The aging spontaneously hypertensive rat (SHR) is a model in which the transition from chronic stable left ventricular hypertrophy to overt heart failure can be observed. Although the mechanisms for impaired function in hypertrophied and failing cardiac muscle from the SHR have been studied, none accounts fully for the myocardial contractile abnormalities. The cardiac cytoskeleton has been implicated as a possible cause for myocardial dysfunction. If an increase in microtubules contributes to dysfunction, then myocardial microtubule disruption by colchicine should promote an improvement in cardiac performance. We studied the active and passive properties of isolated left ventricular papillary muscles from 18- to 24-month-old SHR with evidence of heart failure (SHR-F, n=6), age-matched SHR without heart failure (SHR-NF, n=6), and age-matched normotensive Wistar-Kyoto rats (WKY, n=5). Mechanical parameters were analyzed before and up to 90 minutes after the addition of colchicine (10(-5), 10(-4), and 10(-3) mol/L). In the baseline state, active tension (AT) developed by papillary muscles from the WKY group was greater than for SHR-NF and SHR-F groups (WKY 5.69+/-1.47 g/mm(2) [mean+/-SD], SHR-NF 3.41+/-1.05, SHR-F 2.87+/-0.26; SHR-NF and SHR-F P<0.05 versus WKY rats). The passive stiffness was greater in SHR-F than in the WKY and SHR-NF groups (central segment exponential stiffness constant, K-cs: SHR-F 70+/-25, SHR-NF 44+/-17, WKY 41+/-13 [mean+/-SD]; SHR-F P<0.05 versus; SHR-NF and WKY rats). AT did not improve after 10, 20, and 30 minutes of exposure to colchicine (10(-5), 10(-4), and 10(-3) mol/L) in any group. In the SHR-F group, AT and passive stiffness did not change after 30 to 90 minutes of colchicine exposure (10(-4) mol/L). In summary, the data in this study fail to demonstrate improvement of intrinsic muscle function in SHR with heart failure after colchicine. Thus, in the SHR there is no evidence that colchicine-induced cardiac microtubular depolymerization affects the active or passive properties of hypertrophied or failing left ventricular myocardium.

1. To determine whether diltiazem protects the hypoxic myocardium by reducing contractile work, we have compared the effects of diltiazem and quiescence on left ventricular (LV) papillary muscle subjected to hypoxia. Papillary muscles were obtained from male Charles River CD rats weighing 150-250 g.2. Four groups of muscles were studied: control (N = 6), non-stimulation (N = 10), diltiazem 10(-4) M (N = 6) and diltiazem 10(-4) M plus non-stimulation (N = 10).3. Isolated mt LV papillary muscles were studied in Krebs-Henseleit solution with a calcium concentration of 2.52 mM at 28-degrees-C while contracting isometrically at a stimulation rate of 0.2 Hz. Resting tension and active isometric tension were measured.4. Both diltiazem and quiescence significantly attenuated contracture tension during hypoxia (0.91 +/- 0.10 vs 2.26 +/- 0.49 g/mm2 for diltiazem vs control, and 0.55 +/- 0.18 vs 2.26 +/- 0.49 g/mm2 for quiescence vs control). Recovery of active tension was improved in the diltiazem groups during reoxygenation (4.16 +/- 0.42 vs 3.75 +/- 0.51, 3.53 +/- 0.15 vs 2.90 +/- 0.13, 5.84 +/- 0.33 vs 6.48 +/- 0.29 and 5.98 +/- 0.90 vs 7.67 +/- 0.68 g/mm2 for diltiazem, diltiazem non-stimulation, non-stimulation and control groups).5. The results suggest that the protective effect of diltiazem during hypoxia was due to the reduction in energy demand of the myocardium.

