Introduction: La stomatite prothétique est une condition inflammatoire chronique de la muqueuse buccale recouverte par une prothèse. Cette maladie est considérée comme la lésion buccale la plus fréquente chez les porteurs de prothèses amovibles. Des études récentes sur l'étiologie de la stomatite prothétique suggèrent que des traitements basés sur la réduction de l'inflammation seraient efficaces dans le traitement de cette maladie. Objectifs: Évaluer l'efficacité du brossage du palais dans le traitement de la stomatite prothétique. Méthodes: Quarante-huit participants (âge moyen : 66,0 ± 11,2 ans) avec un diagnostic de stomatite prothétique, ont été sélectionnés à partir d’un examen préalable de 143 individus, afin de participer à cet essai clinique de phase I à deux centres, réalisé selon un devis de type pré-test/post-test à un seul groupe. L'intervention a consisté en un brossage du palais avec une brosse manuelle après chaque repas et avant le coucher. Des examens cliniques et microbiologiques ont été effectués avant le traitement, et à 1 mois et 3 mois de suivi. Des données supplémentaires ont été obtenues par l'utilisation d'un questionnaire validé. Les résultats primaires et secondaires étaient, respectivement, la rémission de stomatite prothétique et la diminution du nombre de colonies de Candida. Des tests statistiques descriptifs et non paramétriques ont été menés pour analyser les données. Résultats: À 3 mois de suivi, 10,4 % des participants ont été guéris et 70,8 % ont eu une amélioration clinique de la stomatite prothétique grâce au brossage du palais. Une réduction statistiquement significative de la surface et de l’intensité de l’inflammation après 3 mois de brossage du palais a été démontrée (p < 0,0001). L’ampleur de l’effet a varié d’un effet modéré à important (0,34 à 0,54) selon la classification utilisée pour le diagnostique de la stomatite prothétique. De plus, le nombre de colonies de Candida, recueillies par sonication des prothèses et par échantillonnage du palais, a diminué de manière statistiquement significative après 3 mois de brossage (p ≤ 0,05). Conclusion: Les résultats de cette étude suggèrent que le brossage du palais est efficace comme traitement de la stomatite prothétique.

Des études antérieures démontrent que les descendants de peuples européens et africains présentent des différences de susceptibilité à certaines maladies infectieuses. Ces différences suggèrent des variations interpopulationnelles de la réponse immunitaire qui résultent probablement de l’adaptation de ces individus aux pathogènes de leur environnement. Nous avons caractérisé la réponse immunitaire chez des descendants de peuples européens et africains à des infections bactériennes. Nous avons infecté des macrophages dérivés de monocytes de 30 Américains d’origine africaine (Africains) et de 31 Américains d’origine européenne (Européens) avec les pathogènes intracellulaires Listeria monocytogenes et Salmonella typhimurium pendant 4 heures, puis nous avons mesuré le niveau d’expression pangénomique des cellules infectées et non infectées par séquençage de l’ARNm. Nous avons estimé le niveau de contrôle de l’infection par les macrophages à 2, 4 et 24 heures post-infection en évaluant le taux de survie des bactéries. Nous avons observé que les Africains présentent significativement moins de bactéries intracellulaires après 4 et 24 heures que les Européens, suggérant que les Africains contrôlent mieux les infections bactériennes. Nous avons identifié des différences interpopulationnelles dans le niveau de sécrétion des cytokines et dans le niveau d’expression de certains gènes, ce qui suggère que les Africains modulent une réponse inflammatoire plus forte que les Européens. Nous avons démontré que plusieurs de ces gènes ont subi des évènements de sélection positive récents seulement chez les Européens. Notre étude a identifié plusieurs gènes candidats susceptibles d’influencer le cours des infections bactériennes chez les humains. Nos résultats indiquent que les différences dans la progression des maladies infectieuses entre les populations européennes et africaines seraient le résultat de la sélection naturelle.

