in children.

The current study is a piece from the original project entitled "Diagnosis of the Artistic Gymnastics Development Program in Brazil". Among others issues, the coaching philosophy of coaches who are developing gymnasts that may be representative of the national team was a matter of analysis. We interviewed 46 coaches from 29 sports institutions in Brazil. As data collection we used a semi-structured interview and for data treatment we adopted the content analysis method of Bardin (2010). We have found out that there is an inconsistent philosophical basis, and in many institutions this has been mixed up with the objectives of the training program or just doesn't exist. This fact needs consideration and criticism, but we know that the coaching philosophy is not developed during the coaching education courses.

Vibrio cholerae Cytolysin (VCC) gehört zur Gruppe der Exotoxine und bildet auf Membranen heptamere transmembrane Poren. VCC wird als protoxin mit einem Molekulargewicht von 79 kDa sezerniert und benötigt die proteolytische Spaltung der N-terminalen Pro-Region um Poren in der Membran zu bilden. Diese Spaltung erfolgt sowohl in Lösung, als auch nach der Bindung an Membranen, aber nur aktiviertes VCC oligomererisiert in eine lytische Pore. Die Kristallstruktur von VCC zeigt, dass das Monomer vier verschiedenen strukturellen Domänen enthält; die cytolytische Domäne, mit der Pre-Stem-Sequenz, der Pro-Region und den beiden C-terminalen Domänen β-Trefoil und β-Prism. Die porenbildende β-Barrel wird aus je einer Pre-Stem Domäne jedes der einzelnen sieben Untereinheiten gebildet. Da sich die porenbildende Region im Monomer zwischen den Domänen β-Prism und β-Trefoil befindet, sind konformationelle Änderungen des Toxins notwendig, um die Insertion dieser Region in die Membran zu ermöglichen. In dieser Arbeit wurde unter anderem der Mechanismus der Porenbildung durch die Konstruktion von Disulfid-Derivaten untersucht. Die Bildung von Disulfidbrücken wurde verwendet, um die porenbildende Region entweder mit der β-Trefoil oder β-Prism Domäne zu verknüpfen. Unter nicht-reduzierenden Bedingungen bindet das Toxin an Membranen und oligomerisiert zu SDS-labilen Oligomeren. Nach der Reduktion der künstlichen Disulfidbrücke erlangen die gebildeten Oligomere SDS-Stabilität und permeabilisieren die Membran. Durch die Zugabe steigender Konzentrationen des VCC-Derivats zu aktivem Toxin, wird die SDS-Stabilität der gebildeten Oligomere stark reduziert. Die Insertion des aktiven Toxins in die Membran wird allerdings nicht verhindert und daher Poren mit reduziertem funktionellen Durchmesser gebildet. Diese Ergebnisse verdeutlichen, dass die Bildung einer Prä-Pore vor der Insertion des Toxins in die Membran erfolgt und zeigt zum ersten Mal ein solches Zwischenstadium für ein β-porenbildendes Toxin, das von Gram-negativen Organismen produziert wird. Diese Ergebnisse deuten auf einen archetypischen Mechanismus der Porenbildung hin. Zusätzlich wurde die Funktion der beiden C-terminalen Domänen untersucht, und daher verschiedene Deletions- und Substitutionsmutanten konstruiert. Die β-Trefoil Domäne ist nicht essentiell für die Bindung des Toxins an Membranen, ist aber für die korrekte Faltung des Toxins notwendig. Die C-terminale β-Prism Domäne vermittelt die Bindung des Toxins an Membranen über Zuckerrezeptoren.