Thyroid hormone is known to affect myocardial glycogen stores and thereby possibly limit anaerobic performance of mammalian cardiac muscle. Thyroid hormone administration (3,5,T-triiodo-L-thyroxine, 300 mu g/kg/day, sc) for 10 days decreased left ventricle (LV) glycogen concentration relative to euthyroid animals (2.78 +/- 0.46 vs. 4.28 +/- 0.29 mg/g of LV (mean +/- SEM)) while increasing the percent of V(1) myosin isozyi-ne, contractile activity and cardiac mass. In contrast, thyroidectomy increased myocardial glycogen stores (8.50 +/- 0.56 mg/g of LV) and shifted the myosin isozyme toward V(3), prolonged contractile activity and decreased LV mass. Thyroxine administration for 3, 7 and 10 days to thyroidectomized animals progressively decreased contractile duration and increased LV mass. Thyroxine administration for 3 or 7 days to thyroidectomized rats did not reduce glycogen stores (7.75 +/- 1.02 and 9.62 +/- 1.16 mg/g of LV, respectively), whereas myocardial glycogen declined to 3.30 +/- 0.58 mg/g of LV after 10 days of treatment. During hypoxia, cardiac muscle from thyroidectomized rats maintained greater active force and developed less contracture relative to euthyroid and, to a greater extent, than hyperthyroid rats. Removal of glucose from the bath decreased anaerobic performance and impaired recovery; however, myocardium from thyroidectomized rats remained more tolerant to hypoxia than the euthyroid group. Overall, the intrinsic LV glycogen content was positively correlated to anaerobic performance. These data demonstrate that the thyroid state profoundly affects myocardial growth, contractility and anaerobic performance of rat myocardium. Although energy demand may affect function during hypoxia, anaerobic substrate reserve (cardiac glycogen concentration) appears to be the primary factor determining tolerance to hypoxic stress. J. Objective: To examine the basis for local wall motion abnormalities commonly seen in patients with ischemic heart disease, computer-controlled isolated muscle studies were carried out. Methods: Force patterns of physiologically sequenced contractions (PSCs) from rat left ventricular muscle preparations under well-oxygenated conditions and during periods of hypoxia and reoxygenation were recorded and stored in a computer. Force patterns of hypoxic-reoxygenating and oxygenated myocardium were applied to oxygenated and hypoxic-reoxygenating myocardium, respectively. Results: Observed patterns of shortening and lengthening closely resemble those obtained from ischemic and non-ischemic myocardial segments using ultrasonic crystals in intact dog hearts during coronary occlusion and reperfusion, and are similar to findings reported in angiographic studies of humans with coronary artery disease. Conclusion: The current study, demonstrating motions of oxygenated isolated muscle preparations which are similar to those in perfused segments of intact hearts with regional ischemia, supports the concept that the multiple motions of both ischemic and non-ischemic segments seen in regional myocardial disease can be explained by interactions of strongly and weakly contracting muscle during the physiologic cardiac cycle.

Purpose - To investigate the participation of contractile state and relaxation in cardiac muscle dysfunction during the transition from stable hypertrophy to cardiac decompensation in aging spontaneously hypertensive rats (SHR). Methods - isolated left ventricular papillary muscle function was studied in SHR with heart failure (SHR-F), in age-matched SHR without evidence of heart failure (SHR-NF), and in nonhypertensive controls Wistar-Kyoto rats (WKY). Muscles were analised in isometric and isotonic contractions in Krebs-Henseleit solution with calcium concentration of 1.25mM at 28°C. Results - Papillary muscles from SHR-F and SHR-NF demonstrated decreased active tension development and shortening velocity relative to normotensive WKY (p<0.05). SHR-F and SHR-NF did not differ. Compared with SHR-NF and WKY, muscle passive stiffness was increased in the failing SHR (p<0.05 versus WKY and SHR-NF). This parameter did not differ between SHR-NF and WKY (p> 0.05). Conclusion - These data suggest that the progression from stable hypertrophy to heart failure is associated with changes in the passive stiffness and is not related to depression of myocardial contractile function.

O presente estudo partindo da perspectiva contextualista e adotando o modelo bioecológico do desenvolvimento buscou identificar as percepções compartilhadas entre pais de crianças com deficiência mental moradoras da Ilha do Combu, região ribeirinha amazônica. Foram acompanhadas as três famílias que tinham filhos cadastrados na Secretaria Municipal de Ensino de Belém e/ou diagnóstico que comprovava a deficiência mental. Os cuidados éticos tomados foram balizados na aprovação do projeto pela UFPA, no esclarecimento dos participantes sobre os objetivos e os riscos inócuos envolvidos assim como o sigilo de suas identidades. A aproximação da pesquisadora ao contexto investigado e às famílias participantes se deu via inserção ecológica. A coleta se dividiu em dois momentos distintos: primeiramente ocorreram visitas sistemáticas às famílias com a utilização do instrumento diário de campo, desse modo a pesquisadora buscou integrar-se ao ambiente estudado via interações sucessivas e regulares tornando-se o mais próximo possível daqueles que o constituem e no segundo momento ocorreu a aplicação do inventário sócio-demográfico e teste de identificação familiar – FIT. Os resultados apontaram condições diferenciadas de desenvolvimento dos filhos relacionadas às percepções compartilhadas pelos pais e as características contextuais presentes na Ilha. As identificações positivas associadas ao bom desempenho dos papéis e atividades estiveram ligadas às percepções de aceitação e pertencimento, aspectos observados na família de Ana. Nas demais famílias (Taciana e Alexandra) estiveram presentes conflitos e dificuldades que ressaltam os seguintes fatores: sobrecarga vivenciada nos cuidados aos filhos, percepções de estranhamento e incompreensão frente à deficiência, conflitos conjugais, rejeição do diagnóstico e identificações negativas. Conclui-se que as percepções compartilhadas pelos pais mostram-se ligadas ao suporte recebido durante o diagnóstico, aos papéis desempenhados na família, às características das crianças e aos aspectos contextuais presentes na cultura ribeirinha que pressupõe o isolamento marcado pela região alagada e conta com uma rede social composta por parentes que vivem na ilha há gerações.