L’interaction du CD40L plaquettaire avec le CD40 exprimé par les mono-lymphocytaires, dont les cellules progénitrices endothéliales (EPCs), médie l’hémostase. Deux sous-types d’EPCs induisent la réparation vasculaire : les early outgrowth cells (EOCs) et les endothelial colony forming cells (ECFCs). Les EOCs expriment des protéines adaptatrices s’associant aux récepteurs du facteur de nécrose tumorale (TRAFs) nécessaires à la signalisation du CD40. L’association des TRAFs au CD40 contribuerait à la fonction antiplaquettaire d’EOCs prétraitées au CD40L, via la libération de prostacycline (PGI2) ou d'oxyde nitrique (NO). Toutefois, la contribution des TRAFs des ECFCs dans la libération de PGI2 et de NO via la régulation des cyclo-oxygénases (COX) et des NO synthases (NOS) demeure inexplorée. Cette étude vise à comprendre le rôle des TRAFs, COX et NOS dans les ECFCs. Nous avons différencié des EPCs via la culture de cellules mononucléaires du sang périphérique (PBMCs) dans un milieu à croissance endothéliale (EGM-2) et révélé, par microscopie optique et confocale, le phénotype monocytaire de nos EOCs et de cellules endothéliales (ECs) de nos ECFCs, incluant leurs caractéristiques endothéliales par cytométrie en flux. L’expression constitutive de l’eNOS, l’iNOS, la COX-1 et faiblement la COX-2 dans nos ECFCs et ECs, des enzymes absentes de nos EOCs, a été décelée par Western Blot. Le profil d'expression des TRAFs dans nos EOCs, ECFCs, PBMCs et ECs a démontré la présence variée du CD40 et celle des TRAF1, 2, 3, 5 et 6, selon le type cellulaire. En conclusion, nous avons révélé la présence de TRAFs, COX et NOS, ainsi que leur expression différentielle dans les EOCs et ECFCs. Des études portant sur l’association des TRAFs au CD40 éclaireront sur les mécanismes intracellulaires impliqués dans la régulation de la synthèse de PGI2 et de NO et la fonction antiplaquettaire des EPCs.

The use of bioluminescence was evaluated as a tool to study Pseudomonas syringae population dynamics in susceptible and resistant plant environments. Plasmid pGLITE, containing the luxCDABE genes from Photorhabdus luminescens, was introduced into Pseudomonas syringae pv. phaseolicola race 7 strain 1449B, a Gram-negative pathogen of bean (Phaseolus vulgaris). Bacteria recovered from plant tissue over a five-day period were enumerated by counting numbers of colony forming units and by measurement of bioluminescence. Direct measurement of bioluminescence from leaf disc homogenates consistently reflected bacterial growth as determined by viable counting, but also detected subtle effects of the plant resistance response on bacterial viability. This bioluminescence procedure enables real time measurement of bacterial metabolism and population dynamics in planta, obviates the need to carry out labour intensive and time consuming traditional enumeration techniques and provides a sensitive assay for studying plant effects on bacterial cells.

This work investigated the role of rpoS in the development of increased cell envelope resilience and enhanced pressure resistance in stationary phase cells of Escherichia coli. Loss of both colony-forming ability and membrane integrity, measured as uptake of propidium iodide (PI), occurred at lower pressures in E. coli BW3709 (rpoS) than in the parental strain (BW2952). The rpoS mutant also released much higher concentrations of protein under pressure than the parent. We propose that RpoS-regulated functions are responsible for the increase in membrane resilience as cells enter stationary phase and that this plays a major role in the development of pressure resistance. Strains from the Keio collection with mutations in two RpoS-regulated genes, cfa (cyclopropane fatty acyl phospholipid synthase) and osmB (outer membrane lipoprotein), were significantly more pressure-sensitive and took up more PI than the parent strains with cfa having the greatest effect. Mutations in the bolA morphogene and other RpoS-regulated lipoprotein genes (osmC, osmE, osmY and ybaY) had no effect on pressure resistance. The cytoplasmic membranes of the rpoS mutant failed to reseal after pressure treatment and strains with mutations in osmB and nlpI (new lipoprotein) were also somewhat impaired in the ability to reseal their membranes. The cfa mutant, though pressure-sensitive, was unaffected in membrane resealing implying that the initial transient permeabilization event is critical for loss of viability rather than the failure to reseal. The enhanced pressure sensitivity of polA, recA and xthA mutants suggested that DNA may be a target of oxidative stress in pressure-treated cells.