Diese Arbeit unterstreicht das Potential von Hybridfunktionalen (B3LYP) für die Untersuchung einer großen Bandbreite von Systemen. Durch die Einbeziehung der exakten Hartree-Fock Austauschenergie kann B3LYP für molekulare und kristalline Systeme eingesetzt werden. Zum Beispiel können stark korrelierte Systeme mit B3LYP erfolgreich erforscht werden. Die elektronische Struktur von PAHs wurde mit B3LYP Hybriddichtefunktionalen untersucht. Mit der ∆SCF-Methode wurden Elektronenbindungsenergien bestimmt, welche die mit UPS gewonnenen experimentellen Resultate bestätigen und ergänzen. Symmetrieeigenschaften der molekularen Orbitale wurden analysiert, um eine Zuordnung und Einschätzung der zugehörigen Signalstärke zu ermöglichen. Während σ-artige Orbitale nur schwer durch UPS-Messungen an dünnen Filmen detektiert werden können, bieten Rechnungen eine detaillierte Einsicht in die verborgenen Teile der Spektren.rnWeiterhin wurden π−π-Komplexe untersucht, welche von verschiedenen Donor- und Akzeptor-Molekülen gebildet werden. Die Moleküle basieren auf polyzyklischen, aromatischen Kohlenwasserstoffen. Für Ladungstransferkomplexe finden DFT Rechnungen ein Minimum in der Oberfläche der potentiellen Energie. Diese attraktive Wechselwirkung wird durch Coulombanziehung verursacht. Allerdings ist die Coulombanziehung nicht die stärkste Wechselwirkung in Ladungstransferkomplexen. Die Einbeziehung von van der Waals-Korrekturen verbessert den intermolekularen Abstand und die Bindungsenergie.rnEine Verkleinerung der intermolekularen Abstände führt zu einer großen Verschiebung der HOMO- und LUMO-Energie.rnAus der Klasse der kristallinen korrelierten Systeme wurden Rb4O6 und FeSe untersucht. Im Falle von Rb4O6 führen Ladungsordnung und Korrelationen zu einem isolierenden Grundzustand. Das hypothetische druckabhängige Phasendiagramm wurde untersucht. Eine Erhöhung des Drucks führt zu einer vergrößerten Bandlücke. Bei etwa 75 GPa wird die Bandbreite W größer als der Bandabstand U und das System nimmt einen homogen gemischt valenten Zustand mit teilweise besetzten π−π-Orbitalen an. Für Drücke ab 160 GPa wird W sehr viel größer als U und das System wird metallisch.rnIm Fall von FeSe finden wir eine korrelierte und isolierende Phase bei hohen Drücken, während das System bei niedrigen Drücken supraleitendes Verhalten zeigt. Die Berechnungen der Elektronenstruktur mit dem Hybridfunktional B3LYP führt zum korrekten halbleitenden Grundzustand in der NiAs- und MnP-Struktur von FeSe. Die Rolle der Korrelationen, der Stöchiometrie und der Nähe zum Magnetismus wird besprochen. Im Speziellen wird gezeigt, dass die Phase mit NiAs-Struktur starke lokale Korrelationen aufweist, was zu einem halbleitenden Zustand in einem weiten Druckbereich führt.

Due to the limited availability of natural oil and the harmful effects of its usage, the industry has focused in searching for sustainable types of raw materials for the production of chemicals. The bioethanol, obtained by fermentation of biomass, has gained particular importance in recent years both as a biofuel, and as a “building block” molecule because it can be considered as a starting reagent to obtain other added value chemical compounds, such as ethylene, acetaldehyde, butadiene and ethyl acetate. The goal of this research was the study of the interaction of ethanol with catalysts based on TiO2-CeOX. Since the electronic properties have implications on the catalytic activity, the idea was to understand if the TiO2-CeOX systems have different reactivity from that of ceria and rutile alone, or an intermediate between them. The study was focused on the characterization of the adsorbed species on the catalysts surface after ethanol adsorption through an in-situ spectroscopic technique (DRIFTS) that allowed us to extract information that could be helpful for the understanding of the processes at the molecular level. The mass spectrometry was used to monitor on-line the desorbed products. Furthermore, reactivity tests in a flow reactor were performed, in order to verify the catalytic behavior of the samples in conditions which are more similar to those applied at an industrial scale. The samples showed to behave in different way depending on the conditions used and the thermal treatment. The particular behavior of the mixed samples with respect to the single oxides is interpreted for each case according to the spectroscopic information collected.

A field study of thermal circulation over very gentle slope is described for a specific day characterised by weak synoptic conditions. The emphasis was on morning and evening transitions, but measurements cover the entire day; therefore a brief analysis is performed to represent the general thermal circulation pattern. Both transition periods are characterised by complex dynamic behaviours. During evening transition, the upslope flow has got through a stagnation condition characterised by wind velocity U<0:5 m=s. Only when the stagnating air has become negative buoyant, the flow is allowed to pour downslope like a slab. Some features of front formation has been found during the transition development, such as delay time of downslope flow start up along the slope, and the presence of positive turbulent kinetic energy at the onset of the motion. Eventually the observed evening transition has followed a mixed mechanisms, with features from different models. Therefore the Rayleigh number seems not to be a good criterion by which parametrise evening transition itself. Morning transition is characterised by destruction of nocturnal temperature inversion and the onset of upslope flow. Inversion destruction can be described in terms of CBL growth at surface and inversion decent from the top of the layer. KH has found to be a good indicator of inversion breakup, if used as parameter to study the inversion breakup in terms of temperature reversal. After the inversion breakup, buoyancy and mechanical productions supply the flow with the necessary energy to start the upslope wind. More quantitative analysis are provided by the study of stability parameters and turbulent kinetic energy budgets. Gradient Richardson number has been used in this terms, finding that a mixed SBL-CBL behaviour dominates the most of the observed layers. Tke budget has shown high turbulent behaviour during morning transition while the evening transition has developed entirely in laminar condition, apart from short intermittent turbulent events.

