The objective of this study was to determine the kinetic parameters of a new formulation that contained 2.25% ivermectin combined with 1.25% abamectin in bovine plasma. The results for 2.25% ivermectin: C-max (37.11 ng/mL +/- 7.42), T-max (16 days +/- 5.29), T-1/2 (44.62 days +/- 53.89), AUC (928.2 ng.day/mL +/- 153.83) and MRT (36.73 days +/- 33.64), and for 1.25% abamectin: C-max (28.70 ng/mL +/- 9.54), T-max (14 days +/- 4.04), T-1/2 (15.40 days +/- 11.43), AUC (618.05 ng.day/mL +/- 80.27) and MRT (20.79 days +/- 8.43) suggest that this combination of 2.25% ivermectin + 1.25% abamectin possesses properties that give this pharmaceutical formula a longer activity time than two of the commercial products tested (1% ivermectin and 1% abamectin), and showed similarity to 3.15% ivermectin.

The effects of the herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) on the central nervous system (CNS) were studied in rats. Behavioural and neurochemical studies were performed. Results show that acute and oral administration of dimethylamine 2,4-D was able to decrease locomotion and rearing frequencies and to increase immobility duration of rats observed in an open-field test. Treatment of rats with p-chlorophenylalanine (PCPA) was unable to change rat's open-field behaviour; 5-hydroxytryptophan (5-HTP) administration not only increased locomotion and rearing frequences but also decreased immobility duration. Pretreatment of the rats with PCPA and 5-HTP decreased and increased dimethylamine 2,4-D effects, respectively. The herbicide was not able to change the striatal levels of dopamine and homovanilic acid but decreased the striatal levels of serotonin (5-HT), as observed for the doses of 100 and 200 mg/kg and increased those of 5-hydroxyindoleacetic acid (5-HIAA) as measured after the 200 mg/kg dose treatment. When the levels of serotonin and 5-HIAA were measured at the brain stem level, only those of 5-HIAA were modified, being increased by diethylamine 2,4-D (60; 100 and 200 mg/kg); this increment on 5-HIAA levels was observed even 1 hr after pesticide administration. Further analysis showed that 2,4-D concentrations chromatographycally detected both in serum and brain of the intoxicated animals were dose-dependent, being found as early as 1 hr after the smaller dose of the herbicide used (10 mg/kg). The results suggest that diethylamine 2,4-D modify 5-HT functional activity within the CNS. Thus, the effects of the herbicide on open-field behaviour of rats could be attributed to a direct or indirect pesticide action on serotoninergic systems.

Structural, electrochemical and spectroscopic data of a new dinuclear copper(II) complex with (+/-)-2-(p- methoxyphenoxy) propionic acid are reported. The complex {tetra-mu-[(+/-)-2-(p-methoxyphenoxy)propionato-O,O']-bis( aqua) dicopper(II)} crystallizes in the monoclinic system, space group P2(1)/n with a = 14.149(1) angstrom, b = 7.495(1) angstrom, c = 19.827(1) angstrom, beta = 90.62(1) and Z = 4. X-ray diffraction data show that the two copper(II) ions are held together through four carboxylate bridges, coordinated as equatorial ligands in square pyramidal geometry. The coordination sphere around each copper ion is completed by two water molecules as axial ligands. Thermogravimetric data are consistent with such results. The ligand has an L' type shape due to the angle formed by the beta-carbon of the propionic chain and the linked p-methoxyphenoxy group. This conformation contributes to the occurrence of a peculiar structure of the complex. The complex retains its dinuclear nature when dissolved in acetonitrile, but it decomposes into the corresponding mononuclear species if dissolved in ethanol, according to the EPR measurements. Further, cyclic voltammograms of the complex in acetonitrile show that the dinuclear species maintains the same structure, in agreement with the EPR data in this solvent. The voltammogram shows two irreversible reduction waves at E-pc = -0.73 and -1.04 V vs. Ag/AgCl assigned to the Cu(II)/ Cu(I) and Cu(I)/Cu degrees redox couples, respectively, and two successive oxidation waves at E-pa = -0.01 and +1.41 V vs. Ag/AgCl, assigned to the Cu degrees/Cu(I) and Cu( I)/Cu( II) redox couples, respectively, in addition to the oxidation waves of the carboxylate ligand.