The purpose of this study was to estimate the alcoholic beverage consumption among undergraduate students of a Dentistry Course and compare the effectiveness of screening methods for the detection of binge drinking. A total of 284 undergraduate students participated in the research. We used the full AUDIT and their reduced forms (AUDIT-3 and AUDIT-C). In addition, a ROC curve was created and its area was calculated. Of the students, 91.0% and 90.6% of the male and female genders, respectively, reported alcohol beverage consumption. Bing-drinking behavior occurred in 69.2% of the men and in 52.4% of the women. The 3 methods showed good discriminatory ability. For the women, there was a significant difference between the AUDIT-3 and AUDIT-C. Both the complete version of AUDIT and its reduced forms presented good discriminatory ability

Neutral-pion pi(0) spectra were measured at midrapidity (vertical bar y vertical bar < 0.35) in Au + Au collisions at root s(NN) = 39 and 62.4 GeV and compared with earlier measurements at 200 GeV in a transverse-momentum range of 1 < p(T) < 10 GeV/c. The high-p(T) tail is well described by a power law in all cases, and the powers decrease significantly with decreasing center-of-mass energy. The change of powers is very similar to that observed in the corresponding spectra for p + p collisions. The nuclear modification factors (RAA) show significant suppression, with a distinct energy, centrality, and p(T) dependence. Above p(T) = 7 GeV/c, R-AA is similar for root sNN = 62.4 and 200 GeV at all centralities. Perturbative-quantum-chromodynamics calculations that describe R-AA well at 200 GeV fail to describe the 39 GeV data, raising the possibility that, for the same p(T) region, the relative importance of initial-state effects and soft processes increases at lower energies. The p(T) range where pi(0) spectra in central Au + Au collisions have the same power as in p + p collisions is approximate to 5 and 7 GeV/c for root sNN = 200 and 62.4 GeV, respectively. For the root sNN = 39 GeV data, it is not clear whether such a region is reached, and the x(T) dependence of the x(T)-scaling power-law exponent is very different from that observed in the root sNN = 62 and 200 GeV data, providing further evidence that initial-state effects and soft processes mask the in-medium suppression of hardscattered partons to higher p(T) as the collision energy decreases.

We present measurements of the J/psi invariant yields in root s(NN) = 39 and 62.4 GeV Au + Au collisions at forward rapidity (1.2 < vertical bar y vertical bar < 2.2). Invariant yields are presented as a function of both collision centrality and transverse momentum. Nuclear modifications are obtained for central relative to peripheral Au + Au collisions (R-CP) and for various centrality selections in Au + Au relative to scaled p + p cross sections obtained from other measurements (R-AA). The observed suppression patterns at 39 and 62.4 GeV are quite similar to those previously measured at 200 GeV. This similar suppression presents a challenge to theoretical models that contain various competing mechanisms with different energy dependencies, some of which cause suppression and others enhancement. The complete surgical removal of disease is a desirable outcome particularly in oncology. Unfortunately much disease is microscopic and difficult to detect causing a liability to recurrence and worsened overall prognosis with attendant costs in terms of morbidity and mortality. It is hoped that by advances in optical diagnostic technology we could better define our surgical margin and so increase the rate of truly negative margins on the one hand and on the other hand to take out only the necessary amount of tissue and leave more unaffected non-diseased areas so preserving function of vital structures. The task has not been easy but progress is being made as exemplified by the presentations at the 2nd Scientific Meeting of the Head and Neck Optical Diagnostics Society (HNODS) in San Francisco in January 2010. We review the salient advances in the field and propose further directions of investigation.