Chlamydophila (C.) abortus is the most common infectious abortigenic agent in small domestic ruminants in Switzerland. In contrast, the knowledge about chlamydiae in wild ruminants is scarce. As interactions between livestock and Alpine ibex (Capra i. ibex) occur on alpine pastures, the question raises if wild ruminants could play a role as carriers of chlamydiae. Thus, we investigated the prevalence of chlamydiae in Alpine ibex in Switzerland. In total, 624 sera, 676 eye swabs, 84 organ samples and 51 faecal samples from 664 ibex were investigated. Serum samples were tested by two commercial ELISA kits specific for C. abortus. Eye swabs, organs and faecal samples were examined by a Chlamydiaceae-specific real-time polymerase chain reaction (PCR). Positive cases were further investigated by the ArrayTube (AT) microarray method for chlamydial species determination. Of 624 serum samples investigated, 612 animals were negative, whereas nine sera (1.5%) reacted positively in one of the two tests and three sera showed an inconclusive result. Eye swabs of seven out of 412 ibex (1.7%) were tested positive for Chlamydiaceae by real-time PCR. By AT microarray, Chlamydophila (C.) pecorum was identified in two animals, Chlamydophila (C.) pneumoniae was detected in one animal and a mixed infection with C. abortus and C. pecorum was found in four animals. Organs and faecal samples were all negative by real-time PCR analysis. In summary, we conclude that C. abortus is not a common infectious agent in the Swiss ibex population. To our knowledge, this is the first description of C. pneumoniae in ibex. Further studies are necessary to elucidate the situation in other species of wild ruminants as chamois (Rupicapra r. rupicapra), red deer (Cervus elaphus) and roe deer (Capreolus c. capreolus) in Switzerland.

Rationale: There is increasing evidence that short-term exposure to air pollution has a detrimental effect on respiratory health, but data from healthy populations, particularly infants, are scarce. Objectives: To assess the association of air pollution with frequency and severity of respiratory symptoms and infections measured weekly in healthy infants. Methods: In a prospective birth cohort of 366 infants of unselected mothers, respiratory health was assessed weekly by telephone interviews during the first year of life (19,106 total observations). Daily mean levels of particulate matter (PM10), nitrogen dioxide (NO2), and ozone (O3) were obtained from local monitoring stations. We determined the association of the preceding week's pollutant levels with symptom scores and respiratory tract infections using a generalized additive mixed model with an autoregressive component. In addition, we assessed whether neonatal lung function influences this association and whether duration of infectious episodes differed between weeks with normal PM10 and weeks with elevated levels. Measurements and Main Results: We found a significant association between air pollution and respiratory symptoms, particularly in the week after respiratory tract infections (risk ratio, 1.13 [1.02-1.24] per 10 μg/m(3) PM10 levels) and in infants with premorbid lung function. During times of elevated PM10 (>33.3 μg/m(3)), duration of respiratory tract infections increased by 20% (95% confidence interval, 2-42%). Conclusions: Exposure to even moderate levels of air pollution was associated with increased respiratory symptoms in healthy infants. Particularly in infants with premorbid lung function and inflammation, air pollution contributed to longer duration of infectious episodes with a potentially large socioeconomic impact.