Purpose - To study the incidence and the etiology of the cardiac lesions in AIDS patients. Methods - The autopsy protocols and the filled slides of the heart from 73 consecutive AIDS patients were reviewed. There were, at least, 2 slides of each heart stained by haematoxylin-eosin; when indicated, Ziehl-Nielsen, Gram and Gomori Grocott stains were used. Results - No cause of death was assigned to the heart. There was involvement of the heart in 66 (90%) cases. Marked atrophy of cardiac fibers with or without lipomatosis was observed in 38 patients. Interstitial infiltrates of myocardium were present in 38 necropsies and in 13 of these cases a probable pathogen was demonstrated: cryptococcus neoforms in three cases and mycobacteria tuberculosis, atypical mycobacteria, toxoplasma gondii, trypanosoma cruzi and cytomegalovirus in two cases each. Bacterial endocarditis was found in 4 autopsies and Kaposi sarcome in one. The pericardium was involved in 22 cases; in 12 there was only non specific mononuclear infiltration. Conclusion - Autopsy examination of the heart from AIDS patients revealed frequent pathologic involvement.

Thermal stability and thermal decomposition of succinic acid, sodium succinate and its compounds with Mn(II), Fe(II), Co(II), Ni(II), Cu(II) and Zn(II) were investigated employing simultaneous thermogravimetry and differential thermal analysis (TG-DTA) in nitrogen and carbon dioxide atmospheres and TG-FTIR in nitrogen atmosphere. On heating, in both atmospheres the succinic acid melt and evaporate, while for the sodium succinate the thermal decomposition occurs with the formation of sodium carbonate. For the transition metal succinates the final residue up to 1180 °C in N 2 atmosphere was a mixture of metal and metal oxide in no simple stoichiometric relation, except for Zn compound, where the residue was a small quantity of carbonaceous residue. For the CO 2 atmosphere the final residue up to 980 °C was: MnO, Fe 3O 4, CoO, ZnO and mixtures of Ni, NiO and Cu, Cu 2O.

Background: Early trauma care is dependent on subjective assessments and sporadic vital sign assessments. We hypothesized that near-infrared spectroscopy-measured cerebral oxygenation (regional oxygen saturation [rSO 2]) would provide a tool to detect cardiovascular compromise during active hemorrhage. We compared rSO 2 with invasively measured mixed venous oxygen saturation (SvO2), mean arterial pressure (MAP), cardiac output, heart rate, and calculated pulse pressure. Methods: Six propofol-anesthetized instrumented swine were subjected to a fixed-rate hemorrhage until cardiovascular collapse. rSO 2 was monitored with noninvasively measured cerebral oximetry; SvO2 was measured with a fiber optic pulmonary arterial catheter. As an assessment of the time responsiveness of each variable, we recorded minutes from start of the hemorrhage for each variable achieving a 5%, 10%, 15%, and 20% change compared with baseline. Results: Mean time to cardiovascular collapse was 35 minutes ± 11 minutes (54 ± 17% total blood volume). Cerebral rSO 2 began a steady decline at an average MAP of 78 mm Hg ± 17 mm Hg, well above the expected autoregulatory threshold of cerebral blood flow. The 5%, 10%, and 15% decreases in rSO 2 during hemorrhage occurred at a similar times to SvO2, but rSO 2 lagged 6 minutes behind the equivalent percentage decreases in MAP. There was a higher correlation between rSO 2 versus MAP (R =0.72) than SvO2 versus MAP (R =0.55). Conclusions: Near-infrared spectroscopy- measured rSO 2 provided reproducible decreases during hemorrhage that were similar in time course to invasively measured cardiac output and SvO2 but delayed 5 to 9 minutes compared with MAP and pulse pressure. rSO 2 may provide an earlier warning of worsening hemorrhagic shock for prompt interventions in patients with trauma when continuous arterial BP measurements are unavailable. © 2012 Lippincott Williams & Wilkins.