While histopathology of excised tissue remains the gold standard for diagnosis, several new, non-invasive diagnostic techniques are being developed. They rely on physical and biochemical changes that precede and mirror malignant change within tissue. The basic principle involves simple optical techniques of tissue interrogation. Their accuracy, expressed as sensitivity and specificity, are reported in a number of studies suggests that they have a potential for cost effective, real-time, in situ diagnosis.

Zusammenfassung In der vorliegenden Arbeit wurden unterschiedliche Eigenschaften von tandem repetitiver DNA (trDNA) analysiert. Drei der untersuchten trDNA-Familien (Cla-Elemente, Alu-Elemente, 1,688-Satelliten-DNA) stammen aus der genomischen DNA von Insekten, während ein trDNA-Cluster artifiziell aus Hsp70 Promotoren hergestellt wurde. Untersucht wurde die Stabilität dieser trDNA-Sequenzfamilien innerhalb eines Plasmidvektors in E. coli bzw. nach Integration der trDNA auch im Genom von D. melanogaster. Der Schwerpunkt der Arbeit liegt jedoch in der Analyse des Einflusses von trDNA auf die Expression eines benachbarten Reportergens in D. melanogaster.Ziel der Untersuchungen zur Stabilität war es, Eigenschaften von trDNA-Clustern und Mechanismen aufzuzeigen, die die Stabilität derselben in E. coli und im Genom von D. melanogaster beeinflussen. Mit Ausnahme der Alu-Elemente zeigen alle trDNA-Familien eine deutliche Instabilität in E. coli. Am Beispiel der Cla-Elemente wurde gezeigt, daß spezifische Eigenschaften der trDNA-Familie, wie etwa die sequenzbedingte Krümmung der Helixachse, keinen Einfluß auf die Stabilität des trDNA-Clusters haben, sondern die Orientierung des trDNA-Clusters innerhalb des Vektors ausschlaggebend ist. Ein entscheidender Faktor könnte die Orientierung des trDNA-Clusters relativ zur Wanderungsrichtung der Replikationsgabel in E. coli sein. Am Beispiel des trDNA-Clusters aus artifiziellen Hsp70 Promotoren konnte gezeigt werden, daß verschiedene Rekombinationssysteme an der Instabilität in E. coli beteiligt sind. Die meisten beobachteten Deletionen von trDNA sind RecA-abhängig. Zusätzlich findet jedoch in einem kleinen Teil der Plasmide auch eine RecA-unabhängige Rekombination statt. Sowohl in E. coli als auch in D. melanogaster wurde als vorherrschender Mechanismus der trDNA-Instabilität die homologe Rekombination identifiziert. TrDNA-Cluster, die in E. coli deutlich instabil sind, können jedoch im Genom von D. melanogaster weitgehend stabil sein. Auch Faktoren, die in E. coli die Stabilität eines trDNA-Clusters beeinflussen, wie etwa die Orientierung, zeigen in D. melanogaster keinen Einfluß auf die Stabilität der trDNA-Cluster. Ergebnisse aus Stabilitätsuntersuchungen in E. coli können damit nicht ohne Überprüfung auf andere Organismen übertragen werden. Im Hauptteil der Arbeit sollte geklärt werden, ob trDNA generell die Expression benachbarter Gene beeinflußt und welche Eigenschaften der trDNA für diesen inhibitorischen oder stimulierenden Effekt verantwortlich sind. Keine der untersuchten trDNA-Familien zeigt einen inhibitorischen Effekt auf ein benachbartes Reportergen. Entgegen dem Modell von Dorer und Henikoff (1994) führen trDNA-Cluster nicht per se zu der Entstehung von Heterochromatin. Die Ergebnisse der vorliegenden Arbeit zeigen, daß sowohl ein trDNA-Cluster aus Cla-Elementen als auch aus Hsp70 Promotor-Elementen eine deutliche Steigerung der Expression des miniwhite-Reportergens bewirken. Diese Steigerung ist für beide trDNA-Familien unabhängig von der chromosomalen Lage des Transgens im Euchromatin. In beiden Fällen ist der Effekt von der Orientierung des trDNA-Clusters abhängig und verstärkt sich mit wachsender Kopienzahl der trDNA-Einheiten. Während eines der trDNA-Cluster aus trDNA-Einheiten besteht, die bekanntermaßen eine Promotoraktivität aufweisen (Hsp70 Promotoren), war für die Cla-Elemente kein Einfluß auf die Expression eines benachbarten Gens bekannt. Damit wurde für eine trDNA-Familie aus Chironomus nachgewiesen, daß sie auf ein benachbartes Gen ähnlich wirkt wie zusätzliche Promotoren. Die experimentellen Befunde unterstützen ein Modell, demzufolge die Cla-Elemente in gleicher Weise wie tandem repetitive Promotoren auf ein benachbartes Gen expressionssteigernd wirken. Die TATA-Box ist für das Modell ein wichtiges Strukturelement, da diese in beiden expressionssteigernden DNA-Sequenzen der trDNA-Cluster vorkommt und ausschließlich in einer Orientierung wirkt. Das Modell besagt, daß durch die Verbindung einer offenen Chromatinstruktur mit korrekt orientierten Bindungsstellen für TBP der Aufbau von vollständigen Transkriptionskomplexen an den tandem repetitiven Promotoren initiiert wird. Einer Perlenschnur ähnlich wären die Transkriptionskomplexe direkt verfügbar, nachdem ein Transkriptionskomplex den Promotor zur Transkription verlassen hat (Abb. 32). Dies würde zu der beobachteten Steigerung der Expression des Reportergens sowohl durch die tandem repetitiven Hsp70 Promotoren als auch durch die Cla-Elemente führen.