The protective effect of two vaccination regimes using Salenvac, a commercially available iron-restricted Salmonella enterica subsp. Enterica serotype Enteritidis PT4 bacterin vaccine, was verified in laying birds. Immunization was intramuscular at 1 day old and again at 4 weeks of age (V2), or at 1 day and 4 weeks with a third dose at 18 weeks of age (V3). Challenge S. Enteritidis (5 to 7.5) x 10(7) colony forming units) was given intravenously at 8, 17, 23, 30 and 59 weeks of age. For all age groups, both vaccination regimes reduced significantly the number of tissues and faecal samples that were culture positive for the challenge strain. For laying birds, fewer eggs (P < 0.001) were culture positive for S. Enteritidis after challenge from vaccinated laying birds ( 56/439 batches of eggs) than unvaccinated birds (99/252 batches). The data give compelling evidence that the vaccine is efficacious and may contribute to the reduction of layer infection and egg contamination.

Abstract Background: Advancing age is linked to a decrease in beneficial bacteria such as Bifidobacterium spp. and reduced aspects of innate immune function. Objectives: We investigated whether daily consumption of a probiotic [Bacillus coagulans GBI-30, 6086 (BC30); GanedenBC30] could improve immune function and gut function in men and women aged 65–80 y, using a double-blind, placebo-controlled crossover design. Method: Thirty-six volunteers were recruited and randomly assigned to receive either a placebo (microcrystalline cellulose) or the probiotic BC30 (1 3 109 colony-forming units/capsule). Volunteers consumed 1 treatment capsule per day for 28 d, followed by a 21-d washout period before switching to the other treatment. Blood and fecal samples were collected at the beginning and end of each treatment period. Fecal samples were used to enumerate bacterial groups and concentrations of calprotectin. Peripheral blood mononuclear cells (PBMCs) were extracted from whole blood to assess natural killer cell activity and lipopolysaccharide (LPS)-stimulated cytokine production. C-reactive protein concentrations were measured in plasma. Results: Consumption of BC30 significantly increased populations of Faecalibacterium prausnitzii by 0.1 log10 cells/mL more than during consumption of the placebo (P = 0.03), whereas populations of Bacillus spp. increased significantly by 0.5 log10 cells/mL from baseline in volunteers who consumed BC30 (P = 0.007). LPS-stimulated PBMCs showed a 0.2 ng/mL increase in the anti-inflammatory cytokine IL-10 28 d after consumption of BC30 (P < 0.05), whereas the placebo did not affect IL-10, and no overall difference was found in the effect of the treatments. Conclusions: Daily consumption of BC30 by adults aged 65–80 y can increase beneficial groups of bacteria in the human gut and potentially increase production of anti-inflammatory cytokines. The objective of this study was to evaluate the effects of different gamma-radiation doses on the growth of Alternaria alternata in artificially inoculated cereal samples. Seeds and grains were divided into four groups: Control Group (not irradiated), and Groups 1, 2 and 3, inoculated with an A. alternata spore suspension (1 x 10(6) spores/mL) and exposed to 2, 5 and 10 kGy, respectively. Serial dilutions of the samples were prepared and seeded on DRBC (dichloran rose bengal chloramphenicol agar) and DCMA (dichloran chloramphenicol malt extract agar) media, after which the number of colony-forming units per gram was determined in each group. In addition, fungal morphology after irradiation was analyzed by scanning electron microscopy (SEM). The results showed that ionizing radiation at a dose of 5 kGy was effective in reducing the growth of A. alternata. However, a dose of 10 kGy was necessary to inhibit fungal growth completely. The interaction between giant bacteriophage DNA and cationic biomimetic particles was characterized from sizing by dynamic light-scattering, zeta-potential analysis, turbidimetry, determination of colloid stability, visualization from atomic force microscopy (AFM), and determination of cytotoxicity against E. coli from colony forming unities counting. First, polystyrene sulfate (PSS) particles with different sizes were covered by a dioctadecyldimethylammonium bromide (DODAB) bilayer yielding the so-called cationic biomimetic particles (PSS/DODAB). These cationic particles are highly organized, present a narrow size distribution and were obtained over a range of particle sizes. Thereafter, upon adding lambda, T5 or T2-DNA to PSS/DODAB particles, supramolecular assemblies PSS/DODAB/DNA were obtained and characterized over a range of DNA concentrations and particle sizes (80-700 nm). Over the low DNA concentration range, PSS/DODAB/DNA assemblies were cationic, colloidally stable with moderate polydispersity and highly cytotoxic against E. coli. From DNA concentration corresponding to charge neutralization, neutral or anionic supramolecular assemblies PSS/DODAB/DNA exhibited low colloid stability, high polydispersity and moderate cytotoxicity. Some nucleosome mimetic assemblies were observed by AFM at charge neutralization (zeta-potential equal to zero).