A Aniba canelilla (H.B.K) Mez, conhecida popularmente como casca-preciosa é uma espécie da família Lauraceae que apresenta ampla distribuição na região amazônica. O chá das folhas e das cascas são utilizados na medicina popular como digestivo, carminativo e antiinflamatório. Neste estudo decidiu-se avaliar se esta atividade é devida a um de seus principais constituintes, o 1-nitro-2-feniletano. A amostra obtida por purificação do óleo essencial de Aniba canelilla possui 97,5% de 1-nitro-2-feniletano foi fornecida pelo Laboratório de Engenharia Química da UFPA. Nos modelos de nocicepção foram realizados os testes da contorção abdominal, placa quente e formalina. Enquanto que nos modelos de inflamação foram realizados a dermatite induzido pelo óleo de croton, edema de pata induzido por dextrana e carragenina e peritonite induzido pela carragenina. No teste de contorção abdominal induzido por ácido acético, o 1-nitro-2-feniletano nas doses de 15, 25 e 50 mg/kg reduziu de maneira significativa o número de contorções abdominais. No teste de placa quente (55 0,5 C), o 1-nitro-2-feniletano nas doses de 50, 100 e 200 mg/kg não induziu alterações no tempo de latência quando comparado ao grupo controle. No teste de formalina, o 1-nitro-2-feniletano nas doses de 25 e 50 mg/kg reduziu de maneira significativa o estímulo álgico na 2 fase do teste. Além disso, a antinocicepção foi revertida pela naloxona na segunda fase. Na dermatite induzida pelo óleo de croton, o 1-nitro-2-feniletano nas doses de 25 e 50 mg/kg reduziu de maneira significativa o eritema em relação ao grupo controle (inibição de 73% e 79%, respectivamente). Nos edemas de pata induzido por carragenina e dextrana, o 1-nitro-2-feniletano foi capaz de impedir o desenvolvimento do edema, nas doses de 25 e 50 mg/kg, em comparação com o grupo controle. Na peritonite induzida por carragenina, o 1-nitro-2-feniletano na dose de 25 mg/kg reduziu o número de células globais e o número de neutrófilos quando comparado ao grupo controle (inibição de 22,55% e 38,13%, respectivamente). Nossos resultados sugerem que o 1-nitro-2-feniletano tem atividade antinociceptiva e antiinflamatória, provavelmente, de origem periférica, além disso, os resultados sugerem que os receptores opióides estão envolvidos no efeito antinociceptivo do 1-nitro-2-feniletano.

Neste trabalho, cristais de sulfato níquel dopados com íons de manganês (NSH: Mn) e cristais de sulfato níquel dopados com de íon de magnésio (NMgSH) foram crescidos e posteriormente caracterizados pelas técnicas de difração de raios X e de espectroscopia Raman. Os resultados obtidos mostraram que os cristais dopados possuem estrutura muito semelhante a do cristal de sulfato de níquel puro (NSH), com uma deformação anisotrópica nas dimensões da célula unitária em relação ao cristal puro. O objetivo do presente estudo foi crescer dois novos monocristais de boa qualidade óptica para serem usados como filtros ópticos de banda passante. Os cristais de sulfato de níquel hexa-hidratado (NHS) são conhecidos por possuírem espectros de transmissão óptica, que tem atraído muita atenção, pois apresentam duas regiões com alta eficiência de transmissão, aproximadamente 80%, sendo a primeira região entre 200 e 350 nm e a segunda entre 400 e 600 nm, e uma alta eficiência de absorção em outras regiões do espectro UV-VIS. Um espectro de transmissão de luz com estas características é semelhante a um filtro óptico. Analises Termogravimetric (TGA) foram realizadas para cristais puros e dopados. A temperatura de decomposição obtida para o NSH foi de 73 ° C, enquanto que os cristais de NSH:Mn e NMgSH apresentam valores de 82 ° C e 86 º C, respectivamente. Como pode ser facilmente percebido, a estabilidade térmica de cristais com o íons de Mn ou Mg em suas estruturas é significativamente maior. A banda de transmissão entre 200 e 350 nm no espectro óptico de NSH foi observada com redução significativa em sua largura nos espectros de transmissão dos cristais dopados restringindo assim a região do espectro conhecida como UVA.

Genotoxicity data on commercial azo dyes and their components remain sparse, despite their widespread use. We have tested the mutagenicity of 2-cyano-4-nitroaniline (CNNA) and 2,6-dicyano-4-nitroaniline (CNCNNA), components of azo dyes such as Disperse Blue 165 and Disperse Red 73, in Ames test strains. Both compounds are extraordinarily potent frameshift mutagens, with much greater activity than structurally similar dihalonitroanilines and halodinitroanilines. Analysis of the responses of strains over-expressing or deficient in bioactivation enzymes shows that bacterial nitroreductase and acetyl CoA: arylamine N-acetyltransferase are important mediators of the mutagenicity of CNNA and CNCNNA.