Die Familie der IL-6-Typ-Zytokine (IL-6, IL-11, CT-1, CNTF, LIF, OSM, BSF-3) ist durch eine vierhelikale Faltung gekennzeichnet. Alle Zytokine dieser Familie agieren über einen Rezeptorkomplex, der als gemeinsame Komponente mindestens ein Molekül gp130 enthält. IL-6 und IL-11 signalisieren über ein gp130-Homodimer, während CT-1, CNTF, LIF und OSM ein Heterodimer aus gp130 und dem strukturell verwandten LIFR oder, im Falle des OSM, auch OSMR verwenden. Die Rezeptoren der vierhelikalen Zytokine sind in ihrem extrazellulären Bereich modulartig aus Ig- und Fibronektin-Typ-III-ähnlichen Domänen aufgebaut. Sie besitzen als gemeinsame Struktureinheit ein zytokinbindendes Modul (CBM) aus zwei Fibronektin-Typ-III-ähnlichen Domänen, die durch vier konservierte Cysteine in der N-terminalen und ein konserviertes WSXWS-Motiv in der C-terminalen Domäne charakterisiert sind. Auf Zielzellen bindet IL-6 an den spezifischen IL-6 Rezeptor, worauf der Komplex aus IL-6/IL-6R mit dem Signaltransduktor gp130 assoziiert. Der IL-6R besteht in seinem extrazellulären Bereich aus drei Domänen. Die N-terminale Ig-ähnliche Domäne ist für die biologische Aktivität nicht notwendig. Die Domänen 2 und 3 bilden das CBM, welches auch in löslicher Form agonistisch wirkt. In der vorliegenden Arbeit wurden die strukturellen und funktionellen Eigenschaften der dritten extrazellulären Domäne des IL-6R untersucht. Das Protein läßt sich effizient in Bakterien exprimieren und in vitro renaturieren. Es konnte gezeigt werden, daß Domäne 3 für die Bindung an IL-6 ausreichend ist, der Komplex aus D3 und IL-6 jedoch nicht mehr mit dem gp130-Molekül assoziieren kann. Da der lösliche IL-6R (bestehend aus D2 und D3) in der Lage ist, an gp130 zu binden und ein biologisches Signal auszulösen, weisen diese Daten der C-terminalen CBM-Domäne (D3) eine ligandenbindende Funktion und der N-terminalen CBM-Domäne eine wichtige Rolle bei der Komplexbildung mit gp130 und Signalinduktion zu. Die gezeigte Expressions- und Renaturierungsstrategie für D3 wurde zur Markierung des Proteins mit 15N und 13C für die mehrdimensionale, heteronukleare NMR-Spektroskopie angewandt. Die hierdurch ermöglichte Strukturaufklärung von D3 als einer eindeutig in die Ligandenbindung involvierten Teilstruktur wird umfassendere strukturelle Informationen über den IL-6R-Komplex liefern, als es die bisherigen Mutations- bzw. Modellbaustudien konnten.