Generalized linear mixed models (GLMM) are generalized linear models with normally distributed random effects in the linear predictor. Penalized quasi-likelihood (PQL), an approximate method of inference in GLMMs, involves repeated fitting of linear mixed models with “working” dependent variables and iterative weights that depend on parameter estimates from the previous cycle of iteration. The generality of PQL, and its implementation in commercially available software, has encouraged the application of GLMMs in many scientific fields. Caution is needed, however, since PQL may sometimes yield badly biased estimates of variance components, especially with binary outcomes. Recent developments in numerical integration, including adaptive Gaussian quadrature, higher order Laplace expansions, stochastic integration and Markov chain Monte Carlo (MCMC) algorithms, provide attractive alternatives to PQL for approximate likelihood inference in GLMMs. Analyses of some well known datasets, and simulations based on these analyses, suggest that PQL still performs remarkably well in comparison with more elaborate procedures in many practical situations. Adaptive Gaussian quadrature is a viable alternative for nested designs where the numerical integration is limited to a small number of dimensions. Higher order Laplace approximations hold the promise of accurate inference more generally. MCMC is likely the method of choice for the most complex problems that involve high dimensional integrals.

This paper considers a wide class of semiparametric problems with a parametric part for some covariate effects and repeated evaluations of a nonparametric function. Special cases in our approach include marginal models for longitudinal/clustered data, conditional logistic regression for matched case-control studies, multivariate measurement error models, generalized linear mixed models with a semiparametric component, and many others. We propose profile-kernel and backfitting estimation methods for these problems, derive their asymptotic distributions, and show that in likelihood problems the methods are semiparametric efficient. While generally not true, with our methods profiling and backfitting are asymptotically equivalent. We also consider pseudolikelihood methods where some nuisance parameters are estimated from a different algorithm. The proposed methods are evaluated using simulation studies and applied to the Kenya hemoglobin data.

This paper proposes a numerically simple routine for locally adaptive smoothing. The locally heterogeneous regression function is modelled as a penalized spline with a smoothly varying smoothing parameter modelled as another penalized spline. This is being formulated as hierarchical mixed model, with spline coe±cients following a normal distribution, which by itself has a smooth structure over the variances. The modelling exercise is in line with Baladandayuthapani, Mallick & Carroll (2005) or Crainiceanu, Ruppert & Carroll (2006). But in contrast to these papers Laplace's method is used for estimation based on the marginal likelihood. This is numerically simple and fast and provides satisfactory results quickly. We also extend the idea to spatial smoothing and smoothing in the presence of non normal response.

Two experiments were conducted with 30 dairy cows each, to study the preference for fresh (Experiment 1) and ensiled (Experiment 2) ryegrass, white and red clover. Both experiments consisted of three choice diets with white or red clover or both, offered with ryegrass, and two diets with ryegrass mixed with white or red clover (40% clover). Cows consumed diets with 37.7% fresh white and 45.9% red clover, and no preference was observed when the cows were offered all three forages. By contrast, cows preferred white and red clover silage (73.0 and 69.2%, respectively) over ryegrass silage (of lower nutritive quality). When offered three forages, cows preferred white (59.8%) over red clover (17.5%) and ryegrass (22.7%). Choice diets resulted in diets similar (fresh forages) or higher in nutrient content and digestibility (silages). Treatments did not affect feed intake and performance. Choices compared to mixed diets with red clover silage were preferable regarding the fatty acid composition of the milk fat. Obviously, only large differences in nutrient and energy concentration facilitate preferences for clovers over ryegrass, which could, depending on clover type, be beneficial in terms of the milk's fatty acid composition.

Data from 59 farms with complaints of udder health problems and insufficient quality of delivered milk that had been assessed by the Swiss Bovine Health Service (BHS) between 1999 and 2004 were retrospectively analysed. Data evaluated included farm characteristics such as farm size, herd size, average milk yield, milking system and housing system, deficits of the milking equipment and the milking practices, and bacteriological results of milk samples from all cows in lactation. The average size of the farms assessed by the BHS was larger than the size of the were evaluated, 42 showed obvious failures which the farm managers could have noticed. Only 5 of the 57 milkers carried out their work according to the generally valid guidelines of the National Mastitis Council. More than 2 basic mistakes were observed in the milking practices of 36 milkers. In 51 farms, mixed infections with several problem bacteria (those present in at least 20 % of the tested cows on a farm) were found. Staphylococcus aureus proved to be the most common problem germ. As the bacteria responsible for the herd problem (the sole problem bacteria detectable on a particular farm) Staphylococcus aureus was detected in 4 farms. The current study revealed that education in the area of milking techniques and milking practices of farmers should be improved in order to reduce the incidence of udder health problems on herd level. Staphylococcus aureus is the most important problem bacteria involved in herds with udder health problems in Switzerland. Staphylococcus aureus might be used in practice as the indicator germ for early recognition of management problems in dairy farms.