Field-scale remediation of oil-contaminated soils from the Liaohe Oil Fields in China was examined using composting biopiles in windrow technology. Micronutrient-enriched chicken excrement and rice husk were applied as nutrition and a bulking agent. The lipase activities of indigenous micro-organisms were analyzed, and three indigenous fungi with high lipase activities was identified. An inoculum consisting of the three indigenous fungi and one introduced (exotic) fungus was applied to four different types of oil-contaminated soils. The results showed that the inoculum of indigenous fungi increased both the total colony-forming units (TCFU) and increased the rate of degradation of total petroleum hydrocarbons (TPH) in all contaminated soils but at different rates. In sharp contrast to other studies, the introduction of exotic micro-organisms did not improve the remediation, and suggests that inoculation of oil-contaminated sites with nonindigenous species is likely to fail. On the other hand, indigenous genera of microbes were found to be very effective in increasing the rate of degradation of TPH. The degradation of TPH was mainly controlled by the compositions of aromatic hydrocarbons and asphaltene and resin. Between 38 to 57% degradation of crude oils (with densities ranging from 25,800 to 77,200 mg/kg dry weight) in contaminated soils was achieved after 53 days of operation. The degradation patterns followed typical first-order reactions. We demonstrate that the construction and operation of field-scale composting biopiles in windrows with passive aeration is a cost-effective bioremediation technology.

Lineage-specific expansion of haematopoietic stem/progenitor cells (HSPCs) from human umbilical cord blood (UCB) is desirable because of their several applications in translational medicine, e.g. treatment of cancer, bonemarrowfailure and immunodeficiencies. The currentmethods forHSPC expansion use either cellular feeder layers and/or soluble growth factors and selected matrix components coated on different surfaces. The use of cell-free extracellular matrices from bone marrow cells for this purpose has not previously been reported. We have prepared insoluble, cell- free matrices from a murine bone marrow stromal cell line (MS-5) grown under four different conditions, i.e. in presence or absence of osteogenic medium, each incubated under 5% and 20% O2 tensions. These acellularmatrices were used as biological scaffolds for the lineage-specific expansion of magnetically sorted CD34+ cells and the results were evaluated by flow cytometry and colony-forming assays. We could get up to 80-fold expansion of some HSPCs on one of the matrices and our results indicated that oxygen tension played a significant role in determining the expansion capacity of the matrices. A comparative proteomic analysis of the matrices indicated differential expression of proteins, such as aldehyde dehydrogenase and gelsolin, which have previously been identified as playing a role in HSPC maintenance and expansion. Our approach may be of value in identifying factors relevant to tissue engineering-based ex vivo HSPC expansion, and itmay also provide insights into the constitution of the niche in which these cells reside in the bone marrow.

O Brasil é o sétimo maior consumidor mundial de águas engarrafadas. Os recipientes mais utilizados, galões plásticos de 20 litros, devem ser submetidos à inspeção individual e posteriormente a sanificação. Recentemente, enfermidades associadas a microrganismos emergentes têm despertado o interesse por novos sanitizantes. Entre estes, o gás ozônio é um dos mais atraentes em virtude da sua segurança e eficácia superiores aos desinfetantes convencionais, não gerando resíduos tóxicos. Neste trabalho, o ozônio foi avaliado como método alternativo na sanificação de galões de água de 20 litros, na cidade de Alfenas, MG. Trinta galões foram avaliados sem tratamento e trinta após a sanificação com água ozonizada (4mg/L/2minutos) quanto à contagem total de microrganismos aeróbios mesófilos heterotróficos, número mais provável (NMP) de coliformes totais e Escherichia coli, Staphyloccocus aureus e Pseudomonas spp. em 100mL de solução enxaguatória. A contagem média de unidades formadoras de colônias (UFC) de microrganismos heterotróficos no estágio de pré-lavagem foi de 5,7/cm² enquanto que o tratamento com a água ozonizada reduziu este valor para 0,003/cm², além de promover a negativação das análises para coliformes Pseudomonas ssp. e somente 13,3% das amostras apresentaram-se positivas para Staphylococcus aureus após a sanificação. Concluiu-se que o tratamento com utilização de ozônio foi eficiente, nas condições testadas.
