3D Computer Graphics (CG) has become the dominant medium for modern animated feature films. It is widely understood that traditional principles of animation developed in the 1930s at the Walt Disney Studio remain applicable to this new medium and heavily influence the range of aesthetic motion styles in contemporary animation. Via a frame-by-frame textual analysis of four animated feature films, this thesis tests and confirms the validity of the principles of animation and expands upon them by reinterpreting the Disney principle of appeal as aesthetic harmony, which delineates the way in which character posing and transitions between poses contribute to the animated motion styles that animators work in today.

Caveolae have been linked to diverse cellular functions and to many disease states. In this study we have used zebrafish to examine the role of caveolin-1 and caveolae during early embryonic development. During development, expression is apparent in a number of tissues including Kupffer's vesicle, tailbud, intersomite boundaries, heart, branchial arches, pronephric ducts and periderm. Particularly strong expression is observed in the sensory organs of the lateral line, the neuromasts and in the notochord where it overlaps with expression of caveolin-3. Morpholino-mediated downregulation of Cav1α caused a dramatic inhibition of neuromast formation. Detailed ultrastructural analysis, including electron tomography of the notochord, revealed that the central regions of the notochord has the highest density of caveolae of any embryonic tissue comparable to the highest density observed in any vertebrate tissue. In addition, Cav1α downregulation caused disruption of the notochord, an effect that was enhanced further by Cav3 knockdown. These results indicate an essential role for caveolin and caveolae in this vital structural and signalling component of the embryo.

A detailed study is presented of the expected performance of the ATLAS detector. The reconstruction of tracks, leptons, photons, missing energy and jets is investigated, together with the performance of b-tagging and the trigger. The physics potential for a variety of interesting physics processes, within the Standard Model and beyond, is examined. The study comprises a series of notes based on simulations of the detector and physics processes, with particular emphasis given to the data expected from the first years of operation of the LHC at CERN.

We present noise measurements of a phase fluorometric oxygen sensor that sets the limits of accuracy for this instrument. We analyze the phase sensitive detection measurement system with the signal ''shot'' noise being the only significant contribution to the system noise. Based on the modulated optical power received by the photomultiplier, the analysis predicts a noise spectral power density that was within 3 dB of the measured power spectral noise density. Our results demonstrate that at a received optical power of 20 fW the noise level was low enough to permit the detection of a change oxygen concentration of 1% at the sensor. We also present noise measurements of a new low-cost version of this instrument that uses a photodiode instead of a photomultiplier. These measurements show that the noise for this instrument was limited by noise generated in the preamplifier following the photodiode. (C) 1996 Society of Photo-Optical Instrumentation Engineers.

The use of reconstructed ceramic surfaces as templates for nanopatterning has been demonstrated recently. This technique differs from the surface decoration by Au on stepped surfaces of alkali halides which has been a topic of intense research in the past. Some of the intriguing aspects related to the physical origin of the phenomena have been considered here. Based on heterogeneous nucleation of Pt vapor on wedged alumina surfaces, it has been shown that the valley sites are the preferred sites for nucleation. However, the hill sites are decorated by the particles in the present study pointing out to a different physical origin for the formation of the nanoparticles. The role of electrostatic energy reduction on the formation of such nanopatterns is discussed.

Visible-light microscopy (VLM) and atomic-force microscopy (AFM) were used to study the progression of grain-boundary grooving and migration in high-purity alumina (Lucalox™). Groove profiles from the same grain boundaries were revisited using AFM following successive heat-treatments. The grooves measured from migrating grain boundaries were found to have asymmetric partial-angles compared to those measured from boundaries that did not migrate during the experiment. For a moving boundary, the grain with the larger partial-angle was consistently found to grow into the grain with the smaller partial-angle. Migrating boundaries were observed to leave behind remnant thermal grooves. The observations indicate that the boundary may be bowing out during the migration process.

Position-dependent gene expression is a critical aspect of the development and behaviour of multicellular organisms. It requires a complex series of interactions to occur between different cell types in addition to intracellular signalling cascades. We used Escherichia coli to study the properties of an artificial signalling system at the interface between two expanding cell populations. We genetically engineered one population to produce a diffusible acyl-homoserine lactone (AHL) signal, and another population to respond to it. Our experiments demonstrate how such a signal can be used to reproducibly generate simple visible patterns with high accuracy in swimming agar. The producing and responding cassettes of two such signalling systems can be linked to produce a symmetric interface for bidirectional communication that can be used to visualise molecular logic. Intracellular feedback between these two cassettes would then create a framework for self-organised patterning of higher complexity. Adapting the experiments of Basu et al. (Basu et al., 2005) using cell motility, rather than a differential response to AHL concentrations as a way to define zones of response, we noted how the interaction of sender and receiver cell populations on a swimming plate could lead to complex pattern formation. Equipping highly motile strains such as E. coli MC1000 with AHL-mediated auto-inducing systems based on Vibrio fischeri luxI/luxR and Pseudomonas aeruginosa lasI/lasR cassettes would allow the amplification of a response to an AHL signal and its propagation. We designed and synthesised codon-optimised auto-inducing luxI/R and lasI/R cassettes as optimal gene expression is crucial for the generation of robust patterns. We still have to complete and test the entire genetic circuitry, although by modelling the system we were able to demonstrate its feasibility. © 2007 The Institution of Engineering and Technology.

Analisa a crise do Parlamento no desempenho de suas funções típicas, a saber, a representativa, a legislativa e a fiscalizadora. Em seguida, desenvolve o conceito de educação legislativa, destacando duas dimensões: os efeitos epistêmicos do desempenho das funções típicas e as atividades de caráter estritamente educativo, como o Parlamento Jovem. Por fim,discute se a educação legislativa pode ser considerada uma resposta para a crise do Parlamento.
